The influence of different winemaking techniques on the extraction of grape tannins

These building blocks are called monomers which consist of catechins, epicatechins, epigallocatechins and epicatechin-gallate. Tannin is important in wine as it contributes to bitterness, mouth feel (astringency) and maturation potential of the wine. Futhermore it has a health benefit as an antioxidant. Anthocyanins are responsible for the colour of red wine. The anthocyanins combine with tannins to form stable polymeric pigments. Due to the importance of tannins and anthocyanins in wine, it is imperitative that different winemaking techniques are used to extract as much of these components as possible and that the analysis is done quickly and accurately. The aim of this study was to evaluate different winemaking techniques and their extraction of tannins and anthocyanins into the wine. Too much tannin extraction can have a negative effect on the sensory quality of the wine. Therefore a second aim was to evaluate the mouth feel properties of a Shiraz wine. A third aim was to compare the two tannin precipitation methods in terms of time efficiency, repeatability and the ease of practice. To investigate the amount of tannin concentration extracted by different winemaking techniques, two cultivars (Cabernet Sauvignon and Shiraz) were used. These treatments included the addition of an enzyme during fermentation [E], cold maceration [CM], post maceration [PM] and the combination of cold and post maceration [CM+PM]. The grapes were harvested in two different climatic areas during the 2008 and 2009 vintages. The two climatic areas were classified according to the Winkler scale as a III (Morgenster) and a IV (Plaisir de Merle). The grapes were harvested at two different ripeness levels in order to evaluate the effect of the different winemaking processes on the extraction of tannins and anthocyanins. One harvest was before (LB) and the other after (HB) the commercial harvest. The results of this study showed significant differences in the phenolic composition of the wines. It was found that the warmer area showed higher tannin concentrations than the cooler area for both cultivars. In the 2008 Cabernet Sauvignon the CM extracted higher concentrations of tannin from the cooler area at both ripeness levels. In the warmer area, CM extracted the highest tannin concentration HB, but the CM+PM extracted the highest tannin concentration from Cabernet Sauvignon at the LB and CM at the HB of the warmer area. In 2009 the PM extracted the highest concentration of tannin at the lower ripeness level, while the E treatment extracted the highest concentration from the warmer area. In the cooler area the CM+PM extracted the highest concentration of tannin at a lower ripeness level, while there were no siginicant differences between the different treatments at the higher ripeness level. The highest anthocyanin concentration was found in the cooler area. The CM treatment was found to have no effect on anthocyanin extraction. Different methods are available to quantify the tannin concentration in wine. Two of the most popular tannin analytical methods are the bovine serum albumin (BSA) and the methyl cellulose precipitable tannin (MCP) methods. The BSA method is a very complex method which uses at least 3 times more reagents than the MCP method. The MCP method only analyzes tannins, while the BSA method analyzes tannins, monomeric pigments (MP), small polymeric pigments (SPP) and large polymeric pigments (LPP). In this study a good correlation was found between the two tannin precipitation methods (R2 – 0.88). There is controversy regarding the variability of these methods. Some scientists found that the two methods show a good correlation with HPLC, while others found that there was no such correlation between the precipitation methods and the HPLC. The MCP method had a practical advantage as