Optimization and control of a large-scale solar chimney power plant

Thesis (PhD (Mechanical and Mechatronic Engineering))-- University of Stellenbosch, 2007. / ENGLISH ABSTRACT: The dissertation builds on previous research (Pretorius, 2004) and investigates the optimization and control of a large-scale solar chimney power plant. Performance results are based on a reference location near Sishen in South Africa and a so-called reference solar chimney power plant, with a 5000 m collector diameter and a 1000 m high, 210 m diameter chimney. The numerical simulation model is refined and used to perform a sensitivity analysis on the most prominent operating and technical plant specifications. Thermo-economically optimal plant configurations are established from simulation results and calculations according to an approximate plant cost model. The effects of ambient wind, temperature lapse rates and nocturnal temperature inversions on plant performance are examined. Various new technologies are investigated for the purpose of controlling plant output according to specific demand patterns. The incorporation of vegetation under the collector roof of the plant and the influence thereof on plant performance is also explored. Results indicate that, through the modification of the collector roof reflectance, collector roof emissivity, ground surface absorptivity or ground surface emissivity, major improvements on plant performance are possible. Introducing thermal insulation or double glazing of the collector roof also facilitates substantial enhancements on plant yield. Simulations predict a notable sensitivity to the ground surface absorptivity value, while variable atmospheric temperature lapse rates and windy ambient conditions may impair plant performance significantly. Furthermore, sand is found to be unsuitable as plant ground type and thermoeconomically optimal solar chimney plant dimensions are determined to be generally larger than plant dimensions employed in previous studies. Good dynamic control of solar chimney power output is established, suggesting that a solar chimney power plant can be implemented as a base or peak load electricity generating facility. Lastly, results predict that vegetation, when provided with sufficient water, will be able to survive under the collector roof but the inclusion of vegetation will however cause major reductions in plant performance. / AFRIKAANSE OPSOMMING: Die proefskrif bou op vorige navorsing (Pretorius, 2004) en ondersoek die optimering en beheer van 'n grootskaalse sonskoorsteen-kragstasie. Uitsetresultate word baseer op 'n verwysingsligging naby Sishen in Suid-Afrika en 'n sogenaamde verwysingskragstasie, met 'n kollektor deursnee van 5000 m en 'n 1000 m hoë, 210 m deursnee skoorsteen. Die numeriese rekenaarmodel is verbeter en gebruik vir die uitvoering van 'n sensitiwiteits-analise op die belangrikste bedryfs- en tegniese kragstasie spesifikasies. Image processing methods for dynamical intracellular processes analysis in quantitative fluorescence microscopy / Méthodes numériques pour l’analyse de processus intracellulaires dynamiques en microscopie quantitative

