These fuels tend to become extinct, so that, in order to guarantee economic, social, and ecological progress,it is necessary to look for renewable alternatives for the generation of energy. Bio-hydrogen production resulting from the biological method stands out due to the possibility of its obtaining from residue. Therefore, this work has the objective of evaluating the efficiency of anaerobic sequential batch reactor with immobilized biomass (AnSBBR) for the production of bio-hydrogen from cassava starch wastewater (CSW). The AnSBBR used in the tests presents mechanical agitation system and total and useful volumes of 6.0 and 4.3 L, respectively. The reactor was kept in a controlled temperature chamber (30 °C). Four experimental conditions (I, II, III, and IV) were carried out in a fed batch with organic volumetric loads (9.0, 13.5, 18.0, and 18.0 g-total sugars L-1d-1) for cycle durations of 4; 4; 3; and 2 h, respectively.Conditions I, II, and IV were inoculated with sludge from an anaerobic reactor from CSW, and condition III was with auto-fermented inoculum. It was found that the inoculum from the anaerobic reactor of the cassava starch effluent influenced the production of biogas in AnSBBR reactors. Condition II showed total sugar conversion of 84%, hydrogen molar productivity of 35.8 mLH2 m-3d-1 and hydrogen yield related to total sugar applied and removed of 2.42 and 4.6 molH2 Kg total sugars-1, respectively. In condition III (18 g-total sugars L-1d-1 and TDH of 3 h) with auto-fermented inoculum, there was no hydrogen production; however, it presented 98.47% of total sugar conversion throughout the operation. As for the intermediate products, there was an increase of acetic, butyric, and propionic acids in the reactor. However, the lactic acid did not increase, probably due to consumption by microorganisms, except in condition IV. / A demanda energética vem sendo atendida, em sua maior parte, por fontes de energia não renováveis advindas, principalmente, de combustíveis fósseis. Esses combustíveis tendem a se extinguir, de forma que se faz necessário, para garantir os avanços econômico, social e ecológico, buscar alternativas renováveis para a geração de energia.A produção de bio-hidrogênio, resultante do método biológico, se mostra de interesse pela possibilidade da sua obtenção a partir de resíduos.Diante disso, esse trabalho teve por objetivo avaliar a influência da carga orgânica na produção de bio-hidrogênio, a partir de água residuária de fecularia de mandioca (ARF) em reator anaeróbio operado em bateladas sequenciais com biomassa imobilizada (AnSBBR). O AnSBBR utilizado nos ensaios apresenta sistema de agitação mecânica e volume total e útil de 6,0 e 4,3L, respectivamente. O reator foi mantido em câmara com temperatura controlada (30 °C). Foram realizadas quatro condições experimentais (I, II, III e IV) em batelada alimentada com cargas orgânicas volumétricas de 9,0; 13,5; 18,0 e 18,0 g-carboidratos totais.L-1d-1), nos tempos de ciclo de 4; 4; 3 e 2 h, respectivamente. As condições I, II e IV foram inoculadas com lodo de um reator anaeróbio tratado termicamente, e a condição III com inóculo obtido por autofermentação da ARF. Concluiu-se que o inóculo proveniente do reator anaeróbio influenciou na produção de biogás em reatores AnSBBR. A condição II apresentou a conversão de açúcar total de 84%, a produtividade molar de hidrogênio de 35,8 molH2 m-3d-1 e o rendimento de hidrogênio em relação ao açúcar total aplicado e removido de 2,42 e 4,6 molH2 Kg-carboidratos totais-1, respectivamente. Na condição III (18 g-carboidratos totais L-1d-1e TDH de 3 h) com inóculo autofermentado não houve produção de hidrogênio; no entanto, houve conversão de açúcar total de 98,47% ao longo da operação. Quanto aos produtos intermediários, ocorreu aumento dos ácidos acético, butírico e propiônico no reator. Todavia, o ácido lático não aumentou, provavelmente devido ao consumo pelos micro-organismos, exceto na condição IV.

