A hallmark of retinal diseases is degeneration of neural cells, leading to subsequent vision loss. For such diseases, replenishment of functional neural cells may be an optimal therapy. Unlike humans, the adult red-spotted newt, Notophthalmus viridescens, possesses the remarkable ability to regenerate a complete retina following its removal or injury. The purpose of this study was to develop a reproducible model of retinal damage and regeneration in the newt to understand the process of retinal regeneration. Intense light, shown in other organisms to be a relevant model of visual cell loss, was tested in the newt and resulted in variable loss of retinal function, correlating with the appearance of apoptotic cells. Due to the variability of damage observed, surgical removal of the retina was used to complement the light-damage model. A novel and non-invasive protocol using full-field electroretinography was developed to assess retinal function in vivo following damage. Measures of retinal function with the electroretinogram protocol successfully showed that photoreceptor function is initially lost and subsequently restored during regeneration. These results enhance our understanding of retinal regeneration in the adult newt and serve as a starting point for further studies aimed at determining the molecular mechanisms involved in the regeneration process.

Avaliação da fotoestabilidade e da fototoxicidade de fotoprotetores contendo associação de filtros solares e vitamina A / Evaluation of photostability and phototoxicity of sunscreen formulations containing a combination of UV-filters and vitamin A

Em função da preocupação com a saúde do consumidor, a maioria das formulações cosméticas tem sido acrescida de filtros solares e substâncias ativas antioxidantes, tais como vitaminas, para a proteção dos danos causados na pele pelos raios solares, entretanto, o uso de associações fotoinstáveis pode levar à formação de intermediários reativos que podem promover dermatites de contato e reações fototóxicas na pele, o que leva à necessidade da comprovação da segurança desses produtos frente a exposição à radiação solar. Há um grande empenho da comunidade científica para a utilização de testes alternativos ao uso de animais de experimentação sendo que o teste de fototoxicidade 3T3 NRU PT, em cultura de fibroblastos tem sido o preconizado para a essa finalidade. Assim, o objetivo deste projeto foi a avaliação da fotoestabilidade e da fototoxicidade de formulações fotoprotetoras contendo diferentes associações de filtros solares e estabilizadores de filtros solares, associados ao retinol e ao palmitato de retinila, por meio do uso de CLAE e cultura de fibroblastos, respectivamente. Para tal, foram desenvolvidas formulações fotoprotetoras contendo diferentes filtros solares que foram acrescidas ou não de retinol ou de palmitato de retinila, e de estabilizadores de filtros solares, que são utilizados para melhorar a sua fotoestabilidade. Nesse estudo, amostras das formulações foram aplicadas em lâminas de vidro e expostas à radiação UVA/UVB, a seguir, foram realizadas análises por CLAE, para dosagem do teor de filtros solares e vitaminas, e foi realizado estudo de espectrofotometria no UV, em que foi determinada a razão da absorção na faixa UVA/UVB. A fototoxicidade das associações de filtros solares, estabilizadores e vitaminas foi avaliada por meio do uso de cultura de fibroblastos 3T3, que foi submetida à radiação UVA para a determinação da viabilidade celular na presença e ausência da radiação. Os resultados obtidos nos sugerem que, no estudo por CLAE, bemotrizinol foi considerado o melhor estabilizador para ambas as vitaminas. No estudo por espectrofotometria no UV, as formulações contendo avobenzona / vitaminas foram consideradas fotoinstáveis, e apenas o bemotrizinol aumentou a fotoestabilidade da avobenzona associada a ambas as vitaminas. No estudo de fototoxicidade, a avobenzona, o dietilexilnaftalato e o retinol apresentaram potencial fototóxico. As associações que continham avobenzona / palmitato de retinila e avobenzona / retinol foram consideradas fototóxicas. A presença do estabilizador dietilexilnaftalato reduziu o potencial fototóxico das associações avobenzona / vitaminas e a presença do estabilizador bemotrizinol reduziu o potencial fototóxico da associação avobenzona / palmitato de retinila. Os resultados encontrados no presente estudo são promissores