Currently, among the various types of waste, electronic waste is considered obsolete in a very short time and require proper disposal. In Brazil, the last year, the reuse of plastic materials has increased. This study aimed to research the importance of mobile phones waste recycling, which have short pace of useful life used as reinforcements, no development of new materials containing recycled thermoplastic polymer (Low Density Polyethylene LDPE) and from the analyzes mechanical and dangerousness of these new materials, so justify the replacement of the importance of some materials with longer life time and easy recyclability, that form provides a reduction of environmental impacts caused by the generated waste were developed six blends of (Low Density Polyethylene recycled and PCABS carcasses - LDPErec/PC-ABS), with composition and grain size variation of cell granulometry housing and also an analysis of pure materials. The results obtained for analysis of diffraction X-ray, density, water absorption, hardness, scanning electronic microscopy and mechanical testing showed one low interaction between polymers, beyond the performance of the amorphous polymer morphology and hence the crystallinity mix, directly affected at the thermal and mechanical properties. As for the hazard analysis of the materials developed, they were not found excess levels of metal toxicants, the limits established by ABNT NBR 10004 / ? crescente a pesquisa por novos materiais e metodologias para a cria??o e inser??o dos mesmos no mercado. Atualmente, dentre os diversos tipos de res?duos, os res?duos eletroeletr?nicos tem aumentado exponencialmente, pois estes tem sido considerados obsoletos em um intervalo de tempo muito curto e necessitam de descarte adequado. No Brasil, nos ?ltimos anos, o reaproveitamento de materiais pl?sticos tamb?m tem aumentado. Neste contexto, este trabalho teve como objetivo pesquisar a import?ncia da reciclagem de res?duos de telefones celulares (carca?as), que possuem curto tempo de vida ?til, utilizados como refor?os, no desenvolvimento de novos materiais, contendo pol?mero termopl?stico reciclado (polietileno de baixa densidade ? PEBD) e, a partir das an?lises mec?nicas e da periculosidade destes novos materiais, justificar a import?ncia da substitui??o de alguns materiais por outros com maior tempo de vida ?til e de f?cil reciclabilidade, de forma que proporcione a redu??o dos impactos ambientais provocados com os res?duos gerados. Para tal, foram desenvolvidas 6 blendas de (Polietileno de baixa densidade reciclado e carca?as de PC-ABS - PEBDrec/PC-ABS), com varia??o da composi??o e granulometria das carca?as de aparelhos celular, e tamb?m a an?lise dos mat?rias puros. Os resultados obtidos pelas an?lises de difra??o de raios-X, densidade, absor??o de ?gua, dureza, microscopia eletr?nica de varredura e ensaios mec?nicos mostraram que a baixa intera??o entre os pol?meros, al?m da atua??o do pol?mero amorfo na morfologia e, consequentemente, na cristalinidade da mistura, afetou diretamente nas propriedades t?rmicas e mec?nicas. Quanto ? an?lise de periculosidade dos materiais desenvolvidos, n?o foram encontrados n?veis excedentes de metais t?xicos, conforme limites estabelecidos pela NBR ABNT 10004.

