This thesis describes factors which influence the qualitative status of new planting of trees in the urban environment. The work is focused in detail on the most common mistakes and deficiencies in the care of new planting of trees and other factors which negatively influence the resulting quality of new planting of trees in the city. The evaluation of 2 075 new tree plantings in the city of Zlín was focused both on the quality of the individual trees and on the evaluation of the factors which influence this condition. The evaluation of the data obtained has shown that the most important factor directly affecting the growth of trees is the quality of finishing and development care and care maintenance of trees. Finding real damage to new plantings in the urban environment can help prevent them.

Spain currently represents one of the most popular tourist destinations in the world. Tourism is its important economic sector that is strongly reflected in the Spanish economy. Therefore an apparent interest could be noticed in maintain Spain competitive in the field of tourism not only in Mediterranean destinations but Europe-wide or world-wide as well. To understand the current state of tourism in this country it is neccessary to result from historical context and understand the current problems and their causes well by thus. Spanish competent institutions do realise this and new products and forms of tourism are being conswquently expanded now. Another impulse is a change of motivation of participants, which could be designated as being different in the 21st century. Consequently the traditional forms of tourism are insufficient to meet their needs. Especially the mainland of Spain is diverse and the great conditions for creating new forms of tourism ? cultural, rural, sports, business ? could be found there. (in English): The diploma thesis follows up on the prior attempts to introduce and make available a unique historical source called the inspection protocol of the Theresian Cadastre for Prague towns from 1725 to 1726. This thesis focuses on the New Town, but in some cases it also compares the obtained data with the Old Town and Lesser Town. The theoretical part discusses a whole range of questions that can be answered based on the New Town inspection protocol. The questions include the total number of houses or their division by taxation, house prices as well as occupation and income of homeowners. The houses exterior is evaluated based on the building condition, the material used and the number of storeys. It is possible to study the size of areas that are not built on thanks to an accurate register of garden size. Moreover, this part also concentrates on beer brewing and consumption in New Town pubs. The small scope of the theoretical part is compensated by a content-rich appendix (database), which is in fact an edition of the source in a nontraditional form. Understanding the communication of new media can contribute to understanding the processes that take place in cyberspace. The aim of this thesis is to decipher the forms of propaganda that operate in it. This work can contribute to the understanding of several forms of current social events, such as public relations, influencing public opinion or political struggle, which have significantly moved into cyberspace. After 2010, substances that have similar effects as opiate drugs and stimulant drugs started to appear in the Czech Republic as well as in other European countries and around the world, however they are not controlled by any legislation because of their "newness". The aim of this study is to evaluate whether under the current circumstances this is a problem that should be solved politically, including the assessment of its extent. The current solutions to this problem focused on its regulation shall be assessed on the basis of the results of this analysis. The thesis also aims to identify different perspectives of the drugs issue experts and to understand their involvement in the process of making the drug policy of the Czech Republic concerning these substances. The first part describes the current situation in the field of new psychoactive substances in the Czech Republic using a qualitative method of the documentary content analysis, and introduces the legislative approaches currently used at both national and international level. The research part employs the qualitative research methods (semi- structured interviews with experts in the field of drugs problems). This thesis deals with the success of the two newly formed parties ANO 2011 and Úsvit přímé demokracie Tomia Okamury in the elections to the House of Commons in the Czech Republic in 2013. These two parties gained a substantial percentage of votes and became part of the government despite the fact, that they were established shortly before the elections. In the theoretical part of this thesis, the formation and success of new parties abroad and in the Czech Republic is analysed. The main focus are the voters voting these parties, so there is an extended part dealing with voting behavior in the Czech Republic. Secondary analysis is the research method used in this thesis; its advantages are discussed in the methodological part. Secondary analysis of multiple high quality data sets is used to compare the voters of the new parties and the voters of other established parties. It turns out there are significant differences between these two groups in the areas of sociodemography, political views and experiences and even outlooks on life. The trend of new parties, which started already in the 2010 elections, is obviously continuing and it seems that it is even broadening.

So why are we building vertical farms in sheet metal halls and using only the potential of racks stacked on top of each other? Let's use the potential of this technology and the great adaptability of plants to the new environment and create a new food source operating locally and in continuous operation. Not only would this benefit from the availability of local and foreign food regardless of the season or the changing climate, but the changes it would bring to the economic gap would have a major impact on reducing intercontinental and intracontinental traffic. This would reduce carbon emissions and reduce agricultural areas that could replace forests that are more environmentally friendly.

Rare diseases represent a heterogeneous group of more than ~7000 different diseases, affecting 3,5-5,9% of the global population. Most rare diseases are genetic, but causal genes are known only in some of them. Many patients with rare diseases remain without a diagnosis, which is crucial for genetic counseling, prevention, and treatment. With the development of new methods of genome analysis, decreasing cost of sequencing, and increasing knowledge of the human genome, a new concept for identifying disease-causing genes was established. It is based on comparing the patient's genetic variability with the genetic variability of the general population. This dissertation describes next-generation sequencing technologies (NGS), bioinformatic analysis of acquired data and their applications in the elucidation of molecular underpinnings of rare genetic diseases. These procedures have led to the identification and characterization of causal genes and gene mutations in autosomal dominant tubulointerstitial kidney disease (SEC61A1, MUC1), autosomal dominant neuronal ceroid lipofuscinosis (CLN6, DNAJC5), neurodegenerative disease of unknown etiology (VPS15), Acadian variant of Fanconi syndrome (NDUFAF6) and spinal muscular atrophy (SMN1). Polycystic kidney disease is a severe genetic disease occurring in both adult and pediatric patients. The basic characteristic of this disease is the development and progressive enlargement of renal cysts gradually replacing functional kidney tissue. This leads to renal failure in many patients. However, renal cysts may also occur in a number of other diseases, including multisystem syndromes. This complicates differential diagnosis in some patients. In our study, we first focused on the diagnosis and characterization of genotypic-phenotypic relationships in patients with polycystic disease arising in childhood, later we extended our study to adult patients and patients with unclear clinical diagnosis. At the same time, we expanded the portfolio of analyzed disorders to a number of diseases in which the phenotype of polycystic kidneys may occur, and noncystic diseases as well. During our project, massive parallel sequencing was used to analyze 149 patients - 128 with cystic and 21 with noncystic clinically diagnosed nephropathies. At the same time, the findings were verified by Sanger sequencing in 176 relatives of our probands. Mutation detection reached 59% in cystic patients, and 43% in non-cystic patients, respectively.
