Importance of arbuscular mycorrhiza fungi on crop production (yield and quality)Zerbib, Jérémie 19 December 2018 (has links)
In its environment, the plant interacts constantly with microorganisms. It is confronted with a variety of beneficial, neutral or pathogenic microorganisms, which are highly dependent on the carbon resources that the plant releases into the soil, giving them a supply of nutrients and energy necessary for their development. The arbuscular mycorrhiza is a mutualistic symbiosis formed by 80% of terrestrial plants from the Ordovician period, i.e. about 450 million years ago. The research presented in this manuscript was focused on two main goals: (i) to gain a better understanding of the mechanisms underlying trophic relationships in the mycorrhizal plant, and (ii) to study the impact of mycorhization on plant production, both in terms of quantity and quality. We investigated the mycorrhizal response of wheat landraces in order to understand if there were differences in a semi-controlled environment (greenhouses) or in an uncontrolled environment (field), and / or depending on the community of mycorrhizal fungi. Our results showed that mycorrhization and the nature of endogenous arbuscular mycorrhizal fungi can affect yield, even in wheat varieties with low mycorrhizal ability. We also highlighted the importance of choosing the criteria used to measure this potential gain. In a second line of research, we were interested in the impact of arbuscular mycorrhiza on the development of Micro-Tom tomato, and the trophic exchanges during the interaction. Multivariate analysis revealed behavioral differences between plants at mature green fruit and red ripening stages depending on arbuscular mycorrhizal fungal strain, as well as potential interaction event between two arbuscular mycorrhiza fungus trains. Our results confirm that the arbuscules are not the only sites of exchange in arbuscular mycorrhiza. The third axis of this work focused on the potential impact of seed production methods of ancient tomato varieties in drought condition, both on the endogenous community of arbuscular mycorrhizal fungi in the field, on the tolerance of the plant to water stress and on crop production in terms of fruit yield and quality. The experimental site was located in the Potager de santé (seed farm managed by Pascal Poot). Our results show no difference, under conditions of water stress in the field, between varieties regularly cultivated on this site and commercial varieties. Then, one may wonder if other factors related to the exploitation site, such as the beneficial microorganisms interacting with the roots of the plants could be at the origin of the increased tolerance to water stress. Work is underway in the laboratory to study this hypothesis. All of this work paves the way for new research aimed at elucidating the molecular mechanisms involved in the regulation of trophic exchanges during arbuscular mycorrhizae and the valuation of the ecosystem services rendered by arbuscular mycorrhizae in the field.

Transformação genética de tomate Micro-Tom e de laranja doce com os genes chitinase type III (PR-8) e constitutive disease resistance protein (CDR-1) de Citrus sinensis / Genetic transformation of Micro-Tom tomato and sweet orange with chitinase type III (PR-8) and constitutive disease resistance protein (CDR-1) genes from Citrus sinensisAnsante, Nathalia Felipe 04 December 2015 (has links)
HLB is currently considered the main disease affecting citrus plants. Given this factor, research for cultivars resistant to this disease is needed. Genetic transformation via Agrobacterium with the use of model plants has been an alternative for checking the gene function in response to pathogens, because these model plants have as characteristic a short life cycle and high power of regeneration. Therefore, the aim of this work was to produce transgenic plants, via Agrobacterium tumefaciens, of Micro-Tom tomato (Solanum lycopersicum L.), and sweet orange, with the genes encoding the PR-8 and CDR-1 proteins isolated from Citrus sinensis. The cotyledons from in vitro germinated Micro-Tom tomato seeds were used as explants source for genetic transformation experiments with PR-8 and CDR-1 genes. These explants were subcultured until the appearance of regenerating shoots and after transgenic plants, which were acclimatized and taken to a greenhouse. The transgenic plants were confirmed by PCR and the number of gene insertions by Southern blot. The plants were grown until T1 generation was obtained. Simultaneously genetic transformation experiments were performed with epicotyl segments from \'Hamlin\' sweet orange seeds germinated in vitro with CDR-1 gene in order to obtain transgenic plants and their characterization. Simultaneously, the standard curve construction was performed by qPCR analysis for identification of Pseudomonas syringae pv. tomato. Thirteen transgenic plants of Micro-Tom tomato with PR-8 gene and three with CDR-1 gene were obtained. The transformation efficiencies were around 0,38 to 1,98%. Three transgenic plants of Micro-Tom tomato with PR-8 gene were characterized by southern blot, and the number of inserts ranged from 1 to 3. Nineteen transgenic \'Hamlin\' sweet orange plants with CDR-1 gene were obtained through genetic transformation experiments, and the transformation efficiency was 2,06 to 5,96%. One plant was characterized, by Southern blot and has one DNA copy number in the plant genome.