O presente estudo investigou a relação entre os níveis plasmáticos da lipoproteína de baixa densidade oxidada (LDLox), as metaloproteinases de matriz (MMP) e seus inibidores teciduais (TIMPs) e o remodelamento vascular em pacientes com LM, e o papel da atividade física nesta relação. Foram estudados 42 homens com LM (? 2 anos), [18 sedentários (S-LM) e 24 fisicamente ativos (A-LM)] e 16 homens fisicamente saudáveis por meio de análise clínica, antropométrica, laboratorial e de espessura íntima-média da carótida (EIM). Todos os participantes estudados eram normotensos, não diabéticos, não fumantes e normolipêmicos. As concentrações plasmáticas de LDLox, MMP-2, MMP-8, MMP-9, TIMP-1 e TIMP-2 foram determinados por ensaio imunoenzimático (ELISA). Os resultados mostraram que a EIM da carótida, razão EIM/diâmetro e as concentrações de LDLox dos A-LM e dos indivíduos fisicamente normais não foram diferentes estatisticamente. Por outro lado, indivíduos com S-LM apresentaram maior EIM, razão EIM/diâmetro e concentrações aumentados de LDLox em comparação com A-LM (p<0,01, p<0,001 e p=0,01, respectivamente) e indivíduos controles (p<0,001 para todos). Os resultados da análise de correlação bivariada, incluindo todos os indivíduos com LM, demonstrou que o EIM de carótida e a razão EIM/diâmetro se correlacionaram apenas com LDLox, MMP-8 e com a relação MMP-8/TIMP-1. Além disso, a análise de regressão ajustada para a presença ou não de atividade física e idade mostrou que a LDLox foi associada à EIM carotídea e com a relação EIM/diâmetro, enquanto que MMP-8 foi associado com o índice EIM/diâmetro em indivíduos com LM. Em conclusão, as concentrações plasmáticas de LDLox e MMP-8 estão associados com aterosclerose carotídea e há interação entre a inatividade física, aterosclerose e LDLox em indivíduos com LM / Abstract: Previous reports have indicated that subjects with chronic spinal cord injury (SCI) exhibit increased cardiovascular risk compared to able-bodied individuals. This study investigated the relationship between plasmatic oxidized low-density lipoprotein (OxLDL), matrix metalloproteinases (MMPs) and tissue inhibitors of MMPs (TIMPs) levels and vascular remodeling in SCI subjects and the role of physical activity in this regard. We studied 42 men with chronic (?2 years) SCI [18 sedentary (S-SCI) and 24 physically active (PA-SCI)] and 16 able-bodied men by clinical, anthropometric, laboratory, and carotid intima-media thickness (IMT) analysis. All enrolled subjects were normotensive, non-diabetics, non-smokers and normolipemic. Plasmatic OxLDL, MMP-2, MMP-8, MMP-9, TIMP-1 and TIMP-2 levels were determined by enzyme-linked immunosorbent assay. The results showed that carotid IMT, IMT/diameter ratio and OxLDL levels of PA-SCI and able-bodied subjects were statistically similar. Conversely, S-SCI subjects exhibited higher IMT, IMT/diameter ratio and OxLDL levels compared to PA-SCI (p<0.01, p<0.001 and p=0.01, respectively) and able-bodied (p<0.001 for all) individuals. Results of bivariate correlation analysis including all injured subjects showed that carotid IMT and IMT/diameter ratio only correlated with OxLDL, MMP-8 and MMP-8/TIMP-1 ratio. Further stepwise regression analysis adjusted for the presence or not of physical activity and age showed that OxLDL was associated with carotid IMT and IMT/diameter ratio, while MMP-8 was associated with IMT/diameter ratio in SCI individuals. In conclusion, plasmatic OxLDL and MMP-8 levels are associated with carotid atherosclerosis and there is an interaction among physical inactivity, atherosclerosis and OxLDL in SCI individuals / Mestrado / Clinica Medica / Mestra em Clínica Médica

Natural antibodies to malondialdehyde adducts in atherosclerosis

