For a drug to have a therapeutic effect, it has to reach its site of action in sufficient quantities. The Pheroid drug delivery system enhances the absorption of drugs in various pharmacological categories and is the focus of this study. A number of patents are registered in various countries to protect its application. Pheroid technology is trademarked, but may for ease of reading, be called Pheroid(s) only. The Pheroid itself is composed of an organic carbon backbone composed of unsaturated fatty acids with some side-chain interactions that result in self-emulsifying characteristics. The resulting vesicles and nano-sponges can entrap hydrophilic, hydrophobic or amphiphilic compounds for biomedical and agricultural application and can be manipulated as to loading ability, mechanical resistance, permeability, size and solubility. Pheroid was investigated for its potential use in the areas of vaccines, peptide drugs, topical products and cosmeceuticals, antimicrobial treatments and agriculture. In all of these areas, the Pheroid has indeed shown applicability: the results showed improved uptake and/or efficacy of the entrapped chemical or biological compounds after administration by a number of administration routes. For oral administration, a precursor format, the pro-Pheroid, was used, wherein the vesicles and/or sponges are formed post-administration. Proof of concept studies on the in vivo absorption and bioavailability, as well as studies on in vitro efficacy of Pheroid-based formulations were carried out for antimicrobials, such as tuberculosis drugs, antimalarials and antiretrovirals. In all cases, the in vitro efficacy of the active compounds was increased, compared to well-known standard drug treatments. In a phase I bio-equivalence study, a Pheroid-containing combination formulation was compared against the comparative market leader. The results demonstrated that the bioavailability of the active compounds in the Pheroid was at least as good but mostly significantly better than that of the comparative medication. In addition, the incidence of side-effects was decreased in the case of the Pheroid formulations. Furthermore, in vitro results indicate that drug resistance can at least partially be negated. Pheroid technology may also be capable of protecting labile drugs such as peptides against degradation and increasing efficacy so that lower dosages can be administered less frequently and with fewer side effects. Based on in vitro and in vivo results, a number of products are currently in development. Prevalência de brucelose bovina entre rebanhos fornecedores de leite de um laticínio no município de Itirapuã, Estado de São Paulo /Medeiros, Marina de Almeida Braga. January 2010 (has links)
Resumo: O estudo teve por objetivo determinar a prevalência da brucelose bovina entre fornecedores de leite de um laticínio situado no Município de Itirapuã, Estado de São Paulo, analisar fatores de risco eventualmente associados à ocorrência de brucelose nos rebanhos estudados e analisar o desempenho de testes sorológicos para detectar a infecção. De uma população de 942 bovinos adultos distribuídos em 55 rebanhos, situados nos municípios de Itirapuã e de Patrocínio Paulista, foi obtida uma amostra voluntária composta por 813 animais, distribuídos em 37 rebanhos cujos proprietários aceitaram participar do estudo. Foi aplicado um questionário, para a obtenção de informações sobre fatores que pudessem estar associados às taxas de prevalência. Foram colhidas amostras de soro sanguíneo de todos animais, as quais foram submetidas ao teste do antígeno acidificado tamponado (AAT), e as amostras positivas nesse teste foram submetidas à reação de fixação de complemento (RFC). Foram obtidas amostras de leite do conjunto de cada rebanho participante do estudo, que foram examinadas pelo teste do anel do leite (TAL). Das 813 amostras de soro sanguíneo colhidas, 26 (3,2%) resultaram positivas na aplicação em série dos testes de triagem (AAT) e confirmatório (RFC), e das 37 propriedades estudadas, 12 (32,4%) apresentaram pelo menos um animal positivo. Os fatores que se mostraram associados à ocorrência de brucelose foram número de animais no rebanho, não utilização da vacina B19 e compra frequente de animais. Não se observou boa concordância entre o TAL, que apontou resultado positivo em quatro (10,8%) dos 37 rebanhos, e os testes sorológicos individuais na detecção de rebanhos infectados / Abstract:The aim of this investigation was to verify the prevalence of bovine brucellosis among milk suppliers of a dairy industry located in the municipality of Itirapuã, state of São Paulo, Brazil, analyze risk factors eventually associated with the occurrence of brucellosis among the herds involved in the study, and compare the performance of serological tests in detecting infected herds. From a population of 942 adult cattle in 55 herds, in the municipalities of Itirapuã and Patrocínio Paulista, a voluntary sample of 813 animals was obtained in 37 herds whose owners agreed in participating