Nous présentons dans la première partie du document une étude portant sur l'imagerie de temps de vie de fluorescence sur structures dynamiques dans le domaine de fréquence (FD FLIM). Une mesure en FD FLIM est définie par une série d'images présentant une variation d'intensité sinusoïdale. La variation d'un temps de vie se traduit par une variation dans la phase de la sinusoïde décrite par l'intensité. Notre étude comporte deux contributions principales: une modélisation du processus de formation de l'image et du bruit inhérent au système d'acquisition (capteur ICCD) ; une méthode robuste d'estimation du temps vie sur des structures mobiles et des vésicules intracellulaires. Nous présentons ensuite une étude en microscopie de fluorescence portant sur la quantification du transport hétérogène dans un environnement intracellulaire dense. Les transitions entre la diffusion Brownienne dans le cytoplasme et les transports actifs supportés par le cytosquelette sont en effet des scénarios très couramment observés dans des cellules vivantes. Nous montrons que les algorithmes classiques de suivi d'objets nécessaires dans ce contexte, ne sont pas conçus pour détecter les transitions entre ces deux types de mouvement. Nous proposons donc un nouvel algorithme, inspiré de l'algorithme u-track [Jaqaman et al., 2008], qui s'appuie sur plusieurs filtrages de Kalman adaptés à différents types de transport (Brownien, Dirigé ...), indépendamment pour chaque objet suivi. Nous illustrons sur séquences simulées et expérimentales (vimentine, virus) l'aptitude de notre algorithme à détecter des mouvements dirigés rares. / We propose in this manuscript a study of the instrumentation required for the quantification in frequency domain fluorescence lifetime imaging microscopy (FD FLIM). A FD FLIM measurement is defined as a series of images with sinusoidal intensity variations. The fluorescence lifetime is defined as the nanosecond-scale delay between excitation and emission of fluorescence. We propose two main contributions in the area: a modeling of the image process and noise introduced by the acquisition system (ICCD sensor); a robust statistical method for lifetime estimation on moving structures and intracellular vesicles. The second part presents a contribution to the tracking of multiple particles presenting heterogeneous transports in dense conditions. We focus here on the switching between confined diffusion in the cytosol and motor-mediated active transport in random directions. We show that current multiple model filtering and gating strategies fail at estimating unpredictable transitions between Brownian and directed displacements. We propose a new algorithm, based on the u-track algorithm [Jaqaman et al., 2008], based on a set of Kalman filters adapted to several motion types, for each tracked object. The algorithm has been evaluated on simulated and real data (vimentin, virus) data. We show that our method outperforms competing methods in the targeted scenario, but also on more homogeneous types of dynamics challenged by density.

Molecular and cellular characterization of apical and basal progenitors in the primate developing cerebral cortex / Caractérisation cellulaire et moléculaire des progéniteurs apicaux et basaux lors du développement du cortex cérébral chez le primate

Le cortex cérébral primate a subi des modifications majeures pendant l'évolution qui ont permis le développement de fonctions cognitives supérieures. Un accroissement massif a eu lieu avec l'extension spécifique des couches supragranulaires et une forte expansion tangentielle. Le cortex primate ne possède pas uniquement davantage de neurones, comparé au rongeur, mais aussi des différences qualitatives. Ceci suggère des différences qualitatives pendant le développement du cortex.Une zone proliférative corticale supplémentaire a été identifiée chez le singe macaque: la zone subventriculaire externe (OSVZ) supposée être impliquée dans l'expansion du cortex primate. Mais les propriétés des précurseurs de l'OSVZ restent mal connues. Des techniques de microscopie en temps réel et d'immunofluorescence ont permis de réaliser une description exhaustive des précurseurs de l'OSVZ et de leurs propriétés chez le singe macaque.Nos résultats mettent en évidence des différences primates/rongeurs majeures. Les observations en temps réel révèlent des capacités prolifératives bien plus importantes des précurseurs. Les précurseurs primates de l'OSVZ présentent des taux de prolifération variables pendant la corticogenèse liés à la cinétique du cycle cellulaire. Nos enregistrements ont permis la génération d'une grande base de données de propriétés et lignages de précurseurs et la mise en évidence d’une diversité morphologique inattendue. 5 types ont été identifiés. Impliqués dans des lignages complexes, chaque type a la capacité de s'auto-renouveler et de générer directement des neurones. Parallèlement, nous avons développé une méthode de classification non supervisée des précurseurs corticaux. Cette technique a identifié les mêmes 5 types de précurseurs.Les résultats de cette thèse apportent de nouveaux éléments dans la compréhension des spécificités de la corticogenèse primate qui contribuent à l'expansion corticale et au développement de capacités cognitives supérieures. / The primate cerebral cortex underwent major modifications during evolution that enabled the development of high cognitive functions. A massive enlargement occurred with the specific expansion of the supra granular layers and the apparition of new frontal areas. Not only quantitative differences are found compared to the rodent but also qualitative differences. This points to potential qualitative differences in primate cortical development. An extra proliferating zone had already been identified during macaque corticogenesis: the outer subventricular zone (OSVZ). This zone is assumed to play a key role in the expansion of the primate cortex but the cellular and functional properties of OSVZ precursors remain elusive. We used quantitative long-term time-lapse video-microscopy (TLV) and immunofluorescence in and ex vivo to perform a detailed and exhaustive description of OSVZ precursor types and proliferative abilities at different stages of macaque cortical development. Our results highlight major rodent/primate differences. TLV observations revealed a much higher proliferative potential of OSVZ compared to the rodent SVZ. We report variable rates of proliferation linked to cell-cycle duration in a stage-specific manner. TLV recordings allowed the formation of a large database of primate precursor properties and lineages. This dataset unravelled an unexpectedly high diversity of OSVZ precursor morphologies. Five precursor types were identified. Involved in complex lineages, each precursor type can self-renew and directly generate neurons. In a parallel approach, we developed an unbiased clustering tool to automatically classify cortical precursors. This technique returned the same five precursor types as the morphological categorization. The results of this PhD thesis provide new insights into primate specificities during corticogenesis that contribute to cortical expansion and to the development of higher cognitive abilities.