Abstract Atherosclerosis is a chronic inflammatory disease and infections potentially contribute to its pathogenesis. Oxidation of LDL (low-density lipoprotein) is a key event in atherogenesis. Lipid peroxidation produces reactive aldehydes such as malondialdehyde (MDA) and MDA-acetaldehyde that form immunogenic adducts on for example LDL particles. This thesis investigated natural IgM antibodies that recognize MDA and MDA-acetaldehyde adducts and bind, through molecular mimicry, to epitopes on oral pathogenic bacteria. In the first study, the induction of cross-reactive IgM antibodies binding to MDA adducts was investigated in mice immunized with periodontopathogen Porphyromonas gingivalis. The effect of immunization on the development of atherosclerosis was analyzed after a high fat diet. Immunization of mice with killed P. gingivalis elevated IgM levels to MDA adducts and reduced atherosclerosis. Mouse monoclonal IgM to MDA modified LDL recognized epitopes on P. gingivalis that were identified as gingipain protease enzyme of this bacterium. The presence of IgM with similar specificity was also demonstrated in human sera. In the second study the effect of immunization with MDA modified LDL in conferring protection against pathogen-accelerated atherosclerosis was investigated. Mice were immunized with MDA modified LDL before challenge with live P. gingivalis. The extent of atherosclerosis after a high fat diet was reduced in immunized mice compared to control groups. Immunized mice had elevated IgM and IgG levels to MDA modified LDL compared to controls. The third study investigated antibody recognition of MDA-acetaldehyde adducts in umbilical cord blood plasma of preterm and full-term newborns. Natural IgM to MDA-acetaldehyde adducts was enriched in umbilical cord blood plasma compared to maternal levels even in preterm neonates. This thesis highlighted the importance of immune recognition of MDA adducts by natural IgM antibodies, and demonstrated that pathogenic bacteria and MDA derived structures are recognized by natural IgM throughout the lifespan. These natural IgM are proposed to modulate the development of chronic inflammatory diseases such as atherosclerosis and periodontitis. / Tiivistelmä Valtimonkovettumatautiin, eli ateroskleroosiin, liittyy matala-asteinen tulehdustila, ja myös infektiot vaikuttavat taudin ilmentymiseen. LDL:n (low-density lipoprotein) hapettuminen on merkittävä valtimonkovettumataudin syntyä edistävä reaktio. Lipidiperoksidaatio tuottaa reaktiivisia aldehydejä, kuten malonidialdehydiä (MDA) ja MDA-asetaldehydiä, jotka muodostavat immuunijärjestelmän tunnistamia rakenteita esimerkiksi LDL-partikkeleihin. Väitöskirjassa tutkittiin luonnollisia IgM-vasta-aineita, jotka tunnistavat MDA- ja MDA-asetaldehydirakenteita, ja jotka rakenteiden samankaltaisuuden takia sitoutuvat myös parodontiittipatogeenien epitooppeihin. Ensimmäisessä työssä MDA-rakenteisiin sitoutuvien IgM-vasta-aineiden ristireaktiota tutkittiin hiirillä, jotka immunisoitiin Porphyromonas gingivalis parodontiittipatogeenillä. Hiirten immunisoiminen inaktivoidulla P. gingivalis bakteerilla lisäsi IgM-vasta-ainetasoja MDA-rakenteita vastaan ja vähensi valtimonkovettumatautia. MDA-LDL:ään sitoutuva hiiren monoklonaalinen IgM-vasta-aine sitoutui P. gingivalis bakteerin proteiineihin, joiden tunnistettiin olevan osia tämän bakteerin gingipain proteaasientsyymistä. Myös ihmisen seerumista osoitettiin IgM-vasta-aineita, joilla oli samantyyppinen sitoutumisspesifisyys. Toisessa työssä tutkittiin MDA-LDL-immunisaation suojaavaa vaikutusta infektion kiihdyttämän valtimonkovettumataudin synnyssä. Hiiret immunisoitiin MDA-LDL:llä ennen infektiota elävällä P. gingivalis bakteerilla. Valtimonkovettumatauti väheni immunisoiduilla hiirillä rasvaruokinnan jälkeen verrattuna kontrolliryhmiin. Immunisoiduilla hiirillä IgM- ja IgG-vasta-ainetasot MDA-LDL:ää vastaan olivat kohonneet verrattuna kontrolliryhmiin. Kolmannessa työssä tutkittiin, tunnistavatko ennenaikaisten ja täysiaikaisten vastasyntyneiden luonnolliset vasta-aineet MDA-asetaldehydirakenteita. IgM-vasta-aineita MDA-asetaldehydirakenteisiin esiintyi jo ennenaikaisesti syntyneiden napaverinäytteissä. MDA-asetaldehydiin sitoutuvien IgM-vasta-aineiden määrä vastasyntyneillä oli korkeampi verrattuna äiteihin. MDA-rakenteet ovat yksi tärkeä kohde immuunijärjestelmän luonnollisille IgM-vasta-aineille, jotka tunnistavat myös taudinaiheuttajabakteereja. Näillä vasta-aineilla voi olla vaikutusta kroonisten tulehdustautien, kuten valtimonkovettumataudin ja parodontiitin syntymekanismeissa.