uma vez que algumas associações consideradas fotoestáveis apresentaram potencial fototóxico, mostrando a importância da realização de ambos os estudos no desenvolvimento / avaliação de formulações fotoprotetoras estáveis e seguras ao uso. / Due to consumer health concern, many cosmetic formulations has been supplemented by UVfilters and antioxidants such as vitamins, for the protection of UV-induced skin damages; however, the use of photounstable combinations can lead to the formation of reactive intermediates which may induce contact dermatitis and phototoxic reactions in the skin, which leads to the necessity of proving the safety of these products before exposure to solar radiation. There is a great effort by the scientific community to promote the alternatives to animal testing, whereas the 3T3 NRU phototoxicity test in cultured fibroblasts has been validated and recommended for this purpose. The objective of this project was to evaluate the photostability and phototoxicity of sunscreen formulations containing different combinations of photostabilizers and UV-filters, combined with retinol and retinyl palmitate, by using HPLC and fibroblast cultures, respectively. For this purpose, sunscreen formulations developed with different UV-filters were added or not of retinol or retinyl palmitate, and photostabilizers, which are used to improve the photostability. Firstly, samples of the formulations were applied on glass slides and exposed to UVA / UVB radiation, then, they were analyzed by HPLC, to determine the concentration of UV-filters and vitamins, and by UV spectrophotometry, to determine the UVA / UVB absorption ratio. Phototoxicity of combinations of UV-filters, photostabilizers and vitamins was evaluated by using the 3T3 fibroblast cultures, which was exposed to UV radiation for the determination of cell viability in the presence and absence of radiation. The obtained results suggested that, in the HPLC study, bemotrizinol was considered the best photostabilizer for both vitamins. In the UV spectrophotometry study, formulations containing avobenzone / vitamins were considered photounstable, and only bemotrizinol increased photostability of avobenzone combined with both vitamins. In the phototoxicity study, avobenzone, diethylhexylnaphthalate and retinol showed phototoxic potential. The combinations containing avobenzone / retinyl palmitate and avobenzone / retinol were considered phototoxic. The presence of the stabilizer diethylhexylnaphthalate reduced the phototoxic potential of avobenzone / vitamins combinations and the presence of bemotrizinol reduces the phototoxic potencial of avobenzone / retinyl palmitate combination. The results of the present study are promissory since some combinations that were considered photostable presented phototoxic potential, showing the importance of performing both studies in the development / evaluation of stable and safe sunscreen formulations.

Synthesis and Encapsulation of a New Zinc Phthalocyanine Photosensitizer Into Polymeric Nanoparticles to Enhance Cell Uptake and Phototoxicity

Efforts to enhance the utility of photodynamic therapy as a non-invasive method for treating certain cancers have often involved the design of dye sensitizers with increased singlet oxygen efficiency. More recently, however, sensitizers with greater selectivity for tumor cells than surrounding tissue have been targeted. The present study provides an approach to the modification of the known photosensitizer zinc phthalocyanine (ZnPc), to enhance its solubility and delivery to cancer cells. Targeting a photosensitizer to the site of action improves the efficacy of the sensitizer in photodynamic therapy. In this work we used PLGA-b-PEG to encapsulate a new zinc phthalocyanine derivative, 2(3), 9(10), 16(17), 23(24)-tetrakis-(4'-methyl-benzyloxy) phthalocyanine zinc(II) (ZnPcBCH3), to enhance uptake into A549 cells, a human lung cancer cell line. ZnPcBCH3 exhibited the same photochemical properties as the parent compound ZnPc but gave increased solubility in organic solvents, which allowed for efficient encapsulation. In addition, the encapsulated dye showed a near 500-fold increase in phototoxicity for A549 cancer cells compared to free dye.

Análogos de micosporinas: síntese e avaliação de parâmetros farmacológicos e toxicológicos / Mycosporines analogues: synthesis and evaluation of pharmacological and toxicological parameters.

