Aspects of noncommutativity and holography in field theory and string theory

Die Arbeit beschäftigt sich mit zwei Themen: den nichtkommutativen Yang-Mills-Theorien und der AdS/CFT-Korrespondenz. Im ersten Teil wird eine teilweise Aufsummation der theta-entwickelten Störungstheorie untersucht. Letztere stellt einen Weg dar, nichtkommutative Yang-Mills-Theorien mit beliebigen Eichgruppen G als Störungsentwicklung im Nichtkommutativitätsparameter theta zu definieren. Es wird gezeigt, daß man im Fall, daß G eine echte Untergruppe von U(N) ist, die ungleich einer U(M) ist (M / This thesis addresses two topics: noncommutative Yang-Mills theories and the AdS/CFT correspondence. In the first part we study a partial summation of the theta-expanded perturbation theory. The latter allows one to define noncommutative Yang-Mills theories with arbitrary gauge groups G as a perturbation expansion in the noncommutativity parameter theta. On diode-pumped solid-state lasers

The research that is presented in this thesis can be divided into two major parts. The first part concerns longitudinally pumped, bulk Er-Yb lasers. In these lasers, the main limitation is the thermal shortcomings of the phosphate glass host material. From the laser experiments and the spectroscopic measurements on crystalline host materials, as well as an investigation to bring further light to the physical background of the involved dynamics, the thesis presents some novel results that contribute to the search for a crystalline replacement. The second part concerns novel laser concepts applied to Yb-doped double tungstate lasers. Different crystal orientations are investigated, such as an athermal orientation for reduced thermal lensing and a conical refraction orientation for complete polarization tuning. Furthermore, the introduction of volume Bragg gratings in the cavity enables wide spectral tuning ranges and extremely low quantum defects. Regarding the first part, the main results are the achievement of 15 % slope efficiency in a monolithic, continuous-wave Yb:GdCOB laser and the achievement of Q-switching of the same laser. The Q-switched pulse durations were around 5-6 ns and the Q-switched slope efficiency was 11.6 %. For both lasers, a maximum output power of 90 mW was obtained, which is close to ordinary glass lasers under similar conditions. A spectroscopic investigation into the Er,Yb-codoped double tungstates was also performed and the results have enabled mathematical modeling of the fluorescence dynamics in these materials. Finally, the temperature dependence of the dynamics in Er,Yb:YAG was studied and the results have given some insight into the physical background of the mechanisms involved. Regarding the second part, different end-pumped Yb:KReW laser cavities were constructed to demonstrate the different concepts. With a laser crystal cut for propagation along the athermal direction at 17º angle clockwise from the dielectric direction Nm, the thermal lens could be reduced by 50 %. In these experiments the maximum output power was 4 W at 60 % slope efficiency. In another cavity incorporating a volume Bragg grating in a retroreflector set-up, the wavelength could be continuously tuned between 997 - 1050 nm. The spectral bandwidth was 10 GHz and the peak output power was 3 W. The same output power could also be obtained at 1063 nm with the grating positioned as an output coupler instead. If, on the other hand, the grating was positioned as an input coupler, 3.6 W output power at 998 nm was obtained at a quantum defect of only 1.6 %. Furthermore, using a crystal oriented for propagation along an optic axis, internal conical refraction could be used to establish arbitrary control of the polarization direction as well as the extinction ratio. Even unpolarized light could be enforced despite the highly anisotropic medium. With this configuration, the maximum output power was 8.6 W at 60 % slope efficiency which equals the performance of a reference crystal with standard orientation. The completely novel concepts of laser tuning with Bragg grating retroreflectors, of low quantum defect through Bragg grating input couplers and of polarization tuning by internal conical refraction can all easily be applied to several other laser materials as well. / QC 20100713

Measurement of cell adhesion forces by holographic microscopy / Mesure des forces d'adhérence cellulaire par microscopie holographique