Physique, chimie et biologie de la filamentation d’impulsions laser femtosecondes en solutions aqueuses / Physics, chemitry and biology of femtosecond laser pulses filamentation in aqueous solutions

La propagation d’une impulsion laser femtoseconde dans un milieu condensé trans- parent tel que l’eau conduit, dans les conditions appropriées, à la manifestation de phénomènes d’optiques non linéaires regroupés sous le terme de filamentation laser. Le faisceau laser correspondant voit alors sa propagation métamorphosée sous la forme de filaments de lumière intense. Au coeur de ces filaments, l’irradiance considérable provoque l’ionisation des atomes du milieu et la génération de plasmas. Produit de manière spontanée et auto-régulée, ces plasmas ont la particularité de combiner une densité importante d’événements d’ionisation avec des effets thermo-mécaniques minimisés. Leurs propriétés intrinsèques font de ces plasma une source d’ionisation singulière tout particulièrement en ce qui concerne les sciences qui s’intéressent à l’étude des effets des radiations ionisantes. Entre autres, les sciences des radiations étudient la physique, la chimie et la biologie de l’action des rayonnements ionisants sur des systèmes d’intérêt biologique. Dans ce contexte, cette dissertation s’intéresse à la filamentation d’impulsions laser femtosecondes proches infrarouges en solution aqueuse. L’eau représentant la compo- sante majeure des systèmes d’intérêt biologique, une solution aqueuse constitue une approximation satisfaisante d’un échantillon biologique plus concret. Tout d’abord, l’étude de la physique de la filamentation laser a permis de mieux appréhender l’interaction des impulsions assujetties au processus de filamentation dans l’eau, primordiale pour l’identification des conditions d’irradiation propices à une meilleure maîtrise des conséquences de la génération des plasmas photo-induits. Les effets d’un rayonnement ionisant en solution aqueuse sont notamment véhiculés au travers de la chimie déclenchée par l’ionisation de l’eau, qui implique une interaction entre les espèces réactives produites et les solutés dilués en solutions. L’étude des conséquences de l’irradiation laser sur des solutés inorganiques a permis d’élucider la nature de cette chimie sous rayonnements. De surcroît, il a été démontré comment la malléabilité qu’offre l’utilisation d’un laser se répercute sur la capacité à moduler les conséquences de l’irradiation. Enfin, l’étude a été étendue à l’irradiation de molécules d’ADN diluées en solution aqueuse. L’analyse détaillée des dommages occasionnés à l’ADN a permis de mettre en exergue la présence de lésions complexes caractéristiques d’une irradiation par un faisceau intense de rayonnements ionisants. / Abstract : The present study is part in a new framework in radiobiology, introduced a decade ago: femtosecond laser-induced "cold" low density plasmas for the highly localized deposition of energy at sub-cellular scales in systems of biological interest. Since in aqueous solutions the action of such plasmas is equivalent to the deposition of a dose by ionizing radiation, plasma-mediated effects on solutes involve the radiation chemistry of water. This chemistry corresponds to the interaction of solutes with radical oxygen species as well as with secondary low energy electrons, produced by the plasma. Here, to better understand the radiation chemistry underlying the generation of low density plasmas in aqueous environments, we harnessed the multi-filamentation of powerful femtosecond laser pulses as a way to achieve a self-regulated production of spatially homogeneous low density plasma foci in water. The "cold" low density plasma micro-channels generated by the filamentation of the femtosecond laser pulses in aqueous solutions constitute a source of dense ionization. We studied the femtosecond laser filamentation in inorganic solutions to account for the radiation-assisted chemistry triggered by laser ionization in aqueous environment. We highlighted that the trivial optical control of the spatio-temporal distribution of light filaments in the irradiated sample resulted in the modulation of the corresponding radical chemistry. We concluded that these spatially and temporally resolved plasmas could be developed as a tool for the unprecedented control of chemistry under ionizing radiation. The addition of a