Prostatacancer är den sjätte mest förekommande cancertypen i världen, och den tredje vanligaste cancertypen bland män. De olika typer av behandlingar som finns idag botar oftast inte sjukdomen. Det är därför viktigt att utveckla bättre behandlingsmetoder. Det är sedan tidigare känt att mangan kan orsaka apoptos i olika celltyper. Det ger en möjlighet att använda mangan för att hämma cancer och det är därför viktigt att veta vilka apoptotiska markörer som är involverade. Syftet med projektet var att undersöka om de sker en ökning av de apoptotiska markörerna kluven Kaspas-3 och kluven PARP efter manganbehandling av prostatacancerceller. Därefter kunna avgöra om manganbehandlingen har orsakat celldöd genom att inducera apoptos. Under projektet odlades prostatacancerceller (PC3) som sedan behandlades med 200 µM mangan under 6, 24 och 48 timmar. Därefter mättes mängden av proteinerna kluven Kaspas-3 och kluven PARP med hjälp av ett sandwich-ELISA kit. En tydlig stegvis ökning av apoptosmarkörerna med inkuberingstid hade förväntats. Det förväntade resultatet erhölls inte, för kluven Kaspase-3 ledde manganbehandlingen t.o.m. till en sänkning av koncentrationen. Det kan ha uppstått problem vid analysen som t.ex dålig lysering av cellerna eller ojämn tillväxt av dem. Det behövs fler studier för att utreda detta och för att undersöka andar potentiella apoptosmarkörer. / Prostate cancer is the sixth most common cancer type in the world, and the third most common cancer type among men. The different types of treatments that are available today do not usually cure the disease. It is therefore important to develop better treatment methods. It has previously been stated that manganese can cause apoptosis in different cell types. It provides an opportunity to use manganese to inhibit cancer and it is therefore important to know which apoptotic markers that are involved. The purpose of the project was to investigate whether an increase in the apoptotic markers cleaved Kaspas-3 and cleaved PARP after manganese treatment of prostate cancer cells. Thereafter, it can determine whether manganese treatment has caused cell death by inducing apoptosis. During the project prostate cancer cells (PC3-cells) were cultivated, then treated with 200 µM manganese for 6, 24 and 48 hours. The amount of the proteins cleaved Kaspas-3 and cleaved PARP was measured by using a sandwich ELISA kit. A clear gradual increase of apoptotic markers with incubation time was expected. The expected result was not obtained, for cleaved Kaspase-3 manganese treatment even decreased the concentrations. There may have been problems with the performance of the analysis such as poor lysis of the cells or uneven growth of the cells. More studies are required to investigate this and other potential apoptotic markers.