Micro-irradiation ciblée par faisceau d'ions pour la radiobiologie in vitro et in vivo / In vitro and in vivo ion beam targeted micro-irradiation for radiobiology

Les microfaisceaux d’ions ont, au cours de ces dernières décennies, montré leur efficacité dansl’étude des effets des rayonnements ionisants sur le vivant notamment concernant les effets des faiblesdoses ou l’étude de l’effet de proximité. Le CENBG dispose depuis 2003 d’un dispositif permettant la micro-irradiation ciblée d’échantillons biologiques vivants. Les applications des microfaisceaux dans ce domainese sont récemment diversifiées et des études plus fines sur les mécanismes de réparation desdommages ADN radio-induits aux échelles cellulaire et multicellulaire sont devenues possibles via lesévolutions en imagerie par fluorescence et en biologie cellulaire. Ces approches ont nécessité une évolutionimportante de l'instrumentation de la ligne de micro-irradiation du CENBG qui a été entièrementredessinée et reconstruite dans un souci d’optimisation d’apport de nouvelles fonctionnalités. Les objectifsde mes travaux ont été i) la mise en service du dispositif, ii) la caractérisation des performances dusystème, iii) la mise en place de protocoles pour l’irradiation ciblée à dose contrôlée aux échelles cellulaireet multicellulaire, in vitro et in vivo, et le suivi en ligne des conséquences précoces de cette irradiation,iv) la modélisation des irradiations afin d’interpréter les observables biologiques au regard des donnéesphysiques calculées.Ces travaux ont permis i) de caractériser les performances du dispositif : une taille de faisceau d’environ2 μm sur cible et une précision de tir de ± 2 μm, de développer des systèmes de détection d’ions pour uncontrôle absolu de la dose délivrée, ii) d’induire des dommages ADN fortement localisés in vitro, et devisualiser en ligne le recrutement de protéines impliquées dans la réparation de ces dommages,iii) d’appliquer ces protocoles pour générer des dommages ADN in vivo au sein d’un organisme multicellulaireau stade embryonnaire, Caenorhabditis elegans.Ces résultats ouvrent la voie vers des expériences plus fines sur la ligne de micro-irradiation ciblée duCENBG pour étudier les effets de l’interaction des rayonnements ionisants avec le vivant, aux échellescellulaire et multicellulaire, in vitro et in vivo. / The main goal of radiobiology is to understand the effects of ionizing radiations on the living.These past decades, ion microbeams have shown to be important tools to study for example the effects oflow dose exposure, or the bystander effect. Since 2003, the CENBG has been equipped with a system toperform targeted micro-irradiation of living samples. Recently, microbeams applications on this subjecthave diversified and the study of DNA repair mechanisms at the cellular and multicellular scales, in vitroand in vivo, has become possible thanks to important evolutions of fluorescence imaging techniques andcellular biology. To take into account these new approaches, the CENBG micro-irradiation beamline hasbeen entirely redesigned and rebuilt to implement new features and to improve the existing ones. My PhDobjectives were i) commissioning the facility, ii) characterizing the system on track etch detectors, and onliving samples, iii) implementing protocols to perform targeted irradiations of living samples with a controlleddelivered dose, at the cellular and multicellular scales, and to visualize the early consequencesonline, iv) modelling these irradiations to explain the biological results using the calculated physical data.The work of these past years has allowed us i) to measure the performances of our system: a beam spotsize of about 2 μm and a targeting accuracy of ± 2 μm, and to develop ion detection systems for an absolutedelivered dose control, ii) to create highly localized radiation-induced DNA damages and to see onlinethe recruitment of DNA repair proteins, iii) to apply these protocols to generate radiation-induced DNAdamages in vivo inside a multicellular organism at the embryonic stage: Caenorhabditis elegans.These results have opened up many perspectives on the study of the interaction between ionizing radiationsand the living, at the cellular and multicellular scales, in vitro and in vivo.