spatial light modulator to control the filamentation process improves significantly our control on the spatio-temporal distribution of the laser-induced plasma channels. From a bundle of entangled random low density plasma channels, usually produced by the non-linear propagation of the powerful laser beam, we were able to obtain a programmable matrix of mono-filaments to achieve a more pervasive and homogeneous energy deposition. This method of irradiation allowed us to perform a detailed analysis to determine, quantify and compare the consequences of the laser irradiation with those of a conventional source of ionizing radiation (Gamma-Rays) on organic molecules (e.g. DNA) desolved in aqueous solutions. We showed that each filament behaves as an independent intense micro beam of ionizing radiation, that is capable of inducing complex DNA damage. We believe that a better understanding of the laser-induced plasma-mediated effects in aqueous solutions of biological interest will further the adoption of such laser-based ionisation sources, and that this unorthodox approach to radiation sciences will open new fields of investigation at the frontiers of radiation and laser-driven chemistry. Moreover, one of the principal conclusions of this thesis argues in favour of a shift of paradigm in radiation sciences, shuch that the consequences of ionising radiation would not only be considered for their injurious effects but also for the fine modulation of the functions of systems of biological interest. This sentiment paves the way for new emerging techniques and applications in biomedical fields.

The Role of Scavenger Receptor-A in Heat Shock Protein 27-mediated Atheroprotection: Mechanistic Insights into a Novel Anti-atherogenic Therapy

Heat shock protein (HSP)27 is traditionally described as an intracellular chaperone and signaling molecule, but growing evidence suggests it is released from immune cells where it plays an anti-inflammatory role during atherogenesis. Previously, the O’Brien lab found that overexpression of HSP27 led to augmented HSP27 serum levels in female apolipoprotein E knockout (ApoE-/-) mice, attenuated atherogenesis, and inhibited macrophage foam cell formation via physical binding with scavenger receptor (SR)-A. However, the precise mechanism of atheroprotection remained elusive. This thesis sought to ascertain the mechanism(s) by which HSP27 prevents foam cell formation, and determine if SR-A, a key receptor involved in the uptake of lipid into macrophages, plays an important role in HSP27-mediated atheroprotection. Pre-treatment of human macrophages with recombinant HSP27 (rHSP27) inhibited acytelated low density lipoprotein (acLDL) binding and uptake independent from receptor competition effect. Reduction in uptake was associated with attenuation of expression of SR-A mRNA, total protein, and cell surface expression. To explore the signaling mechanism by which HSP27 modulated SR-A expression it was hypothesized that nuclear factor-kappa B (NF-kB), a major regulator of many atherosclerosis gene programs, is altered by extracellular HSP27. Indeed, rHSP27 markedly activated NF-kB signaling in macrophages. Using an inhibitor of NF-kBsignaling there was an attenuation of rHSP27-induced inhibition of SR-A gene and protein expression, as well as lipid uptake, suggesting that SR-A expression is regulated by NF-kB activation. Lastly, to investigate if SR-A is required for HSP27-mediated atheroprotection in vivo, ApoE-/- and ApoE-/-SR-A-/- mice fed a high fat diet were treated with rHSP25, the mouse orthologue of HSP27, or PBS for 3 weeks. While rHSP25 therapy equally reduced serum cholesterol levels in the mouse cohorts, aortic atherogenesis, assessed using en face and sinus cross-sectional analyses, was attenuated in ApoE-/- mice but not ApoE-/-SR-A-/- mice. In conclusion, rHSP27 inhibits foam cell formation by downregulating SR-A expression. This effect may be associated with NF-kB activation. Reductions in atherosclerotic burden by rHSP27 require SR-A, and are independent of changes in serum cholesterol levels, highlighting the importance of macrophage lipid uptake in atherogenesis. Results presented in this thesis demonstrate that SR-A is a major target for HSP27 atheroprotection in the vessel wall, and provide an impetus for further studies that investigate the potential therapeutic value of HSP27.