it could be performed in half the time required for the BSA method. This has a significant impact in scenarios where a high sample throughput is required although it only measures total tannin. The phenolic composition and mouth feel of the wine was strongly influenced by the climatic area. In the warmer area the effect of tannin concentration on mouth feel was much less than in the cooler area. The wine made of riper grapes, was more grippy, bitter and numbing than the wines made from greener grapes. The E treatment was especially associated with a dry, grippy sensation. / AFRIKAANSE OPSOMMING: Druif en wyn fenole bestaan uit flavanole wat weer die boublokke is van tanniene. Hierdie boublokke, wat bekend staan as monomere, betsaan uit katesjiene, epikatesjiene, epigallokatesjiene an epikatesjien-gallaat. Tanniene is belangrik in wyn aangesien dit bydra tot bitterheid, mondgevoel (vrankheid) asook die verouderingspotensiaal van wyn. As antioksidante hou dit ook gesondheidsvoordele in. Antosianiene dra by tot die kleur van rooiwyn. Antosianiene kombineer met tanniene om meer stabiele polimeriese pigmente te vorm. As gevolg van die belangrikheid van tanniene en antosianiene is dit van uiterse belang dat verskillende wynmaak tegnieke gebruik word om ekstraksie in die wyn te bevoordeel en dat die analitiese metode so vinnig en akkuraat as moontlik gedoen word. Die eerste doel van hierdie studie was om die ekstraksie van tanniene en antosianiene deur middel van verskillende wynmaak tegnieke te evalueer. Te veel tanniene in die wyn kan negatiewe sensoriese kwaliteit tot gevolg het. Daarom is die tweede doel om die sensoriese kwaliteit van Shiraz wyn te evalueer. Die derde doel van hierdie studie was die twee tannien presipitasie metodes met mekaar te vergelyk in terme van die moeilikheidsgraad van die metode, tyd doeltreffendheid en herhaalbaarheid. Verskillende wynmaak tegnieke (ensiem byvoegings [E], koue maserasie [CM], verlengde dopkontak [PM] en ‘n kombinasie van koue maserasie en verlengde dopkontak [CM+PM]) is vergelyk ten opsigte van tannien en antiosianien ekstraksie. In 2008 en 2009 is twee kultivars (Cabernet Sauvignon en Shiraz) in twee verskillende klimatologiese areas gepars. Hierdie areas is geklassifiseer in die Winklerskaal as ‘n IV (Plaisir de Merle) en ‘n III (Morgenster). Om die effek van die verskillende wynmaak tegnieke op die ekstraksie van antosianiene en tanniene te vergelyk, is hierdie twee kultivars by twee verskillende rypheidsgrade geoes. Die eerste oes was net voor kommersiële oes (LB) en die tweede oes het net na kommersiële oes (HB) plaasgevind. In beide 2008 (LB en HB) en 2009 (LB) het CM die meeste antosianiene geëkstraheer, terwyl geen behandeling ‘n effek gehad het by HB. Twee van die mees populêre tannien analitiese metodes is die BSA (bovine serum albumien) en die MCP (metielsellulose presipitasie) metodes. Die BSA metode is ‘n baie meer ingewikkelde metode waarvoor drie keer meer reagense gebruik word as vir die MCP metode. Maar waar die MCP net tanniene ontleed, ontleed die BSA metode tanniene, monomere (MP), klein polimeriese pigmente (SPP) en groot polimeriese pigmente (LPP). Die herhaalbaarheid van die metodes het redelike kontroversie veroorsaak, waar sommige navorsers bevind het dat die BSA metode nie so herhaalbaar is soos eers bevind is nie. Die MCP metode het ’n praktiese voordeel aangesien dit in die helfde van die tyd van die BSA metode uitgevoer kan word. Dit het ‘n groot impak indien ‘n groot hoeveelheid monsters ontleed moet word. Die fenoliese samestelling en mondgevoel word sterk beïnvloed deur die klimatologiese area. In die warmer area was die effek van tannien konsentrasie op mondgevoel kleiner as in die koeler area. Die wyn van ryper druiwe het meer harder, verdowingseffek en bitter nasmaak gehad as by die wyn van groener druiwe. Die ensiem behandeling was meer geassossieerd met droë mond gevoel.