Micosporinas são substâncias pertencentes a um grupo de compostos naturalmente encontrados em algas e fungos, que são utilizadas por estes organismos como mecanismo de defesa. São potentes bloqueadores químicos de radiação ultravioleta (UV), atuando principalmente na faixa espectral da radiação UVA, com absorção máxima variando entre 300 e 360 nm, dependendo de sua composição estrutural. Quimicamente, são caracterizadas por um cromóforo ciclohexenona ou ciclohexenimina conjugado com o nitrogênio substituinte de um aminoácido ou aminoálcool. Em alguns membros deste grupo também foi evidenciada uma moderada atividade antioxidante, podendo estes atuarem como antioxidantes naturais em organismos marinhos. Os processos sintéticos utilizados, com o objetivo de obtenção de forma não natural destas moléculas, se basearam em princípios de química verde e evitaram a utilização de solventes orgânicos bem como a produção de intermediários tóxicos. As reações envolvendo uma dicetona cíclica e diferentes aminoácidos foram testadas por diferentes metodologias \"verdes\". A partir destes processos foram obtidos 11 análogos das micosporinas, dos quais 9 tiveram seus potenciais farmacológicos e toxicológicos avaliados. Para verificação do potencial farmacológico das moléculas, foi determinado um espectro de absorção de radiação ultravioleta para cada um dos compostos, que posteriormente tiveram seus potenciais antioxidantes verificados através das atividades sequestrantes de radicais DPPH e de radical superóxido. Os espectros obtidos dos compostos demonstram uma absorção máxima variando de 255 a 309 nm para os diferentes compostos. No ensaio antioxidante de DPPH, dois compostos atingiram a IC50, em concentrações de 1,597 e 1,387 mM, e no ensaio de sequestro de radical superóxido, todos os compostos testados atingiram a IC50, em concentrações que variam de 0,204 a 1,164 mM. O potencial citotóxico das moléculas foi testado em culturas celulares de fibroblastos 3T3, através da incorporação das substâncias em diferentes concentrações e verificação de viabilidade celular, pelo método de redução de MTT, após exposição por 24 horas. O ensaio fototóxico procedeu da mesma forma, porém com a exposição das células a radiação ultravioleta concomitantemente à aplicação dos compostos. Os resultados obtidos demonstraram que nenhum dos compostos pode ser considerado citotóxico, porém um dos compostos é ativado com a exposição a radiação UVA, sendo assim, considerado fototóxico. / Mycosporines are substances belonging to a group of compounds naturally found in algae and fungi, which are used by these organisms as a defense mechanism. They are potent chemical blockers of ultraviolet radiation (UV), acting mainly in the spectral range of UVA, with maximum absorption ranging from 300 to 360 nm, depending on its structural composition. Chemically, they are characterized by a cyclohexenone or cyclohexenimine chromophore conjugated with the nitrogen substituent of an amino acid or an amino alcohol. Some mycosporines present a moderate antioxidant activity; in this case, they can act as natural antioxidants in marine organisms. Our synthesis was based on green chemistry principles and avoided the use of organic solvents and the production of toxic intermediates. The reactions involving a cyclic diketone and different amino acids were tested by different clean methods. Eleven mycosporines analogues were obtained. Nine of them had their pharmacological and toxicological potential evaluated. In order to verify the pharmacological potential of molecules, we determined the UV absorption spectrum of each mycosporine analogue. In addition, we evaluated their antioxidant activity by DPPH and superoxide radical scavenging potential methods. The compounds spectra show an absorption maximum ranging from 255 to 309 nm for different compounds. In the DPPH assay, two compounds reached the IC50 at concentrations of 1.597 e 1.387 mM. In the superoxide scavenging assay, all compounds tested reached the IC50 at concentrations ranging from 0.204 to 1.164 mM. The cytotoxic potential of these molecules was tested in cell cultures (3T3 fibroblasts) through the incorporation of different concentrations of the analogues and checking cell viability by MTT reduction method. The phototoxic test was conducted similarly, but adding the exposure of cells to UV radiation after addition of mycosporines analogues. The results showed that none of the compounds can be considered cytotoxic, but one of the compounds is activated by exposure to UVA radiation, becoming phototoxic.