The Use of Lignin Derivatives to Improve Selected Paper Properties

<p>Ved består huvudsakligen av tre typer av polymerer, cellulosa, hemicellulosa och lignin. Lignin bildas i naturen genom enzymatiskt initierad oxidativ koppling av tre olika typer av fenylpropan-enheter. Dessa bygger genom olika kol-kol- och kol-syre-bindningar upp en amorf tredimensionell polymer. När kemisk massa tillverkas bryts lignin ner och löses ut i kokluten. Luten innehåller de förbrukade kokkemikalierna och bränns generellt i en sodapanna för att regenerera kemikalierna och producera ånga. Sodapannan är emellertid dyr. Därför har den blivit produktionsbegränsande på många massabruk. Att avlägsna en del av ligninet från avluten vore därför önskvärt och att finna ekonomiskt intressanta produkter baserade på lignin från svartlut är därför ett viktigt forskningsområde .</p><p>Ett lämpligt område för ligninprodukter vore som tillsatts i oblekt massa. Oblekt massa används till stor del för tillverkning av kraftliner, topp- och bottenskikten på wellpapp. När lådor av wellpapp lagras i containrar som färdas över haven, förändras den relativa luftfuktigheten. Detta gör att lådorna kollapsar lättare än om de skulle ha lagrats vid konstant luftfuktighet, även en hög sådan. Detta är på grund av det så kallade mekanosorptiva- eller accelererade krypfenomenet. Genom tillsatts av våtstyrkemedel till kraftliner eller behandla den med hydrofoba ämnen, finns indikatoner på att mekanosorptiva effekten skulle kunna minska.</p><p>För att försöka minska den effekten har ett lågmolekylärt kraftlignin, som utvunnits med hjälp av tvärsflödesfiltrering av svartlut och svavelsyrafällning, använts. Genom derivatisering av detta lignin med linolja erhölls ett hydrofobt ligninderivat som uppvisar strukturella likheter med biopolymeren suberin. När detta suberinlika ligninderivat tillsätts till massa verkar det mekanosorptiva krypet minska. När lågmolekylärt lignin används tillsammans med ligninradikalinitiatorerna lackas eller mangan(III) i kraftlinermassa erhålls dessutom en våtstyrka på ca 5% av torrstyrkan. Efter aminering av detta lignin gav en tillsatts till kraftlinermassan en våtstyrka på upp till 10% av torrstyrkan. Det finns indikationer på att det mekanosorptiva krypet samtidigt minskar när dessa behandlingar görs som ger upphov till ökad våtstyrka.</p> / <p>Wood consists mainly of three types of polymers; cellulose, hemi cellulose and lignin. Lignin is formed in nature through enzymatic initiated oxidative coupling of three different kinds of phenyl propane units. These form by various carbon-carbon and carbon-oxygen bonds, an amorphous three-dimensional polymer. As chemical pulp is produced, lignin is degraded and dissolved into pulping liquors. These liquors contain the spent cooking chemicals and are generally burnt in a recovery boiler to regenerate cooking chemicals and produce steam. However, the recovery boiler is expensive. Hence, it has become the bottleneck for production in many pulp mills. Removal of some lignin from the spent cooking liquor would, for that reason, be desired and valuable products based on lignin from cooking liquors are searched for.</p><p>One suitable area for lignin products would be as additive in unbleached pulp. A major product from unbleached pulp is kraftliner, the top and bottom layers of corrugated board. When boxes of corrugated board are stored in containers travelling overseas the relative humidity is varying. This makes the boxes collapse more easily than if they were stored at constant humidity, even a high one. This is due to the so called mechano-sorptive or accelerated creep phenomenon. By addition of wet strength additive to kraftliner or treating it with hydrophobic compounds there are indications on that the mechano-sorptive effect would decrease.</p><p>Trying to decrease this effect, low molecular weight kraft lignin has been used. It was obtained by cross-flow filtration of black liquor and precipitation by sulphuric acid. By derivatisation of this lignin by linseed oil, a hydrophobic lignin derivative was obtained, similar in structure to units in the biopolymer suberin. As this suberin-like lignin-derivative was added to pulp the mechano-sorptive creep seemed to be lowered. Furthermore, when the low molecular weight lignin was used together with the lignin radical initiators laccase or manganese(III) in kraftliner pulp, a wet strength of about 5% of dry strength was obtained. An amination treatment of this lignin and addition to kraftliner pulp resulted in a wet strength of up to 10% of dry strength. There are indications of that the mechano-sorptive creep also decreases as these treatments, resulting in increased wet strength, are made.</p>

The aim of my diploma thesis was to explore the influence of manganese ions put into nickel nitrate structure. The attention was focused mainly on the stabilization of nickel nitrate a modification, which could not change into b modification. The theoretical part of my thesis focused on applied materials, kinds of acumulators, basic parameters of acumulators and the description of chosen methods of measurement employed. It contained the electrodeposition, the impedance spectroscopy and the cyclic voltammetry. The task of the experimental part was to measure the effect of manganese ions put into potassium hydroxide structure. Proportions of Ni (nickel nitrate) : Mn (manganese nitrate) 10:0, 7:3, 5:5, 6:1 and 6:4 were blended for the measurement. By means of the electrodeposition, there was a thin layer of nickel hydroxide with an admixture of manganese ions from nickel nitrate and manganese nitrate dilution put on the nickel plate. After the spread of the thin layer on the nickel plate, the measurement by the method of cyclic voltammetry followed. The next task consisted in trying to add KOH (potassium hydroxide) anionic and cationic surfactant into the dilution.