Les forces mécaniques, générées par la cellule jouent un rôle crucial dans l'adhésion cellulaire, qui est un processus commun à un grand nombre de lignées cellulaires. Afin de mesurer la champ des forces pendant l'adhérence cellulaire, nous utilisons la microscopie de force de traction, où la cellule adhère à la surface plane d'un substrat souple dans le plan. Les forces sont calculées à partir du champ de déplacement mesuré à l'intérieur du substrat sous la cellule. Nous avons construit le microscope, dans lequel nous utilisons des billes sphériques en polystyrène pour mesurer le champ de déplacement. Les positions des marqueurs sont obtenues en analysant I' image interférentielle des particules. Avec cette technique, nous atteignons une précision nanométrique sur le champ de déplacement des particules, ce qui nous permet d'améliorer la résolution en force de ce type de microscope. Les premières mesures ont été effectuées avec la lignée de cellules cancéreuses SW 480. / Mechanical forces, generated by the cell plays crucial role in cell adhesion - common process for different cell lines. ln order to measure the force map during cellular adhesion, we use Traction Force Microscopy (TFM), where cell adheres to the soft substrate in 20 plane, and the forces are calculated from measured displacement field inside the substrate underneath the cell. We built the microscope, where instead of using fluorescent markers, we use spherical polystyrene beads in order to measure the displacement field. Positions of the markers are obtained by analyzing the interference pattern caused by the beads in bright-field light. With this technique, we reach nanometer accuracy of the microsphere position determination, that, respectively, influence accuracy of the calculated force field. With the microscope first measurements were performed with cancer cell line SW 480.

Communication synaptique et non-synaptique entre neurones et cellules précurseurs d’oligodendrocytes dans le cortex somatosensoriel / Synaptic and non-synaptic communication between neurons and oligodendrocyte precursor cells in the somatosensory cortex

Les cellules précurseur d'oligodendrocytes (CPOs) représentent la majeure source d'oligodendrocytesmyélinisants durant le développement post-natal. Ces progéniteurs, identifiés par l'expression du protéoglycane NG2, sont non seulement extrêmement abondants avant la myélinisation, mais ils persistent aussi dans le cerveau mature. À l'instar d'autres cellules non-neuronales, elles expriment un large panel de canaux ioniques et de récepteurs pour des neurotransmetteurs. Cependant, ils sont uniques de part leur capacité à recevoir de véritables contacts synaptiques neuronaux glutamatergiqueset GABAergiques. Durant cette thèse, nous avons caractérisé les propriétés électrophysiologiques des CPOs durant le développement post-natal du cortex en champ de tonneaux de la souris (premier mois post-natal). En effectuant des enregistrements de patch-clamp, des analyses par RT-PCR sur cellule unique et des analyses pharmacologiques, nous avons observé que la courbe I-V à rectification sortante devient linéaire durant le développement, résultant d'une régulation positive de l'expression des canaux potassiques de type Kir4.1. Dotés de ces canaux, les CPOs adultes sont capables de détecter les augmentations locales de potassium extracellulaire générées par l'activité neuronale. Cette régulation positive développementale des canaux Kir4.1 dans les CPOs révèle que ces cellules ont un gain de fonction durant le développement, leur conférant la capacité de communiquer avec les neurones via un mécanisme non-synaptique lié au potassium. Ce changement développemental soutient aussi l'idée que les CPOs sont probablement plus que des progéniteurs. Dans la deuxième partie de cette thèse, nous nous sommes intéressés à l'étude des patrons de connectivité du réseau GABAergique interneurones-CPOs dans le cortex en champ de tonneaux jeune (deuxième semaine post-natale). Dans un premier temps, nous avons tiré avantage de la haute précision latérale et axiale de la photolyse holographique en mono-photon pour stimuler les interneurones GABAergiques avec une résolution à l'échelle de la cellule, de manière à évoquer un potentiel d'action. Nous avons ensuite utilisé cette technique pour cartographier la connectivité entre interneurones et CPOs. Nous avons trouvé que la probabilité de connexion des CPOs est près de moitié moins que celle des cellules pyramidales, et implique plutôt une microcircuitrie locale. De plus, en effectuant des enregistrements pairés, nous avons observé que les CPOs sont contactés transitoirement par des interneurones à décharge rapide et à décharge régulière. Ces connections se caractérisents pour la présence d'un ou deux sites de libération uniquement. Étonnamment, les sites post-synaptiques contenant des récepteurs GABAA avec la sous-unité γ2 sont principalement connectés par les interneurones à décharge rapide, indiquant que ces cellules constituent une afférence spécifique auprès des CPOs. Ici nous décrivons pour la première fois l'émergence de réseaux corticaux spécifiques entre neurones et cellules non-neuronales. / Oligodendrocyte precursor cells (OPCs) are the main source of myelinating oligodendrocytes during postnatal development. These progenitors, identified by the expression of the proteoglycan NG2, are extremely abundant before myelination, but also persist in the mature brain. Similarly to other non-neuronal cells they express a wide range of ionic and ligand-gated ion channels. However, they are unique by their ability to receive truly glutamatergic and GABAergic synaptic contacts from neurons. During this thesis, we characterized the electrophysiological properties of OPCs during the postnatal development of the mouse somatosensory cortex (post postnatal month). By performing patch-clamp recordings, single-cell RT-PCR analyses and pharmacological approaches, we found that outwardly rectifying I-V curves become linear during development, as the result of an upregulation of Kir4.1 potassium channels. Endowed with these channels, adult OPCs are able to sense local extracellular potassium increases generated by neuronal activity. This developmental upregulation of Kir4.1 channels in OPCs revealed that these cells gain physiological properties during development, conferring them the capacity to communicate with neurons, via a non-synaptic potassium-mediated mechanism. This developmental change also supports the view that OPCs are probably more than simple progenitors. In the second part of this thesis, we were interested in study the connectivity patterns underlying the GABAergic interneuron-OPC network in the young somatosensory cortex (second postnatal week). First, we took advantage of the high lateral and axial precision of one-photon holographic photolysis to stimulate GABAergic interneurons at a single cell resolution in order to evoke an action potential. We then used this technique to map the connectivity between interneurons and OPCs. We found that the connectivity probability of OPCs was around half less than that of pyramidal cells and involved more local microcircuits. In addition, by performing paired-recordings, OPCs showed to be transiently contacted by fast-spiking (FSI) and non-fast-spiking (NFSI) interneurons, through single or double release sites. Interestingly, postsynaptic sites containing GABAA receptors with the γ2 subunit were predominantly connected by FSI, indicating that these cells provide a specific input to OPCs. Here we described for the first time the emergence of specific cortical network between neurons and non-neuronal cells. In conclusion, this thesis contributed to get a better understanding of the different modes of communication between neurons and OPCs and the establishment of new signaling mechanisms used by neurons to control the activity of these precursors.

