Ecotoxicidade do cobre e possíveis biomarcadores no reservatório Guarapiranga. / Copper ecotoxicity and possible biomarkers at Guarapiranga reservoir.

Yoel Rodríguez Pinto 06 June 2016 (has links)
Copper is an essential ion for plants, animals and for human health. Although the crucial role of Cu in several enzymatic processes, this heavy metal can exert adverse toxicological effects, when present in high concentrations in water. The determination of waters physic chemistry parameters, trophic index, histological alterations, hepatosomatic index, Cu bioaccumulation in livers and gills and oxidative stress enzymes like, CAT, SOD and GPX were done. Our results show that the higher values of Cu concentration were in the dissolved Cu and in the rainy season. The exposition o the fishes to a higher Cu concentrations altered the gills; aneurism, lamellar fusion, basal epithelium lifting. In the liver the alterations were, vacuolization and necrosis among others. The livers were the greatest bioaccumulation organs, and the other hand the hepatosomatic index decreased with higher Cu concentrations. There was no significant difference in the enzyme activities with the exception of CAT that increased with Cu increase, nevertheless in general, the enzyme activities of SOD, CAT and GPx were altered some how after 120 hours of Cu treatment, with water sample from 4 points located at Guarapiranga Reservoir.

In Brazil and around the world, the fish-related industry produces a great amount of waster of high biological value. Due to the lack an adequate use, this residual material is considered a problem from the sanitary, environmental and production efficiency perspectives. With the objective of minimizing environmental problems and increasing fish production efficiency, the elaboration and the physical-chemical and nutritional characterization of the acid silage (AS), biological silage (BS) and enzymatic silage (ES) was performed with the use of trash fish and Nile tilapia (Oreochromis niloticus) processing waste (Experiment 1). The AS, BS, and ES values (dry matter basis) found were, respectively, 25.21; 34.58 and 25.01 g.100g-1 of dry matter, 54.25; 33.00 and 54.50 g.100g-1 of crude protein, 12.45; 12.25 and 12.17 g.100g-1 of lipids; 8.03, 7.33 and 8.58 g.100g-1 of calcium and 4.71; 2.86 and 4.85 g.100g-1 of phosphor. In general, leucin (3.50; 2.41 and 3.31 g.100g-1 to AS, BS and ES, respectively) and lysine (3.33; 2.41 and 3.22 g.100g-1 to AS, BS and ES, respectively) were the essential amino acids (EAA) with the highest concentrations. According to their chemical score, all silages were tryptophan-deficient when compared to the EAA requirements for Nile tilapia according to the NRC (1993). However, considering that amino acids are only a limitation when less than 30% of the minimal requirements for fish in general, these products are not deficient in any EAA. In a second experiment, the silage produced in Experiment 1 was biologically evaluated by the determination of the apparent digestibility coefficient (ADC) for energy, nutrients and amino acids. The ADC values found were: crude protein, 92.01; 89.09 and 93.66%; crude energy, 86.39; 84.53 and 89.09%; dry matter, 82.52; 78.98 and 82.96%; calcium, 81.72; 73.99 and 80.27%; phosphor, 77.86; 79.21 and 81.46% of AS, BS and ES, respectively. The average amino acids ADC was 91.83; 90.76 and 94.61% of AS, BS and ES, respectively. Based on the results of the experiments, we may conclude that all prepared silages are potentially viable for the use in balanced fish diets. It is recommended, the use of AS for the easiness in its elaboration.

Off flavor detection and control in nile tilapia (Oreochromis niloticus) through depuration and smoking.