In this work, we focused on the expression of the AmpC group β- lactamase DHA-1 and its negative regulator AmpR, as well as the porins OmpK35 and OmpK36 and on effect of inoculum. We used well-characterized Klebsiella pneumoniae strains in this study. Plasmids obtained from these strains were also transformed into different wild-type Klebsiella pneumoniae strains, which were typed by pulsed-field gel electrophoresis (PFGE) and multi-locus sequence typing (MLST). Gene expression analysis was performed by RT-PCR using specific primers and TaqMan probes. In most strains, expression was dependent on the presence of an inducer. The highly resistant strain showed a different expression pattern, but the expression of blaDHA-1 remained inducible by cefoxitin. Different regulation was also observed in the transformants. Based on our data, we suggest that the previously described regulatory pathway for AmpC is not generally suitable, and we propose that there are more...

Assessment of the Bacterial Growth Potential of Reverse Osmosis Produced Chlorinated Drinking WaterFelix, Alejandra Ibarra 07 1900 (has links)
Reverse Osmosis (RO) filtration is capable of producing high quality drinking water with an ultra-low nutrient level. Therefore, a very low bacterial growth potential (BGP). BGP is a key bioassay to evaluate microbial quality and the biological stability of drinking water.
Current methods to assess BGP in drinking water need to be adapted to the wide variety of water types due to results could highly vary from each, providing unreliable insights to the biological stability of the assessed water.
This study evaluates the application of an FCM-based BGP assay for RO produced chlorinated drinking water. The approach combines (i) the standardization of a quenching agent concentration, (ii) the impact of sample pre-treatment such as filtration and pasteurization on the BGP of RO produced chlorinated water, (iii) the effect of different inoculums (an indigenous community and a mixture with bottled water) on the bacterial growth and their longevity after being stored, (iv) the use of BGP to assess the performance of carbon filters in removing chlorine and (v) the use of BGP to assess the effect of the addition of magnesium on bacterial growth. The results showed that high concentrations of sodium metabisulphite (> 7.5 mg/L) decrease the pH levels of the water,thus, inhibiting bacterial growth. Filtration had a significant effect on BGP values (2.62 x10^5 intact cells/mL) in comparison to pasteurization (9.02 x 10^4 intact cells/mL), when compared to the control. Using a mixture of water types as inoculum might provide a better insight to bacterial growth potential in water due to a higher consumption of nutrients. BGP demonstrated to be a sensitive tool to test the performance of carbonfilters applied to remove chlorine and its applicability to evaluate the biological stability of RO produced chlorinated drinking water. The concentrations of magnesium chloride tested in this study did not have a significant effect on cell numbers.

Schlechtend.:Fr. f.sp. cepae (H.N. Hans.) W.C. Snyder & H.N. Hans. (Focep) has
been identified as the leading cause of harvest and storage losses. This pathogen is of
world-wide importance and causes Fusarium basal rot of onions (Allium cepa),
affecting all onion growth stages. No information is available on the evolution,
genetic diversity, molecular detection and inoculum sources of the South African
Focep population.
Similar to what is the case for South Africa, limited information is available
on Focep in other regions of the world. World-wide, four vegetative compatibility
groups (VCGs) and two single-member VCGs (SMVs) have been identified among
two Japanese and 19 Colorado (USA) isolates. This polyphyletic origin of Focep
suggested by VCG analyses was confirmed through molecular analyses of isolates
from a few countries. Only the mating type (MAT)1-1 idiomorph has been reported
for Focep isolates from Welsh onion (Allium fistulosum).
The development of sustainable management strategies of Focep is dependent
on knowledge of (i) the genetic diversity and evolution of Focep, (ii) whether high
throughput molecular methods can be developed for identifying the most virulent and
widespread Focep genotypes and (iii) the role of seedlings and seeds as primary
inoculum sources, and the Focep genotypes associated with these growth stages.
Therefore, the three main aims of the current study were to investigate the
aforementioned three aspects.
