Studium probiotických bakterií mléčného kvašení produkujících antimikrobiální látky / Study of Probiotic Lactic Acid Bacteria Producing Antimicrobial CompoundsTurková, Kristýna January 2014 (has links)
The sixty-eight strains isolated from breastfed full-term infant feces and from another sources were identified using genus-specific polymerase chain reaction (PCR) for Lactobacillus, species-specific PCRs, multiplex PCR, pheS PCR, rep-PCR, RAPD-PCR and 16S rDNA sequencing into Lactobacillus species or group of species. Seven strains produced antimicrobial proteinaceous substances in the supernatants. Antimicrobial proteinaceous substances of three strains were tested on temperature, pH a detergent stability. All tested strains produced temperature-stable antimicrobial proteinaceous substances. Antimicrobial activity was not influenced by detergents with exception of SDS. Presence of genes for production of bacteriocins (acidocin B, gassericin A, gassericin T, gassericin K7A and gassericin K7B) were detected in DNA of fourteen strains using PCR and DNA/DNA hybridization. Selected PCR products were sequenced and analyzed using BLAST algorithm and CLUSTAL W2 programme. The sequences of specific PCR products in DNA of two strains had 100% similarity with the sequences from the database GeneBank. Selected strains of Lactobacillus acidophilus group were tested for the surveillance in gastrointestinal tract, for the production of antimicrobial substances, for the adhesion on Caco-2 cells and for the presence of genes of antibiotic resistance. DNA of strains was tested using specific primers on the presence of genes for histidine-decarboxylase, tyrosine-decyrboxylase and linoleate isomerase. The gene for histidine-decarboxylase production was detected in DNA of seven strains, for tyrosine-decarboxylase production in DNA of one strain and for linoleate isomerase in DNA of four strains. Imunomagnetic separation of the cells was optimized. Magnetic particles functionalized with streptavidin and the anti-Lactobacillus antidote was used for the separation of the cells of Lactobacillus rhamnosus LOCK 0900 from MRS medium, UHT milk and from the yogurt. The IMS-PCR was used for detection of imunomagnetic separated bacterial cells.

Identifikace organismů pomocí analýzy nukleotidových denzitních vektorů / Identification of Organisms Based on Analysis of Nucleotide Density VectorsMaděránková, Denisa January 2015 (has links)
Most methods for analysis of genomic data work with symbolic sequences. Numerically represented genomic sequences can be analyzed by signal processing methods. A new method of numerical representation of DNA sequences, nucleotide density vectors, is proposed in this thesis. Usability of this method for purposes of molecular species identification is tested on DNA barcoding sequences. DNA barcoding is modern and popular methodology based on comparison of short mitochondrial DNA sequences. Beside species identification by proposed method based on nucleotide density vectors, higher taxa rank identification (e.g. families) was also tested. Furthermore, dendrograms were constructed from standardly used evolutionary distances and distances between nucleotide density vectors and the dendrograms were compared.

Matematický model kotle na biomasu pro účely řízení / Mathematical Model of Biomass Boiler for Control PurposesMáša, Vítězslav January 2010 (has links)
he thesis focuses on building of dynamic mathematical model of biomass boiler as a controlled system and the use of this model in practise. At the first part of the thesis the area of energy production using biomass is introduced and the current development at the area of the automatic control of biomass boilers with medium energy output (in units of MW) is described. Although the main topic of the thesis is the construction of model of biomass boiler for control purposes, thesis deals with technology of boilers and principles of its inner processes as well. Creation of the model comes out not only from these important findings, but also from experimental data collected during measurements in real operation. Heat and mass balance calculations were made according to these data and they serve to precise static properties of experimental unit for biomass combustion. Central part of the thesis presents development of the final model that resulted from balance calculations and from step responses of the system obtained by measuring. Built dynamic model is compared with experimental data through simulations in Simulink and verified. At the next part the model of controlled system is completed with control system and closed-loop control circuit is validated and verified by simulation. Then the design of new controller configuration, which improves the quality of control considerably, is presented. At the final part of the thesis, possibilities of other use of the model of boiler are given, both in control theory and in industrial practice.

Identifikace aerodynamických charakteristik atmosférického letadla z výsledků letových měření / Aerodynamic Characteristics Identification of Atmospheric Airplane from Flight Measurement ResultsZikmund, Pavel Unknown Date (has links)
The thesis deals with aerodynamic characteristics identification from flight measurement. The topic is part of flight mechanic – handling qualities. The first theoretic part consists of three identification methods description: Error equation method, Output error method and Filter error method. Mathematical model of an airplane is defined and restricted to the motion with 3 degree of freedom. There is also introduced simulation of flight measurement for identification software validation. Practical part is focused on experiment preparation, execution and evaluation. The airplane VUT 700 Specto had been chosen to carry out flight tests. The airplane was modified to the new electric powered VUT 700e Specto after first measurement flights with combustion engine. Data record from on-board measurement unit was completed by telemetric data from autopilot and remote control system. Flight tests were carried out in stabilised mode of autopilot in symmetric flight. The results were confronted with analytical analysis results and DATCOM+ software parameter estimation.

This project deals with monitoring cars approaching the crossroads. Describes various methods of detection and discussing their problems. Primary goal is surveillance during the day in different weather conditions, but method of detection cars during the night and low light is also introduced. The most widely used algorithms are implemented using the OpenCV library. Important part is testing different algorithms and also variety of lighting conditions, camera locations and settings.

Identifikace počítače na základě časových značek paketů / Computer Identification Based on Packet's TimestampsKrba, Martin January 2012 (has links)
Basic way how to identify a device in computer network is by MAC address and IP address. Main goal of this work is to create an application capable of clear identification of devices in computer network regardless change of their MAC address or IP address. This is done by exploiting tiny deviations in hardware clock known as clock skew. They appear in every clock based on quartz oscillator. Using clock skew is beneficial, because there is no need of any changes in fingerprinted device nor their cooperation. Accessing these values is done by capturing packets with timestamps included. Application of this method is very wide, for example computer forensics, tracking the device using different access points or counting devices behind router with NAT.

Rozpoznávání podle sítnice oka / Recognition by RetinaStružka, Jaroslav January 2010 (has links)
This thesis deals with recognition by retina (identification, verification). In introduction we describe information about human eye, its diseases with focus on retina impact. Further (in main part) we conduct SW analyses based on biometry retina requirements and design of SW application for retina recognition. It is based on processing pipeline design (sequential application of image filters). This pipeline mostly contains filters focused on edge detection, adaptive threshold and skeletonisation. Finally, basic SW functions includes users registration (enroll), identification, verification. In conclusion we discuss experimental results and success of designed SW in practical application.

Risk Management Support System / Risk Management Support SystemKubíček, Jan January 2010 (has links)
The work provides theoretical base of project management. It describes the current approach to project management as a combination of multiple processes. The process of risk management is described with special care. It also discuss some of the different approaches to risk management. \\ The work suggests new way of handling risk management, that combines risk management and data mining. Data mining approach is also used to mine quantitative risk values. This approach was successfully implemented and tested. Tests showed that this approach is very useful for omitted risk identification. Unfortunately it is also not recommendable for mining quantitative risk values.