Fotoestabilidade e fototoxicidade de avobenzona e palmitato de retinila associados a diferentes fotoestabilizadores / Photostability and phototoxicity of avobenzone and vitamin A associated with different light stabilizers

Os filtros solares formam a base da indústria cosmética de proteção solar. Estas substâncias podem absorver ou refletir a radiação UV. Por essa radiação provocar danos à pele, fotoenvelhecimento, câncer, é necessário o uso de protetores solares. Com isso, nas últimas décadas, as formulações cosméticas passaram a ser acrescidas dessas substâncias, que protegem a pele do consumidor contra a radiação UV. As vitaminas também passaram a ser adicionadas à a formulações fotoprotetoras para atuarem como antioxidantes. No entanto, os filtros sofrem degradação na presença de luz (fotodegradação), além disso, sua combinação com vitaminas pode levar à formação de intermediários reativos os quais podem provocar dermatites de contato e reações fototóxicas na pele. A comunidade científica tem se empenhado para a utilização de testes alternativos ao uso de animais de experimentação, sendo que o teste de fototoxicidade 3T3 NRU PT (Neutral red uptake), em cultura de fibroblastos, tem sido o preconizado para essa finalidade. Assim, o objetivo deste trabalho é avaliar a fotoestabilidade e fototoxicidade da avobenzona e palmitato de retinila associadas a diferentes fotoestabilizadores, por meio do uso de cromatografia liquida de alta eficiência (CLAE) e cultura de fibroblastos em monocamadas, respectivamente. Para tal, foram desenvolvidas formulações fotoprotetoras contendo os filtros solares avobenzona (AVO) e metoxicinamato de etilexila (MTX) acrescidos ou não de palmitato de retinila (Pr), e de fotoestabilizadores (tris tetrametilhidroxipiperidiol - TTMHP, etilhexil metoxicrileno - EHMCR e Tris-bifenil Triazina - TBFT), que foram utilizados para melhorar a sua fotoestabilidade. Os resultados de fotoestabilidade por CLAE demonstraram que a avobenzona, o metoxicinamato de etilexila e o palmitato de retinila foram considerados fotoinstáveis, porém a adição do fotoestabilizador EHMCR melhorou a fotoestabilidade das mesmas. Já o fotoestabilizador TBFT e TTHMP melhoraram a fotoestabilidade da associação de filtros sem a presença do palmitato de retinila. As análises por espectrofotometria demonstraram que a inclusão do TBFT na associação de filtros (FB3) e a inclusão do EHMCR na associação de filtros e o palmitato de retinila (FB2Pr), melhoraram a fotoestabilidade de suas respectivas formulações. A fototoxicidade das associações de filtros solares, estabilizadores e palmitato de retinila foi avaliada por meio do uso de cultura de fibroblastos 3T3, que foi submetida à radiação UVA para a determinação da viabilidade celular na presença e ausência da radiação seguindo recomendações da OECD TG 432. Os resultados obtidos nos sugerem que, no estudo de fototoxicidade, a AVO e TBFT apresentaram potencial fototóxico. As associações contendo os filtros AVO e MTX, acrescidos ou não de Pr (C1C e C1PrC) foram consideradas fototóxicas. Quanto à adição dos fotoestabilizadores, observou-se que a adição dos fotoestabilizadores TBFT e EHMCR à associação de AVO, MTX também foi considerada fototóxica (C2 e C3, respectivamente). Entretanto a adição do fotoestabilizador EHMCR à associação AVO, MTX e Pr, foi considerada não fototóxica. Já a inclusão dos fotoestabilizadores TTMHP e TBFT à associação contendo AVO, MTX, Pr (C3Pr e C4Pr) foram consideradas fototóxicas. / Sunscreens form the basis of cosmetic sun protection industry. These substances can absorb or reflect UV radiation. The radiation causes UV-induced damages to the skin, photoaging, cancer; therefore the use of sunscreen is necessary. In recent decades, the cosmetic formulations are now added of those substances which protect the consumer\'s skin against UV radiation. Vitamins also began to be added to the photoprotective formulations to act as antioxidants. However, the filters are degraded in the presence of light (photodegradation), moreover, the combination with vitamins can lead to the formation of reactive intermediates which