Modulating the p53 response to DNA damage by applying different perturbations types

Der Tumorsuppressor p53 spielt eine wichtige Rolle bei der Aufrechterhaltung der zellulären Homöostase und verhindert die Bildung und Entwicklung von Krebs. Frühere Studien auf Einzelzellebene haben gezeigt, der p53 nach dem Auftreten von DNS Schäden in einer Serie gleichförmiger Pulse im Kern akkumuliert. Um den Einfluss von Temperatur, dem Zustand des NFκB Weges oder dem Vorhandensein von Wachstumsfaktoren auf die p53 Dynamiken nach DNS-Schäden zu untersuchen, wurden in dieser Arbeit A549 und MCF10A p53 Reporter-Zelllinien verwendet. Um Daten mit hoher zeitlicher und räumlicher Auflösung auf Einzelzellebene zu erhalten, wurde Zeitraffer-Fluoreszenzmikroskopie verwendet. Überraschenderweise zeigten A549 Zellen, die mit γ-Strahlung behandelt wurden, eine höhere p53 Akkumulation und ein verlängertes Zeitintervall zwischen den p53 Pulsen, wenn sie bei 30°C inkubiert wurden im Vergleich zu Zellen unter physiologischen Bedingungen. Hingegen zeigten Zellen bei 40°C eine höhere p53 Pulsfrequenz. Außerdem wurde die p53 Zielgen-Expression durch die Änderungen in der Dynamik beeinflusst. In den beiden Zelllinien, A549 und MCF10A, wurde die p53 Dynamik durch Inhibierung des NFκB Signalweges verändert, nicht aber durch dessen Aktivierung mittels TNF. So verlängert die Inhibierung des NFkB Signalweges das Zeitintervall zwischen p53 Pulsen, was sich auch in der Expression von p53 Zielgenen wiederspiegelt. Weiterhin konnte in MCF10A Zellen durch Experimente mit verschiedenen Medien-Bedingungen gezeigt werden, dass p53 Dynamiken vor allem durch die Anwesenheit von EGF und Hydrocortison geprägt werden. So führt EGF zu einem pulsierenden p53 Verhalten, während Hydrocortison alleine die p53 Antwort vollständig aufhebt.. Diese Studie macht deutlich, wie wichtig es ist, das Verhältnis von zellulärem Zustand und p53 Antwort systematisch zu untersuchen, um Krebstherapien wirksamer zu machen / The tumor suppressor p53 plays important roles in maintaining cellular homeostasis and in preventing the formation and development of cancer. Previous studies on p53 activation after DNA damage have reported that it shows a series of regular discrete pulses of protein accumulation over time at the single cell level. In this work A549 and MCF10A p53 reporters cell line were used to investigate how p53 dynamics after DNA damage were affected by changes in temperature, by changes in the state of the NFκB pathway and changes in the provided growth factors. Time-lapse florescent microscopy was used to obtain single cell data with high temporal and special resolution at the single cell level. Surprisingly A549 cells treated with γ-irradiation showed higher level of p53 accumulation and increased time between p53 pulses when imaged at 30°C than cells imaged under physiological conditions. Cells imaged at 40°C showed instead higher p53 pulse frequency. P53 target gene expression was also affected by these changes in dynamics. In both A549 and MCF10A cells, p53 dynamics were changed by NFκB pathways inhibition but not activation via TNFα. Upon inhibition of the NFκB pathway the timing between p53 pulses was increased leading to changes also in p53 target genes expression. In MCF10A cells experiments done under different medium conditions proved that p53 dynamic in this cell line was shaped mainly by the presence of EGF and hydrocortisone that are usual components of the media. EGF leads to a more pulsatile p53 behavior while hydrocortisone completely abrogates the p53 response. These discoveries pointed out the need to study more systematically the relationship between the p53 response to a given stress and the cellular state in order to make cancer therapies more effective.

