Reclaiming the city: housing for inner-city Johannesburg

The way in which a city is spatially laid out effects the natural environment of the planet (for example: pollution and the depletion of natural resources) as well as the social environment (the community and daily life) of its residents. Through the exploration of various modern urban planning theories, I will begin to look at some different approaches to urban planning. This document favours the compact city approach which advocates higher densities, mixed use development, public transport and community living. This approach relates to initiatives currently underway in the inner city of Johannesburg. This document is about the exploration of how people live in the city and the issues surrounding housing in the urban context. The proposed architectural project is a housing scheme located in Newtown, Johannesburg. Central concepts include: urban regeneration, inner city living, visual variety in the urban realm, street edge conditions and public to private hierarchies

Evolution of the Lower Cretaceous Chifeng Half-Graben Basins, Inner Mongolia, China

As a result of complex extensional tectonics in northeast China and southern portions of Mongolia, some workers have interpreted the Cretaceous Maanshan Uplift and associated Chifeng basins as metamorphic core complex. Previous work has relied solely upon kinematic indicators to determine the structural origin of these basins. To fully understand the creation of these basins, the sedimentiological record was analyzed in this study. The early Cretaceous sedimentary fill of these basins was analyzed to determine if it is synextensional in nature, and if so what manner of extension was in progress during that deposition. The Chifeng basins are filled with four distinct facies associations and are floored by Late Jurassic and Early Cretaceous volcanics of intermediate and felsic composition. The facies associations observed are interpreted as lacustrine/fluvial deposits, alluvial fan conglomerates, distal fan deposits with fluvial deposits, and hanging wall derived fluvial deposits. These facies are composed of sediment shed from the footwall and hanging wall of the master faults and volcanic deposits. Paleocurrent indicators, primarily in the form of imbrication, along with clast count data show provenance directly off the structure separating the two basins and from the eastern margin of the southeast basin. The distribution of facies, as well as paleocurrent data, provenance data, and structural geometry, implies that these basins are of half-graben origin and that no sedimentological evidence exists of an Early Cretaceous metamorphic core complex near Chifeng.

Defining Inner Ear Cell Type Specification at Single-Cell Resolution in a Model of Human Cranial Development

Indiana University-Purdue University Indianapolis (IUPUI) / Inner ear development requires the complex interaction of numerous cell types arising from multiple embryologic origins. Current knowledge of inner ear organogenesis is limited primarily to animal models. Although most mechanisms of cellular development show conservation between vertebrate species, there are uniquely human aspects of inner ear development which remain unknown. Our group recently described a model of in vitro human inner ear organogenesis using pluripotent stem cells in a 3D organoid culture system. This method promotes the formation of an entire sensorineural circuit, including hair cells, inner ear neurons, and Schwann cells. Our past work has characterized certain aspects of this culture system, however we have yet to fully define all the cell types which contribute to inner ear organoid assembly. Here, our goal was to reconstruct a time-based map of in vitro development during inner ear organoid induction to understand the developmental elements captured in this system. We analyzed inner ear organoid development using single-cell RNA sequencing at ten time points during the first 36 days of induction. We reconstructed the on-target progression of undifferentiated pluripotent stem cells to surface ectoderm, pre-placodal, and otic epithelial cells, including supporting cells, hair cells, and neurons, following treatment with FGF, BMP, and WNT signaling modulators. Our data revealed endogenous signaling pathwayrelated gene expression that may influence the course of on-target differentiation. In addition, we classified a diverse array of off-target ectodermal cell types encompassing the neuroectoderm, neural crest, and mesenchymal lineages. Our work establishes the Inner ear Organoid Developmental Atlas (IODA), which can provide insights needed for understanding human biology and refining the guided differentiation of in vitro inner ear tissue. / 2024-08-02

Determinants for Stop-Transfer and Post-Import Pathways for Protein Targeting to the Chloroplast Inner Envelope Membrane