In the first aim of the study, the genetic diversity and evolution of Focep was
investigated using a collection of 79 F. oxysporum isolates from South Africa (27
Focep and 33 non-pathogenic isolates) and Colorado (19 Focep isolates). VCG
analyses revealed the presence of six VCGs, four among the Colorado Focep isolates
(VCGs 0421, 0422, 0423 and 0424) and two among the South African bulb-associated
isolates (VCGs 0425 and 0426). VCG 0421 and VCG 0425 were the two main VCGs
in Colorado and South Africa, respectively. Four SMVs and one heterokaryon selfincompatible
(HSI) isolate were also identified. The polyphyletic nature of Focep in South Africa and Colorado was shown through a combined translation elongation
factor 1α (EF-1α) and mitochondrial small-subunit (mtSSU) phylogeny. The
phylogeny divided the Focep isolates into two main clades, of which one contained
the two main VCGs (0421 and 0425), SMVs and non-pathogenic isolates. The
second, ancestral clade contained the HSI isolate, VCGs 0422, 0423 and 0424, and
non-pathogenic isolates. Unlike the clade containing the two main VCGs, which were
highly virulent toward onion bulbs, the ancestral clade contained isolates that were
mostly moderately virulent. The incongruence of the EF-1α and mtSSU datasets with
an intergenic spacer (IGS) region data set, and the presence of both MAT idiomorphs
within the same isolate for some isolates, suggested possible exchange of genetic
material between isolates.
The second aim of the study was to develop molecular methods for identifying
the two main Focep VCGs (0425 and 0421), using DNA fingerprinting methods and
sequence-characterized amplified region (SCAR) markers. These techniques were
first developed using the F. oxysporum isolates from the first aim, and were then used
to investigate the prevalence of VCG 0425 among 88 uncharacterized F. oxysporum
isolates from onion bulbs in South Africa. Two random amplified polymorphic DNA
primers provided two diagnostic amplicons for VCG 0425, but attempts to develop
SCAR markers from these amplicons were unsuccessful. In contrast, an interretrotransposon
amplified polymorphism (IRAP) fingerprinting method enabled the
developed of a multiplex IR-SCAR polymerase chain reaction method that detected
the VCG 0421, 0425 and SMV 4 isolates as a group. Fingerprinting and SCAR
marker testing of the 88 uncharacterized F. oxysporum isolates from South Africa (65
Focep and 23 non-pathogenic) confirmed that VCG 0425 is the main VCG in South
Africa associated with mature onion bulbs, since 63 of the Focep isolates had the
molecular characteristics of VCG 0425.
The third aim of the study was to determine whether seed and seedling
transplants are inoculum sources of Focep, and whether the same genotype (VCG
0425) that dominated on mature bulbs could be detected from these sources. Focep
isolates were obtained from seven of the 13 investigated onion seed lots, as well as
from onion seedling transplants that were collected from all five onion nurseries in the
Western Cape. Focep seedling infection more than doubled from the 6-week growth stage to the 14-week growth stage. Seed infections by Focep were low, but the
seedborne nature of Focep was confirmed by showing that a green fluorescent protein
labelled Focep transformant could be transmitted from infected soil to onion seed via
the onion bulbs and seedstalks. It is thus clear that commercial seed and seedlings are
inoculum sources of Focep. However, the Focep genotypes on seed and seedlings are
different from those in mature bulbs and were not dominated by VCG 0425.
Furthermore, most (≤ 60%) of the seed and seedling isolates were moderately
virulent, as compared to the mostly highly virulent isolates from mature bulbs. / AFRIKAANSE OPSOMMING: In die Wes-Kaapse uiebedryf in Suid-Afrika is Fusarium oxysporum

geïdentifiseer as die vernaamste oorsaak van oes- en opbergingsverliese. Hierdie
patogeen is van wêreldwye belang; dit veroorsaak Fusarium-bolvrot van uie (Allium
cepa) en affekteer alle plantgroeistadia. In Suid-Afrika is daar geen inligting
beskikbaar oor die evolusie, genetiese diversiteit, molekulêre opsporing en
inokulumbronne van die Focep-populasie nie.
Soortgelyk aan wat die geval in Suid-Afrika is, is daar beperkte inligting
beskikbaar oor Focep in ander wêrelddele. Wêreldwyd is daar vier vegetatiewe
versoenbaarheidsgroepe (VVGe) en twee enkellid VVGe (ELVe) geïdentifiseer onder
twee Japannese en 19 Colorado (VSA) isolate. Hierdie veelvuldige oorsprong van
Focep wat deur VVG-analise voorgestel was, is deur die molekulêre analises van
isolate uit ’n paar ander lande bevestig. Slegs die paringstipe (PT)1-1 idiomorf is vir
Focep-isolate uit Walliese-tipe uie (ook bekend as ‘lenteuie’ in Suid Africa) (Allium
fistulosum) berig.