Nitrogênio e tipos de substratos no monitoramento nutricional, na produtividade e na qualidade do tomateiro cultivado em ambiente protegido climatizado / Nitrogen and substrate in nutritional management, yield and quality of tomato growth in climated greenhouseRafael Campagnol 12 February 2015 (has links)
Vegetables production in soilless media is a technique that can increase yield, saving resources and facilitate the nutritional management of plants. However, for that to be possible, its necessary to know how the factors affect the plants growth in this system. The aim of this study was to evaluate the effects of different nitrogen rates of nutrient solution (NS) (60, 80 100, 120 and 140% of the standard) in two type of soilless media (S1: coconut fiber and S2: pine bark as base) for the mini tomato Sweet Grape growth. Were evaluated during the growing season pH, electrical conductivity (EC) and the nitrate content (NO3-drained) and potassium (K+ drained) in the solution drained from the vessels, SPAD index and the nitrate (NO3-pecíolo) and potassium (K+ petiole) content on petiole sap from the leaves and the nitrogen content in leaf dry mater (Nleaf). Were determined the number of fruit per plant (NFPP) and the average length of stems (ALS). The harvested fruits were sorted, counted and weighed to obtain the average mass of large fruits (AMLF), medium fruits (AMMF) and small fruits (AMSF), total weight of fruits per plant (TWFPP), total weight of large fruits (TWLF) medium fruits (TWMF), small fruits (TWSF) and cracked fruits (TWCF), total number of fruits per plant (TNFPP), total number of large fruits (TNLF), medium fruits (TNMF), small frutis (TNSF) and cracked fruits (TNCF). Fruit samples were collected in three periods and evaluated for soluble solids (SS), titratable acidity (TA) and concentration of ascorbic acid (AA). The pHdrained values decreased with increasing N levels in nutrient solution. The effects of N on pHdrained intensified throughout the growing season due to the accumulation of salts in the root medium, especially in S1. The CEdrained increased with N rate and was higher in S2. The increase in N rate increased the levels of NO3-drained on both substrates. The K+ drained was higher at 140% N dose in S1. SPAD index and Nleaf values increased with higher N rates in nutrient solution and reduced throughout the growing season. The NO3 - petiole increased with increasing dose of N in NS. The MMFG was higher with the addition of 140% N in relation to the dose of 60%. The increase in N rate increased the TNLF, TNMF, TNSF, TNCF and TNFPP. The biggest TNLF was obtained in S2 while the largest TNFPP, TNSF and TNCF occurred in S1. The TWLF, TWMF, TWSF, TWCF and TWFPP increased with increasing N levels in NS. The highest values of TWLF and TWSF were obtained at 140% N level, 331.05 and 1249.82 g plant-1, respectively. The biggest TWMF was obtained at 120% N level in NS, however, not differing of the 100 and 140% N levels. The SS, TA and AA fruits were not influenced by N and substrate types.

Characterization of tomato root-endophytic fungi and analysis of their effects on plant development, on fruit yield and quality and on interaction with the pathogen Verticillium dahliaeAndrade Linares, Diana Rocío January 2011 (has links)
Non-mycorrhizal fungal endophytes are able to colonize internally roots without causing visible disease symptoms establishing neutral or mutualistic associations with plants. These fungi known as non-clavicipitaceous endophytes have a broad host range of monocot and eudicot plants and are highly diverse. Some of them promote plant growth and confer increased abiotic-stress tolerance and disease resistance. According to such possible effects on host plants, it was aimed to isolate and to characterize native fungal root endophytes from tomato (Lycopersicon esculentum Mill.) and to analyze their effects on plant development, plant resistance and fruit yield and quality together with the model endophyte Piriformospora indica.
