Segmentace vrstev sítnice ve snímcích optické koherentní tomografie / Intra-retinal layer segmentation in optical coherence tomography images

This work describes optical coherence tomography, it's principle and methods for automatic segmentation intra-retinal layers in OCT images . There is proposed algorithm for boundary detection of two intra retinal layers, RFNL and RPE layer based on thresholding and second algorithm for detection of seven layers, which is based on graph search algorithm. In summary is compared accuracy against manualy segmented layers and there is also proposed solution for problematic parts.

The aim of this work is to introduce the principle of scanning in optical microscopy. This thesis deals with problem of parameters of display process and their influence on the results and measurements of these parameters. In detail there is analyzed the depth of field of the optical system and there are established some possibilities of theoretical and experimental determination. Part of this thesis is the realization of the measurement work place and program providing experimental depth of field based on different measurement methods. The conclusion is devoted to the practical part, which has the task to monitor settings and optical systems and their impact on depth of field.

Mazání náhrad kyčelního kloubu / Lubrication of hip joint replacements

This thesis deals with the experimental study of lubrication of hip joint replacements. The influence of the synovial fluid composition and the head material to the lubricating film thickness were asssesed by using a pendulum hip joint simulator. Both of these effects were assesed under static and dynamic loading conditions. The experimental results showed that the movement of the head against the cup is unnecessary for the separation of the rubbing surfaces by a layer of adsorbed proteins. Composition of the synovial fluid then significantly influences the thickness of the lubricating film. Two opposite extremes have been observed during tests with two model synovial fluids. The first fluid formed a very strong lubricating film. The second sample almost prevent adsorption of proteins and consequently formed only very thin lubricating film. The influence of the material is noticeable when comparing the results obtained with a metal head and ceramic heads. Metal head formes under the same conditions thicker lubricating film. This evidence may be partly influenced by different diametrical clearances, which were in the case of ceramic heads higher.

The aim of this thesis is to formulate the design of optical access network in designated area. The design is focused on the physical network layer and it covers optical fibers, cable protectors and links, optical hubs, racks and active components. The thesis also docu- ments network architecture, the configuration of active components and provides results from monitoring of selected network segments. The thesis consists of the theoretical part and practical part. In the theoretical part, there is an introduction to optical networks, their configurations (FTTH, FTTB, FTTC, FTTN), relevant networking standards and technologies. The practical part of the thesis covers design proposal of a network located in Slavičín. The design reuses infrastructure and technologies of existing network. The proposed design follows FTTH architecture which has benefits of low maintenance cost and it also fulfills GPON standard.

This master thesis deals with determination of temperature field in a real circuit breaker. This issue is solved by analysis of measured emission spectra at selected points of the circuit breaker. Spectral line ratios and Boltzmann plot are used for calculation of temperature. These methods are based on elemental analysis of plasma. The main result is finding significant changing of chemical composition of plasma beyond determination of temperature field.

Taking into account dynamic aspects of behavior of studied molecules in solution via conformational averaging therefore seems to be crucial for spectral interpretation. The first of studied models, histidine, plays an important role in metallo-enzymatic reactions and peptide folding, due to its imidazole ring. ROA spectra of His at different pH, His complexed with Cu2+ and dipeptides His- Gly and Gly-His were recorded on the spectrometer built at the Institute of Physics of the Charles University as a first step of the subsequent study. The second studied system, a cyclic hexapeptide c-(Phe-D-Pro-Gly-Arg-Gly-Asp), serves as a convenient model for β- hairpin and anti-parallel β-sheet. It was previously studied by means of VCD and IR. The thesis deals with physico-chemical properties of the MADS box, binding domain of transcription factors, which are important for the formation of complexes with the DNA regulatory segment bearing the CArG box. The study was performed also on model oligopeptides, selected segments of the MADS box and their analogues with a point mutation. A wide range of spectroscopic techniques was employed, namely absorption, circular dichroism, fluorescence and Raman spectroscopies. Advanced approaches including multivariate methods were used for data processing. The three tyrosines of the MADS box located in amino-acid vicinities of different charge and hydrophobicity, were used as intrinsic spectroscopic probes. The obtained characteristics of the MADS box and its segments structural arrangement, flexibility and acid-base equilibria are the main results of the work.

Multielementární chemická analýza popílků ze zpracování druhotných surovin olova / Multielement analysis of flue gas residues from secondary lead smelting

Multielement analysis of flue gas residues from secondary lead smelting was tested using seven decomposition methods on four different flue gas residues and certified reference materials (CRM) CPB-2 (Pb concentrate, CCRMP). The studied samples contained both silicates and also high contents of phases with Pb, primarily Pb chlorides and sulphates. Solutions prepared by various decomposition methods were analysed using ICP OES (Al, As, Ba, Be, Bi, Ca, Cd, Co, Cr, Cu, Fe, K, Li, Mg, Mn, Mo, Na, Ni, P, Pb, Sb, Se, Si, Sn, Sr, Ti, Tl, V, Zn). Greater yields were found for most of the elements by combined decomposition (with fusion) and decomposition by sintering. Nonetheless, both methods exhibited major losses of volatile elements (As, Bi, Sb, Tl). Thus, one of these methods must always be combined with a method of decomposition in a closed system. The work also demonstrates that, for investigation of untraditional waste geological materials (e.g. flue residues from high- temperature industrial processes), it will be necessary in the future to produce new certified reference materials with anomalous chemical and mineralogical compositions, which are not currently available on the market.
