Cette thèse présente un système bioélectronique prometteur, l’Hynet. Ce Réseau Hybride (vivant-artificiel) est conçu pour l’étude du comportement à long terme des cellules électrogénératrices, comme les neurones et les cellules betas, en deux aspects : l’individuel et en réseau. Il est basé sur une boucle fermée et sur la communication en temps réel entre la culture cellulaire et une unité artificielle (Matériel, Logiciel). Le premier Hynet utilise des Matrices d’électrodes (MEA) commerciales qui limitent les performances spatiotemporelles du Hynet. Une nouvelle Matrice d’électrodes intelligente (iMEA) est développée. Ce nouveau circuit intégré, analogique et mixte, fournit une interface à forte densité, à forte échelle et adaptative avec la culture. Le nouveau système améliore le traitement des données en temps réel et une acquisition faible bruit du signal extracellulaire. / This thesis presents a promising new bioelectronics system, the Hynet. The Hynet is a Hybrid (living-artificial) Network, developed to study the long-term behavior of electrogenic cells (such as Neurons or Beta-cells), both individually and in a network. It is based on real-time closed-loop communication between a cell culture (bioware) and an artificial processing unit (hardware and software). In the first version of our Hynet, we use commercial Multielectrode Arrays (MEA) that limits its spatiotemporal performances. A new Intelligent Multielectrode Array (iMEA) is therefore developed. This new analog/mixed integrated circuit provides a large-scale, high-density, and adaptive interface with the Bioware, which improves the real-time data processing and the low-noise acquisition of the extracellular signal. / Esta dissertação de doutorado apresenta um sistema bioeletrônico auspicioso, o Hynet. Esta Rede Híbrida (viva e artificial), é concebida para o estudo do comportamento à longo prazo de células eletrogeneradoras (como neurônios ou células beta), em dois aspectos : individual e em redes. Ele é baseado na comunicação bidirecional, em laço fechado e em tempo real entre uma cultura celular (Bioware) e uma unidade artificial (Hardware ou Software). Um primeiro Hynet é apresentado, mas o uso de Matrizes de Eletrodos (MEA) comerciais limita a performance do sistema. Finalmente, uma nova Matriz de Eletrodos Inteligente (iMEA) é desenvolvida. BECCS (Bioenergy Carbon Capture and Storage) is an important part of measures to achieve zero net emissions globally by 2050, as the technology can create carbon sinks. However, the technology is very energy-intensive and expensive, and affects the existing systems at implementation. The purpose of this study is to investigate the possibility of implementing BECCS at Karlstad Energy's biofuel-fired CHP-plant, Heden 3. The goal is, by simulation in CHEMCAD, to generate energy consumption key figures for two different separation technologies (MEA-MonoEthanolAmine and HPC-HotPotassiumCarbonate) with 90 % separation rate in three different operating cases. In addition, the systemic impact on Heden 3 will be determined by analyzing three different scenarios. In the first scenario fuel consumption is kept unchanged and steam to the carbon capture system is extracted before the turbine. In the second scenario fuel supply increases to meet the district heating needs of the existing system and steam to the carbon capture system is extracted before the turbine. In the third scenario fuel supply is kept unchanged and steam is extracted from the turbine. In addition, the study investigates various transport options for storage of carbon dioxide and finally calculate the total carbon sink Karlstad Energy can contribute to. The results show that production of electricity is reduced by 65-87 % after implementation of MEA and 151-238 % for HPC in the first scenario. Without heat utilization in the carbon capture system, heat production is reduced by 66-86 % with MEA and 54-76% for HPC. In the second scenario, a fuel supply increase by 134 % is required to meet the needs, which corresponds to more than twice the boiler capacity and results in a reduced production of electricity by 247 %. In the third scenario, production of electricity is reduced by 104 % at maximum load with HPC. The HPC system has high-quality heat to utilize, probably enough to meet the district heating needs without increasing the boiler power. But heat optimization opportunities need to be further explored in order to be able to express something to a greater extent. The MEA process does not offer the same opportunities for heat utilization. As the CHP-plant have heat as the main product, HPC would be a more suitable alternative despite the high load on the electricity production. The performance of the carbon dioxide plant seems to vary between different operating cases and it can be concluded that the variation is related to the flue gas composition rather than being load dependent. Transport of carbon dioxide by train