Die ontwikkeling van volhoubare bestuurstrategieë vir Focep steun op kennis
van (i) die genetiese diversiteit en evolusie van Focep, (ii) of hoë-deurset molekulêre
metodes ontwikkel kan word vir die identifisering van die mees virulente en
wydverspreide Focep-genotipes en (iii) die rol van saailinge en saad as primêre
inokulumbronne, en die Focep-genotipes wat met hierdie groeistadia geassosieer
word. Daarom was die hoof doelstellings van hierdie studie om die bogenoemde drie
aspekte te bestudeer.
Om die eerste doel van die studie te bereik is die genetiese diversiteit en
evolusie van Focep bestudeer deur gebruik te maak van ‘n versameling van 79 F.
oxysporum-isolate uit Suid-Afrika (27 Focep en 33 nie-patogeniese isolate) en uit
Colorado (19 Focep-isolate). VVG-analises het die teenwoordigheid van ses VVGe
aangetoon – vier onder die Colorado Focep-isolate (VVGe 0421, 0422, 0423 en 0424)
en twee onder die Suid-Afrikaanse bol-geassosieerde isolate (VVGe 0425 en 0426).
VVG 0421 en VVG 0425 was die twee hoof VVGe in onderskeidelik Colorado en Suid-Afrika. Vier ELVe en een meerkernige self-onversoenbare (MSO) isolaat is ook
geïdentifiseer. Die veelvuldige oorsprong van Focep in Suid-Afrika en Colorado is
ook aangetoon deur ‘n gekombineerde translasie verlengings faktor 1α (VF-1α) en
mitokondriale klein-subeenheid (mtKSE) filogenie. Dié filogenie het die Focepisolate
in twee groepe verdeel, waarvan die een groep die twee hoof VVGe (0421 en
0425), ELVe en nie-patogeniese isolate bevat het. Die tweede, basal groepering het
die MSO-isolaat, VVGe 0422, 0423 en 0424, en nie-patogeniese isolate bevat. In
teenstelling met die eersgenoemde groepering wat hoogs virulente isolate van uiebolle
bevat het, het die basale groepering isolate bevat wat meestal matig virulent was. Die
inkongruensie van die VF-1α en mtKSE-datastelle met ‘n intergeen-gespasieerde
(IGS) area datastel – asook die teenwoordigheid van beide PT-idiomorwe binne
dieselfde isolaat by sommige isolate – het op ’n moontlike uitruiling van genetiese
materiaal tussen isolate gedui.
Die tweede doel van die studie was om molekulêre metodes te ontwikkel vir
die identifisering van die twee hoof Focep VVGe (0425 en 0421) deur gebruik te
maak van DNA-vingerafdrukke en nukleotied-gekarakteriseerde geamplifiseerde area
(NKAA) merkers. Hierdie tegnieke is ontwikkel deur van die F. oxysporum-isolate
van die eerste doelstelling gebruik te maak en is daarna gebruik om die frekwensie
van VVG 0425 onder 88 ongekarakteriseerde F. oxysporum-isolate van uiebolle in
Suid-Afrika te ondersoek. Twee gerandomiseerde geamplifiseerde polimorfiese DNS
(RAPD) merkers het twee diagnostiese nukleotiedbasis-areas vir VVG 0425 gelewer,
maar pogings om NKAA-merkers uit hierdie geamplifiseerde nukleotiedbasis-areas te
onwikkel was onsuksesvol. In teenstelling hiermee het ‘n inter-retrotransposon
geamplifiseerde polimorfisme (IRAP) vingerafdrukmetode die ontwikkeling van ‘n
multipleks IR-NKAA polimerase kettingreaksiemetode moontlik gemaak wat die
VVG 0421-, VVG 0425- en ELV 4-isolate as ’n groep aangedui het.