Denise Oliveira Biato 11 March 2005 (has links)
Currently, economic losses are a reality due to the fish off flavor provoked by the handling used in the culture and by the water quality of the fish tanks, which are not always satisfactory. Microbiological, physical-chemical and sensory evaluations of the fish were carried out in order to detect probable alterations that would compromise the fish quality. The off flavor detection, as well as, the suggestion for depuration before distribution and sale, and the use of the smoking as a way of disguising this problem were the objectives of this research. Microbiological, physical-chemical and biological analyses of the waters in the fish and the depuration ponds were performed to monitor the quality of the fish. The fish– Nile Tilapias (Oreochromis niloticus) from the area of Arthur Nogueira, in the State of São Paulo – was submitted to the depuration (3, 5 and 7 days). The fish submitted to the depuration and the control fish were compared in order to find the best depuration period. No significant differences (p>0.05) for centesimal composition, non-protein nitrogen - NNP, total base volatile nitrogen BNVT, and pH were found among the depuration periods. The depuration process provoked a more intense weight loss in the first three days for depuration. Through the sensory analyses, we could verify that the control fish presented a greater off flavor intensity than the others. The three-day depuration showed a similar statistical efficiency (F Test) than that of the five-day treatment, but differed from that of the 7-day treatment. As for the flavor attribute, the 3, 5, and 7-day treatments did not show a significant difference, statistically. The water in the fish pond presented eutrophization problems and presence of geosmine, whereas the water in the depuration pond was clear and geosmine-free. The fish that did not go through depuration (control) were submitted to smoking. Fresh fish fillets were compared to the smoked ones. As to the centesimal composition, we verified that they were significantly affected (F Test). As to non-protein nitrogen – NNP, total base volatile nitrogen – BNVT and pH, there were no significant changes (p>0.05) either among the treatments. Through sensory analysis, we verified that the fresh fish presented a more intense off flavor than the smoked ones. As to odor and flavor, there was a significant (p>0.05) difference among the treatments. The depuration process was efficient in the elimination of the off-flavor in the tilapia, for, according to our analyses, we could conclude that the ideal period for this elimination was 5 days of depuration, carried out in a pond with clean running water. When compared to the fresh fish, the smoked fish was better accepted by the panelists. The smoking process is a way of disguising the off flavor; it is a simple, inexpensive process, which can be readily adopted by producers a way of adding value to the fish.

THE EFFECT OF CANNABIDIOL (CBD) ON THE PHYSIOLOGY AND IMMUNOLOGY OF NILE TILAPIA (OREOCHROMIS NILOTICUS) IN-VITRO AND IN-VIVO

Asif Mortuza (10700823) 26 April 2021 (has links)
<p>As the human population increases and the demand for aquaculture increases, aquaculturists are coming up with new ways to mitigate stress in fish to increase their production. Cannabidiol (CBD) is an up and coming nutraceutical that may have potential to reduce stress in not only humans but also other vertebrates such as fish. In this project the effect of CBD on the stress physiology and immunology of Nile tilapia was evaluated both <i>in-vitro</i> and <i>in-vivo</i>. In the <i>in-vitro</i> study, spleen cell proliferation was conducted to observe the effect of CBD on fish T and B cells and were compared to mouse T and B cell proliferation. In the <i>in-vivo</i> study, the fish were reared in a recirculating aquaculture system. The effect of CBD on the stress physiology of the fish in short term and long term were evaluated. Based on the short-term acute study, a longer chronic study was designed where tilapia were fed with and without CBD (0.001% of feed weight) and with and without hydrocortisone stress hormone (0.01% of body weight) every day for four weeks. This experiment compared the various growth and feed utilization parameters as well as physiological and immunological parameters such as, plasma cortisol, blood glucose and protein levels, liver and spleen somatic indices (HSI and SSI, respectively), and lysozyme activity of the fish. From our current research, CBD shows potential in stress modulation and in immune modulation. It may have different effects based on the species, whether they need to enhance their immune response or reduce inflammation to be healthy. It also seems to have had different effect on different parts of the immune system. Hematological parameters were not significantly affected by acute stress. CBD did not make any substantial difference in growth. However, in the presence of stress, CBD was able to lower lysozyme activity down to the normal control levels. By administering the proper dosage of CBD on a case by case basis, health benefits can be achieved. Further investigation into the matter may not just be useful in stress mediation in aquatic organisms but may also have implications in human medicine as well.</p>