Multi-Criteria Decision Modeling for Best Value Selections in Target Value Design Integrated Project Delivery

Integrated Project Delivery (IPD) combined with Target Value Design (TVD) is a better way to deliver value for the client than traditional guaranteed maximum price (GMP) methods. With traditional GMP delivery methods, the interests of the parties are often at odds. The goal of IPD is to align all party interests in order to achieve a win-win scenario. Due to the aligning nature of IPD and the fact that each party’s success is dependent on achieving the project objectives as a whole; a non-biased, transparent, decision-making process is necessary in order to deliver the project objectives within the constraints of the TVD. Thus delivering the expected value for the client and ensuring that all parties achieve project success. The need for this transparent decision-making process is compounded by the fact that a “target” based system rapidly declines to a less than optimal state if there is no unbiased decision-making process in place. If we treat the entire lifespan of a project as the complex system that it is, we can begin to take advantage of the hierarchical nature of complex systems. The goal of this paper is to show that by modeling the life span of a project through a multi-criteria decision making model, built on a hierarchical framework will allow you to find a non-inferior solution to your TVD. I’m proposing to use Hierarchical Holographic Modeling (HHM) as the framework for an Analytical Hierarchical Process (AHP) multi-criteria decision-making model complete with post-optimality analysis as the preferred project management method.

Trojrozměrné zobrazování v holografickém mikroskopu pomocí koherenční brány / Coherence-gate assisted three-dimensional imaging by holographic microscope

Tato diplomová práce se zabývá výzkumem na téma vlivu prostorové koherence osvětlení. Účelem je určit schopnost osové lokalizace při zobrazení Koherencí řízeným holografickým mikroskopem (CCHM) v závislosti na různé prostorové koherenci světelného zdroje. Osová lokalizace je v tomto případě zkoumána jako kvalita rozlišení drobných detailů trojrozměrného vzorku, umístěných nad sebou. Teorie zobrazení holografickým mikroskopem a teorie rozptylu v nehomogenních prostředích je shrnuta v první části práce, v rozsahu nutném pro pochopení části praktické. Základní princip fungování mikroskopu a přesný popis jeho uspořádání je zde podrobně popsán. Proběhl mechanický návrh stavební úpravy mikroskopu tak, aby bylo možno využívat kondenzorovou optiku s vysokou numerickou aperturou a omezenými optickými vadami. Několik různých přístupů, které by mohly vést ke zlepšení zobrazovacích vlastností mikroskopu, bylo navrženo a vyzkoušeno a jsou zde popsány i s jejich výhodami a nevýhodami. Pro experimentální část práce byl vyroben modelový vzorek. Závislost osové lokalizace na prostorové koherenci osvětlení byla demonstrována pomocí simulace a následně ověřena experimentálně, pozorováním vyrobeného modelového vzorku. Experimentální výsledky potvrzují základní principy vycházející ze zmíněné teorie. Na závěr jsou navržena možná vylepšení, pro budoucí zpřesnění výsledků.