在DNA微陣列基因的實驗中資料包括數千個cDNA 序列,為了要篩選出有差異表現基因,同時針對大量基因個數作假設檢定。若無適當地調整個別檢定問題中的誤差率,則將會膨脹整體的誤差率。在多重假設檢定中為了讓整體誤差率(familywise error rate, FWE)控制在設定水準下,必須調整個別假設檢定之個別型一誤差率CWE的檢定準則,此為多重比較方法(multiple comparison procedures:MCP)。然而當多重比較的個數增加時,控制整體誤差率FWE之傳統的多重比較方法會是過於嚴格的標準,不容易推翻虛無假設,使得檢定的結果太過保守。為了解決此現象,Benjamini and Hochberg(1995) 建議另一種錯誤率:錯誤發現率(false discovery rate:FDR)。錯誤發現率定義為在被拒絕之虛無假設中錯誤拒絕的比例之期望值。而Benjamini and Hochberg(1995)也在文中提出一個得以控制錯誤發現率的多重比較方法,稱為BH方法。本篇論文將詳盡地介紹CWE、FWE和FDR三種誤差率,並提出-修正BH的方法,稱為BH( )。我們將透過電腦模擬驗證出新的修正BH方法之表現比原BH方法有較高的檢定力,且從實例的結果中發現BH( )比原BH方法能檢測出更多的顯著個數。 關鍵字:個別型一誤差率(CWE);整體誤差率(FWE);多重比較方法(MCP); 錯誤發現率(FDR)。 / cDNA microarray technology provides tools to study thousands of genes simultaneously. Since a large number of genes are compared, using a conventional significant test leads to the increase of the type I error rate. To avoid the inflation, the adjustment for multiplicity should be considered and a multiple comparison procedure (MCP) that controls the familywise error rate (FWE) is recommended. However, the conservativeness of a MCP that controls FWE becomes more and more severe as the number of comparisons (genes) increases. Instead of FWE, Benjamini and Hochberg (1995) recommended to control the expected proportion of falsely rejecting hypotheses—the false discovery rate (FDR)—and developed a MCP, which has its FDR under control. In this paper, the error rates CWE, FWE and FDR are fully introduced. A new MCP with FDR controlled is developed and its performance is investigated through intensive simulations. Cette thèse a été effectuée dans le cadre des projets DISTOR (Européen) et HTPSTOCK (Français) qui visent à concevoir et étudier de nouveaux composites graphite/sel destinés au stockage de l'énergie thermique à haute température (>200°C). Elle est scindée en deux parties distinctes. <br />La première partie détaille les travaux relatifs à l'élaboration et la caractérisation thermique de ces nouveaux composites. On y présente les différentes voies d'élaboration des composites (dispersion, compression uniaxiale, isostatique) associées aux différents types de graphite (graphite naturel expansé (GNE), graphite synthétique) investigués au cours de cette thèse. On y retrouve également les résultats liés à la campagne de caractérisation thermique de ces composites permettant de mettre en exergue l'impact du graphite sur les propriétés conductrices des matériaux étudiés. A partir de ces résultats, des études de modélisation de l'évolution de la conductivité thermique ont pu être entrepris afin d'approfondir la compréhension de l'effet du graphite (quantité, taille de particules) sur la conductivité effective des composites.<br />La deuxième partie présente essentiellement les dispositifs de caractérisation thermique et les modèles thermocinétiques associés qui ont du être développés et adaptés aux spécificités des matériaux nouvellement élaborés. Cela concerne principalement les matériaux élaborés par compression, matériaux difficilement reproductible et possédant des propriétés orthotropes. La caractérisation de ce type de matériaux s'avère très délicate et pour le moins fastidieuse. C'est pourquoi nous nous sommes attachés à développer et adapter les moyens de caractérisation existants afin de permettre la caractérisation thermique complète d'un matériau conducteur orthotrope à partir d'une seule mesure sur un seul échantillon

L'objectif de cette thèse est d'étudier un système énergétique intégré dans l'enveloppe des bâtiments permettant de préchauffer l'air neuf.L'originalité du travail repose sur le fait que ce mur est équipé de matériau à changement de phase (MCP).Celui-ci a pour rôle de stocker l'énergie solaire captée en façade puis de la déstocker en préchauffant l'air neuf de ventilation. Notre étude est constituée de deux grandes phases, une phase expérimentale et une phase numérique.La phase expérimentale consiste à effectuer des essais en laboratoire, afin de connaître le comportement du système étudié sous sollicitations