The Purification and Identification of Interactors to Elucidate Novel Connections in the HEK 293 Cell Line

The field of proteomics studies the structure and function of proteins in a large scale and high throughput manner. My work in the field of proteomics focuses on identifying interactions between proteins and discovering novel interactions. The identification of these interactions provides new information on metabolic and disease pathways and the working proteome of a cell. Cells are lysed and purified using antibody based affinity purification followed by digestion and identification using an HPLC coupled to a mass spectrometer. In my studies, I looked at the interaction networks of several AD related genes (Apolipoprotein E, Clusterin variant 1 and 2, Low-density lipoprotein receptor, Phosphatidylinositol binding clathrin assembly protein, Alpha-synuclein and Platelet-activating factor receptor) and an endosomal recycling pathway involved in cholesterol metabolism (Eps15 homology domain 1,2 and 4, Proprotein convertase subtilisin/kexin type 9 and Low-density lipoprotein receptor). Several novel and existing interactors were identified and these interactions were validated using co-immunopurification, which could be the basis for future research.

Temperamento em bovinos é definido como a reação dos animais em resposta ao contato com o ser humano, geralmente atribuído ao medo ocasionado no manejo. Animais agitados são mais difíceis de manejar nas fazendas. Estudos na literatura mostram que características de temperamento influenciam o desempenho produtivo dos rebanhos, devido à sua correlação com outras características dentre elas, ganho de peso diário, taxa de prenhez, qualidade e rendimento de carcaça. Todavia, estudos visando o melhor entendimento dos mecanismos biológicos, e da arquitetura genética destas características são escassos. Com a evolução das tecnologias de genotipagem em alta densidade com marcadores de polimorfismo único (SNP), observou-se um aumento de pesquisas nas áreas de seleção genômica (GS) e associação genômica (GWAS). Entretanto, devido ao alto custo que a genotipagem em larga escala apresenta, torna-se inviável a genotipagem para todos os animais candidatos à seleção. Porém, existe a possibilidade de utilizar painéis de baixa densidade de SNP e inferir estes genótipos desconhecidos para painéis de alta densidade. Neste contexto foram desenvolvidos dois estudos. No primeiro, o objetivo foi identificar regiões cromossômicas associadas com características de temperamento em animais da raça Nelore. As características avaliadas foram: velocidade de saída (VS) e mediana do escore composto (EC_mediana), em que 599 e 575 animais foram utilizados, respectivamente. Todos os animais foram genotipados com Illumina BovineHD BeadChip (800K), e para o GWAS dois modelos estatísticos foram aplicados. O primeiro modelo utilizado foi genômico de única etapa (ssGBLUP), em que os efeitos dos SNP são derivados da predição dos valores genéticos genômicos dos animais. O segundo modelo foi linear misto, similar ao modelo anterior, porém várias análises, SNP por SNP (regressão simples) foram realizadas. Nos dois modelos foram incluídos os efeitos de grupo de contemporâneo como fixo, idade do animal como covariável e efeito poligênico do animal e ambiente permanente como aleatórios. Os componentes de variância foram estimados pelo método de máxima verossimilhança restrita. Os coeficientes de herdabilidade apresentaram baixa magnitude com estimativas de 0,02 e 0,05 para VS e EC_mediana, respectivamente. Diferentes regiões cromossômicas foram associados com as características estudadas nesta população, de acordo com os modelos utilizados, contribuindo para o entendimento