in the study. A questionnaire about risk factors that could be associated to the prevalence rates was applied. Serum samples of all the 813 animals were collected and tested by the rose Bengal plate test (RBPT); positive samples were then tested by the complement fixation test (CFT). Milk samples were taken from each can at the industry and tested by the milk ring test (MRT). Of the 813 serum samples, 26 (3.2) tested positive in the serial application of RBPT and CFT, and of the 37 herds, 12 (32.4%) had at least one positive animal. Factors that showed association with the prevalence rate were number of animals in the herd, lack of vaccination with strain 19 and frequent introduction of animals. Embolie dans les plantes : dynamique de l'invasion d'air dans des réseaux hydrauliques naturels et artificiels sous pression négative / Embolism in plants : dynamics of air invasion in natural and artificial hydraulic networks under negative pressureBienaime, Diane 07 October 2016 (has links)
Pour assurer le transport de la sève des racines vers les feuilles, les plantes vasculaires génèrent de très fortes dépressions dans le liquide, pouvant atteindre -200 bar, au niveau des feuilles. Cette dépression « tire » sur la colonne d'eau contenue dans l'appareil vasculaire de l'arbre. La cohésion de l'eau maintient la sève sous forme liquide. Cet état métastable peut se rompre : des bulles de cavitation apparaissent. Elles créent un « bouchon » d'air dans le réseau hydraulique de la plante et gênent la circulation de la sève. C'est ce que l'on appele l'embolie. Si ce phénomène se généralise, il peut provoquer la mort de la plante.Ce travail de thèse est consacré à 'invasion d'air dans des réseaux hydrauliques naturels ou artificiels initialement à pression négative. Nous avons d'abord étudié l'embolie dans les feuilles. Nous avons développé une technique novatrice permettant de relever la propagation spatiale de l'embolie dans le réseau hydraulique des feuilles. Nous montrons que l'embolie, quelque soit l'espèce, se propage par à-coups des plus grosses nervures aux plus petites.Afin de comprendre les lois physiques sous-jacentes, nous utilisons deux systèmes modèles. Nous réalisons d'abord des réseaux artificiels dans un hydrogel reproduisant les caractéristiques de la circulation de la sève ascendante. Après la relaxation de la tension dans le réseau par l'apparition de la bulle, nous observons des oscillations de surface et une croissance lente de la bulle, liée à l'évacuation de l'eau à travers l'hydrogel. Cette croissance peut atteindre un régime quasi-stationnaire. Ce systèmes ne nous permettant pas de reproduire toutes les caractéristiques géométriques du xylème, nous présentons une modélisation informatique reposant sur l'analogie entre réseaux hydrauliques et électrocinétique. Nous reproduisons les caractéristiques du xylème dans lequel circule la sève : les éléments conducteurs sont reliées par les ponctuations, des valves protégeant la plante de l'embolie. Nous retrouvons les à-coups caractéristiques de la propagation de l'embolie dans les feuilles.Enfin, nous discutons l'application des résultats précèdents dans le cas du bois et nous présentons quelques résultats obtenus sur du pin sylvestre. / To assure the transport from the roots to the leaves, vascular plants create strong depressions in the sap, next to -200 bars. This depression pulls the water column contained by the tree vascular system. The water cohesion keeps the sap under liquid state. This metastable state can breaks: cavitation bubbles appear. They create an air plug inside the plant hydraulic network and impede sap flow. This phenomena called embolism could lead to the plant death by preventing the sap transport.This thesis is dedicated to the air invasion into hydraulics networks under negative pressure. First, we study the leaf embolism. We developed a new technique which allows us to record the spatial propagation of embolism in leaves hydraulic network. We show that the embolism propagates by steps from biggest veins to smallest veins.Next, in order to understand the underlying physical laws, we use two model systems. We build artificial networks in a hydrogel which mimics the sap flow characteristics. After the relaxation of the negative pressure in the network by the nucleation of a bubble, we observe surface oscillations and the slow growth of the bubble. This growth is linked to the water transport through the hydrogel and can reach a stationary regime.As we are not able to reproduce all the characteristics of the leaf network with the hydrogel, we create a computer modeling based on the Ohm analogy between hydraulics networks and electrical circuits. We reproduce the specific features of the xylem which transport the sap: the conduits are linked by pits, small valves which limit the progression of the embolism. We were able to recover the distinctiveness steps in embolism.Finally, we discuss the application of the preceding results to wood and we present some results on Pinus sylvestris.