thermiques. Ces essais ont été réalisés grâce à un prototype du mur, instrumenté et installé entre deux cellules avec des conditions climatiques contrôlées.Le but de la phase numérique est de mettre en place un modèle ID permettant de simuler le comportement thermique du mur et en particulier celui du MCP. Ce modèle a été validé en comparant les résultats numériques avec ceux obtenus expérimentalement.Pour modéliser le changement de phase nous avons utilisé les paramètres thermophysiques du matériau obtenu par caractérisation expérimentale réalisée dans notre laboratoire. Nous avons ensuite utilisé la méthode de la capacité variable pour simuler le comportement de la paroi stockeuse du mur. Dans la dernière partie du travail numérique le modèle a été utilisé afin de montrer l'influence de quelques paramètres permettant d'optimiser les gains énergétiques.La simulation dynamique du système a été effectuée grâce au logiciel TRNsys, qui nous a permis d'effectuer des bilans énergétiques et d'estimer l'efficacité du système pour des climats variés. / The objective of this thesis is to study an integrated energy system in the building envelope for fresh air preheating. The originality of the work is that the wall is equipped with phase-change material (MCP) packed into briquettes. The main role ofthe wall is to preheat the fresh air (coming from outside) by destocking the solar energy captured in sunny periods.Our study consists of two phases, an experimental phase and a numerical one. The experimental phase involves a series of tests that allow studying the thermal behaviour of the system under thermal stress. These tests were done in laboratory through a prototype of the wall which is instrumented and installed between two airconditioned cells.The purpose of the numerical phase is to develop a one-dimensional model to simulate the thermal behaviour of the wall and especially that of MCP. This model has been validated by comparing numerical results with those obtained experimentally. To model the phase changing we used the thermophysical parameters of the same material obtained by experimental characterization conducted in our laboratory.We used a variable capacity method whose principle is to vary the heat capacity as a function of temperature in order to simulate the phase changing of the wall. The last part of the numerical work is the exploitation of the model, the aim is to determine the optimal configuration of the wall that provides maximum energy savings. Dynamic simulation of the system was performed using the TRNSYS. This one is equipped with weather files which allow carrying out heat balances and the estimation of the system efficiency for various climates.

The identification of polymerized and oxidized alpha-1 antitrypsins (ATs) induced by cigarette smoke as proinflammatory factors in the pathogenesis of emphysema

Chronic Obstructive Pulmonary Disease (COPD) is a chronic inflammatory disease, characterized by progressive and largely irreversible airflow limitation due to alveolar destruction (emphysema), small airway narrowing, and chronic bronchitis. It is one of the leading causes of morbidity and mortality worldwide and in the UK, it may affect approximately 1.5 per cent of the population; and up to one in eight emergency admissions may be due to COPD,corresponding to over one million bed days, with some 24160 people in the UK dying as a result of COPD in 2005 (Burden of Lung Disease 2nd Edition,British Thoracic Society 2006). Most cases of COPD are triggered by chronic inhalation of cigarette smoke.However, some people do not suffer from COPD even if they smoke for many years. COPD cannot be cured, and patients usually live with poor life quality. Treatments include giving up smoking, medication and oxygen therapy. Genetic factors contribute to the development of COPD. In Northern Europe,Z-AT homozygotes (342Glu Lys) develop emphysema in their third or forth decade. One explanation is AT deficiency because they form inactive polymers. However, this