can cause contact dermatitis and phototoxic skin reactions. The scientific community has been engaged in the use of alternative assays to replace animal experimentation, and the 3T3 NRU phototoxicity test (Neutral Red Uptake) in fibroblasts culture, has been recommended for this purpose. The objective of this study is to evaluate the photostability and phototoxicity of avobenzone and vitamin A associated with different photostabilizers, through the use of high performance liquid chromatography (HPLC) and culture of fibroblast in monolayers, respectively. For this, were developed photoprotective formulations containing avobenzone (AVO) and ethylhexyl methoxycinnamate (MTX) with or without the presence of vitamin A palmitate (Pr), and photostabilizers (Tris Tetramethylhydroxypiperidinol Citrate - TTMHP, ethylhexyl methoxycrylene - EEMC, Tris-Biphenyl Triazine - TBFT ), which were used to enhance the sunscreens photostability. The results of photostability by HPLC demonstrated that avobenzone, ethylhexyl methoxycinnamate and retinyl palmitate were considered photounstable, but the addition of the EHMCR improved their photostability. The TBFT and TTHMP improved the photostability of the association of sunscreens without the retinyl palmitate. Results by spectrophotometry demonstrated that the TBFT inclusion in the association of sunscreens (FB3) and EHMCR inclusion with the association of sunscreens and retinyl palmitate (FB2Pr) improved the photostability of their respective formulations. Phototoxicity of sunscreens combinations, photostabilizers, and vitamin A was evaluated by using 3T3 fibroblasts culture, which has been subjected to UVA radiation for the determination of cell viability in the presence and absence of radiation, according to OECD TG 432. The results suggest that, in phototoxicity study, AVO and TBFT presented phototoxic potential. Combinations containing AVO and MTX filters, added or not of Pr (C1C and C1PrC) were considered phototoxic. Regarding the addition of the photostabilizers, it was observed that the addition of TBFT and EHMCR photostabilizers to the combination of AVO, MTX was also considered phototoxic (C2 and C3). However, the addition of EHMCR to AVO, MTX and Pr was considered non-phototoxic. The inclusion of the TTMHP and TBFT photostabilizers in the combination containing AVO, MTX, Pr (C3Pr and C4Pr) were considered phototoxic.

Avaliação da performance de formulações fotoprotetoras associadas a mangiferina e naringenina: fotoestabilidade e fototoxicidade / Performance of photoprotective formulations containing mangiferin and naringenin: photostability and phototoxicity evaluation

Atualmente, devido ao conhecimento dos danos causados pela exposição da pele à radiação UV, existe uma tendência em utilizar, além dos filtros solares convencionais, substâncias naturais com potencial antioxidante. Entretanto, o uso de associações fotoinstáveis, pode, além de comprometer a capacidade fotoprotetora, levar à formação de intermediários reativos que podem ocasionar dermatites de contato e reações fototóxicas na pele. Dessa forma, o objetivo deste trabalho foi avaliar a fotoestabilidade e fototoxicidade de formulações fotoprotetoras contendo diferentes associações de filtros solares acrescidas ou não dos polifenóis mangiferina e naringenina. Para o estudo de fotoestabilidade, amostras das formulações foram aplicadas em lâminas de vidro e expostas à radiação UVA e, a seguir, foram feitas análises por cromatografia líquida de alta eficiência (CLAE), para dosagem do teor de filtros solares e antioxidantes, e por espectrofotometria, para determinação da razão UVA/UVB. A fototoxicidade foi avaliada por meio do uso de cultura de fibroblastos 3T3, submetida ou não à radiação UVA, para determinação da viabilidade celular. Os resultados de fotoestabilidade por CLAE demonstraram que a naringenina, o metoxicinamato de etilexila e avobenzona foram considerados fotoinstáveis. Além disso, foi demonstrado que a adição de mangiferina à associação contendo avobenzona e naringenina, proporcionou um aumento da fotoestabilidade das mesmas. A presença do dietilamino