Risk management in company AT CAR, s.r.o. with focus on its branch Brno is the main topic of this thesis. As for the risk management itself, the most important tasks are identification, analysis and reducing the possible risks. These processes are described and discussed in the theoretical part of the thesis, while it also introduces all important terms related to this topic. In the analytical part, the risks are identified using appropriate methods such as SLEPTE analysis, Porter’s Five Forces and 7S model. The risks identified are thereafter evaluated using the scoring method. In the last part, recommendations and measures for reducing the risk are presented.

The diploma thesis deals with risk management in a selected company, which is ČSAD LOGISTIK Ostrava, a. s. This thesis is divided into two parts, where the first part deals with the analysis of the current situation, which is the transport sector in the Czech Republic and its relevant legislation. Subsequently also deals with the terms related to the identification, analysis and elimination of risks. In the second part is the theoretical knowledge is applied in such a way, that from the results obtained with What IF and FMEA analyzes is later created a suggested plan for risk prevention and its elimination.

The work is aimed at the center of the village, which is a square that, in size comparable to that of the Square of Freedom in Brno, is an exhibition of problems: considerable deterioration of the building fund, underground use of the public space, which is moreover unreadable and unreachable. The proposed solution divides the area of the square into three smaller public spaces that have a different character and function. Mariánské námětí is the torso of the existing one, which has been earmarked for the building of a new municipal office and library, is characterized by town houses and the location of services and public amenities. The second area is a semi-detached cottage. It is dominated by walnuts and water. The proposed buildings are oriented towards the library and the wine house. The third such defined area is the Wine Market, which is the crossroads, the scattered area of the surrounding houses, like a school, shops, or wine house. A large part of the COOP building has a sports complex with a halo and a swimming pool. There are solved especially the buildings of the town hall with a library and a wine house.

Identifikace a verifikace osob pomocí záznamu EKG / ECG based human authentication and identification

In the past years, utilization of ECG for verification and identification in biometry is investigated. The topic is investigated in this thesis. Recordings from ECG ID database from PhysioNet and our own ECG recordings recorded using Apple Watch 4 are used for training and testing this method. Many of the existing methods have proven the possibility of using ECG for biometry, however they were using clinical ECG devices. This thesis investigates using recordings from wearable devices, specifically smart watch. 16 features are extracted from ECG recordings and a random forest classifier is used for verification and identification. The features include time intervals between fiducial points, voltage difference between fiducial points and PR intervals variability in a recording. The average performance of verification model of 14 people is TRR 96,19 %, TAR 84,25 %.

Izolace a identifikace DNA probiotických bakterií v komplexních matricích / Izolation and identification of DNA from probiotic bacteria in complex matrices

Nowadays, probiotic lactic acid bacteria (LAB) and bifidobacteria are plentifully exploited in food processing industry. LAB and bifidobacteria are important part of microflora of gastro intestinal tract (GIT). Probiotics (most often just lactobacilli and bifidobakteria) can be supplied to the GIT of the organism like food complements. Species identification is therefore very important. New methods of identification of LAB and bifidobacteria are based on analysis of DNA. Mostly exploited method is polymerase chain reaction (PCR). In my diploma work, genus and species specific PCRs were used for identification of different species of bacteria of genus Lactobacillus and Bifidobacterium in complex matrices of six food supplements (Zenflo, Linex Forte, Probian, Nutra Bona, GS lactobacillus Forte, Pangamin Bifi plus). Total DNA was isolated from crude lysates of cells present in tablets by magnetic particles coated by carboxyl groups . The preparation of cell lysates was optimalised. Different amounts of lysozyme (3 mg/ml, 10 mg/ml), time of incubation at laboratory temperature (1,5 hour, 3 hour) and time of incubation with SDS and proteinase K at 55 °C (1 hour, 3 hour, over all night) were tested. Isolated DNA was quantified and checked in PCR. Primers specific for genus Lactobacillus and Bifidobacterium and for species Lb. acidophilus, Lb. casei, Lb. rhamnosus and Lb. plantarum and B. animalis, B. bifidum, B. infantis, B. longum were used, respectively. All identified bacteria were in accord with the data declared by producer in 3 food supplements (Zenflo, Linex Forte and Pangamin Bifi Plus). The genus indentification was in accord with declaration of producer in other food products only (Probian, Nutra Bona, GS Laktobacily Forte).

Kvasinky a víno / Yeasts and wine

This thesis deals with isolation and identification wine yeasts from grapes and must. For analysis was used white wine Sauvignon that was grown and producing after needs ecological agriculture. Remove samples were processed in laboratory and by the help of dilution method were obtained pure culture isolated yeasts. In the following step, by the application of commercial kit UltraCleanTM Microbial DNA Isolation Kit we were able to isolated individual DNA that it was used to the next analysis. Isolated DNA was amplification by PCR method with ITS1 and ITS4 primers. PCR products were detected on agarose gel. Amplification samples were chopped five restriction endonucleases: HaeIII, HinfI, TaqaI, AluI and MseI. Chopped DNA was detected by the same way as PCR products and it was compared with restriction patterns of collection yeasts. In the next step it was compared genetic similarity of isolated yeasts by using BioNumerics software. As a criterion it was used Pearson coefficients and UPGMA clastering analysis. The result is dedrogram of genetics similarity isolated yeasts.

The aim of thesis is focused on isolation of DNA in quality for polymerase chain reaction (PCR) and the identification of probiotic bacteria. From six probiotic supplements for children were isolated PCR-ready DNAs using magnetic carriers P(HEMA-co-GMA). Isolated DNA was amplified by genus-specific and species-specific primers. DNAs of Lactobacillus, Bifidobacterium and Streptococcus genera were identified as: L. acidophilus, L. rhamnosus, L. casei, B. bifidum, B. longum ssp. longum, B. breve, B. longum ssp infantis, B. animalis and S. thermophilus. The identification corresponded with the data declared by the producers.