cannot explain why bronchoalveolar lavage fluid (BALF) from Z-AT homozygotes with emphysema contains more neutrophils than BALF from individuals with emphysema and normal AT (M-AT). Inhaling pollutants which include smoking (cigarettes, pipes, cigars, etc.) and other fumes such as those found in many industrial work environments probably also plays a role in an individual’s development of COPD. Previously, it has been shown that the polymeric conformer of AT is present in BALF from Z-AT homozygotes and that it is a chemoattractant for neutrophils in vitro (Parmar JS, 2002). These findings have been confirmed by others (Mulgrew AT, 2004). However, it is unknown where the polymers form and if 4 they are chemotactic in vivo. My colleague Dr Carl Atkison† showed that polymers of Z 1-AT are present in the alveolar wall of Z-AT homozygotes with emphysema, which accounts for 20% of the total AT from lung homogenates.These Z-AT individuals also have an excess of neutrophils in the alveolar wall compared with M-AT homozygotes. Furthermore, neutrophils and polymeric AT co-localize in the alveolar wall (Mahadeva R, 2005). To investigate whether there was a direct relationship between polymers of Z-AT and the excess neutrophils, polymers of AT were instilled into the lungs of wild-type mice (Mahadeva R, 2005). This produced a significant increase in neutrophil influx into the lungs compared with instillation of the native protein.Examination of the time course demonstrated that the influx of neutrophils was closely linked to the presence of polymeric AT. The mechanism of neutrophil recruitment in this mouse model was subsequently shown to be a direct chemotactic effect rather than stimulation of IL-8 homologues or other CXC chemokines. Oxidized AT (Ox-AT) promotes release of human monocyte chemoattractant protein-1 (MCP-1) and IL-8 from human lung type epithelial cells (A549) and normal human bronchial epithelial (NHBE) cells. Native, cleaved, polymeric AT and secretory leukoproteinase inhibitor (SLPI) and oxidized conformations of cleaved, polymeric AT and SLPI did not have any significant effect on MCP-1 and IL-8 secretion. These findings were supported by the fact that instillation of Ox-AT into murine lungs resulted in an increase in JE (mouse MCP-1) and increased macrophage numbers in the bronchoalveolar lavage fluid. The effect of Ox-AT was dependent on NF- B and activator protein-1 (AP-1)/JNK. These findings have important implications. They demonstrate that the oxidation of methionines in AT by oxidants released by cigarette smoke or inflammatory cells not only reduces the anti-elastase lung protection, but also converts AT into a proinflammatory stimulus. The release of oxidants by these inflammatory cells oxidizes AT, perpetuating the cycle, potentially contributing to the pathogenesis of COPD. Furthermore, this demonstrates that molecules such as oxidants, anti-proteinases, and chemokines, rather than acting independently, collectively interact to cause emphysema (Li Z, 2009). To investigate the molecular basis for the interaction between Z-AT and Ox-AT associated with cigarette smoking, female mice transgenic for normal (MAT)or Z-AT on CBA background were exposed to cigarette smoke (CS). Transgenic mice for Z-AT developed a significant increase in pulmonary polymers following acute CS exposure. Increased levels of neutrophils in CSZ lungs were tightly correlated with polymer concentrations. Oxidation of human plasma Z-AT by CS or -chlorosuccinimide greatly accelerated polymerization, which could be abrogated by antioxidants. The results showed that cigarette smoke accelerated polymerization of Z-AT by oxidative modification, which in so doing further reduced pulmonary defense and increased neutrophil influx into the lungs. These novel findings provided a molecular explanation for the striking observation of premature emphysema in ZZ homozygote smokers, and raised the prospect of anti-oxidant therapy in ZAT related COPD (Alam S, 2011).