has the lowest carbon dioxide emissions and requires the least number of cargoes for transport from Karlstad to storage in Norway. However, this is not relevant at present because of the lack of rail connection to the plant. Total carbon sink is approximately 127 000 tonnes per year if the boiler capacity is assumed to be unchanged. / BECCS (Bioenergy Carbon Capture and Storage) är en viktig del av åtgärder i målet om att nå nollnetto utsläpp år 2050 globalt, då tekniken kan skapa kolsänkor. Tekniken är dock mycket energikrävande och dyr, och påverkar de befintliga systemen vid implementering. Syftet med den här studien är att undersöka möjligheten att implementera BECCS på Karlstad Energis biobränsleeldade kraftvärmeverk, Heden 3. Målet är att, genom simulering i CHEMCAD, ta fram förbrukningsnyckeltal för två olika avskiljningstekniker (MEA-MonoEtanolAmin och HPC-HotPotassiumCarbonate) med 90 % avskiljningsgrad vid tre olika driftfall. Dessutom ska systempåverkan på Heden 3 fastställas genom analys av tre olika scenarier. I första scenariot hålls bränsleförbrukningen oförändrad och ånga till koldioxidavskiljningssystemet tappas av innan turbinen. I det andra scenariot ökar bränsletillförseln för att tillgodose fjärrvärmebehovet i det befintliga systemet och ånga till koldioxidavskiljningssystemet tappas av innan turbinen. I det tredje scenariot hålls bränsletillförseln oförändrad och ånga extraheras från turbinen. Därtill undersöks i studien olika transportmöjligheter till lagringsplats av koldioxiden och slutligen beräknas den totala kolsänkan Karlstad Energi kan bidra med. Resultaten visar att elproduktionen i det första scenariot reduceras med 65-87 % för MEA och för HPC 151-238 %. Utan värmeutnyttjande från koldioxidavskiljningssystemen reduceras värmeproduktionen med 66-86 % med MEA och 54-76 % med HPC. I det andra scenariot krävs att bränsletillförseln ökar med 134 % för att tillgodose behoven vilket motsvarar mer än dubbla panneffekten och innebär en reducerad elproduktion på 247 %. I det tredje scenariot reduceras elproduktionen med 104 % vid maximal last med HPC.  I HPC-systemet finns högvärdig värme att utnyttja, sannolikt tillräckligt mycket för att kunna uppfylla fjärrvärmebehovet utan att öka panneffekten. Men värmeoptimeringsmöjligheter behöver undersökas ytterligare för att kunna uttrycka något i större omfattning. I MEA-processen finns inte samma möjligheter till värmeutnyttjande. Eftersom kraftvärmeverket har värme som främsta produkt skulle således HPC vara ett lämpligare alternativ trots den höga belastningen på elproduktionen. Koldioxidanläggningens prestanda förefaller variera mellan olika driftfall och med en enklare undersökning kunde slutsatsen dras att variationen har ett samband med rökgassammansättningen snarare än att det är ett lastberoende. Transport av koldioxid med tåg har lägst koldioxidutsläpp och kräver minst antal laster för transport från Karlstad till lagring i Norge. Detta är dock inte aktuellt i dagsläget på grund av avsaknaden av räls in till verket. Den totala kolsänkan är cirka 127 000 ton per år om pannan antas köras oförändrat.

Tiefdruckverfahren zur Herstellung von Katalysatorschichten für (PEM) Brennstoffzellen

Diese Dissertation befasst sich mit der industrienahen Herstellung von Katalysatorschichten für Polymer-Elektrolyt-Membran-Brennstoffzellen mit Hilfe des Tiefdrucks als Fertigungsverfahren. Um die Anforderungen an die Katalysatorschicht hinsichtlich der Schichtdicke zu erreichen, wird ein Linienraster für den Tiefdruck entwickelt. Das patentierte und verifizierte Designkonzept des Linienrasters ermöglicht es, trotz Tinten mit geringem Feststoffgehalt hohe Trockenschichtdicken zu erzeugen. Aufgrund des verwendeten Tiefdruckrasters sind Optimierungsschritte an der Fertigungsanlage notwendig, um eine hohe Schichtqualität zu erreichen. Schließlich werden kontinuierlich und industrienah Katalysatorschichten gefertigt, die als Membran-Elektroden-Einheit in einer Polymer-Elektrolyt-Membran-Brennstoffzelle erfolgreich eingesetzt werden. / This work presents an industrial close manufacturing process of active electrodes for Polymer Electrolyte Fuel Cells utilizing an adapted gravure printing process. To meet the requirements of the electrodes regarding the layer thickness (weight) and quality a novel line screen with maximized dipping volume for gravure printing was developed and investigated. A design rule for this kind of screens was realized and verified by a successful manufacturing of electrodes with different dried layer thicknesses. Due to the rough structure and the high dipping volumes of these line screens an adaption and optimization of the machinery and the whole process was necessary to achieve high quality electrodes. Finally, it is shown that it is possible to manufacture continuiously in an industrial close roll-to-roll process platinum loaded electrodes, working successful as cathode in a Membran-Electrode-Assembly.