hidroxibenzoil hexil benzoato (DHHB) na formulação contendo avobenzona e naringenina promoveu melhora da fotoestabilidade da naringenina, no entanto não houve melhora da fotoestabilidade da avobenzona. As análises espectrofotométricas demonstraram que as formulações contendo a associação de avobenzona e DHHB (associação 3) apresentaram uma proteção no UVA superior às demais associações, ou seja, que continham avobenzona e DHHB isoladamente. O estudo de fototoxicidade mostrou que a avobenzona e o DHHB apresentaram potencial fototóxico. Quando as associações de filtros solares e antioxidantes foram analisadas, foi observado que a associação de avobenzona e naringenina, combinada ou não ao DHHB, apresentou potencial fototóxico. Dessa forma, esta não é recomendada para o desenvolvimento de formulações fotoprotetoras. A associação DHHB / avobenzona, por apresentar elevada proteção UVA, pode ser vantajosa para o desenvolvimento de fotoprotetores, entretanto a sua associação com antioxidantes deve ser utilizada com cautela e analisada caso a caso. A naringenina não é indicada para fotoprotetores contendo avobenzona, uma vez que esta associação foi considerada fotoinstável e fototóxica. Já a utilização da mangiferina pode ser considerada mais segura, e, além disso, a sua elevada atividade antioxidante pode complementar a eficácia de fotoprotetores. / Nowadays, due to the knowledge of UV-induced skin damage, there is a tendency to use natural substances with antioxidant potential, beyond conventional UV-filters. However, the use of photounstable combinations can compromise the photoprotective capacity and lead to formation of reactive intermediates that can cause contact dermatitis and phototoxic skin reactions. Thus, the objective of this study was to evaluate the photostability and phototoxicity of sunscreen formulations containing different UV-filter combinations supplemented or not with the polyphenols mangiferin and naringenin. For photostability studies, samples of the formulations were spread onto glass plates, exposed to UVA radiation and then analyzed by HPLC to determine the UV-filters and antioxidants concentrations, and also by spectrophotometry to determine the UVA/UVB ratio. Phototoxicity was evaluated by using the 3T3 fibroblast cultures, which was exposed to UVA radiation for the determination of cell viability. The results of photostability by HPLC analysis showed that naringenin, ethylhexyl methoxycinnamate and avobenzone were considered photounstable. Furthermore, it was demonstrated that the addition of mangiferin combined with avobenzone and naringenin, provided an increase in their photostability. The presence of DHHB in the formulation containing avobenzone and naringenin promoted an improvement in naringenin photostability, what was not observed for avobenzone. The spectrophotometric analysis showed that the formulations containing the combination of avobenzone and DHHB (combination 3) presented higher protection against UVA than the other combinations, i.e. the combinations containing avobenzone and DHHB, separately. The phototoxicity study showed that avobenzone and DHHB presented phototoxic potential. When UV-filters and antioxidants combinations were analyzed, it was observed that the combination of avobenzone and naringenin, combined or not with DHHB, presented phototoxic potential. Therefore, this is not recommended for the development of sunscreens. The combination DHHB / avobenzone, due to its high UVA protection, may be suitable on the development of sunscreens, however its combination with antioxidants should be used with caution and analyzed case-by-case. Naringenin is not indicated for sunscreens containing avobenzone, since this combination was considered photounstable and phototoxic. Moreover, the use of mangiferin can be considered safer than naringenin and, besides that, its high antioxidant activity can improve the photoprotective effects of sunscreens.

Análogos de micosporinas: síntese e avaliação de parâmetros farmacológicos e toxicológicos / Mycosporines analogues: synthesis and evaluation of pharmacological and toxicological parameters.

