In the present thesis called "The modern analytical methods for detection of aminoacids asociated with malignant prostate tumours" sarcosine has been studied as a potential tumor marker which has significance for the diagnosis of aggressive forms of prostate cancer. The effect of addition of sarcosine prostate line PC3 has been also studied as well. Next studied area were the the interactions with amino acid sarcosine, sodium chloride, uric acid and uracil. We have primarily focused also on the development of methods for the detection of sarcosine in urine samples of patients with Cap and prostate cells.

1 Abstract Charles University in Prague Faculty of Pharmacy in Hradec Králové Department of Pharmaceutical Botany Author: Vendula Ježková Supervisor: RNDr. Jitka Vytlačilová, Ph.D. Title of diploma thesis: Evaluation of natural substances using in vivo tests with Artemia salina Biological activity evaluation of substances is important because of toxic effect's assessment on chemical and physiological functions of organisms and their parts. For acute toxicity evaluation was used crustacean Artemia salina in this study. They were tested aqueouses extracts isolated from various plant drugs of Salviae folium, Apii semen, Coptidis rhizoma, Evodiae fructus, Zanthoxyli radix and Ziziphi fructus and two isolated alkaloids galanthamine and huperzine A. For each extract and alkaloid was calculated value of lethal concentration LC50. Results of this study showned, that the extract of Coptis chinensis, LC50 = 0,134 ± 0,004 mg/ml, had most toxic effects. Further the extract's toxicity decreased in the order Ziziphus jujuba, LC50 = 0,817 ± 0,165 mg/m > Zanthoxylum nitidum, LC50 = 1,154 ± 0,015 mg/ml > Salvia officinalis, LC50 = 2,074 ±0,462 mg/ml > Evodia rutaecarpa, LC50 = 2,166 ± 0,244 mg/ml > Apium graveolens, LC50 = 4,041 ± 0,108 mg/ml. The goal of the Thesis is the analysis of maritime transportation of dangerous goods. Thesis is divided into 4 Parts. Chapter 1 describes development of dangerous goods regulations with the emphasis on maritime safety and environmental aspects. Chapter 2 is based on the analysis of IMDG Code, whose regulations have the fundamental importance for the carriages of dangerous goods by sea. Chapter 3 is dedicated to the basic duties of the parties included in the maritime transportation of dangerous goods- carriers on the one side and shippers on the other side. Final Chapter is related to the maritime carriage of oil - one of the dangerous substances with the strategic importance.

Criminological Aspects of Drug-Related Crime The thesis deals with the topic of criminological aspects of drug-related crime. Due to the breadth of the issue, the work is focused only on the so-called primary drug crime, which is understood as a criminal activity consisting in violating the regulations governing the handling of narcotic drugs and psychotropic substances. Specifically, it deals with crime associated with the unauthorized handling of cannabis. The main goal of this work is to answer the question whether the criminal law regulation of unauthorized handling of cannabis chosen by the legislator is appropriate and respects in a reasonable way the specific criminological aspects related to cannabis. The first part focuses on explaining some basic terms that are used in the work. The second part deals with the interpretation of terms that are specifically related to the issue of illicit handling of cannabis. Also it includes the issue of the harmfulness of cannabis use and describes the state and trends of crime associated with cannabis use. The subject of this thesis was the determination of the profile of volatile aromatic compounds of varietal wines and the subsequent statistical analysis, aiming to find varietal markers of selected varieties – individual substances, their typical contents, mutual correlations. The HS-SPME-GC-MS technique was used to determine the aromatic profile. The main focus of this work was the analysis of the varieties Pálava, Hibernal, Johanniter and Solaris. The data obtained were statistically processed using descriptive and multivariate statistical methods. On the basis of these analyses, models were developed which, from the contents of 14 selected compounds of the aromatic profile, allow the Piwi varieties Hibernal, Johanniter and Solaris to be distinguished from each other and from the Pálava variety and, for the Pálava variety wines, to determine their origin in the Znojmo or Mikulov wine sub-region.

This diploma thesis deals with the comparison of aroma compounds in goat and cow milk. The content of aroma active substances in samples of goat and cow milk, which were analyzed by HS-SPME-GC-MS method, is monitored. In the theoretical part, milk is characterized, its formation in the dairy mammary gland is described, its composition including an overview of aromatic active substances, physical and chemical as well as its sensory properties, milk defects and differences between cow's and goat's milk are described. Further, gas chromatography in combination with solid phase microextraction technique in tandem with mass spectrometry is described. In the experimental part, 13 samples of goat and 8 samples of cow milk were subjected to HS-SPME-GC-MS analysis. Samples were analyzed fresh and after heat treatment. Altogether, 78 volatile substances were identified in the samples. Ketones were the most numerous, aldehydes, alcohols and carboxylic acids were the most abundant chemical groups. There were differences between samples in number as well as in content of the identified compounds, 64 in goat milk and 48 in the cow milk.