MEDICINAL BENEFITS OF SEA CUCUMBERS FROM THE WATERS OF THE EASTERN UNITED STATES

Eaint Honey Aung Win (13163001) 27 July 2022 (has links)
<p>Sea cucumbers have been found to contain bioactive compounds such as saponin, fucoidan, frondoside, and glycosides that have pharmacological properties like antitumor, antibacterial, anti-inflammation, and antihyperglycemic activity. Although several species of sea cucumbers have been studied and reared for the food and medicinal industries, not much research has been conducted on the species in the waters of the Eastern United States. In this research, physiological and immunological parameters of coelomic fluid from <em>Cucumaria</em> <em>frondosa</em>, <em>Isostychopus</em> <em>badionotus</em>, and <em>Pentacta</em> <em>pygmaea</em> were compared to find the most promising candidate with these properties and pharmacological benefits. We found that <em>C. frondosa</em> was the species with the best immunological and physiological parameters among the three studied. <em>C. frondosa</em> illustrated that its coelomic fluid contains the highest concentrations of cells and lysozymes that had the highest activity. Using <em>C. frondosa</em>’s tissue extracts and coelomic fluid, the ability of the extracts and coelomic fluid to inhibit murine melanoma cells (B16-F10) and modulate T-lymphocytes <em>in vitro</em> were investigated. Although no significant differences were seen statistically, the experiments illustrated that T-lymphocytes were highly activated at higher concentrations (0.001g/uL-0.0002g/uL) for tissue extracts and at lower concentrations (0.000008g/uL) for coelomic fluid. On the other hand, melanoma cells were inhibited highest at lower concentrations (0.000008g/uL-0.0000016/uL). In addition to these studies, the antibacterial activity of <em>C. frondosa</em> extract was tested on ten pathogenic bacterial species. Antibacterial activity of the <em>C. frondosa</em> extract was not seen in this experiment. However, hemolytic activity by compounds present in <em>C. frondosa</em> extracts was seen in blood agars culturing <em>Streptococcus pneumoniae</em> and <em>Enterococcus faecalis</em> in our experiment. Lastly, an <em>in vivo </em>study was conducted to see if <em>C. frondosa</em> extract can modulate stress in Nile tilapia. In our experiment, we observed that <em>C. frondosa</em> extract was able to enhance the activity of one of the parameters, phagocytic capacity significantly. However, we are not able to conclude that <em>C. frondosa</em> extract was able to mitigate chronic stress from the results obtained. Overall, observing the results from the projects, we cannot conclude that <em>C. frondosa</em> extracts illustrated pharmacological properties. Extensive studies are recommended and required to use <em>C. frondosa</em> extract for medicinal purposes. </p>

The trial aimed to evaluate the apparent strain digestibility coefficients (ADC) of dry matter (DM), gross energy (GE), crude protein (CP), starch, lipids, crude fiber (CF), ashes (AS) and essential and non-essential amino acids (AA) of in natura bean flour (FI) and extruded bean flour (FE) for juvenile Nile tilapia using two digestibility markers: chromium oxide (Cr2O3) or acid insoluble ash (AIA). It was elaborated a pelleted reference diet (RD) containing 35.19% CP and 17.78 MJ / kg GE, the test diets consisted of 70% RD and 30% of FI or FE. Groups of 15 individuals (150.16 ± 20.32 g) were distributed in six digestibility aquariums of 200 L, in a 2 x 2 factorial scheme, two processing methods (FI or FE) and two markers (Cr2O3 or AIA) were evaluated, with two replicates, composing the faeces pool of each diet tested. The highest ADC referring to the interaction (processing x marker) were found for DM, GE, CP, As and essential AA as: arginine (Arg), histidine (His), isoleucine (Ile), leucine (Leu), lysine (Lys), methionine (Met), phenylalanine (Phe), threonine (Thr) and for non-essential AAs: aspartic acid (Asp), cysteine (Cys), glutamic acid (Glu), glycine (Gly), serine (Ser) and tyrosine (Tyr). The CDA of the starch and the essential AA tryptophan (Trp) and valine (Val) was higher in FE compared to FI, different from lipids, which presented better results for FI. For GE and nonessential amino acid alanine (Ala) there was no influence of dietary treatments and markers. Regardless of the processing, Met, His and Arg were the limiting AAs for diets containing bean flours. In conclusion, FE proved to be a good source of energy and nutrients, however, the low levels of Met and Cys should be considered for the dietary requirements of AA containing sulfur. The AIA marker proved to be unreliable for Nile tilapia.