Etude d'un nouveau type de RNase P spécifique des eucaryotes chez Arabidopsis thaliana / Characterisation of a novel type of eukaryote-specific RNase P in Arabidopsis thalianaGutmann, Bernard 14 September 2012 (has links)
La RNase P est impliquée dans la maturation des ARNt en libérant l’extrémité 5’ leader des précurseurs d’ARNt. Jusqu’à récemment, il était admis que cette enzyme soit universellement conservée en tant que complexe ribonucléoprotéique. La rupture avec le modèle établi est venue avec l’identification d’une RNase P uniquement protéique dans les mitochondries humaines et chez les plantes. Ces protéines, nommées PRORP (PROteinaceous RNase P), présentent trois paralogues chez Arabidopsis, qui sont localisés soit dans les organelles (PRORP1), soit dans le noyau (PRORP2 et 3). Des tests d’activité in vitro montrent que les protéines PRORP possèdent seules une activité RNase P. L’étude de lignées ADN-T indique que la fonction des protéines PRORP est essentielle et la fonction de PRORP2 et 3 est redondante. L’analyse des lignées de dérégulation montre que les protéines PRORP possèdent une diversité de substrat et que la RNase MRP, une autre ribonucléoprotéine, n’est pas impliquée dans la maturation des ARNt. Ainsi les protéines PRORP seraient bien les seules enzymes impliquées dans la maturation de l’extrémité 5’ des ARNt chez Arabidopsis. / RNase P is involved in the maturation of tRNA precursors by cleaving their 5’ leader sequences. Until recently this enzyme was considered to be universally occurring as a ribonucleoprotein complex. The breakthrough from the existing model came with the identification of protein-only RNase P in human mitochondria as well as in plants. These proteins that we called PRORP (PROteinaceous RNase P) have three paralogs in Arabidopsis, which are localised in organelles (PRORP1) and nuclei (PRORP2 and 3). We have shown that PRORP proteins have RNase P activity in vitro as single proteins. In vivo the functions of PRORP proteins are essential and the function of PROPR2 and 3 are redundant. PRORP down-regulation mutants, show that PRORP proteins have a variety of other substrates and RNase MRP, another ribonucleoprotein, is not involved in the tRNA maturation. Results show that PRORP proteins would be the only enzymes responsible for RNase P activity in Arabidopsis.

Les insectes phytophages causent des dommages importants aux cultures. Ces insectes sont principalement contrôlés par l’utilisation d’insecticides mais les effets néfastes des composés utilisés sur la santé humaine et l’environnement ainsi que l’apparition de populations résistantes à ces composés imposent de trouver des stratégies de lutte alternatives. Sélectionner des plantes pour leur résistance aux insectes pourrait être une solution intéressante. Un frein majeur au développement de cette stratégie est le fait qu’un grand nombre de génotypes doit être criblé pour identifier des sources de résistance et que le phénotypage est souvent complexe et limite les possibilités de criblage à grande échelle. Pour lever ces verrous, il est important de comprendre les mécanismes à l’origine des résistances. Une fois ces mécanismes précisés dans le cadre de l’interaction entre une culture et un ravageur, l’identification de traits clés de cette interaction permet d’envisager l’utilisation de biomarqueurs de la résistance qui peuvent fortement simplifier le phénotypage. Ce type d’approche a été conduit au cours de cette thèse dans le cadre de l’interaction entre le colza (Brassica napus) et l’un de ses principaux ravageurs, le méligèthe (Brassicogethes aeneus). Des travaux antérieurs avaient mis en évidence l’existence de certains composés biochimiques présents dans le périanthe des boutons floraux et corrélés au niveau d’attaque de différents génotypes de colza. Ces résultats avaient été obtenus au laboratoire et il était nécessaire de les confirmer dans des conditions de culture réalistes. Le premier objectif de cette thèse était donc de développer une méthode permettant de cribler au champ différents génotypes de colza pour leur résistance au méligèthe afin de valider ou non le potentiel des composés biochimiques identifiés précédemment comme biomarqueurs mais aussi d’en identifier de nouveaux. Le second objectif de ce travail était de mieux comprendre l’écologie alimentaire du méligèthe afin d’identifier de nouvelles cibles potentielles pour la sélection de plantes résistantes. Nous avons mené plusieurs expérimentations sur le terrain qui nous ont permis de développer une méthode de crible au champ des génotypes de colza pour leur résistance au méligèthe. Cette méthode a mis en évidence l’existence de différences de résistance fortes et robustes entre génotypes parmi une gamme de 19 génotypes testés. Les biomarqueurs potentiels précédemment identifiés n’ont pas été validés au champ mais deux autres composés semblent influencer la résistance au méligèthe et pourraient être utilisés comme futurs biomarqueurs. Ces expériences