Fifty one new fungal strains were isolated from desinfected tomato roots of four different crop sites in Colombia. These isolates were roughly characterized and fourteen potential endophytes were further analyzed concerning their taxonomy, their root colonization capacity and their impact on plant growth. Sequencing of the ITS region from the ribosomal RNA gene cluster and in-depth morphological characterisation revealed that they correspond to different phylogenetic groups among the phylum Ascomycota. Nine different morphotypes were described including six dark septate endophytes (DSE) that did not correspond to the Phialocephala group. Detailed confocal microscopy analysis showed various colonization patterns of the endophytes inside the roots ranging from epidermal penetration to hyphal growth through the cortex. Tomato pot experiments under glass house conditions showed that they differentially affect plant growth depending on colonization time and inoculum concentration.
Three new isolates (two unknown fungal endophyte DSE48, DSE49 and one identified as Leptodontidium orchidicola) with neutral or positiv effects were selected and tested in several experiments for their influence on vegetative growth, fruit yield and quality and their ability to diminish the impact of the pathogen Verticillium dahliae on tomato plants. Although plant growth promotion by all three fungi was observed in young plants, vegetative growth parameters were not affected after 22 weeks of cultivation except a reproducible increase of root diameter by the endophyte DSE49. Additionally, L. orchidicola increased biomass and glucose content of tomato fruits, but only at an early date of harvest and at a certain level of root colonization. Concerning bioprotective effects, the endophytes DSE49 and L. orchidicola decreased significantly disease symptoms caused by the pathogen V. dahliae, but only at a low dosis of the pathogen.
In order to analyze, if the model root endophytic fungus Piriformospora indica could be suitable for application in production systems, its impact on tomato was evaluated. Similarly to the new fungal isolates, significant differences for vegetative growth parameters were only observable in young plants and, but protection against V. dahliae could be seen in one experiment also at high dosage of the pathogen. As the DSE L. orchidicola, P. indica increased the number and biomass of marketable tomatoes only at the beginning of fruit setting, but this did not lead to a significant higher total yield. If the effects on growth are due to a better nutrition of the plant with mineral element was analyzed in barley in comparison to the arbuscular mycorrhizal fungus Glomus mosseae. While the mycorrhizal fungus increased nitrogen and phosphate uptake of the plant, no such effect was observed for P. indica.
In summary this work shows that many different fungal endophytes can be also isolated from roots of crops and, that these isolates can have positive effects on early plant development. This does, however, not lead to an increase in total yield or in improvement of fruit quality of tomatoes under greenhouse conditions.

Previous issue date: 2016-03-31 / With the use of new farming techniques, add high productive potential genetic material arises, the need to assess the nutritional requirements of tomato producing areas. Thus, in order to evaluate the accumulation of dry matter and nutrients by ‘Caeté’ tomato, and its response to nitrogen and phosphorus doses, two experiments were conducted. The first was conducted in Fazenda Terra Santa, municipality of Quixeré-CE, between the months of June and September 2013, in an area of commercial plantations, which were evaluated the accumulation dry matter and nutrients by sampling tomato plants to 14, 28, 42, 56, 70, 84 and 99 days after transplanting (DAT). The second experiment was conducted at Fazenda Boágua, municipality of Baraúna-RN, from August to October 2014. The experimental design was a randomized block with four replications. The treatments consisted of five nitrogen rates (0, 60, 120, 240 and 360 kg ha-1 of N) and five doses of phosphorus (0, 75, 150, 300 and 600 kg ha-1 P2O5). The total estimated production of dry matter was 912.84 g plant-1, with the largest accumulations of nutrients in the fruiting stage, the fruits being the preferred drains. The macro and micronutrients accumulation order by tomato plants at 99 DAT was respectively K> N> Ca> S> Mg> P and Fe> Mn> B> Zn> Cu. The nitrogen, phosphorus and potassium were accumulated in large quantities in fruits. For micronutrient accumulation was higher in the vegetative to the fruit. Nitrogen and phosphorus doses that provided the highest commercial yields were 45 and 311 kg ha-1, respectively. Nitrogen and phosphorus in the diagnostic sheet increased with the addition of N rates and P2O5 to the ground. Tomato 'Caeté' fertilized with 60 and 150 kg ha-1 of N and P2O5, accumulated 36.30 and 2.83 kg ha-1 of nitrogen and phosphorus in the fruits, respectively / Com o emprego de novas técnicas de cultivo, acrescidas de materiais genéticos de alto potencial produtivo, surge a necessidade de se avaliar os requerimentos nutricionais do tomateiro nas regiões produtoras.