Vingerafdruktoetsing en NKAA-merkertoetsing van die 88 ongekaraktariseerde F.
oxysporum isolate van Suid-Afrika (65 Focep en 23 nie-patogenies) het bevestig dat
VVG 0425 die hoof VVG in Suid-Afrika is wat met volwasse bolle geassosieer word,
aangesien 63 van die Focep-isolate die molekulêre eienskappe van VVG 0425 gehad
het. Die derde doel van die studie was om vas te stel of saad en saailinge
inokulumbronne van Focep is, en of dieselfde genotipe (VVG 0425) wat op volwasse
bolle dominant is, waargeneem kon word op hierdie bronne. Focep-isolate is verkry
van sewe van die 13 uiesaadlotte asook van uiesaailinge wat in al vyf
uiesaailingkwekerye in die Wes-Kaap versamel is. Focep-saailinginfeksie was meer
as dubbel in die 14-week groeistadium as wat dit in die 6-week stadium was.
Saadinfeksies deur Focep was laag, maar die saadgedraagde aard van Focep is
bevestig deur aan te toon dat ’n Focep-transformant wat met ‘n groen fluoreserende
proteïen geëtiketeer is, van geïnfekteerde grond na uiesaad oorgedra kon word via die
uiebolle en -saadstele. Dit is dus duidelik dat kommersiële saad en saailinge as
inokulumbronne van Focep dien. Die Focep-genotipes op saad en saailinge verskil
egter van dié in volwasse bolle en is nie deur VVG 0425 gedomineer nie. Verder was
die meeste (≤ 60%) saad- en saailingisolate matig virulent, in teenstelling met die
meestal hoogs virulente isolate uit volwasse bolle.

Spatial distribution and temporal progress of Huanglongbing in areas with strict disease management / Distribuição espacial e progresso temporal de Huanglongbing em áreas com manejo rigoroso da doençaPazolini, Kelly 20 March 2019 (has links)
Citriculture is an important agroindustrial activity for Brazil and citrus production is concentrated in São Paulo State. However, the activity is facing a Huanglongbing (HLB) epidemic which is considered to be the main threat to global citrus production. In spite of many management efforts, the incidence of HLB continues to increase since its detection in São Paulo State, in 2004. The difficulty of handling HLB is associated with the migration of the vector, Diaphorina citri, which acquires the bacteria in external inoculum sources and the limited effect of the strict HLB management performed by farms (eradication of symptomatic trees and frequent vector control) on the primary dissemination of the disease. Thus, epidemiological studies can facilitate the understanding of the complex pathosystem and help in the decision of more efficient control measures. The objectives of this study were: 1) to determine the spatial pattern of HLB in 24 citrus blocks under strict management of the disease and 2) to study the temporal progress of the disease and the spatial distribution of HLB and the vector in a large scale property that carried out the strict management of the disease and its association with external inoculum sources. To verify the spatial pattern of HLB, modified Ripley\'s K-function and kernel density estimation maps were used. The results showed that the regular distribution pattern of HLB symptomatic trees prevailed in the blocks studied and a weak aggregation of approximately 58 m was observed at the edges of the blocks. The largest aggregation distances and averages of D. citri/trap occurred in blocks located at the periphery of the properties. The regular spatial pattern and the concentration of disease and vector in the periphery of the properties evidenced the importance of primary dissemination. For the study of the disease temporal progress, the Gompertz and logistic models were adjusted to the annual progress of HLB for 177 blocks. The distribution of HLB progress rates and incidence (for 177 blocks) and average D. citri/trap (for 296 blocks) were visualized on gradient maps. The disease progress rate values obtained with the Gompertz model (0.04 to 0.28 per year), average HLB incidence (0.11) and average D. citri/trap (0.016) were low compared with literature data. The variables presented the highest values for blocks located at the periphery of the property, rather than for blocks surrounded by citrus. Many noncommercial plants, potentially serving as source of inoculum, were found near blocks with the highest rates of disease progress, HLB incidence and average D. citri/trap. The results demonstrated the importance of primary dissemination in areas that carry out a strict HLB management. Therefore, in order to reduce new infections, HLB management must effectively reduce external sources of inoculum and regional management should include, in addition to commercial orchards, noncommercial plants. / A citricultura é uma importante atividade agroindustrial para o Brasil e grande parte da produção se concentra no estado de São Paulo. Entretanto, a produção de laranja enfrenta, atualmente, uma epidemia de Huanglongbing (HLB), doença considerada a principal ameaça à citricultura mundial. Apesar de muitos esforços no manejo, a incidência de HLB continua aumentando desde a sua detecção no estado de São Paulo, em 2004. A dificuldade de manejo de HLB está associada à migração do inseto vetor, Diaphorina citri, que adquire a bactéria em fontes de inóculo externas e ao efeito limitado do manejo rigoroso da doença realizado pelas fazendas (erradicação de plantas sintomáticas e controle frequente do vetor) sobre a disseminação primária da doença. Assim, estudos epidemiológicos podem facilitar o entendimento do complexo patossistema e auxiliar na decisão de medidas de controle mais eficientes. Os objetivos desse trabalho foram: 1) determinar o padrão espacial de HLB em 24 blocos cítricos sob o manejo rigoroso da doença e 2) estudar o progresso temporal da doença e a distribuição espacial de HLB e do vetor em uma propriedade de larga escala que realizou o manejo rigoroso da doença e sua associação com fontes externas de inóculo. Para verificar o padrão espacial de HLB, utilizou-se a analise função-K de Ripley modificada e mapas de densidade de kernel. Os resultados demonstraram que o padrão de distribuição regular de plantas sintomáticas de HLB prevaleceu nos blocos estudados e uma agregação fraca de aproximadamente 58 m foi observada nas bordas dos blocos. As maiores distâncias de agregação e médias de D. citri/armadilha ocorreram em blocos localizados na periferia das propriedades. O padrão espacial regular e a concentração da doença e do vetor na periferia da propriedade evidenciaram a importância da disseminação primária na área. Para o estudo do progresso temporal, os modelos Gompertz e logístico foram ajustados ao progresso anual da doença de 177 blocos. A distribuição das taxas de progresso e incidência de HLB (para 177 blocos) e a média de D. citri/armadilha (para 296 blocos) foram visualizadas em mapas de gradiente. Os valores encontrados de taxa de progresso da doença, obtida com o modelo de Gompertz (de 0,04 a 0,28 por ano), incidência média de HLB (0,11) e média de D. citri/armadilha (0,016) foram baixos em comparação com dados da literatura. As variáveis apresentaram os maiores valores para blocos localizados na periferia da propriedade do que para blocos rodeados por citros. Uma grande quantidade de plantas não comerciais, e potencialmente servindo como fonte de inóculo, foi encontrada perto de blocos que apresentam as maiores taxas de progresso e incidências de HLB e médias de D. citri/armadilha. Os resultados encontrados demonstraram a importância da disseminação primária em áreas que realizam o manejo rigoroso de HLB. Portanto, para reduzir novas infecções, o manejo do HLB precisa reduzir efetivamente as fontes externas de inóculo e o manejo regional deve incluir, além de pomares comerciais, plantas não comerciais.