A comparative study of natural contamination with aflatoxins and fumonisins in selected food commodities from Botswana and Zimbabwe

Mycotoxins are toxic secondary metabolites produced by filamentous fungi. Aflatoxins and fumonisins are among the most toxic mycotoxins. They are a significant risk factor for a cocktail of chronic health conditions including cancer of the liver, oesophagus and kidney, teratogenicity, neural tube defects, interference with lipid metabolism, a weakened immune system and a negative impact on micronutrient absorption in both man and animals. This study compared natural contamination of peanuts, peanut butter and sorghum from Gaborone, Botswana and Bulawayo, Zimbabwe with aflatoxins and fumonisins. In total 34 peanut samples, 34 sorghum samples and 11 peanut butter samples were collected randomly from retail shops and informal markets in the two cities. Fungal contamination was determined using standard mycology methods. Aflatoxin and fumonisin contamination was determined using HPLC-FLD. A. flavus/parasiticus species were detected in 66% and 100% of randomly analysed peanut samples from Bulawayo and Gaborone respectively and 27% (3/11) of peanut butter samples from Bulawayo. 67% of randomly analysed sorghum samples from Bulawayo showed A. flavus/parasiticus and Fusarium species contamination while none of the randomly analysed sorghum samples from Gaborone showed any fungal contamination. Furthermore aflatoxins were not detected in any of the sorghum samples; however 61% (11/18) of the Bulawayo sorghum samples showed fumonisin contamination (Range: 8 – 187 ng/g). Three of the peanut samples from Bulawayo were contaminated with aflatoxins (range: 6.6 – 622 ng/g) and no aflatoxins were detected in Gaborone peanuts. All 11 peanut butter samples from Bulawayo were contaminated with aflatoxins (Mean: 73.5 ng/g, Range: 6.8-250 ng/g) and AFB1 was the most prevalent. These preliminary results indicate that peanut butter and peanuts from Bulawayo are contaminated with high levels of aflatoxins. Stricter policing of regulations should be implemented to ensure compliance by manufacturers and public health interventions implemented in vulnerable communities. / Life & Consumer Sciences / M. Sc. Sistemas de PEMFC são capazes de gerar energia elétrica com alta eficiência e baixa ou nenhuma emissão de poluentes, porém questões de custo e durabilidade impedem sua ampla comercialização. Nesse trabalho foi desenvolvido um MEA com eletrocatalisadores à base de paládio. Foram sintetizados e caracterizados eletrocatalisadores Pd/C, Pt/C e Ligas PdPt/C com diferentes razões entre metais e carbono. Foi realizado um estudo da razão entre ionômero de Nafion e eletrocatalisador para formação de triplas fases reacionais de máximos desempenhos, criado um modelo matemático para transpor esse ajuste para eletrocatalisadores com diferentes razões entre metal e suporte, considerando os aspectos volumétricos da camada catalisadora, e então realizado um estudo da espessura da camada catalisadora. Para as caracterizações foram utilizadas as técnicas de Difração de Raios-X, Microscopias Eletrônicas de Transmissão e de Varredura, Energia Dispersiva de Raios-X, Picnometria a Gás, Porosimetria por Intrusão de Mercúrio, Adsorção de Gás, segundo as equações de BET e BJH, Análise Termo Gravimétrica e feitas as determinações de diâmetros de partículas, de áreas de superfície específica e de parâmetros de rede. Todos os eletrocatalisadores foram usados no preparo de MEAs que foram avaliados em célula unitária de 5 cm2 entre 25 e 100 °C a 1 atm; e a melhor composição foi avaliada também a 3 atm. No estudo dos metais para as reações, visando reduzir a platina aplicada aos eletrodos, sem perdas de desempenho, foram selecionados Pd/C para ânodos e PdPt/C 1:1 para cátodos. A estrutura de MEA desenvolvida utilizou 0,25 mgPt.cm-2 e resultou em densidades de potência de até 550 mW.cm-2 e potências de até 2,2 kWe por grama de platina. A estimativa realizada mostrou