Post-dispersal seed predation in a conifer-broadleaf forest remnant : the importance of exotic mammals

Despite extensive international acceptance of the critical role of mammalian post-dispersal seed predation in many plant communities, in New Zealand we have limited knowledge of these predators’ influence on plant recruitment in our forests. The principle objective of my thesis was to determine the importance of exotic mammals as post-dispersal seed predators in a New Zealand conifer-broadleaf forest remnant. To address this goal, I used a series of field-based experiments where the actions of different post-dispersal seed predators were separated by wire-mesh exclosures. My study was conducted at Mount Peel Forest Park Scenic Reserve, South Canterbury, New Zealand. Being a human modified conifer forest currently dominated by broadleaf species, it is typical of forest remnants in New Zealand. This presented an opportunity to study a wide range of both potential post-dispersal seed predators and broadleaf tree species. My findings indicate that exotic mammals are not only post-dispersal seed predators at Peel Forest, but are responsible for the majority of post-dispersal predation events observed. Ship rats (Rattus rattus) were the dominant post-dispersal seed predators, while brushtail possums (Trichosurus vulpecula), house mice (Mus musculus) and native invertebrates were also important post-dispersal seed predators for several tree species. Through use of time-lapse video and cafeteria experiments I found that exotic mammalian seed predators, when compared to native invertebrate seed predators, preyed upon larger-seeded plant species and were responsible for considerable seed losses of several tree species. However, exotic mammalian seed predators do share several foraging characteristics with native invertebrate seed predators, as predators foraged in similar habitats and responded in a similar way to changes in seed density. In investigating if post-dispersal seed predation by mammals had a flow-on effect to plant recruitment, I observed natural seedling densities at Peel Forest were significantly higher in the absence of mammalian seed predators, but I found no evidence that the presence of mammals significantly altered the overall species richness. At the community level, I did not find an interaction between habitat and exotic mammals, however I present evidence that for individual plant species a significant mammal : habitat interaction occurred. Consequently, even though my cafeteria experiment implied there was no significant difference in the overall amount of seed preyed upon within different habitats, the less favourable microsite conditions for germination under an intact continuous canopy allows mammals to exacerbate habitat-related patterns of seed mortality and have a noticeable effect on seedling establishment. In an effort to validate the use of manipulative experiments to predict the long-term effect of post-dispersal seed predation on plant dynamics, I attempted to link results of my cafeteria experiment with observed seedling abundance at Peel Forest. Seven tree species were used in this comparison and a strong correlation was observed. This result shows that the level of post-dispersal seed predation determined in the cafeteria experiment provided a good predictor of the effect of mammalian post-dispersal seed predation on seedling establishment. To fully gauge the impact of mammalian post-dispersal seed predators on seedling establishment, the relationship between these seed predators and the type of recruitment limitation experienced by a plant species was also investigated. By using a combination of seed addition, plot manipulations and seed predator exclusion I was able to investigate this relationship. I found evidence that seed limitation at Peel Forest is positively correlated with seed size, and that while mammalian post-dispersal seed predators can further reduce plant recruitment of plant species experiencing seed limitation, the influence of mammals in determining plant recruitment was limited for plant species experiencing microsite limitation. My study has proven that exotic mammals are now the dominant post-dispersal seed predators at Peel Forest, the amount of seed preyed upon varies among plant species, and post-dispersal seed predation by mammalian species can lead to differences in seedling richness and abundance. I proved that the influence of exotic mammals on seedling establishment is also linked to habitat structure and recruitment limitations. When combined these observations suggest that exotic mammalian post-dispersal seed predators may play an important role in determining landscape abundance and distribution of plants at Peel Forest.