All samples of diets and individuals were held between August and November 2013 in the reservoir cages. Individuals throughout the cultivation, as well as take in the natural food, were feed a control 40 and 32% crude protein for three stages of cultivation. For sampling isotopes dorsal muscle samples were taken from 15 individuals captured, collected food samples 15, periphyton and seston (local water filtration). All samples were dried in an oven at a temperature between 55-60 °C for a period ranging from 36-72 hours, and ground in a mortar and pestle manual to a fine powder. were performed sweeps on material samples filters. The powder samples were stored in tin capsules for conducting the analysis of stable isotopes 15N and 13C for tissue samples and diets which were performed at UC Davis Stable Isotope Laboratory Facility, University of California - USA. The field was monitored the limnological variables with the use of a multisensor. In laboratory Limnologia and Water Quality - LIMNOAQUA, analyzes were performed of growing water, in order to characterize the conditions of abiotic cages. The results of the ratios of C and N isotopes for diets and diets of fish and the contribution rates were analyzed using the R software, version 3.2.2. For data of stable isotopes of carbon and nitrogen, for samples with normal distribution and homogeneous variance was applied one-way ANOVA and Tukey test a posteriori. Where the ANOVA assumptions were not satisfied we used the nonparametric Kruskal-Wallis test and paired Wilcoxon. The values of the isotopic ratios were analyzed individually by period. Regarding the values of the contribution rates of the diets were analyzed together for each period. It was used Mixsiar Mixture Model to determine the relative contribution of diets. Limnological variables did not vary during the study period, only variations were observed for total phosphorus concentrations in the final cultivation stage and chlorophyll-a in the entire study period. Samples of feed showed significant differences in isotopic composition of 13C, and the values of 15N in tissue samples from fish between periods I and II, occurring distinct discriminating rates in diet-tissue interface for both isotopic elements of C and N in all diets. The waste feed supplied contributed to the high availability of natural foods, especially for diet based on seston. The periphyton diet (26.6%) had reduced contribution compared the diets of seston (39.2%) and fish feed (34.3%) for fish reared in Santa Cruz. In this context, it can be concluded that natural diet - seston and periphyton - (65.8%) contributed to the growth of Nile tilapia cultured in cages, changing the paradigm that only provided food is assimilated by the fish, contributing so almost exclusively on the growth of this species / A presente pesquisa investigou a contribuição relativa dos alimentos naturais e dietas comerciais assimilados na nutrição de tilápia do Nilo (Oreochromis niloticus) cultivada em tanques-rede no Reservatório de Santa Cruz-Apodi/RN.

The fingerlings of tilapia (Oreochromis niloticus), commun carp (Cyprinus carpio) and tambaqui (Colossoma macropomum) were submitted to the herbicide roundup in the following concentrations: 0.0 (control); 18,06; 19,10; 20,14; 21,18 and 22,22 mg.L-1, 0.0 (control); 13,89; 14,86; 15,83; 16,81 and 17,78 mg.L-1, and 0.0 (control); 18,06; 19,10; 20,14; 21,18 and 22,22 mg.L-1, respectively, three for 96 hours. The LC50 - 96h for O. niloticus, C. carpio and C. macropomum was 21,63, 15,33 and 20,06 mg.L-1 of the herbicide roundup, respectively. The results show that this herbicide is classified as slightly toxic to the three species. The values of dissolved oxygen, pH and temperature recorded in the aquarium control and aquarium experimental of the three fish species have remained without significant variations during the tests, which reduces the possibility of death caused by sudden variations of these parameters during the 96 hours the experiment. The values of LC50 between different species of fish were observed, noting that the species O.niloticus, C. carpio and C. macropomum showed no expressive differences. The values of environmental risk of Roundup were calculated to obtain more stringent parameters in assessing the dangerousness of those on nontargets. The risk of environmental contamination by Roundup for the Nile tilapia, common carp, and tambaqui are low for the lowest application rate (1 L.ha-1) and depths (1.5 and 2.0 m). The dilution of 100%, the highest recommended dose (5 L.ha-1) and depths (1.5 and 2.0 m) the risk is moderate for the three species.
