Lokality nové rezidenční výstavby v Praze a struktura jejich obyvatel / New Residential Localities in Prague and the Structure of Their Inhabitants

This thesis deals with the analysis of housing construction in Prague in the years 2000-2009, and evaluation of socio-demographic structure of the population in the selected new residential areas. First is with the use of the database of the completed dwellings in the respective period described the deployment of new housing stock according to the character of the building in various concentric zones of the city, and on this basis the most important areas of new residential construction are identified. In the main empirical part is through the analysis of migration data at five selected case sites studied the relationship between socio- demographic structure of population and the selected/preferred types of housing. The analysis of statistical data is further supplemented by the data collected through field investigation.

We will discuss here the issue of manipulation in the digital era of photojournalism. At first we will describe the relation between photography and its referent inculding its ability to represent the reality. Depicting the contrast between analogue and digital photography we will point out the independent position of digital photography from its referent, which is creating better possibilities for manipulation and decreasing the credibility of photography. Then we will analyze the text The Dusk of Photojournalism? that is reflecting these perspective and offering the solution increasing the position of the relation between photography and its referent. By contrast we will analyze the text called Photojournalism is dead, long live photojournalism. Why digitization can increase the credibility of journalistic photography? which considers this perspective as obsolete and offers us the new perspective of the relation between photography and its viewers, that could increase the credibility of photography in photojournalism. According to these two major perspectives we will point out whether it is possible to consider this option of photography - viewers relation as a relevant alternative. This thesis deals with archaeological evidence of medieval pottery products which were discovered during the emergency archaeological excavation in the area of former Jiřího z Poděbrad barracks on Náměstí Republiky square in Prague. This area is nowadays called Palladium shopping centre. The first part of the thesis deals with real findings. These are construction debris of burning kilns and historical evolution of its construction. The second part of the thesis compares results of excavation with historical datas. The last part of the thesis evaluates the extraordinarily huge file of ceramic artefacts from waste areas, especially defective pieces which complement the picture of medieval pottery production.

Karel Janko, Ph.D. ABSTRACT: Subject of this thesis is to test several hypotheses about the evolution of asexual reproduction in model group of fish family Cobitis and its mutual competition among sexual and asexual forms, which touches one of the oldest unresolved issues of biology. Specifically, the work deals with the accumulation of non-synonymous mutations, which accelerated accumulation in the genome of clonal lineages theoretically leads to increased extinction compared with sexually reproducing populations (so-called. The theory of Muller's ratchet and Kondrashov's hatchet). This thesis is based on a normalized cDNA sequencing data from oocytes and liver tissue, which has served as a base matrix (generated based on non-normalized cDNA data) for transcriptome sequencing (RNAseq). The rapid antidepressant effect of ketamine changed the direction of the research of potential antidepressants and its effect was also evaluated in this thesis. However, the main focus of this thesis is a new methodological approach to the research of depressive disorder. The main interest lies with the evaluation of automated monitoring of behaviour in this research. The first aim of this thesis was to evaluate the antidepressant effect of ketamine in the forced swimming test using software enabling automated monitoring of behaviour. The second aim was to meassure the change in phosphorylated Mammalian target of rapamycin (mTOR), using Enzyme-Linked ImmunoSorbent Assay (ELISA). The last, but the most important aim of this thesis was to implement the utilization of Phenotyper boxes in the automated behavioural evaluation of the olfactory bulbectomy model of depressive disorder and also evaluate the effect of ketamine in this model. Ketamine did not show an antidepressant effect in forced swimming test, however this observation could be influenced by chosen dose and mouse strain. Sensitivity of the test to chosen experimental protocol shows insufficient validity of this test. Observed change in level of phosphorylated mTOR corresponded with the behavioural results. Cystic fibrosis (CF) is genetically conditioned, autosomal recessive disease that occurs in the European population with a prevalence of about 1:2500 - 1:1800. In this disease we observe a mutation of the CTFR gene with subsequent fault in chloride channels. Such afflicted individuals usually suffer from chronic respiratory problems, pancreatic insufficiency, high concentration of chloride ions in sweat and obstructive azoospermia. Genetic testing of CFTR gene is indicated in individuals who meet the CF clinical picture and a positive sweat test (increased concentration of chlorides in the sweat). Genetic testing of the CFTR gene is usually done by using commercial kits detecting the most common mutations of the CFTR gene in the Czech Republic. If the testing results are negative, it is further performed an MLPA method that captures the larger deletions and duplications of gene, eventually a sequencing of all exons is. Despite the well-established algorithm of the testing, some patients suffering from symptoms of CF are left without genetic findings. Thanks to development of next generation sequencing, it is possible to make the diagnosis of CF more effective and uncover the variants that were not captured by previous methods.

German Foreign Fighters in the Yugoslav Wars

The aim of the thesis is to identify those German citizens, who were fighting in the Yugoslav wars, determine their background, actions on the ground and post-war trajectories, as well as suggest probable motivations for joining the combat in the way they did. The thesis raises the question, how these German war volunteers can be best described and if they are somehow specific within their subgroup of predominantly Western anti-Yugoslav foreign fighters. I argue that Nir Arielli, one of the few scholars dealing with the role of Westerners in the conflict, falls short by qualifying them merely as "meaning seekers" and thus overlooks the multitude of political connections and references, first and foremost among the Germans. I will offer a critique by pointing out differences and nuances, especially in origin- based motives, ideological underpinning and perspectives on the conflict. In doing so, I will raise the question of what we do know about the political situation in both Germany and Croatia in the early 1990s, and how each of that might that have facilitated decisions to join combat abroad. In the last few years new parties were rising in the post-communist bloc rapidly. Moreover, many of them were successful in the parliamentary elections. Party systems were slowly stabilizing and now they are threatened by new influences. Success of new parties is changing current politics in the central and eastern Europe. These parties are connected not only by the date of foundation but also by the way they act against current establishment. Despite their different strategies, level of success and their creation, their quick unexpected success is interesting. What is the structure of the electorate? This work is focused on identifying the structure of electorate of the new parties in central and eastern Europe. It is divided into two parts. First one, summarizes the theoretical concepts concerning the study of the new parties and clearly identifies what kind of new parties will be considered in this work. For this designation are used theories by Paul Lucardie, Allan Sikk a Krystyna Litton. Second part is quantitative research, based on advanced statistical methods. Specialized supervisor: Prof. Dr. Antje Gohla Title of diploma thesis: Establishment of screening methods to find new regulators of the activity of phosphoglycolate phosphatase This work deals with the siRNA-based genomic screening for the modification of phosphoglycolate phosphatase (PGP) activity. 235 proteins were affected by transient transfection of siRNAs in vitro. Each siRNA was used in duplicates and the control was carried out by two control siRNAs. After downregulation of protein by siRNA PGP activity was evaluated, whether any modifications of PGP activity have occurred. PGP was the main research target. The main goal of this study before the screening was to set up a method, to create a reliable protocol. The whole study was 96 plate well. It was necessary to find the right conditions to measure PGP activity in two cell types (HEK AD 293 and Hep G2). Subsequently, optimal conditions were set up to influence expression of the protein.