一般而言,壽險公司會在保險契約生效前就支付保單相關之費用,例如核保與承保之成本,並且公司會預期未來保險期間內可以填補上述費用;但若保戶於保險期間內早期解約或是解約情形嚴重,將使壽險公司難達到損益兩平之目標而招受損失,影響公司預期盈收,進而增加公司資金調度上之困難。因此,對於長期穩健經營之壽險公司而言,瞭解各保險解約率變動情形對於公司之財務規劃相當重要,以期降低危害公司之風險。 本文期望藉由台灣保險事業發展中心之實證資料蒐集與相關分析,探討影響台灣壽險業生死合險及不還本終身壽險解約之因素以及其解約率之特性,進而建立與利差及保單年度相關之解約率模型,以期能準確地估計台灣壽險公司生死合險解約率與不還本終身壽險解約率。除此之外,本研究將所建構之解約率模型應用於公司未來現金流量分析,以蒙地卡羅法模擬各險種保單準備金之分配,瞭解各種解約率假設對於公司未來現金流量之影響,進而瞭解解約率參數假設對於準備金風險之評估扮演重要角色。 / In general, the life insurance companies would pay the expenses with respect to the insurance policies before the validity of insurance contracts such as underwriting and insuring costs. If the policyholders are early-surrendered or over-surrendered during the policy period, then it will make the insurance companies hard to achieve their break-even goal and result in affecting the companies’ surplus as well as management of their capital. Thus, for the long-term and stable life insurance companies, it is extremely important to understand the changes of lapse rate in order to reduce the financial risk damage before making any financial decisions. In this article, we expect to focus on the causes and the features of lapse rate changes by collecting and analyzing the empirical data of endowment and whole life insurance in Taiwan from Taiwan Insurance Institute. Based on our analysis, we could build the lapse rate model concerning the relation between the lapse rate and interest rate difference or policy year for estimating the endowment lapse rate and whole life insurance lapse rate accurately. Moreover, we apply the lapse rate model to company’s cash flow analysis. We employ the Monte Carlo simulation to simulate the policy reserve distribution, and we find out that the lapse rate assumption plays an important role in the policy reserve evaluation.