Chloroplast biogenesis relies on the import of thousands of nuclear encoded proteins into the organelle and proper sorting to their sub-organellar compartment. The majority of nucleus-encoded chloroplast proteins are synthesized in the cytoplasm and imported into the organelle via the Toc-Tic translocation systems of the chloroplast envelope. In many cases, these proteins are further targeted to subcompartments of the organelle (e.g. the thylakoid membrane and lumen or inner envelope membrane) by additional targeting systems that function downstream of the import apparatus. The inner envelope membrane (IEM) plays key roles in controlling metabolite transport between the organelle and cytoplasm, and is the major site of lipid and membrane biogenesis within the organelle. In contrast to the protein import and thylakoid targeting systems, our knowledge of the pathways and molecular mechanisms of protein targeting and integration at the IEM are very limited. Previous reports have led to the conclusion that IEM proteins are transferred to the IEM during protein import via a stop-transfer mechanism. Recent studies have shown that at least two components of the Tic machinery (AtTic40 and AtTic110) are completely imported into the stroma and then re-inserted into the IEM in a post-import mechanism. This led me to investigate the mechanisms and pathways involved in the integration of chloroplast IEM proteins in more detail. I selected candidates (AtTic40 for post-import and IEP37 for stop-transfer) that are predicted to have only one membranespanning helix and adopt the same IEM topology to facilitate my analysis. My studies confirm the existence of both stop-transfer and post-import mechanisms of IEM protein targeting. Furthermore, I conclude that the IEP37 transmembrane domain (TMD) is a stop-transfer signal and is able of diverting AtTic40 to this pathway in the absence of AtTic40 IEM targeting information. Moreover, the IEP37 TMD also functions as a topology determinant. I also show that the AtTic40 targeting signals are context dependent, with evidence that in the absence of specific information in the appropriate context, the AtTic40 TMD behaves as a stop-transfer signal. This is an indication that the stop-transfer pathway is the default mechanism of protein insertion in the IEM.

In the past, pastoral-therapeutic support was mainly approached only from the functioning of the left hemisphere of the brain. This implies that pastoral therapists mainly interacted with people in therapy at an intellectual-analytical level under modernistic influence. Since man's destitution and need changes along with time, it is essential for pastoral therapists/counsellors to supplement and adapt their modus operandi and strategies if they wish to remain relevant regarding their pastoral dealings with people. In this study the researcher attempted to formulate from the Scriptures a basis theory regarding the role of music as a whole-brain strategy in inner change. For the sake of a more holistic approach, a meta-theoretical investigation was also undertaken. To confirm the basis and meta-theoretical information, researcher entered the living reality of seven individuals during which a qualitative research was done. Attending to the inner life of an educator: the human dimension in education

My dissertation is a selection of essays that reflect upon human potential, particularly but not exclusively, within educational environments. I offer theory and practices that suggest that under the right conditions educators and students will move towards the far reaches of their own creative capacities. I offer my own experience and practice as an exemplar of possibilities. I make proposals about educators and education of educators that represent a paradigm shift from centralizing curriculum and content to focusing on care, nurturance, subjective and inter-subjective understanding, and development of educators. The reader is invited to see educators as central, and is encouraged towards the possibility that educators must be supported, encouraged, and cared for in order to support emergence of their vitality, first for themselves and subsequently for students. I outline an approach that puts human beings in educational environments first in practical and specific ways. Integration of personal experience and curriculum material is explicated. The importance of personal inner work for educators is highlighted. Inner Work is characterized as a personal and spiritual process. The claim that educators need to have group facilitation skills is made and evidence offered. Philosophical and theoretical background from education, eastern and western philosophy, humanistic and transpersonal psychology, process-oriented methods, and counselling psychology are drawn upon. The approach is holistic and systemic. The human is viewed as important but not separate from other living beings or the environment. The values of presence, care, and deep democracy underlie the ideas. The importance of relationality and I-Thou connection are explicated. The writing and research draws on a variety of qualitative approaches, including, living inquiry, autobiography, and self-study, as well as conceptual, narrative, poetic, auto-ethnographic, heuristic, and analytic methods. The material, personal, and ephemeral are investigated as integrated parts of the Dao-Field of education and life.
