Studium antioxidačnej kapacity

Híc, Pavel January 2012 (has links)
This thesis dealing with the study of antioxidant capacity was developed between 2006-2011 at the Department of Post-Harvest Technology of Horticultural Products, Faculty of Horticulture, Mendel University in Brno. The aim of this thesis was the evaluation of photo-chemiluminescence method and its optimization for determining the antioxidant capacity by the Bux Photochem device. The assessment of impact of selected substances and treatment of experimental material on measured values of antioxidant capacity. The other aim of this thesis was the monitoring of development of antioxidant capacity of the grapes during ripening period and during the process of alcoholic fermentation. The basic principles of measuring the antioxidant capacity by photo-chemiluminescence method are provided by compliance of the methodology with an emphasis on the same treatment of the device. The work showed that the dilution of the observed samples affects the antioxidant capacity measured by Photochem Bux PCL. The solutions of the reference wine and the antioxidant rutin showed higher antioxidant capacity at the samples with higher dilution level. At the parallel extraction by non-polar and polar extractants and the subsequent summation of the measured values of TAC was shown that the resulting value was higher than at the consecutive extractions. Therefore the summation of the values is not appropriate for parallel extractions of antioxidants obtained from the extractants with various polarities. During the extractions 75% methanol was evaluated as the best extractant of antioxidants present in grapes. It is necessary to homogenize the sample before extraction. All the extraction operations must be finished in the shortest time possible. It was shown that all the extraction operations apart from used extractant and re-extraction have demonstrable effects on degradation of antioxidants extracted from the samples. Therefore it is necessary to describe all the extraction procedures perfectly. Very suitable extraction procedure is a re-extraction. There is a big difference in concentrations of extracted substances and extractant occurring during re-extraction, so the extraction is very effective. It was observed that one of the factors causing the degradation of antioxidant compounds during the extraction procedures is the presence of atmospheric oxygen. Sulphites present in wine do not affect the value of antioxidant capacity measured by photo-chemiluminescence method. Conversely, FRAP method has a positive response to the presence of sulphites in wine, so the values measured by this method are affected by the sulphites content. The impact of alcohol content on antioxidant capacity measured by photo-chemiluminescence method was not demonstrated. The monitoring of the antioxidant capacity development in grapes during the ripening period showed that the white varieties have no characteristic development. The blue varieties showed that during the ripening period the values of antioxidant capacity were gradually increasing up to three times the first measurement (46 days before the harvest). The antioxidant capacity of grape juice was increasing during the alcoholic fermentation only if the juice was left in the contact with skins. There was no increase of antioxidant capacity observed after the pressing, although the fermentation process was highly reductive and took place in closed containers.

Modelování parametrů spalovacích motorů

Dobrovolný, Milan January 2015 (has links)
Master thesis deals with the link between the internal and external parameters of the combustion engine. Its point is not to exactly define this links but only check if this links exist and what is their character. First part of the thesis is dedicated to present state of the mixture preparation systems of the petrol engines. Here are described the ways of the lambda regulation and its effect of the specific fuel consumption. Further are described types of injection systems and at the end is described the way for the determination of the ignition advance. Second part is focused on the chosen mathematics-statistics methods which are interesting and useful for the solving this problem. Last part is paid to the verification of the existence the links of the operating parameters and attempt of their definition in numeric way using the Maple software and chosen mathematics-statistics methods.

Možnosti využití NIR spektroskopie pro stanovení vybraných parametrů u brambor

Balarinová, Alena January 2015 (has links)
Current trends in analytical chemistry is the introduction of instrumental methods that are quick and economical. This process is obviously not avoid potato field of chemistry, using optical methods - NIR spectroscopy. This thesis therefore focuses on the analysis of basic nutrition and most of all the determined parameters of potato tubers (dry matter, starch, N-substances and reducing sugars) using laboratory reference methods and their subsequent transfer to this new analytical method by creating appropriate calibration models. It is also part of the work is devoted to the statistical comparison of both analytical methods in order to determine whether these methods can replace each other. The measurement has been found that some parameters NIR spectroscopy can be replaced, while in others the conversion is not suitable. Specifically parameters starch, N-substances or dry matter had correlation coefficients (R) 0.901; 0.806 and 0.977, meaning satisfactory calibration models. Whereas on parameters carbohydrates, carbohydrates after hydrolysis with the values of correlation coefficients 0.769 and 0.786 is impossible to apply the method of NIR spectroscopy. Models for determining these are not suitable.

Srovnání metod měření zrnitostního složení půd: laserová granulometrie versus pipetovací metoda

Chmelíková, Ludmila January 2015 (has links)
Thesis aims to define determination of particle size distributions loamy and sandy soils measured by two methods of measurement: laser granulometry and pipette method and comparison of measured results. First part of thesis is focused on theoretical comparison of two methods of measurements, predominantly on potential problems connecting with their comparing and suitable setting of device parameters to obtain accurate results. Comparison between laser granulometry and pipette method was carried out for 90 loamy and 30 sandy soil samples. Analyzed samples were measured using a Malvern Mastersizer 2000. Comparison of laser granulometry and pipette methods demonstrated differences of results, which were different for the various samples. Despite the variance of the measured results, was found for the clay fraction the correlation 0, 92 between methods for loamy soils, for sandy soils was found the correlation of 0.90 for the sand fraction.

Závislost vybraných antagonistů na početnosti Lymantria dispar / Dependence of selected antagonists on Lymantria dispar abundance

Jelínková, Zuzana January 2016 (has links)
The aim of this study was to investigate the dependence of selected antagonists to the abundance of Lymantria dispar. The aim of the study was to determine whether these natural enemies can affect the abundance of L. dispar. I have focused on South Moravia, where L. dispar occurs. In 2015 the Turček method reviewed a total of 3,600 trees in seven locations with the historical appearance of outbreaks of gypsy moth. Density egg clutches were extremely low, about 0.019 for each new clutch per tree, which corresponds to the latent state. On the studied locations egg clutches was collected, measured and recorded in notebooks. A load with at least one egg in each area was removed with the top 5 cm of litter and a top layer of mineral soil. The removed soil was put into pots in which were stored late instar caterpillars for the detection of E. maimaiga. The cups were inserted along with the caterpillars in the refrigerator for three days at 15°C without food. Aafter three days the caterpillars were taken away individually and placed on an artificial diet. An inspection was performed and a their mortality was monitored. The presence of E. maimaiga was examined microscopically for 325 dead caterpillars, but no pathogen was detected. In the laboratory, the individual egg clutches were placed upon a Petri dishes with filter paper and the number of eggs counted with a stereo microscope. Subsequently, the eggs were left to hatch and were analyzed for the presence of parasitoids. A total of 1 246 species of egg parasitoids Anastatus japonicus was found in six locations, taking in donations females predominated. Egg parasitism was on average around 5%. Flight activity A. japonicus was quite long and covered the period 24 May 2015 to 28 July 2015. In the absence of E. maimaiga and low egg parasitism at the studied locations we do not expect any strong influence of the studied antagonists on population density and outbreak cycle L. dispar in South Moravia.