本文針對附保證年金型投資商品進行評價,其中被保險人脫退因素除受到死亡解約因素之外,對經濟環境影響因素產生解約問題,如利率攀升、經濟成長率、失業率等亦須考慮。附保證年金型投資商品公帄價值為保險公司販賣投資型年金商品須對負債面進行評價,以確保被保險人之權益,保險商品價值除因投資市場環境變動造成投資商品價值累積變動之外,對於被保險人因應市場環境轉變造成脫退問題亦影響保險公司對於投資型商品準備金價值評估,本篇依照Kolkiewicz & Tan(2006)之研究,假設附保證年金型投資商品評價方式,除投資標的受到市場變動影響外,對於經濟環境變動造成被保險人解約狀況亦考慮於核保模型中,因脫退因素考慮層面過廣,故本篇主要以死亡、經濟環境變動劇烈與利率上升導致解約因素為主要考慮狀態。 本研究推導之模型主要得出下列結果:(1)附保證年金型商品的公帄價格以保險年期的影響最大,其次為風險性資本市場長期帄均波動,而死亡率影響附保證投資年金型商品主要由風險性資本市場價值決定。(2)契約初始為主要解約期間,當解約力持續增加至一定值,契約後期解約率將趨於帄坦,本研究推估契約前期經濟市場波動易造成被保險人解約狀況,故解約程度增加。(3)主要投資型商品風險價差問題影響因素為長期市場波動程度,因風險價差之衡量主要考慮風險因子變動因素導致與公帄價值或期初保費差距,依照模型假設變動因子以風險性資產價值波動程度影響最巨,其次為保險期間,因此歸納出風險價差因子主要變動來源為風險性資產價值。 / In this paper, the goal is to evaluate fair value of guaranteed annuity-type investment products. In addition to death factors, the insured terminate by other reasons, such as interest rates raising, economic growth rate, and unemployment rate. Accordance with the liabilities side, the reserve of guaranteed annuity-type investment products must match it’s fair value. There is a question how to accurately evaluate fair value of guaranteed annuity-type investment products. The price of guaranteed annuity-type investment products is affected by two parts. One is cumulative index price change in value of investment goods, the other one is withdrawal rates. Kolkiewicz & Tan’s research assume guaranteed annuity-type investment products evaluation methods which is affected by market environment and termination status of the insured. The results show that (1) The major impact on fair value of guaranteed annuity-type investment products is mainly from the period of the insurance contracts. The secondary effect is long-term average risk capital market volatility. (2) The main terminate time is the beginning of the contracts. When the lapse rates continued to increase to a certain value, lapse rate tends to smooth.(3) The major impact on risk spread of guaranteed annuity-type investment products is mainly from long-term market volatility. To sum up, the major changes in sources of risk spreads factor are from asset value.

Live Single Cell Imaging and Analysis Using Microfluidic Devices

Today many cell biological techniques study large cell populations where an average estimate of individual cells’ behavior is observed. On the other hand, single cell analysis is required for studying functional heterogeneities between cells within populations. This thesis presents work that combines the use of microfluidic devices, optical microscopy and automated image analysis to design various cell biological assays with single cell resolution including cell proliferation, clonal expansion, cell migration, cell-cell interaction and cell viability tracking. In fact, automated high throughput single cell techniques enable new studies in cell biology which are not possible with conventional techniques. In order to automatically track dynamic behavior of single cells, we developed a microwell based device as well as a droplet microfluidic platform. These high throughput microfluidic assays allow automated time-lapse imaging of encapsulated single cells in micro droplets or confined cells inside microwells. Algorithms for automatic quantification of cells in individual microwells and micro droplets are developed and used for the analysis of cell viability and clonal expansion. The automatic counting protocols include several image analysis steps, e.g. segmentation, feature extraction and classification. The automatic quantification results were evaluated by comparing with manual counting and revealed a high success rate. In combination these automatic cell counting protocols and our microfluidic platforms can provide statistical information to better understand behavior of cells at the individual level under various conditions or treatments in vitro exemplified by the analysis of function and regulation of immune cells. Thus, together these tools can be used for developing new cellular imaging assays with resolution at the single cell level. To automatically characterize transient migration behavior of natural killer (NK) cells compartmentalized in microwells, we developed a method for single cell tracking. Time-lapse imaging showed that the NK cells often exhibited periods of high motility, interrupted with periods of slow migration or complete arrest. These transient migration arrest periods (TMAPs) often overlapped with periods of conjugations between NK cells and target cells. Such conjugation periods sometimes led to cell-mediated killing of target cells. Analysis of cytotoxic response of NK cells revealed that a small sub-class of NK cells called serial killers was able to kill several target cells. In order to determine a starting time point for cell-cell interaction, a novel technique based on ultrasound was developed to aggregate NK and target cells into the center of the microwells. Therefore, these assays can be used to automatically and rapidly assess functional and migration behavior of cells to detect differences between health and disease or the influence of drugs. The work presented in this thesis gives good examples of how microfluidic devices combined with automated imaging and image analysis can be helpful to address cell biological questions where single cell resolution is necessary. / <p>QC 20130927</p>