Micosporinas são substâncias pertencentes a um grupo de compostos naturalmente encontrados em algas e fungos, que são utilizadas por estes organismos como mecanismo de defesa. São potentes bloqueadores químicos de radiação ultravioleta (UV), atuando principalmente na faixa espectral da radiação UVA, com absorção máxima variando entre 300 e 360 nm, dependendo de sua composição estrutural. Quimicamente, são caracterizadas por um cromóforo ciclohexenona ou ciclohexenimina conjugado com o nitrogênio substituinte de um aminoácido ou aminoálcool. Em alguns membros deste grupo também foi evidenciada uma moderada atividade antioxidante, podendo estes atuarem como antioxidantes naturais em organismos marinhos. Os processos sintéticos utilizados, com o objetivo de obtenção de forma não natural destas moléculas, se basearam em princípios de química verde e evitaram a utilização de solventes orgânicos bem como a produção de intermediários tóxicos. As reações envolvendo uma dicetona cíclica e diferentes aminoácidos foram testadas por diferentes metodologias \"verdes\". A partir destes processos foram obtidos 11 análogos das micosporinas, dos quais 9 tiveram seus potenciais farmacológicos e toxicológicos avaliados. Para verificação do potencial farmacológico das moléculas, foi determinado um espectro de absorção de radiação ultravioleta para cada um dos compostos, que posteriormente tiveram seus potenciais antioxidantes verificados através das atividades sequestrantes de radicais DPPH e de radical superóxido. Os espectros obtidos dos compostos demonstram uma absorção máxima variando de 255 a 309 nm para os diferentes compostos. No ensaio antioxidante de DPPH, dois compostos atingiram a IC50, em concentrações de 1,597 e 1,387 mM, e no ensaio de sequestro de radical superóxido, todos os compostos testados atingiram a IC50, em concentrações que variam de 0,204 a 1,164 mM. O potencial citotóxico das moléculas foi testado em culturas celulares de fibroblastos 3T3, através da incorporação das substâncias em diferentes concentrações e verificação de viabilidade celular, pelo método de redução de MTT, após exposição por 24 horas. O ensaio fototóxico procedeu da mesma forma, porém com a exposição das células a radiação ultravioleta concomitantemente à aplicação dos compostos. Os resultados obtidos demonstraram que nenhum dos compostos pode ser considerado citotóxico, porém um dos compostos é ativado com a exposição a radiação UVA, sendo assim, considerado fototóxico. / Mycosporines are substances belonging to a group of compounds naturally found in algae and fungi, which are used by these organisms as a defense mechanism. They are potent chemical blockers of ultraviolet radiation (UV), acting mainly in the spectral range of UVA, with maximum absorption ranging from 300 to 360 nm, depending on its structural composition. Chemically, they are characterized by a cyclohexenone or cyclohexenimine chromophore conjugated with the nitrogen substituent of an amino acid or an amino alcohol. Some mycosporines present a moderate antioxidant activity; in this case, they can act as natural antioxidants in marine organisms. Our synthesis was based on green chemistry principles and avoided the use of organic solvents and the production of toxic intermediates. The reactions involving a cyclic diketone and different amino acids were tested by different clean methods. Eleven mycosporines analogues were obtained. Nine of them had their pharmacological and toxicological potential evaluated. In order to verify the pharmacological potential of molecules, we determined the UV absorption spectrum of each mycosporine analogue. In addition, we evaluated their antioxidant activity by DPPH and superoxide radical scavenging potential methods. The compounds spectra show an absorption maximum ranging from 255 to 309 nm for different compounds. In the DPPH assay, two compounds reached the IC50 at concentrations of 1.597 e 1.387 mM. In the superoxide scavenging assay, all compounds tested reached the IC50 at concentrations ranging from 0.204 to 1.164 mM. The cytotoxic potential of these molecules was tested in cell cultures (3T3 fibroblasts) through the incorporation of different concentrations of the analogues and checking cell viability by MTT reduction method. The phototoxic test was conducted similarly, but adding the exposure of cells to UV radiation after addition of mycosporines analogues. The results showed that none of the compounds can be considered cytotoxic, but one of the compounds is activated by exposure to UVA radiation, becoming phototoxic.
