Goal of this dissertation's thesis is to familiarize the reader with the issues connected with automatic optimization and its implementation in desired company, as a substitute to its current solution. The main objective of this thesis is therefore considered to be consideration and evaluation of benefits software solution can bring to HOPI, with full meeting of its requirements using its resources. In theoretical part of thesis will look at all the necessary information the reader will need to fully understand the theory, as well as the issueo of creating and optimizing great number of routes with a large fleet. Company HOPI will be introduced, as well as all software solutions, which were used in this work, or are mentioned with references. Practical part will describe the process of planning in both version, actual and software one. Form gained information and consultations with management of HOPI, conclusions will be drawn about benefits and difficulties of each solution. Ideal procedure for consideration will be drawn and verdict about software solution will be reached. In conclusion, thesis will summarize all gained knowledge, which will lead to final assessment and it will establish the best options of solutions for HOPI.

Koordinace provozu PST transformátorů v propojeném regionu / Coordination of operation of PST transformers in the interconnected region

This master’s thesis deals with phase-shifting transformers (PST) and coordination of electrically close devices in an interconnected region. Theoretical research is performed in the work, describing methods of power control, function and design of PST, as well as actual methods of PST coordination in the rest of the world. Based on the research there is an optimalisation algorithm designed in MATLAB, which computes recommended tap petting of PST based on the network configuration. In the second part of this thesis there are described components of this algorithm and also presented the results of three demonstrative scenarios. Algorithm is able to solve minor problems in the network; more complex problems are beyond its solving ability. For an improved functionality there is a necessity to further expand this algorithm by a network redispatch solving algorithm.

The aim of this master thesis is optimization of the rotor in the terms of efficiency. An analysis of the effects of rotor parameters is performed by RMXPRT. A genetic algorithm is created to find the optimal solution. Solutions are evaluated and adjusted according to the results. Measurements on the actual machine are performed and evaluated to verify the functionality of the simulation models.

Mgr. Jan Fránek 2011: The optimization the production of selected medicinal substances The diploma thesis dealt with optimization of production of fingolimod by hydrogenation. During the optimization of production, there was reduction of benzylic hydroxygroup and nitrogroup of nitrofingotriol. Reduction of two different groups caused dividing process of hydrogenation into two phases. The original intention was to optimize production only in the autoclave, but the need of using acidic catalytical medium in first phase led to dividing the production into reduction in the reactor and following catalyzed hydrogenation, already in neutral conditions, in the autoclave. The way of preparation of active substance by using an autoclave had not been explored in detail before. Evaluation of optimization is a part of results. There are also production charges on pilot plant included in results. Because the results of procedure of the pilot unit were not uniform, it was necessary to continue in optimization even after finishing the production in pilot plant. Following procedure modified according to the latest experiments was passed to the operating process.

This diploma thesis deals with the determination of nicotine in different products. The theoretical part summarizes review on nicotine, smoking and opportunities how to quit. In the practical part a method for the determination of nicotine by HPLC / PDA was optimized. As the most suitable stationary phase was selected a Kinetex 5u C18 100A 150 x 4.6 mm column, as the optimal mobile phase was chosen a pure methanol with a flow rate of 1 ml min-1 and a temperature of 25 °C. For the analysis of nicotine were chosen: 18 kinds of cartridges for electronic cigarettes, two kinds of nicotine gum, nicotine spray, nicotine pastilles, nicotine orodispersible film and ten species of classic cigarettes. For each type of product the most appropriate method for extracting nicotine and its subsequent analysis by HPLC / PDA was found. For tobacco 24 hour extraction in methanol and 10s ultrasound was selected. The nicotine spray and electronic cigarette refills without flavours were only diluted with methanol. Flavoured refills were first diluted by sodium hydroxide and then with methanol. For chewing gums, pastilles and nicotine film extraction with 5% sodium hydroxide was chosen. In this study also new experimental nicotine product was designed. Nicotine has been encapsulated in alginate-starch material to form small gel particles. As the most suitable medium for storage the water medium was determined.
