Este trabalho apresenta, numa primeira etapa, uma avaliação sobre as vidas da Cooperativa Tritícola Carazinho Ltda. e da Cooperativa Tritícola Mista Alto Jacuí Ltda. Na etapa seguinte, procede a uma comparação entre ambas cooperativas, objetivando indicar quais foram os fatores que colaboraram para o fracasso da primeira e para o sucesso da segunda. Para tanto, realiza uma busca das referências bibliográficas e documentais sobre os fundamentos do Cooperativismo e do contexto econômico, partindo de meados dos anos 1950 até os dias atuais. Além disso, há a reconstrução da história dessas cooperativas através das atas das assembléias gerais ordinárias e extraordinárias e de entrevistas com dirigentes, ex-dirigentes e produtores. Esse processo visa demonstrar a consonância das decisões tomadas pelos dirigentes e produtores de ambas cooperativas com as diversas transformações econômicas operadas no país, a partir de meados de 1950. Por fim, ocorre uma análise de alguns indicadores contábeis e procedimentos administrativos e negociais apresentados por estas cooperativas.

My dissertation deals with the issue of quitting smoking and keeping permanent abstinence in relation to the most significant motivation factors. My particular objective was to specify the most important motivation factors as well as to confirm what effect the price of cigarettes has on giving up smoking with the 25-50 year old generation of South Bohemian geographic classification. A quantitative research was carried out via an internet questionnaire. It contained 14 questions concerning identification, treatment, motivation and finances. 63 respondents filled in the questionnaire.

Which product attributes lead consumers to prefer startups’ products over established companies’ products in the specialty product category? : A study of the electric vehicle market in Germany

Purpose – The purpose of this study is to investigate which product attributes lead consumers to prefer startups’ products over established companies’ products in the specialty product category. The research object is the electric vehicle market in Germany and the product attributes that are researched are alignable attributes, non-alignable attributes and the price. Design/Methodology/Approach – Quantitative data was collected via an internet questionnaire by means of the non-probability sampling techniques convenience and snowball sampling. The data of 408 members of the German Generation Y (age 17 – 38) was used to test six hypotheses by applying ANOVA and multiple linear regression analysis. Findings – It was found that established companies possess a pioneer advantage in comparison to startups for the specialty product ‘electric vehicle’. This advantage can be overcome when startups differentiate their electric vehicles by implementing superior alignable attributes, a valuable non-alignable attribute or a lower price. Superior alignable attributes had the strongest positive influence on consumers’ preferences towards the startup’s electric vehicle, followed by the lower price and a valuable non-alignable attribute. Limitations/Implications – This study is limited to investigating consumers’ preferences without focusing on the reasons behind the preferences. Further, the category of specialty products is represented by only one example, namely the electric vehicle industry. Practical Implications – Startups can benefit from the results by adopting differentiation strategies that were found to be successful in overcoming pioneer advantage. Originality/Value – This study contributes to pioneer advantage literature by researching how startups can successfully overcome pioneer advantage in the specialty product category.

Desempenho de defensas rodoviárias sujeitas ao impacto de automóveis. / Performance of road restraint systems subjected to automobiles impact.

As defensas rodoviárias têm a finalidade de redirecionar com segurança um veículo fora de controle de uma colisão frontal ou trajetória mais perigosa. Secundariamente, a absorção de energia através da deformação do sistema de contenção ou outro método de dissipação da energia de impacto é ressaltado como um benefício em prol da segurança dos ocupantes. Considerando as premissas anteriores e os índices de severidade ao ocupante ASI, THIV e PHD, os desempenhos de uma defensa metálica e outras defensas de concreto são então classificados. Além disso, investiga-se o mecanismo de falha das conexões parafusadas e a sua influência no resultado do impacto de um automóvel leve contra uma defensa metálica. Importância é dada à modelagem do comportamento do material da defensa metálica sob baixas e altas taxas de deformação. Dessa forma, a necessidade de avaliar as propriedades dinâmicas do material motivou o projeto e construção de uma Barra de Hopkinson para testes de tração e compressão até 5000s-1. Adicionalmente, um modelo analítico simplificado da defensa metálica sob ação de uma força estática foi equacionado para validar um modelo em elementos finitos. Um estudo paramétrico da falha da conexão parafusada foi levantado através de diversas simulações estáticas e também do impacto de um veículo leve em uma defensa metálica de acordo com a norma EN 1317. Os resultados das simulações de impacto utilizando um modelo de veículo leve e diferentes sistemas de contenção foram comparados em função dos índices de severidade ao ocupante, indicando que a defensa metálica é mais segura do que uma defensa de concreto com perfil New Jersey ou vertical. / The main function of a road guardrail is to redirect an out of control vehicle avoiding a frontal collision or a dangerous veering off the road trajectory. Secondarily, the energy absorption measured by barrier deformation or any other energy dissipation method is beneficial to the car occupant safety. Considering the previous tasks and some severity occupant index like ASI, THIV and PHD, the performance of a guardrail and other concrete barriers are classified. Besides, it is investigated the fail mechanism of bolted connections and its influence on the guardrail impact result of an automobile. Special attention was taken into the material behavior modeling under low and high strain rates. Thereby, the need of accessing dynamic material properties motivated the design and manufacture of a Hopkinson Bar set-up for tension and compression tests up to 5000s-1. In addition, a simplified analytical model of a guardrail subjected to a static force was settled in order to validate a guardrail finite element model. A parametric study of the bolted connection failure was performed through several impact simulations of a light car model and a guardrail according to EN 1317 standard. The impact simulation results using a light vehicle car model and different barriers types were compared through the occupant severity index showing that the guardrail is a safer system than concrete barrier with New Jersey or vertical profile.