Preventivní a kurativní využití kineziotapu při zatížení palce ruky v ambulantní terapeutické péči / Kinesio taping in treatment and prevention of thumb overload in outpatient physiotherapy

Title : Kinesio taping in treatment and prevention of thumb overload in outpatient physiotherapy Purposes : The main purpose of this thesis was to evaluate a degree of pain decrease in thumb joint after kinesio tape application. The differences in grip strength were concurrently evaluated to objectify previous results. Hand dynamometer was used to support this claim. Methods : Data for the experiment were obtained by use of hand dynamometer measurement and record of a pain degree according to chosen pain scale. Each person was measured right before defined strain and immediately after it (daily work in outpatient therapy according to therapist's working shift). Each therapist was asked to record a degree of actual pain following the instructions during strain. All the results were processed and statistically analyzed. Results : The results show that kinesio tape is successful in decreasing of thumb joint pain. This was confirmed both during and after the strain. Study and test of micro-channel plates used in the dual ion spectrometer of the MMS mission by NASA

The Magnetospheric Multiscale mission led by NASA has been designed to study the micro-physics of Magnetic Reconnection in Earth's magnetosphere by using four identical spacecrafts with instruments with high temporal and spatial resolutions. Among these instruments are the Dual Ion Spectrometers (DIS) engineered to measure the 3D distribution of ion flux in space. The detector assembly of the DIS consists of Micro-Channel Plates (MCP) mounted in Chevron configuration. Centre d'Etude Spatiale des Rayonnements (CESR), Toulouse is responsible for the provision and testing of all fifty MCP pairs for this mission. The goal of the work was to participate in the testing and characterization of the first prototype of the MCPs. It was achieved by understanding the working and characteristics of the MCPs in general and getting familiar with the detector assembly of the DIS i.e. the MCP pair and the detector circuit board in particular. To perform the testing, it was necessary to understand the testing system as well. These topics are described in this report along with the testing procedure and the data analysis. The testing procedure was developed eventually after facing several problems during the testing. MCP pair characteristics like pulse height distributions, gain, resistance and the MCP operating voltages for the mission were determined on analyzing the data. Crosstalk was found in the circuit board of the detector assembly and has also been discussed. / Validerat; 20101217 (root)

Plusieurs solutions existent pour la réalisation de ces objectifs, et les travaux présentés tout au long de cette thèse concernent un composant solaire particulier à la construction externe du bâtiment, appelé cheminée solaire. Cette thèse de doctorat porte sur l'analyse expérimentale et numérique des dispositifs de stockage d'énergie, sous forme de matériaux à changement de phase (PCM), afin d'optimiser les performances de cette technologie solaire. Le but de cette étude est de caractériser l’impact des panneaux Rubitherm RT44 PCM sur une cheminée solaire en laboratoire et in situ afin de permettre une comparaison avec la version classique. De plus, un modèle numérique a été développé et testé dans le but d'obtenir un outil numérique capable de représenter le comportement d'une cheminée solaire. Enfin, une optimisation à deux objectifs du modèle numérique de cheminée solaire intégrée PCM a été réalisée afin de déterminer certains des paramètres optimaux de ce type de technologie afin d’obtenir le flux d’air sortant le plus élevé possible, tout en maintenant une température suffisamment élevée dans la cheminée atteindre la gamme de fusion des PCM. / The current global warming conditions have led nations across the world to commit into energetic sustainability and greenhouse gas emission reduction. Being the third greatest energetic consumer, the building represents a major key towards energy efficiency and global temperature stabilization. Several solutions exist for the accomplishment of these goals, and the works presented throughout this dissertation concerns a particular external building solar-driven component known as solar chimney. This PhD thesis focuses on the experimental and numerical analysis of energy storage devices, in the form of Phase Changing Materials (PCMs), for the optimisation of the performance of this solar technology. The aim of this study is to characterize the impact of Rubitherm RT44 PCM panels on a solar chimney under laboratory and in-situ conditions to carry out a comparison against the classic version. Additionally, a numerical model was developed and tested in the interest of obtaining a numerical tool capable of representing the behaviour of a solar chimney. Finally a bi-objective optimization of the PCM integrated solar chimney numerical model was carried out in order to determine some of the optimal parameters of this type of technology to obtain the highest exiting air flow, all while maintaining a high enough temperature across the chimney to reach the fusion range of the PCMs.