La décision conditionnelle en droit administratif français / The conditional decision in French Administrative law

La décision conditionnelle constitue un objet délaissé de la recherche en droit administratif français. Injustement perçue comme une affaire privée de spécialistes du droit des actes administratifs unilatéraux, elle n’occupe les écrits des publicistes que dans la mesure où la réalisation de la condition porte atteinte à la garantie des droits acquis. Cette réserve explique pour partie la faible contribution de la doctrine à la construction d’une définition de la décision conditionnelle, autonome du droit civil des obligations, et les difficultés corrélatives à déterminer les contours de cette notion mal identifiée. Pourtant, les orientations prises par le juge administratif et, dernièrement, par le codificateur, rendent nécessaire la recherche d’une définition clarifiée de la décision conditionnelle en droit administratif français. Depuis longtemps, le juge administratif range la décision conditionnelle dans la catégorie des actes administratifs unilatéraux et met en œuvre les principes qui les gouvernent. Or, de son côté, la doctrine ne parvient pas à l’intégrer pleinement dans la catégorie de l’acte unilatéral ou du contrat.Le visage inhabituel qu’elle présente, aux yeux des juristes, force la réévaluation des classifications traditionnelles.La doctrine malmène ses catégories binaires, dans le but de souligner la particularité de cette décision unilatérale qui ressemble à s’y méprendre au contrat. Il en résulte une contradiction inévitable entre la présentation doctrinale de la décision conditionnelle et les données du droit positif. Pour la dénouer, il apparaît nécessaire de renouveler l’appareil théorique jusqu’alors mobilisé. En prenant appui sur les données pertinentes du droit positif, il a été possible de définir la condition comme une norme juridique à part entière, à la fois dépendante et distincte des autres dispositions de la décision, et dont la fonction se limite à moduler les effets dans le temps de la norme principale qu’elle prend pour objet. À travers ces éléments de définition, transparaît un régime juridique cohérent et unifié / Conditional decisions remain much overlooked in the legal scholarship on French administrative law. Wrongfully considered as the exclusive province of the specialist on unilateral administrative acts, conditional decisions have received the attention of public lawyers only in so far as they come to present a risk for the protection of aquired rights. This goes some way to explaining both the scarcity of doctrinal contributions to a definition of the concept of a conditional decision – autonomous from the paradigm of the obligations in French civil law –, and the difficulties encountered in trying to bring some clarity to the subject by attempting to determine precisely the boundaries of the notion. Nevertheless, recent turns taken by the administrative courts, and more recently by codifiers, do call for a clear and consistent definition. The case law has for long considered these legal acts as unilateral acts, thus subjecting conditional decisions to the corresponding legal principles. Despite this, the legal scholarship on the question has never managed to fully integrate conditional decisions in either branch of the main binary subdivision – contracts and unilateral acts –, ill-using both in order to stress the highly unusual character of the notion (for instance underlining the peculiar relevance of an analysis of this unilateral decision in contractual terms). This unusual object therefore forces us to reevaluate our traditional classifications, which otherwise systematically result in a severe inconsistency between substantive law and its doctrinal exposition. In order to do so, the theoretical tools and approaches used up to now must be renewed. This doctoral dissertation aims to reconcile the positive law and its doctrinal exposition, thus proposing a clear definition of the condition itself as a legal norm in its own right – both dependant on, and distinct from, the other provisions of the decision it is attached to – the function of which is to modulate the effects over time of the main decision. These proposed elements of definition come to reveal a unified and coherent legal status of the conditional decision in administrative law.