Previous issue date: 2016-11-28 / Banco Santander / Composting is a solution for recycling organic waste, transforming them into compounds that can be used as nutrients for any plant species, with emphasis on the vegetable group. The objective of the present study was to take advantage of biodegradable solid waste, making possible the production of organic fertilizers by means of the composting technique, with a view to their application in the cultivation of cherry tomatoes. The research was based on two experiments. The first was developed through an action research in the Community Association Recycling for Life, located in the municipality of Mossoró/RN. The composting technique was developed according to the windrow method, with the construction of five piles. Controlled until the final phase of composting, parameters such as: temperature, humidity, pH, total organic carbon, total nitrogen, and C/N ratio. Analyzes of humic substances, macro and micronutrients, chemical contaminants (Cd, Ni and Pb), total and thermotolerant coliforms, Salmonella, visual observations of maturation and suggestion of packaging and labeling of the organic compounds produced were performed. The second experiment was carried out in a protected environment, in the West Field of the Federal Rural Semi-Arid University - UFERSA, Mossoró/RN, in order to observe the effects of the organic compounds produced in the cultivation of cherry tomatoes. The experimental design used was in randomized blocks, with five replicates and five treatments, the organic fertilizers produced in the first experiment were combined with coconut fiber (1:1). In the culture were evaluated growth, production, post-harvest quality, nutrition and microbiological analysis. The stabilized organic compounds reached satisfactory results with C/N values between 12/1 to 16/1, presenting as good soil conditioners with potential to improve the physicochemical characteristics of a soil that presents nutritional deficiency. The levels of chemical contaminants evaluated were well below those established in the main European, American and normative legislation, showing that they are within the parameters of quality for the organic compounds that will have applications as fertilizers and soil conditioners. The compounds produced showed absence of total coliforms, thermotolerant and Salmonella. In the crop, the organic production was satisfactory to favor a product of quality free of nutritional and healthy deficiency due to the absence of pathogens. The T5 treatment presented a better result with higher phytomass and average productivity, indicating that significant and diversified amounts of food residues influence the amount of nutrients that will be available to the crop. The results allow us to affirm that the application of cattle manure and dietary residues in small scale composting originated a product with adequate nutritional characteristics and did not alter the microbiological quality of the compounds and concomitantly of the cultivated tomatoes, resulting in the production of healthy fruits.

Receptor-like kinases (RLKs) represent a large family of transmembrane proteins that play important roles in cellular signaling perception and propagation in plants. In Arabidopsis thaliana, the RLK superfamily is made-up of over 600 proteins and many of these RLKs, mainly those bearing leucine-rich repeats (LRR), have been considered as excellent targets for engineering superior crops with enhancement of yield and resistance to biotic and abiotic stresses. The LRRII-RLK subfamily is particularly relevant due to the dual function of its members in both development and defense. In spite of the relevance of the RLK family and the completion of the tomato genome sequencing, the tomato RLK family has not been characterized and a framework for functional predictions of the members of the family is lacking. In this investigation we disclosed a complete inventory of the members of the tomato RLK family. To generate a complete list of all members of the tomato RLK superfamily, we performed a phylogenetic analysis using the Arabidopsis RLKs as a template. A total of 647 RLKs were identified in the tomato genome, which were organized into the same RLK subfamily clades as Arabidopsis. Only eight of 58 RLK subfamilies exhibited specific expansion/reduction compared to their Arabidopsis counterparts and only six proteins were lineage-specific in tomato, indicating that the tomato RLKs share functional and structural conservation with Arabidopsis. We also characterized the LRRII-RLK family by phylogeny, genomic analysis, expression profile and interaction with the virulence factor from begomoviruses, the nuclear shuttle protein (NSP). The LRRII subfamily members from tomato and Arabidopsis were highly conserved in both sequence and structure. Nevertheless, the majority of the orthologous pairs did not display similar conservation in the gene expression profile, indicating that these orthologs may have diverged in function after speciation of tomato and Arabidopsis common ancestor. Based on the fact that members of the Arabidopsis RLK superfamily (NIK1, NIK2, NIK3 and NsAK) interact with the begomovirus nuclear shuttle protein (NSP), we examined whether the tomato orthologs of NIK, BAK1 and NsAK genes interacted with NSP of Tomato Yellow Spot Virus (ToYSV). The tomato orthologs of NSP interactors, SlNIKs and SlNsAK, interacted specifically with NSP in yeast and displayed an expression pattern consistent with the pattern of geminivirus infection. In addition to suggesting a functional analogy between these phylogenetically classified orthologs, these results expand our previous observation that NSP-NIK interactions are neither virus-specific nor host-specific. Therefore, NIK-mediated antiviral signalling is also likely to operate in tomato, suggesting that tomato NIKs may be good targets for engineering resistance against tomato-infecting begomoviruses.