Efeito de tratamentos pré e pós-colheita na qualidade de rosas de corte / Effect of pre and postharvest treatments in quality of cut rose

O objetivo do trabalho foi o desenvolvimento de tecnologias para melhoria da qualidade e vida de vaso em rosas (Rosa hybrida.) Vega. Os tratamentos précolheita foram realizados com aplicação de silício (0, 200, 400 e 800 mg L-1) por meio de fertirrigação via solo e via foliar. Foram realizadas seis aplicações semanais de silicato de potássio (12% Si) em rosas Vega, distribuídas em blocos casualisados com quatro repetições, e seis avaliações pós-colheita (dias 0, 2, 4, 6, 8 e 10). Após a colheita, foram avaliados: comprimento de haste, diâmetro de haste e de pedúnculo, comprimento de botão e a massa das hastes. As flores foram armazenadas em vasos com água deionizada a 20±1°C e 65±5% UR, sendo avaliadas: variação da massa fresca, conteúdo relativo de água (CRA) de pétalas e folhas, atividade de peroxidase, abertura floral, curvatura do pedúnculo, turgescência e escurecimento de pétalas, e coloração de pétalas. A aplicação de 400 mg L-1 de silício proporcionou maior comprimento de haste, maior CRA e cromaticidade das pétalas e menor atividade de peroxidase. A aplicação foliar também apresentou maior comprimento de botão, CRA de folhas, e luminosidade de pétalas, e a aplicação via solo também apresentou maior diâmetro de haste, maior massa inicial, menor variação de massa fresca e maior abertura floral, indicando que a aplicação de silício pode contribuir para melhorar a qualidade de rosas Vega. Os tratamentos pós-colheita foram realizados com aplicações de 0,1 mM de salicilato de metila (MeSa), 1 L L-1 de 1-metilciclopropeno (1-MCP), e 1 mM de ácido salicílico (AS). Após a aplicação dos tratamentos por 12 horas, as flores foram mantidas em vasos com água deionizada, armazenadas a 25±1°C e 65±5% U R. O delineamento experimental foi inteiramente casualisado, com quatro repetições por tratamento e seis avaliações (dias 0, 2, 4, 6, 8 e 10). Os tratamentos com AS e 1-MCP reduziram a atividade respiratória, produção de etileno, perda de massa e a curvatura do pedúnculo, tendo permitido maior manutenção do conteúdo relativo de água das pétalas, maior abertura floral, maior luminosidade das pétalas, e maior atividade de peroxidase. O AS também proporcionou melhor manutenção da turgescência, do conteúdo relativo de água das folhas, e menor escurecimento das pétalas. / The objective of this study was the development of technologies to increase the quality and vase life of cut roses (Rosa hybrida.) Vega. The pre-harvest treatments were the application of silicon (0, 200, 400 and 800 mg L-1) via foliar spray and root feeding, in quality of cut roses. Six weekly applications of potassium silicate (12% Si) were performed on roses Vega, distributed in a randomized block design with four replicates per treatment, and six evaluations (days 0, 2, 4, 6, 8 and 10). After harvest, the stem length, stem diameter, peduncle diameter, bud length and mass of the stems were evaluated. The flowers were kept in vases with deionized water, stored at 20±1°C and 65±5% RH, with evaluati ons of loss of fresh weight, leaf and petal relative water content (RWC), peroxidase activity, flower opening, bent neck, turgidity, petal darkening, and color of petals. The application of 400 mg L-1 of silicon provided increase in stem length, in petal RWC, petal chromaticity and reduced peroxidase activity. The application via foliar spray also increased bud length, the leaf RWC and the petal luminosity. The root feeding also increased the stem diameter, the floral opening, the initial mass, and reduced the fresh weight loss, indicating that the application of silicon can improve the quality of roses Vega. The postharvest treatments were the application of 0,1 mM of methyl salicylate (MeSa), 1 L L-1 of 1-methylcyclopropene (1-MCP), and 1 mM of salicylic acid (SA), in postharvest quality conservation of Vega roses. After the application of treatments for 12 hours, the flowers were kept in vases with deionized water, stored at 25±1°C and 65±5% RH. The experimental design was completely randomized, with four replications and six evaluations (days 0, 2, 4, 6, 8 and 10). The treatments with SA and 1-MCP reduced respiratory activity, ethylene production, loss of fresh weight and bent neck; and enable better maintenance of petal relative water content, largest flower opening and higher petal luminosity. SA also provided better maintenance of turgidity and leaf relative water content and less petal darkening.