au champ ont aussi permis de montrer que la biochimie des périanthes était très dépendante des conditions environnementales, ce qui pourra compliquer le travail de sélection. Par ailleurs, des expériences de développement réalisées en conditions contrôlées ont montré que le méligèthe utilisait le nectar présent dans les fleurs de colza mais que celui-ci ne semblait pas affecter son développement. Le pollen semble, jouer un rôle important sur son développement mais les larves parviennent à se développer sans son apport. Les expériences sur le comportement alimentaire du méligèthe adulte ont montré que cet insecte avait un patron d’attaque spécifique sur les inflorescences de colza en s’alimentant avant tout sur les boutons de petite taille (donc moins riches en pollen par rapport aux gros). Il semble que la disponibilité mais surtout l’accessibilité du pollen médiée par le périanthe joue un rôle essentiel dans le comportement de cet insecte. Ce travail a permis de mieux comprendre le comportement alimentaire du méligèthe et d’identifier des traits importants pour son développement. Ce travail montre que des résistances modérées existent au sein du colza et qu’elles pourraient être utilisées pour la sélection variétale. Il pointe aussi les limites des approches basées sur des biomarqueurs biochimiques. / Herbivorous insects cause important yield losses to crops. These insects are mainly managed through insecticides but negative effects of used compounds on the human health and the environment as well as the development of resistant pest populations impose finding alternative strategies to manage them. Breeding plants for enhanced resistance to insects is an attractive strategy. This kind of strategy was already implemented to manage insects but examples of its utilisation remain rare. The main limitation of this strategy is that a large number of genotypes needs to be screened through generally complex phenotyping methods to identify resistances. To circumvent these issues, an approach based on the understanding of plant defence mechanisms and the utilisation of biochemical biomarkers linked to resistance could be interesting. During this PhD, this kind of approach was developed on the oilseed rape (Brassica napus) and one of its main pests, the pollen beetle (Brassicogethes aeneus). Previous studies identified chemical compounds present in the perianth of flower buds and correlated to oilseed rape resistance to pollen beetle. However these studies were carried out in the laboratory and need to be validated in field conditions. Moreover, information on pollen beetle feeding ecology are still lacking while they could help identifying new targets for breeding. The first objective of this PhD is to develop a method allowing to screen resistance to the pollen beetle in the field. This method will allow to confirm the potential biomarkers previously identified and to look for additional biomarkers. The second objective of the present work was to better understand key steps in the interaction between the pest and its host plant to identify potential new target traits for resistance. For this purpose, the importance of food sources present in flowers such as pollen and nectar on pollen beetle development and the factors impacting the feeding pattern of adults on the inflorescence were investigated. We conducted field experiments in two different sites and for two consecutive years and propose a method allowing to screen oilseed rape resistance to pollen beetle in the field. Using this method, we were able to identify genotypes with moderate levels of resistance among a set of 19 genotypes. Previously identified biochemical biomarkers were not correlated with plant resistance in the field but new markers were identified (i.e. quinic acid and arginine). Our field experiments also showed that plant composition is highly variable according to the environment and this variability could affect usefulness of these markers during plant breeding programs. Experiments under controlled conditions also showed that pollen beetle used nectar for feeding but that it did not seem to affect its development. Pollen, on the other hand, seemed to have a more important impact but was not indispensable to pollen beetle development. The study of the pollen beetle feeding behaviour showed that this insect has a surprising feeding pattern on oilseed rape inflorescences and that small buds are more used for feeding than large buds that contain more pollen. It seems that accessibility and to a lesser extend availability of feeding resource explain this pattern and that the perianth has a major role on this preference. These experiments allowed to better understand the pollen beetle feeding ecology and to identify plant traits important for its development. Our work showed that moderate levels of resistance are present in oilseed rape and could be used in breeding programs. Limitations of approaches based on biochemical biomarkers are discussed.