Previous issue date: 2015-06-03 / Coordenação de Aperfeiçoamento de Pessoal de Nível Superior - CAPES / Photoselective screens are a recent approach in protected cultivation, which can promote benefits in addition of the protective functions of nettings. These nets not only scatter but spectrally-modify the transmitted light, in order to promote desired physiological responses and improve the penetration of the light into the inner plant canopy. Tomato is the second most important horticulture crop in the world. Traditionally, tomato is cultivated in the open field, but recently there has been an increase of the protect cultivation area aiming to protect plants from climate adversities. This study aimed to investigate whether a combination of growth regulator, increased plant density and the effect of red screen-shade net could improve tomato yield, quality and promote desired vegetative development. The experiment was conducted from May 21st to November 1st, 2014, in Goiânia, Brazil (16º35´47´´S, 49°16´47´´W, 730 m). The hybrid Ivete from Sakata Seeds® were cultivated in 16x12x 2.2 m3, 40% red shading screenhouse and field plots as control. Leaf area index, trusses height, and total height were measured. The total number, total weight, diameter and number of lost fruits were totaled. Nutritional analyses were also performed: physical (Fresh mass, total solid soluble - TSS, titritable acidity - TA, SST/AT ratio and pH) and nutritional parameters (vitamin C and lycopene) in two moments of collecting of fruits (98 and 129 days after transplantation). All the measures were carried by official methods. Results showed that highest yields were obtained using 3 plants per m2 in both environments. The highest total fruit production occurred in open field conditions. However, the total fruit lost in the open field conditions was greater. Fruits produced in an open field were more acidic and had greater titratable acidity (0.37% citric acid) compared to fruits from screenhouse (0.33 % citric acid) at 3 plant per square meter. In addition, fruits grown in the field had greater TSS content (6.6 °Brix) than tomatoes grown in a protected environment (5.4 °Brix). Fruits produced in the open field had greater TSS:TA ratios than those produced in a protected environment. Significantly higher lycopene content was observed in plastic-house tomatoes integrated with red shade netting technologies (47.0 μg g−1) using 2.3 plants per m² than in open field-grown tomatoes (29.6 μg g−1). The results provide useful data for detecting differences among environment variation in tomato composition and red colour shade nets. In addition, the photoselective red screen combined with the density of 3 plants per m² promoted higher yield of commercial fruits.
tomateiro e promover desenvolvimento vegetativo desejado.

FundaÃÃo de Amparo à Pesquisa do Estado do Amazonas / The grafting technique has aroused special interest among vegetable producers around the world, this because it allows to obtain quick responses in areas that already have problems with pests and vegetables disease, especially soils, and allows the producer to continue using the same production system, with small change and adaptation of their seedling production system.
Despite the foregoing, few results in the literature showing the productive and physiological
behavior of grown tomato grafts on different rootstocks. Based on the above, the aim of this study was to evaluate yield and physiological characteristics of tomato 'Santa Clara', grafted on rootstocks prepared from different cultivars and species of Solanaceae family. The study was
conducted in the open field area and Plant Growth Laboratory, located on the Pici campus, belonging to
the Federal University of Ceara . It was used a randomized complete block design (RBD), being seven the treatments, seven types of rootstocks, ie cherry tomato 'Vermelho'; tomato 'GuardiÃo', aubergine 'Comprida Roxa ',
eggplant 'Comprido Grande Rio', coconna(S. sessiliflorum), pimentoeiro 'Casca Dura Ikeda', besides the tomato 'Santa Clara' used in autograft system. In addition, to explain possible effects of grafting in the evaluated characteristics was also evaluated the tomato 'Santa Clara' ungrafted. Differences were observed for most traits. In general, all the rootstocks used were efficient in providing conditions for the complete phenological development used graft. For the conditions of this work, the rootstock cherry tomato 'Vermelho' and ungrafted plants were the treatments that showed the best production results, and may be indicated as sustainable alternative production in low altitude and latitude production areas.