Metal acetylacetonates of the general formula [M(acac)3] (MIII=Cr, Mn, Fe, Co) are among the best investigated coordination compounds. Many of these first-row transition metal complexes are known to have unique electronic properties. Independently, photophysical research with different β-diketonate ligands pointed towards the possibility of a special effect of the 2,4,6-trimethylphenyl substituted acetylacetonate (mesacac) on the electron distribution between ligand and metal (MLCT). We therefore synthesized and fully characterized the previously unknown octahedral title complex. Its solid-state structure shows a Jahn-Teller elongation with two Mn−O bonds of 2.12/2.15 Å and four Mn−O bonds of 1.93 Å. Thermogravimetric data show a thermal stability up to 270 °C. High-resolution mass spectroscopy helped to identify the decomposition pathways. The electronic state and spin configuration of manganese were characterized with a focus on its magnetic properties by measurement of the magnetic susceptibility and triple-zeta density functional theory (DFT) calculations. The high-spin state of manganese was confirmed by the determination of an effective magnetic moment of 4.85 μB for the manganese center.

Metallophores from selected actinobacteria for metal extraction and phytomining of strategic elements

Different aspects of interaction of metals other than iron with metallophore mixtures were elucidated. Metal chelation besides iron and greater than with pure desferrioxamine B is documented by enhanced performance of soil mineral dissolution following application of diverse metallophore mixtures. New insights from genome to product of metallophores from Gordonia rubripertincta CWB2 under different cultivation conditions and under metal stress, particularly induced by rare earth elements are given. This evaluation led to increase by a factor of 13. Structure analyses showed that the strain produces citrate and different desferrioxamines. A robust RP-HPLC-based method quantified metal complexation. Further metal immobilization methods for metallophores for metal extraction from solutions with low concentrations were established. Se ofrecen nuevos conocimientos desde el genoma hasta el producto de los metanóforos de Gordonia rubripertincta CWB2 en diferentes condiciones de cultivo y bajo estrés por metales, especialmente inducido por elementos de tierras raras. Esta evaluación condujo a una sobreproducción de un factor de 13. El análisis estructural muestra que la cepa produce citrato y diferentes desferrioxaminas. Un método robusto basado en RP-HPLC cuantificó la complejación de metales. Se establecieron otros métodos de inmovilización de metales para la extracción de metales a partir de soluciones poco concentradas. Processing-structure-mechanical property relationships in high carbon medium manganese steels with austenitic microstructure

A balance between strength and ductility has been one of the most important considerations in the steel industry. Austenitic steel or multi-phase steel with retained austenite has plasticity-enhancing mechanisms, which can make it achieve high strength and good formability. Due to the occurrence of twinning-based mechanisms in high Mn steels, they have improved strength without sacrificing ductility. However, high Mn steels with extraordinary mechanical properties has not been used in mass production because of its high material cost together with welding problems and so on. As a consequence, many researchers have attempted to decrease the Mn concentration of high Mn twinning-induced plasticity steels without significant sacrifice of the mechanical properties. In the present work, a novel medium Mn steel with high C is designed with the aim of obtaining comparable mechanical properties as high Mn TWIP steel. In addition to Mn, C is also common effective austenite stabilizing element. C and Mn both increase the SFE of austenite. It should be possible to substitute at least some of the Mn in high Mn steels with C and still retain the TWIP effect. If the reduction in Mn content is not compensated for by the addition of other alloying elements, the microstructure will additionally contain some ferrite or martensite. The problem with C concentration is that it will result in the formation of carbide during the cooling process. As long as the carbide formation is suppressed, the formation of ferrite/martensite in medium Mn steels can be inhibited by an increase in the C concentration. In such cases, a soft and formable austenitic microstructure can be achieved by quenching from high austenitization temperatures to retain austenite with appropriate mechanical stability. The precipitation and dissolution of cementite in austenitic medium Mn high C steels capable of deformation-induced twinning were analyzed based on the associated length changes. Al addition was found to significantly retard the kinetics of cementite precipitation, indicating its usefulness in the design of cementite-free austenitic medium Mn steels with high C concentrations. Furthermore, Al addition changes the morphology of intragranular cementite from plate-shaped to equiaxed. The tensile properties of alloy were also examined in the present study. The present contribution discusses the mechanical properties of a bulk medium Mn high C steel with special alloying additions to oppose the precipitation of cementite. In particular, it aims to justify the mechanical properties based on crack nucleation and growth mechanisms. The reported mechanical properties enable a comparison with those of the well-known high Mn and Hadfield steels.