Previous issue date: 2007-03-14 / Conselho Nacional de Pesquisa e Desenvolvimento Científico e Tecnológico - CNPq / The Fusarium rot, caused by the fungus Fusarium pallidoroseum, is an important postharvest disease of melon (Cucumis melo) fruits in the Brazil. This work aimed to analyze the influence of the inoculation method (pulverization, drop deposition, pulverization with wound, drop deposition with wound, and sub-epidermal injection), wound age (0, 12 and 24 hours), humidity (with and without moist chamber), temperature (10, 15, 20, 25, 30 e 35°C) and inoculum concentration (101, 102, 103, 104, 105, 106 conidia/mL-1) of three F. pallidoroseum isolates (CF-589, CF-685 and CF-687) on the severity of the Fusarium rot in melon fruits type Cantaloupe (cv. Torreon) and Honeydew (cv. Orange Flesh). The three F. pallidoroseum isolates had provoked symptoms of Fusarium rot in the evaluated situations, but they had not differed significantly (P=0,05) between itself how much to the severity levels of disease, as well as had not been evidenced significant interactions with melon’s cultivars. On the other hand, in all the experiments had been evidenced significant differences in the disease levels between two melon’s cultivar, with cv. Torreon presenting levels of disease’s severity significantly higher to the evidenced on Orange Flesh cultivar. It did not haveseverity significantly higher to the evidenced on Orange Flesh cultivar. It did not have development of injuries in the fruits when the inoculations had been carried without wound. The inoculation for atomization of the conidia’s suspension propitiated the biggest injuries in the submitted fruits at wounds. The inoculation for sub-epidermal injection caused lesser injuries than methods of atomization or drop with wound. It had the reduction of the severity of the disease in the fruits with the increase of the age of the wound. The injuries had been significantly lesser in the wounded fruits 24 hours before the inoculation than in those wounded fruits with 12 hours before or immediately before the inoculation. The free water presence in the surface of the fruits was unnecessary for the beginning of the infection’s process for F. pallidoroseum isolates, even so the injuries has been bigger in the fruits submitted to the humid chamber. The temperature significantly influenced the severity of the Fusarium rot, being that the optimum temperatures estimated for the disease’s development were 23,9 °C e 23,0 °C, respectively for Orange Flesh e Torreon cultivar. The severity of the disease increased with the increment in the inoculum concentration of F. pallidoroseum. The biggest injuries had been observed in the concentration of 1x106 conidia/mL-1 in the twocultivars, but same in lesser inoculum concentration (1x101 conídios/mL-1), they registered considerable levels of disease. The inoculum concentration necessary to generate 50% of the maximum size of the injury Orange Flesh cultivar was four times higher than that demanded for Torreon cultivar. / A podridão-de-fusário, causada pelo fungo Fusarium pallidoroseum, é uma importante doença em pós-colheita de frutos de meloeiro (Cucumis melo) no Brasil. Este trabalho teve como objetivos avaliar a influência dos métodos de inoculação (atomização, gota, atomização com ferimento, gota com ferimento e injeção subepidérmica), da idade do ferimento (0, 12 e 24 h), da umidade (sem e com câmara úmida), da temperatura (10, 15, 20, 25, 30 e 35 °C) e da concentração de inóculo (1x101, 1x102, 1x103, 1x104, 1x105 e 1x106 conídios.mL-1) de três isolados de F. pallidoroseum (CF-589, CF-685 e CF-687) na severidade da podridão-de-fusário em frutos de meloeiro dos tipos cantaloupe (cv. Torreon) e honeydew (cv. Orange Flesh). Os três isolados de F. pallidoroseum provocaram sintomas da podridão-de-fusário nas situações avaliadas, mas não diferiram significativamente (P=0,05) entre si quanto aos níveis de severidade da doença, bem como não foram constatadas interações significativas com as cultivares de meloeiro. Por outro lado, em todos os experimentos foram constatadas diferenças significativas nos níveis de doença entre as duas cultivares de meloeiro, com a cultivar Torreon apresentando níveis de severidade da doença significativamente superiores aos constatados na cultivar Orange Flesh. Não houvedesenvolvimento de lesões nos frutos quando as inoculações foram realizadas sem ferimento. A inoculação por atomização da suspensão de conídios propiciou as maiores lesões nos frutos submetidos a ferimentos. A inoculação por injeção subepidérmica ocasionou lesões menores que os métodos de atomização ou gota com ferimento. Houve a redução da severidade da doença nos frutos com o aumento da idade do ferimento. As lesões foram significativamente menores nos frutos feridos 24 horas antes da inoculação do que naqueles feridos com 12 horas antes ou imediatamente antes da inoculação. A presença de água livre na superfície dos frutos foi desnecessária para o início do