Nové přístupy pro voltametrické stanovení tumorových biomarkerů a antidot v moči / New approaches for voltammetric determination of tumour biomarkers and antidotes in urine

This Ph. D. thesis presents new methods for the determination of selected clinically relevant electrochemically active compounds. The first part deals with development of determination of tumour biomarkers homovanillic acid (HVA) and vanillylmandelic acid (VMA) in human urine with the use of hollow-fibre based liquid-phase microextraction (HF-LPME) and differential pulse voltammetry (DPV) at cathodically pre-treated boron doped diamond electrode (BDDE). Optimum conditions for HF-LPME-DPV of HVA and VMA were as follows: butyl benzoate as supported liquid membrane formed on porous polypropylene hollow-fibre, 0.1 mol L−1 HCl as donor phase and 30 min extraction time. Optimum acceptor phases were 0.1 mol L−1 phosphate buffer of pH 6 with ionic strength set to 0.55 mol L−1 for HVAand 0.1 mol L−1 NaOH for VMA, respectively. HF-LPME-DPV concentration dependencies for HVA and VMAwere linear in the range from 0.4 to 100 µmol L−1 and 0.5 to 100 µmol L−1 . Limits of quantification (LOQ)/detection (LOD) were 1.2/0.4 µmol L−1 for HVA and 1.7/0.5 µmol L−1 for VMA, respectively. This Diploma thesis deals with problematics of new enterprises and factors, which are critical for them and for their success or failure in their early years of entrepreneurship (5 years). The main aim of this diploma thesis is to identify these factors and suggest recommendation, which will lead to better ability of surviving and increases their probability of surviving. For research in this thesis will be used data of the small and medium sized companies (100). These data are collect randomly within electrical and electronic engineering industry. Needed companies were founded in websites ebd.cz and merk.cz. Basic data were gained from basic financial statements of companies, which their posted on website justice.cz. These financial statements are balance sheet and Profit and Loss statement. The data were processed using software power BI, which is using in business intelligence. The second aim of this diploma thesis is also to verify selected bankruptcy and solvency models on chosen data.

The aim of the task was to design a building situated in South Moravia near the Nové Mlýny dam with a view of Pálava hills. The house is designed with respect to the environment as extremely efficient building. Part of the ecological concept is also water management and purification. The primary function of this object is an environmental education aimed at primary schools and its students. There are two classrooms available, both of them designed for 27 children, and a lecture hall for 66 students. There is an accommodation for 60 persons including dining room arranged in the object. Also there is room for cultural events in the area, a separate building that provides refreshment and bicycle or boat rental.

Genom enterovirů z dětské stolice: kombinace next-generation a klasického Sangerova sekvenování / Enterovirus genomes in stool: a combination of the next generation and Sanger sequencing

This diploma thesis deals with a development of a strategy for data evaluation generated by next-generation sequencing. Using bioinformatics tools such as Galaxy, Velvet and Enterovirus genotyping tool new aproach of data processing was optimized. There were 22 samples analyzed which of 10 were grown on cell culture. Remaining 12 were obtained from real stool samples. All samples were taken from children at the highest genetic risk of type 1 diabetes. All of them were enterovirus positive. Enteroviruses and their following infections have been suspecting to be involved in ehiology of type 1 diabetes for a long time. That's a disease resulting to an absolut insulin deficiency due to autoimmune destruction of pancreatic beta cells. Genetic components seems to be relatively well defined (the HLA, INS, STLA4, PTPN22, CTLA4, IFIH1 and numerous other genes), the environmental part of the etiology remains obscured. We were able to assemble 22 genomes de novo. However, there were numerous gaps among the particular contigs. For the first nine samples these gaps were complemented by Sanger sequencing. There is a rise of new phenomenon in drug use in the last few years. There has been increase in the incidence of new synthetic drugs, or the so-called "legal highs" and their use in the general population became more frequent. Legal highs are substances that mimic the effects of illegal drugs, but they are not controlled by the United Nations Single Convention on Narcotic Drugs of 1961 or the UN Convention on Psychotropic Substances of 1971. These substances can produce comparable risks and harms simmilar to the illegal drugs (EMCDDA, 2009c). The aim of research conducted for the purpose of this thesis is to reveal how the risks associated with the use of these products are percieved by their users, whether their legal status has any influence on the perception of their safety and how do the users minimize risks that may be associated with the use of these substances Qualitative research carried out for the needs of graduate work used data obtained by two methods. The first method was a content analysis of online discussion forums of new synthetic drugs users. 16 online discussion forums with a total number of 832 posts were analyzed. In the introduction of theoretical part of diploma thesis is tabularly summed up the systém of birds classification. Other parts are focused not only on general issues and commonly used themes in school textbooks, popular literature and educational media, but also to highlight specific features, characteristic only for the birds. These themes are historical and evolutional questions, cover morphology, the effect of the senses, body structury and function of organs. In detail is analyzed the issue of body composition and skeletal system of birds. In the final sections are mentioned specialities in the behavior and communication between birds and their habits. The practical part includes a SWOT analysis of available textbooks for the four-year gymnasium with regard to the chapters and information about the animal class of birds. The data analyzed in the textbook can only be considered as basic with need to extend and complement by the teacher.