Ce travail vise à développer un modèle générique capable de mieux rendre compte et de prédire les transferts de radionucléides dans le système sol/solution du sol/plante. La première partie de ce travail a été consacrée à l’analyse critique des modèles disponibles dans la littérature pour décrire l’adsorption du césium par les minéraux argileux (principal processus contrôlant sa disponibilité dans les sols). Cette analyse a débouché sur la formulation d’un nouveau modèle mécaniste combinant deux approches: la complexation de surface et l’échange d’ions. Cette approche, a été testée afin de modéliser l’adsorption du Cs sur plusieurs substrats argileux naturels. Ce travail a permis de valider le modèle proposé et de démontrer qu’il constitue un avantage majeur par rapport aux différents modèles existants. La deuxième partie a été consacrée, à la réalisation d’une série d’expérimentations, conduite en milieux contrôlés sur des systèmes dynamiques, et la modélisation de la (bio)disponibilité du Cs dans ces systèmes. A la suite de ces essais, les interactions entre solide et solution observées ont pu être correctement reproduites à partir du modèle proposé en prenant en compte la fraction argileuse du sol uniquement. Ces simulations ont également été comparées aux simulations obtenues à partir des modèles empirique (Kd) et cinétique (EK). Enfin, le développement d’un outil numérique permettant de coupler la description des interactions géochimiques au transfert vers la plante (approche cinétique) a permis de reproduire correctement les essais réalisés en Rhizotests couplant sol, solution et plante et de mieux caractériser la fraction du Cs disponible pour les plantes. / The overall objective of this work is to develop a generic model able to better account and predict the transfer of radionuclides in the soil / soil solution / plant. The first part of the work was devoted to the critical analysis of the models available in the literature to describe the cesium adsorption on clay minerals (the process that mainly controls its availability in the soils). This analysis enabled us to propose a new mechanistic model combining two approaches: surface complexation and cation exchange. Our approach has been tested in order to model the adsorption of Cs on several natural clay substrates, in a wide range of Cs concentrations and physicochemical conditions. This work allowed to validate the proposed mechanistic model and to demonstrate that it constitutes a major advantage over the various existing models. The second part was devoted, to the performing of a series of experiments, carried out in controlled environments on dynamic systems and modeling the (bio)availability of Cs in these systems. Following these tests, the observed interactions between solid and solution could be correctly reproduced with the proposed model taking into account only the clay fraction of the soil. These simulations were also compared with simulations obtained using a empiric (Kd) kinetic models (E-K approach). Finally, the development of a numerical tool for coupling the description of geochemical interactions with transfer to the plant (Michaelis-Menten approach) allowed to reproduce adequately the trials carried out in Rhizotests coupling soil, solution and plant, and to better characterize of the Cs fraction available for plants.