que houve uma redução de até 64,5 % nos custos em relação à estrutura de MEA previamente conhecida. Em função da temperatura e pressão de operação foram obtidos valores a partir de R$ 3.540,73 para o preparo de MEAs para cada quilowatt instalado. Com base em estudos recentes, concluiu-se que o custo do MEA desenvolvido é compatível às aplicações estacionárias de PEMFC. / PEMFC systems are capable of generating electricity with high efficiency and low or no emissions, but durability and cost issues prevent its large commercialization. A study of the ratio between catalyst and Nafion Ionomer for formation of high performance triple-phase reaction was carried out, a mathematical model to implement this adjustment to catalysts with different relations between metal and support taking into account the volumetric aspects of the catalyst layer was developed and then a study of the catalyst layer thickness was performed. X-ray diffraction, Transmission and Scanning Electron Microscopy, X-ray Energy Dispersive, Gas Pycnometry, Mercury Intrusion Porosimetry, Gas adsorption according to the BET and BJH equations, and Thermo Gravimetric Analysis techniques were used for characterization and particle size, specific surface areas and lattice parameters determinations were also carried out. All catalysts were used on MEAs preparation and evaluated in 5 cm2 single cell from 25 to 100 °C at 1 atm and the best composition was also evaluated at 3 atm. In the study of metals for reactions, to reduce the platinum applied to the electrodes without performance losses, Pd/C and PdPt/C 1:1 were selected for anodes and cathodes, respectively. The developed MEA structure used 0,25 mgPt.cm-2, showing power densities up to 550 mW.cm-2 and power of 2.2 kWnet per gram of platinum. The estimated costs showed that there was a reduction of up to 64.5 %, compared to the MEA structures previously known. Depending on the temperature and operating pressure, values from US$ 1,475.30 to prepare MEAs for each installed kilowatt were obtained. Taking into account recent studies, it was concluded that the cost of the developed MEA is compatible with PEMFC stationary application.

An Analysis of Wireless High-speed Data Services for Cellular CDMA Systems

The interest in the development of wireless high-speed data services is in response to the strong market demand for high-speed wireless Internet access. Current standards aim at delivering a peak data rate greater than 2Mbps on the forward link. Since data services and voice services are fundamentally different, new concepts were introduced in the design of the forward data channel. In addition, methods of evaluating the performance of a cellular CDMA system have to be revisited. This thesis proposes a method which can be used to find the forward link peak and average data rates, throughput and coverage of a cellular CDMA system which is capable of delivering high-speed wireless data. A summary of changes in design philosophy and recent advances in technologies which enable high-speed wireless data delivery are presented. The proposed method takes into account major aspects commonly found in the forward data channel and applies the generalized Shannon capacity formula for multi-element antenna (MEA) systems. The analysis focuses on the physical layer and is flexible enough to be adapted to various propagation environments, antenna configurations, multicode allocations, user distributions and cell site configurations. Sample numerical results for various multicode allocations are shown using a system model with two-tier interfering cells with one transmit antenna and two receive antennas operating under a frequency selective slow fading channel with propagation environment described by the Recommendation ITU-R M. 1225 indoor office, outdoor to indoor and pedestrian and vehicular test environments. Different transmit / receive antenna configurations and multicode allocations and their impact on the average data rate is also explored.