processo de infecção pelos isolados de F. pallidoroseum, embora as lesões tenham sido maiores nos frutos submetidos à câmara úmida. A temperatura influenciou significativamente a severidade da podridão-de-fusário, sendo que as temperaturas ótimas estimadas para o desenvolvimento da doença foram 23,9 °C e 23,0 °C para Orange Flesh e Torreon, respectivamente. A severidade da doença aumentou com o incremento na concentração de inóculo de F. pallidoroseum. As maiores lesões foramobservadas na concentração de 1x106 conídios.mL-1 nas duas cultivares, mas mesmo na menor concentração de inóculo (1x101 conídios.mL-1) foram registrados consideráveis níveis de doença. A concentração de inóculo necessária para gerar 50% do tamanho máximo da lesão na cultivar Orange Flesh foi mais de quatro vezes superior à exigida pela cultivar Torreon

Produção de hidrogênio em reator anaeróbio de leito fixo / Hydrogen production using up-flow anaerobic packed bed reactorFernandes, Bruna Soares 16 May 2008 (has links)
O hidrogênio é estudado como alternativa ao uso de combustíveis fósseis para geração de energia, uma vez que é um combustível renovável, apresenta alta concentração de energia por unidade de massa e não gera gases causadores do efeito estufa. Entre os processos de produção de hidrogênio destaca-se o processo fermentativo, pois é um processo de baixo custo quando comparado com outros processos e possibilita unir tratamento de efluente e geração de energia. Neste sentido, este trabalho teve como proposta estudar parâmetros envolvidos no processo de produção fermentativo do \'H IND.2\'. O trabalho envolveu três etapas. Na primeira etapa, foi estudada a produção de hidrogênio a partir de sacarose empregando reatores anaeróbios de leito fixo de fluxo ascendente. Na primeira fase, comparou-se o desempenho de diferentes matérias suportes (argila, carvão vegetal e polietileno) e tempos de detenção hidráulica (TDH) (0,5 e 2h). Na segunda fase, testaram-se diferentes porosidades (50, 75 e 91%) do leito de polietileno TDH de 0,5 h. Os resultados mostraram que TDHs menores e maiores porosidades promovem maiores e contínuas produções de \'H IND.2\'. Na segunda fase, avaliou-se a produção de \'H IND.2\' a partir de quatro inóculos: metanogênico tratamento termicamente e três provenientes de biomassa aderidas aos materiais suportes empregados na primeira etapa. Todos inóculos produziram \'H IND.2\'. Na terceira etapa, avaliou-se a viabilidade de produzir \'H IND.2\' a partir de diferentes águas residuárias (sacarose, esgoto sanitário, vinhaça e glicerina). Houve conversão de hidrogênio a partir de todas as águas residuárias e a vinhaça mostrou ser o efluente mais promissor para esta finalidade. As análises biológicas mostraram baixa diversidade de fungos e bactérias, porém todos associados com o processo de formação de \'H IND.2\'. A varredura dos parâmetros estudados neste trabalho proporcionou o entendimento do processo, assim como, o mapeamento das variáveis adequadas para o projeto e viabilidade da aplicação de reatores desenvolvidos para geração de hidrogênio. / The hydrogen obtained by fermentative production is studied as an alternative process to provide energy instead of fossil fuel application. Moreover, hydrogen is a renewable fuel, has high energy content per unit weight (122 kJ/g), generates clean energy without pollution and produces no greenhouse gases. The fermentative process has low cost when it is compared with traditional process and photosynthetic process, because hydrogen can be produced from wastewater by anaerobic treatment process. For that reason, the aim of this research was to study some parameters involved in the hydrogen production by fermentative process. Three steps were developed. In the first step, it was studied the hydrogen production from sucrose using up-flow anaerobic packed-bed reactor, this step was divide in two phases. In the first phase three support materials (clay beads, vegetal coal and polyethylene) and two hydraulic retention times (0.5 and 2 h) were tested. In the second phase three porosities (50, 75 and 91%) of polyethylene bed were tested. The results demonstrated that the low HRT and high porosities provided high hydrogen production, although, the support materials did not show significant difference in the hydrogen production and in the biomass developed. In the second phase, four inocula were used in order to produce hydrogen: thermal pre-treated methanogenic sludge; and the others three came from the reactors used in the first phase. All inocula were able to produce hydrogen. In the third step hydrogen production was obtained from three wastewaters (domestic wastewater, vinasse and glycerol) and a control (sucrose) in batch reactors. The wastewaters and control produced hydrogen and the vinasse showed the highest production. This research makes available the comprehension on the influence of the different parameters in processes projected for hydrogen production and it makes viable to apply in full-scale.
