This thesis is focused on the determination of antioxidants and antioxidant activity. In the first work DMPD method was optimized to determine the antioxidant activity of antioxidants standards and the reaction was monitored depending on the time, temperature and concentration. In the second work automated determination of antioxidant activity of phenolic compounds standards rutin, quercitrine and standards of phenolic acids gallic acid and ferulic acid was tested using five different methods used in the determination of antioxidant activity using an automated pipetting machines and spectrophotometer. In the third work phenolic profile and total antioxidant capacity (TAC) was determined in apricot cultivars with methods high frequency liquid chromatography with tandem electrochemical and UV spectrometric detection and these two methods were compared. In the fourth work electrochemical methods used for the determination of phenolic compounds were described. In the fifth work oxidative stress induced by silver ions in the Staphylococcus aureus bacterial culture was investigated using spectrophotometric and electrochemical methods.

The aroma compounds suitable for food flavouring are mainly of plant origin. It is necessary to take into account the possible allergic effects of some of them. The theoretic part of this thesis is focused on aroma compounds and their biological effects. It also describes methods useful for their isolation and analysis with a nearer intent on the methods used in this thesis - solid phase microextraction and gas chromatography. The aim of the experimental part was to establish, optimise and validate the method for determination of allergic aroma compounds chosen and subsequently to enable its using in wide spectrum of food and other products.

Thesis "Legislation of handling of sources of danger to the environment" Abstract The thesis deals with material sources of danger to the environment in terms of legal forms of environmental protection from negative impact of these sources. The topic was narrowed at the beginning of the thesis to waste and chemicals so now it represents these two huge and significant groups of threats to the environment. The reason to narrow the topic was existence of a very large scale of different sources of environmental danger and that could cause shattering of the topic during writing the thesis. The thesis describes in individual chapters various forms of waste and chemicals handling in mutual relationship with legal obligations of subjects of law. The thesis at the same time attempts to evaluate valid legislation in terms of sufficiency of legal obligations and whether these obligations are clear and specific enough to enable high level of protection of the environment and human health (at least). Czech waste management legislation is massively influenced by directive of European union waste law, so the thesis compares intention of union legislator (if it is available) with the results of czech transposition. This Diploma thesis contains information about cryomaceration and using of dry ice in wine technology. Base on that we could compare differences in basic wine parameters or amount of fenolic and sensoric contrasts in mature wines. In theoretical part of this thesis are describe principles of cold maceration or cryomaceration, but main focus was on maceration method with dry ice. In next part we oriented on calculating total costs and usefulness of dry ice. Final part is all about maceration and squeezing of blue grapes. In the beginning of experimental part is described variety Frankovka in general, which was used to this experiments. In following article is explained how the experiment was handled, what practices were used for it and why I chose analytic and sensoric method for this experiment. In Final part are presented graphs, results, conclusions we made in this experiment.