In this work, the problem of detecting neuronal spikes or action potentials (AP) in noisy recordings from a Microelectrode Array (MEA) is investigated. In particular, the spike detection algorithms should be less complex and with low computational complexity so as to be amenable for real time applications. The use of the MEA is that it allows collection of extracellular signals from either a single unit or multiple (45) units within a small area. The noisy MEA recordings then undergo basic filtering, digitization and are presented to a computer for further processing. The challenge lies in using this data for detection of spikes from neuronal firings and extracting spatiotemporal patterns from the spike train which may allow control of a robotic limb or other neuroprosthetic device directly from the brain. The aim is to understand the spiking action of the neurons, and use this knowledge to devise efficient algorithms for Brain Machine Interfaces (BMIs). An effective BMI will require a realtime, computationally efficient implementation which can be carried out on a DSP board or FPGA system. The aim is to devise algorithms which can detect spikes and underlying spatio-temporal correlations having computational and time complexities to make a real time implementation feasible on a specialized DSP chip or an FPGA device. The time-frequency localization, multiresolution representation and analysis properties of wavelets make them suitable for analysing sharp transients and spikes in signals and distinguish them from noise resembling a transient or the spike. Three algorithms for the detection of spikes in low SNR MEA neuronal recordings are proposed: 1. A wavelet denoising method based on the Discrete Wavelet Transform (DWT) to suppress the noise power in the MEA signal or improve the SNR followed by standard thresholding techniques to detect the spikes from the denoised signal. 2. Directly thresholding the coefficients of the Stationary (Undecimated) Wavelet Transform (SWT) to detect the spikes. 3. Thresholding the output of a Teager Energy Operator (TEO) applied to the signal on the discrete wavelet decomposed signal resulting in a multiresolution TEO framework. The performance of the proposed three wavelet based algorithms in terms of the accuracy of spike detection, percentage of false positives and the computational complexity for different types of wavelet families in the presence of colored AR(5) (autoregressive model with order 5) and additive white Gaussian noise (AWGN) is evaluated. The performance is further evaluated for the wavelet family chosen under different levels of SNR in the presence of the colored AR(5) and AWGN noise. Chapter 1 gives an introduction to the concept behind Brain Machine Interfaces (BMIs), an overview of their history, the current state-of-the-art and the trends for the future. It also describes the working of the Microelectrode Arrays (MEAs). The generation of a spike in a neuron, the proposed mechanism behind it and its modeling as an electrical circuit based on the Hodgkin-Huxley model is described. An overview of some of the algorithms that have been suggested for spike detection purposes whether in MEA recordings or Electroencephalographic (EEG) signals is given. Chapter 2 describes in brief the underlying ideas that lead us to the Wavelet Transform paradigm. An introduction to the Fourier Transform, the Short Time Fourier Transform (STFT) and the Time-Frequency Uncertainty Principle is provided. This is followed by a brief description of the Continuous Wavelet Transform and the Multiresolution Analysis (MRA) property of wavelets. The Discrete Wavelet Transform (DWT) and its filter bank implementation are described next. It is proposed to apply the wavelet denoising algorithm pioneered by Donoho, to first denoise the MEA recordings followed by standard thresholding technique for spike detection. Chapter 3 deals with the use of the Stationary or Undecimated Wavelet Transform (SWT) for spike detection. It brings out the differences between the DWT and the SWT. A brief discussion of the analysis of non-stationary time series using the SWT is presented. An algorithm for spike detection based on directly thresholding the SWT coefficients without any need for reconstructing the denoised signal followed by thresholding technique as in the first method is presented. In chapter 4 a spike detection method based on multiresolution Teager Energy Operator is discussed. The Teager Energy Operator (TEO) picks up localized spikes in signal energy and thus is directly used for spike detection in many applications including R wave detection in ECG and various (alpha, beta) rhythms in EEG. Some basic properties of the TEO are discussed followed by the need for a multiresolution approach to TEO and the methods existing in literature. The wavelet decomposition and the subsampled signal involved at each level naturally lends it to a multiresolution TEO framework at the same time significantly reducing the computational complexity due the subsampled signal at each level. A wavelet-TEO algorithm for spike detection with similar accuracies as the previous two algorithms is proposed. The method proposed here differs significantly from that in literature since wavelets are used instead of time domain processing. Chapter 5 describes the method of evaluation of the three algorithms proposed in the previous chapters. The spike templates are obtained from MEA recordings, resampled and normalized for use in spike trains simulated as Poisson processes. The noise is modeled as colored autoregressive (AR) of order 5, i.e AR(5), as well as Additive White Gaussian Noise (AWGN). The noise in most human and animal MEA recordings conforms to the autoregressive model with orders of around 5. The AWGN Noise model is used in most spike detection methods in the literature. The performance of the proposed three wavelet based algorithms is measured in terms of the accuracy of spike detection, percentage of false positives and the computational complexity for different types of wavelet families. The optimal wavelet for this purpose is then chosen from the wavelet family which gives the best results. Also, optimal levels of decomposition and threshold factors are chosen while maintaining a balance between accuracy and false positives. The algorithms are then tested for performance under different levels of SNR with the noise modeled as AR(5) or AWGN. The proposed wavelet based algorithms exhibit a detection accuracy of approximately 90% at a low SNR of 2.35 dB with the false positives below 5%. This constitutes a significant improvement over the results in existing literature which claim an accuracy of 80% with false positives of nearly 10%. As the SNR increases, the detection accuracy increases to close to 100% and the false alarm rate falls to 0. Chapter 6 summarizes the work. A comparison is made between the three proposed algorithms in terms of detection accuracy and false positives. Directions in which future work may be carried out are suggested.