da arquitetura genética dessas características. No segundo, o objetivo foi avaliar a acurácia de imputação também nesta população, usando dois painéis de baixa densidade (3K e 6K) e um de média densidade (50K) imputados para o painel de alta densidade (800K). Os animais genotipados com 800K foram divididos em população de validação e população de referência. Na população de validação, os animais tiveram seus genótipos mascarados para os chips Illumina 3K, 6K e 50K. A imputação de 3K, 6K e 50K para 800K foi realizada utilizando os softwares de imputação fastPHASE e Beagle. O software fastPHASE apresentou maiores valores de acurácia de imputação em comparação ao Beagle. O painéis 6K e 50K apresentaram maiores valores de acurácia. / Temperament in cattle is generally defined as the reaction of an animal in response to contact with human, usually attributed to the fear. Animals agitated are harder to manage in farms. Studies in the literature reported that temperamental traits have been found to influence the productive performance of herds, due to its correlation with other traits such as carcass quality, daily gains, pregnancy rate and feed efficiency. However, almost nothing is known regarding the genetic landscape controlling temperamental animal variation in cattle. With the advances in genotyping technologies for high density genotyping platforms to markers of single nucleotide polymorphisms (SNP), there was an increase in research in the areas of genomic selection (GS) and genome-wide association studies (GWAS). However, genotyping many animals with a high-density marker panel can be expensive and economically unfeasible. An alternative in this context is to use a lower density marker panel on a larger number of animals, and impute the missing genotypes. In this context two studies were developed. In the first study, the objective was to identify chromosomal regions associated with temperament traits in Nellore cattle. The temperamental traits evaluated in this study were: exit velocity (VS) and the median score temperament (EC_mediana) assessed in 599 and 575 animals, respectively. All animals were genotyped with Illumina BeadChip BovineHD (800K), and two GWAS statistical models were applied. The first model used was genomic single step (ssGBLUP) in which the effects of SNP are derived from the genomic prediction values of the animals. The second was linear mixed model, similar to the previous model, but a series of models, one for each SNP (single regression), were performed. In both models were included the effects of contemporary group as fixed, the animal\'s age as a covariate effect and polygenic animal and permanent environment as random effect. Variance components were estimated by restricted maximum likelihood. The heritability coefficients showed low magnitude estimative of 0.02 and 0.05 for VS and EC_mediana, respectively. Different chromosomal regions were associated with the traits studied in this population, according with the models used, and moreover to contribute in the understanding of the genetic architecture of these traits. In the second study, the objective was to evaluate the accuracy of imputation in a same population, using two panels of low-density (3K and 6K) and one medium-density (50K) panel markers imputed up to higher density of 800K. The animals genotyped with 800K markers panel were split into a reference population and validation population. The validation population the animals had markers masked to Illumina chip 3K, 6K e 50K. Imputation from 3K, 6K and 50K up to 770K markers was performed using fastPHASE and Beagle. The software fastPHASE had higher imputation accuracy compared to Beagle. The 6K and 50K panels showed higher accuracy.