Contribution to the experimental and numerical characterization of phase-change materials : consideration of convection, supercooling, and soluble impurities / Contribution à la caractérisation expérimentale et numérique des matériaux à changement de phase : Prise en compte de la convection, de la surfusion et d'impuretés solubles

Au cours des deux dernières décennies, le contexte économique a changé de manière significative en raison de la hausse des prix de l'énergie. Le bâtiment étant devenu le principal secteur consommateur d'énergie, la réduction de celle-ci est devenue un objectif économique, sociétal et environnemental. Ce sujet mobilise de nombreux travaux de recherche. Les Matériaux à Changement de Phase (MCP) représentent une solution innovante qui pourrait contribuer à améliorer la performance énergétique des bâtiments. Ils sont principalement utilisés pour la régulation de température, et leur forte capacité de stockage est un moyen de réduire la consommation d'énergie. Notre étude vise à caractériser, via une approche expérimentale et numérique, le comportement d'un PCM (l’Octadécane). Pour cela, nous avons développé et mis en œuvre un modèle numérique qui corrobore les résultats expérimentaux, et ainsi améliore la prédiction de la performance du MCP considéré.Dans ce travail, notre principale préoccupation est de mettre en évidence les erreurs ou simplifications présentes dans le modèle numérique traditionnel pouvant entraîner un écart global par rapport au comportement réel du MCP. Ces différences conduisent à une estimation erronée des temps de fusion et de la quantité d'énergie stockée. L'amélioration significative de notre modèle est la prise en compte de la convection naturelle, de la surfusion, et l'utilisation des courbes réelles d'enthalpie du MCP considéré. La relation température-enthalpie réelle tient compte de la présence d'une fraction d'impuretés solubles dans le matériau. L’originalité de ce travail est de traiter ces phénomènes physiques via la méthode de Boltzmann réseau (connue sous l'acronyme LBM) avec des fonctions de distribution doubles couplée à une formulation enthalpique. Une telle approche permet de passer outre la non-linéarité des équations régissant l'écoulement et le transfert de chaleur. Sa simplicité de mise en œuvre et son caractère local permettent d'affiner le modèle. Ainsi, on peut couvrir les problèmes de changement de phase, y compris ceux pouvant avoir lieu dans des matrices poreuses ou fibreuses. Ce dernier point a été couvert dans cette thèse.Enfin, il s'est avéré que l'approche numérique adoptée ici pour traiter les problèmes de changement de phase corrobore à la fois nos résultats expérimentaux et ceux disponibles dans la littérature. / Over the past two decades, the economic context has changed significantly due to the rise in energy prices. The building sector has become the main consumer of energy. Thereby, reducing the latter is now an economic, societal and environmental necessity. Accordingly, this topic mobilizes many researches. Phase Change Materials (PCMs) represent an innovative solution, which could improve buildings' energy performance. They are primarily used for temperature regulation, and their high storage capacity can reduce energy consumption.Our study aims at characterizing, via a complementary approach of experimental and numerical simulation, the behavior of a PCM (n-Octadecane). For this, we have developed and implemented a numerical model that corroborates the experimental results, and hence improves the prediction of the PCM performance.In this work, our main concern is to highlight the common errors or simplifications taken in the traditional numerical model, which can result in an overall discrepancy compared to the actual behavior of PCMs. Those discrepancies lead to wrong estimation of the fusion times and amount of energy stored. The major improvement of our model is the consideration of the natural convection, the supercooling, and the use of real enthalpy curves of the considered PCM. The actual temperature-enthalpy relationship takes into account the presence of a fraction of soluble impurities in the material. The originality of this work is to handle these physical phenomena via a lattice Boltzmann method (known by the acronym LBM), which leans on double distribution functions and coupled with the enthalpy formulation. Such an approach overcomes the non-linearity in the governing equations of fluid flow and heat transfer. Its simplicity and local character allow adding complexity to the model. Thereby, one can cover up the phase change problems, including those, which may occur in heterogeneous matrices. This last point has been also covered in this thesis.Finally, it turned out that the approach implemented here for phase change problems supports both, our experimental results and those available in the literature.