Structural studies of the Staphylococcus aureus ribosome / Etudes structurales du ribosome de Staphylococcus aureus

Le ribosome est une machinerie cellulaire importante impliquée dans la synthèse protéique de toute cellule vivante. Par conséquent, le ribosome est l'une des principales cibles des antibiotiques naturels, qui sont capables de tuer les cellules bactériennes en bloquant la synthèse protéique. Toutefois, certaines bactéries sont résistantes à ces antibiotiques en raison de petites modifications au niveau de leurs ribosomes. Entre autres, Staphylococcus aureus (S. aureus) est un agent pathogène responsable de nombreuses infections graves chez l’Homme. Les structures cristallines d'antibiotiques en complexe avec des ribosomes de bactéries non-résistantes, non-pathogènes, Gram négatives ont fourni un aperçu sans précédent des mécanismes d'action de ces antibiotiques. Cependant, aucune structure de ribosome de bactéries pathogènes, hautement résistantes, Gram positives telles que S. aureus n’a encore été identifiée.Dans cette étude, nous présentons la première structure de ribosome de S. aureus à haute résolution (3.9 Å) résolue par cryo-microscopie électronique (cryo-ME). Nous mettons en évidence plusieurs caractéristiques de l'organisation des ribosomes spécifiques des bactéries Gram-positives. Nous décrivons également le protocole de purification et de cristallisation du ribosome de S. aureus pour de futures études de cryo-ME et de cristallographie aux rayons X.Tous les résultats obtenus dans ces travaux, faciliteront la description à l’échelle atomique du ribosome de S. aureus et ses complexes fonctionnels’ ’dans un futur proche. La combinaison des méthodes de cristallographie aux rayons X et de cryo-ME aidera à atteindre cet objectif. Les résultats obtenus serviront de base pour le développement de nouveaux composés contre la bactérie pathogène et extrêmement résistante qu’est S. aureus. / The ribosome is a large cellular machinery that performs the protein synthesis in every living cell. Therefore, the ribosome is one of the major targets of naturally produced antibiotics, which can kill bacterial cells by blocking protein synthesis. However, some bacteria are resistant to these antibiotics due to small modifications of their ribosomes. Among them, Staphylococcus aureus (S. aureus) is a severe pathogen that causes numerous infections in humans. The crystal structures of complexes of antibiotics with ribosomes from Gram-negative non-pathogenic non-resistant bacteria have provided unparalleled insight into mechanisms of antibiotics action. However, the structure of the ribosome from Gram-positive pathogenic and highly resistant bacteria such as S. aureus was still unidentified.In this study we present the first high resolution structure of the ribosome from S. aureus solved at 3.9 Å by cryo-electron microscopy (cryo-EM). We demonstrate several features of the ribosome organization which are unique for Gram-positive bacteria. We also describe the protocol of purification and crystallization of S. aureus ribosome for future cryo-EM and X-ray crystallography studies.All the results obtained in this work will help to describe S. aureus ribosome and its functional complexes at the atomic level in the nearest future. The combination of X-ray crystallography and cryo-EM methods will help to achieve this aim. The obtained results will provide a foundation for the development of new compounds against the pathogenic and extremely resistant bacteria S. aureus.

Simulation to Build Empathy in Adolescents With Autism Spectrum Disorders: a Video Modeling Study

Since a deficit in empathy is not only characteristic among individuals with autism spectrum disorder (ASD) but categorically used in defining ASD, it is of utmost importance to explore educational avenues to build prosocial skills among this group. This study sought to explore the primary research question: What impact does the implementation of an empathy-focused video modeling intervention have on frequency of empathic behaviour among adolescents with ASD? The secondary research questions examined were: In what ways does employing a video modeling simulation intervention using the Model Me Kids Friendship program impact the ability of adolescents with ASD to demonstrate empathic behaviour? How do adolescents with ASD express or speak about their empathic behaviour following participation in a simulation intervention using Model Me Kids Friendship? How do the Educational Resource Facilitators (teaching assistants) perceive the same individuals’ empathic behaviour following participation in a simulation intervention using Model Me Kids Friendship (MMF)? This mixed methods study explores 1 particular video modeling simulation program as a focused approach to building empathic behaviour among adolescents with ASD. The theoretical framework presented blends theory of mind, simulation theory, and psychological theories of empathic behaviour including the inherent motor, cognitive, and emotional components. Individuals with ASD may not learn empathic behaviours solely through observation as typically developing children do, but findings suggest that through video simulation, practice may, in fact, lead to increased empathic behaviour. The quantitative findings were not significant but did show increase in motor empathy behaviour ratings among intervention group participants. Support for video modeling as a vehicle to teach empathic behaviour was provided by qualitative data collected over the course of 4 months contextualizing specific examples of empathic behaviour exhibited by participating teens with ASD. There is a link made between high levels of systematizing among teens on the spectrum (the drive to analyze and build a system) and video modeling as a means to foster empathic behaviour, thereby supporting an increased use of video simulation strategies to teach social skills among this group.

The aim of the essay is to problematize the main characters' conception of identity in André Aciman's novel Call Me by Your Name. Due to Elio's inability to see himself and Oliver as continuously constructing and shifting bodies, the essay claims that the end of their relationship as they knew it during their summer in Italy was inevitable. By employing a critical discourse analysis and deconstructive approach, the way the characters position themselves during and after that summer is examined and critiqued. It is found that what contributes to the end of their relationship is their struggle with the multitude of aspects of their identities that are considered to be flaws or insufficiencies. Therefore, certain interests and aspects of their lives are kept a secret. In consequence, this secrecy hinders a full understanding of the other and reveals their idealization as wishful thinking, rather than reality.