Investigating the Mechanisms involved in Traffic-Generated Air Pollution: Mediated Disruption of the Blood-Brain Barrier in a Wild Type Mouse Model using a Pharmaceutical Intervention Approach

This study investigated whether oxLDL and/or angiotensin (Ang) II signaling pathways mediate traffic-generated air pollution- exposure induced alterations in blood-brain barrier (BBB) integrity and permeability in a healthy wild type (C57Bl/6) mouse model; additionally, whether these outcomes are exacerbated by a high fat-diet investigated. An environmentally relevant concentration of a mixture of vehicle engine exhaust (MVE) was used. To investigate the hypotheses, 12 wk old male C57Bl/6 mice on either a high fat (HF) or low fat (LF) diet were randomly assigned to inhalational exposure of either filtered-air (FA) or 30 µg PM/m3 diesel exhaust + 70 µg PM/m3 gasoline exhaust (MVE) for 6 hr/day for 30 days. Additionally, we examined mechanisms involved in MVE-mediated alterations BBB integrity using a novel BBB co-culture in vitro model, consisting of mouse primary cerebral vascular endothelial cells on an apical transwell and astrocytes in the basal compartment, which was treated with plasma from the mice on our exposure study. Our in vivo exposure study results showed that MVE inhalation resulted in increased circulating plasma oxLDL and Ang II, compared to FA controls. Additionally, we observed increased cerebral microvascular expression of oxLDL receptors, LOX-1 and CD-36, and Ang II receptor subtype 1 (AT1) in MVE-exposed C57Bl/6 mice, which was further exacerbated with consumption of an HF diet. Increased signaling of both Ang II and oxLDL was associated with decreased BBB integrity, as evidenced by the concurrent reduction in expression of tight junction (TJ) protein claudin-5 and increased permeability of sodium fluorescein (Na-F) from the blood into the cerebral parenchyma. Our results suggest that possible mechanisms involved in oxLDL and/or Ang II-mediated alterations in BBB integrity include oxidative stress and upregulated expression and activity of matrix metalloproteinase (MMP)-9, which is associated with degradation of TJ proteins in the BBB. Our in vitro BBB co-culture results confirm our in vivo findings, as we observe increased BBB permeability (TEER) and decreased integrity (decreased expression of TJ proteins) in the endothelial (apical) layer when treated with plasma from MVE-exposed mice, which was further exacerbated when treated with plasma from MVE-exposed mice on an HF diet. Pre-treatment of the endothelial cells with the AT1 receptor antagonist, Losartan, prior to applying plasma, resulted in attenuation of the alterations observed in endothelial integrity in the BBB co-culture treated with plasma from either MVE+LF or MVE+HF animals. These results suggest Ang II – AT1 signaling mediate, at least in part, the alterations in the BBB integrity observed after exposure to MVE. Moreover, we observed that treatment of the endothelial (apical) layer with plasma from MVE-exposed animals resulted in increased production of inflammatory mediators interleukin-6 (IL-6) and transforming growth factor-β in the astrocyte media (basal compartment). Additionally, these same astrocytes also displayed increased production of angiotensin-converting enzyme (ACE) and also AT1 receptor mRNA expression, while showing decreased expression of the aryl hydrocarbon receptor (AhR) and glutathione peroxidase (GPx). Collectively, these results suggest that exposure to the ubiquitous environmental air pollutant, vehicle engine emissions, results in increased oxLDL and Ang II signaling in the cerebral microvasculature, which is associated with decreased vessel integrity and increased oxidative stress and inflammatory signaling in the CNS. The observed detrimental outcomes are even further exacerbated when coupled with the consumption of an HF diet.

Binding of the Monomeric Form of C-Reactive Protein to Enzymatically-Modified Low-Density Lipoprotein: Effects of Phosphoethanolamine

Background: The 5 subunits of native pentameric C-reactive protein (CRP) are dissociated to generate the monomeric form of CRP (mCRP) in some in vitro conditions, both physiological and non-physiological, and also in vivo. Many bioactivities of mCRP generated by urea-treatment of CRP and of mCRP generated by mutating the primary structure of CRP have been reported. The bioactivities of mCRP generated by spontaneous dissociation of CRP are largely unexplored. Methods: We purified mCRP generated by spontaneous dissociation of CRP and investigated the binding of mCRP to enzymatically-modified low-density lipoprotein (E-LDL). Results: mCRP was approximately 60 times more potent than CRP in binding to E-LDL. In the presence of the small-molecule compound phosphoethanolamine (PEt), at 37 °C, the binding of mCRP to E-LDL was enhanced <2-fold, while the binding of CRP to E-LDL was enhanced >10-fold. In contrast, PEt inhibited the binding of both CRP and mCRP to pneumococcal C-polysaccharide, another phosphocholine-containing ligand to which CRP and mCRP were found to bind. We have not investigated yet whether PEt alters the structure of CRP at 37 °C. Conclusions: Combined data suggest that the targeting of CRP with the aim to monomerize CRP in vivo may be an effective approach to capture modified forms of LDL.