This diploma thesis deals with the issue of motility analysis in leukemia cells. An accurate description of the cell movement and the detection of differences in motility under experimental conditions can be obtained by quantitative analysis of cell motility using time-lapse recording. The first part of this work describes various types of tumor cell migraton. The second part focuses on methods of analysis of cell motility in tissue culture using time-lapse recording, which include image acquisition and processing. Part of this chapter describes a coherence-controlled holographic microscope, which was used in the practical part and for which an insert was designed to ensure the exact and stable position of the individual chambers. The last part is focused on the research of leukemic cell motility, which is concluded by a discussion of the obtained results. The appendix contains a published study included acknowledgement to the author of this diploma thesis for participation in the project.

Studium chování opticky zachycených částic nám umožňuje porozumět základním fyzikálním jevům plynoucím z interakce světla a hmoty. Předkládaná práce podává vysvětlení zesílení tažné síly působící na opticky svázané částice ve strukturovaném světelném poli, tzv. tažném svazku. Ukazujeme, že pohyb dvou opticky svázaných objektů v tažném svazku je silně závislý na jejich vzájemné vzdálenosti a prostorové orientaci, což rozšiřuje možnosti manipulace hmoty pomocí světla. Následně se práce zaměřuje na levitaci opticky zachycených částic ve vakuu. Představujeme novou metodologii na charakterizaci vlastností slabě nelinearního Duffingova oscilátoru reprezentovaného opticky levitující částicí. Metoda je založena na průměrování trajektorií s určitou počáteční pozicí ve fázovém prostoru sestávajícím z polohy a rychlosti částice a poskytuje informaci o parametrech oscilátoru přímo ze zaznamenaného pohybu. Náš inovativní postup je srovnán s běžně užívanou metodou založenou na analýze spektrální hustoty polohy částice a za využití numerických simulací ukazujeme její použitelnost i v nízkých tlacích, kde nelinearita hraje významnou roli.

Mikroskopie časově proměnných biologických objektů / Microscopy of Time Variable Biologic Objects

The subject of the PhD thesis is the application of a transmission digital holographic microscope (DHM) which was designed and constructed in the Laboratory of optical microscopy at the IPE BUT for the research of live cells dynamics. First part of the work is concerned with theoretical description of the microscope imaging properties dependent on the coherence of illumination. It is supplemented with experiments of imaging of a model and a real biological specimen. The following part describes construction modifications and innovations of the microscope and its equipment that enabled the utilization of the microscope for live cells observations. In the experimental part the methodology of live cells preparation and DHM imaging was worked out. The methodology was verified by the observation of cell dynamics during an apoptosis induced by the cytostaticum cis-platinum. Further experiments examined the dynamics of live cells in standard conditions and during a deprivation stimulus. A novel method of holographically reconstructed phase, named \uva{dynamic phase differences}, was set up to evaluate quantitative changes of cell mass distribution during the experiments. Depending on the degree of malignancy and density of cell outgrowth, various schemes of cancer cells behaviour during a specific reaction were revealed using this method. For the quantitative analysis of the DHM phase imaging, a suitable statistical characteristic and an interpretation of the measured data were proposed. Both of them were successfully applied for the comparison of cell motility of two cell types: parental and progeny cell lines. On the basis of the proposed processing, hypotheses describing the reaction mechanism of tumour cells to stress life conditions were established. In the conclusions we summarize our findings and suggestions for the construction and the applications of a new generation of the transmission DHM.

Koherencí řízená holografická mikroskopie v opticky rozptylujících prostředích / COHERENCE-CONTROLLED HOLOGRAPHIC MICROSCOPY IN DIFFUSE MEDIA

This thesis deals with imaging through diffuse media in coherence-controlled holographic microscope (CCHM) developed in IPE FME BUT. The mutual coherence function as well as the signal dependence on the lateral mutual shift between both arms of the CCHM are calculated. Both functions are related to each other. The latter dependence is measured experimentally. A principle of imaging with CCHM through diffuse media with both ballistic and diffuse light is explained by a simple geometrical model. This model is then verified experimentally by imaging a sample through diffuse medium. The point spread function (PSF) of CCHM for imaging through diffuse media is then calculated. Results of PSF calculation are proved experimentally.