En raison du risque d’un approvisionnement insuffisant en ressources primaires des terres rares et des impacts environnementaux générés par les zones minières, le concept de l’agromine paraît être une solution très prometteuse. Il permet d’extraire des métaux lourds à partir de sols pollués ou de friches industrielles, par une méthode respectueuse de l’environnement, grâce à la culture de plantes hyperaccumulatrices. Une fois la culture réalisée, des procédés hydrométallurgiques sont développés afin d’extraire des plantes les métaux ayant une valeur économique importante. Le présent travail vise à développer des procédés hydrométallurgiques pour l’extraction des terres rares à partir d’une plante hyperaccumulatrice appelée Dicranopteris dichotoma. Cette fougère est connue pour sa capacité à accumuler les terres rares, notamment les légères, dans sa partie aérienne. Différentes voies d’extraction, puis de séparation des terres rares des autres éléments, ont été étudiées. Dans un premier temps, des lixiviations de la biomasse sèche ont été mises en oeuvre. Les rendements de lixiviation selon la nature de l’extractant ont ainsi pu être comparés. Pour cette voie, une précipitation est ensuite effectuée, suivie d’une calcination pour obtenir les oxydes de terres rares. Dans un second temps, la lixiviation de la biomasse sèche a été intensifiée par un procédé d’échange d’ions au cours duquel les terres rares sont fixées sur la résine. Après avoir percolé sur la résine des solutions qui permettent d’éliminer des impuretés, l’élution permet d’obtenir une solution concentrée de terres rares. Enfin, une troisième voie d’extraction est réalisée à partir des cendres de D. dichotoma, après une étape de combustion. Cette voie repose sur une lixiviation alcaline des cendres permettant l’élimination des impuretés solubles dans ce milieu. Une dissolution du résidu est ensuite effectuée, suivie d’une précipitation sélective des terres rares. Les trois voies étudiées s’avèrent en fait complémentaires et la combinaison de certaines étapes peut s’avérer prometteuse, notamment pour éliminer certaines impuretés. L’étude de ces procédés et de leur combinaison mérite d’être poursuivie afin d’améliorer les rendements d’extraction et la pureté du produit final. Il s’agira ensuite de développer un procédé à l’échelle pilote puis industrielle / Due to the risk of primary resource supply of rare earths and the environmental impacts generated by mining areas, the concept of agromining seems to be a very promising solution. It allows the extraction of heavy metals from polluted soils or industrial wastelands, by an environmentally friendly method, thanks to the cultivation of hyperaccumulating plants. Once the culture is completed, hydrometallurgical processes are developed in order to extract from plants the metals having a significant economic value. The present work aims to develop hydrometallurgical processes for the extraction of rare earths from a hyperaccumulator plant called Dicranopteris dichotoma. This fern is known for its high ability to accumulate rare earths, especially light ones, in its aerial part. Different extraction and separation ways of rare earths from other elements have been studied. At first, leaching of dry biomass was implemented. The leaching yields according to the nature of extracting solutions could thus be compared. For this approach, precipitation is then performed, followed by calcination step to obtain rare earth oxides. In a second approach, the leaching of dry biomass was intensified by an ion exchange process during which, the rare earths are fixed on the resin. After percolating solutions on the resin in order to eliminate impurities, an elution step leads to obtain a concentrated solution of rare earths. Finally, a third extraction process is carried out from the ashes of D. dichotoma, after a combustion step. This approach is based on an alkaline leaching of the ash allowing the elimination of soluble impurities in this medium. The residue is then dissolved, followed by rare earths selective precipitation. These three approaches studied, are actually complementary and the combination of certain steps can be promising, especially to eliminate some impurities. The study of these processes and their combination deserves to be pursued in order to improve the extraction yields and the purity of final product. It will then develop a pilot scale and industrial process

De uma população de 942 bovinos adultos distribuídos em 55 rebanhos, situados nos municípios de Itirapuã e de Patrocínio Paulista, foi obtida uma amostra voluntária composta por 813 animais, distribuídos em 37 rebanhos cujos proprietários aceitaram participar do estudo. Foi aplicado um questionário, para a obtenção de informações sobre fatores que pudessem estar associados às taxas de prevalência. Foram colhidas amostras de soro sanguíneo de todos animais, as quais foram submetidas ao teste do antígeno acidificado tamponado (AAT), e as amostras positivas nesse teste foram submetidas à reação de fixação de complemento (RFC). Foram obtidas amostras de leite do conjunto de cada rebanho participante do estudo, que foram examinadas pelo teste do anel do leite (TAL). Das 813 amostras de soro sanguíneo colhidas, 26 (3,2%) resultaram positivas na aplicação em série dos testes de triagem (AAT) e confirmatório (RFC), e das 37 propriedades estudadas, 12 (32,4%) apresentaram pelo menos um animal positivo. Os fatores que se mostraram associados à ocorrência de brucelose foram número de animais no rebanho, não utilização da vacina B19 e compra frequente de animais. Não se observou boa concordância entre o TAL, que apontou resultado positivo em quatro (10,8%) dos 37 rebanhos, e os testes sorológicos individuais na detecção de rebanhos infectados / The aim of this investigation was to verify the prevalence of bovine brucellosis among milk suppliers of a dairy industry located in the municipality of Itirapuã, state of São Paulo, Brazil, analyze risk factors eventually associated with the occurrence of brucellosis among the herds involved in the study, and compare the performance of serological tests in detecting infected herds. From a population of 942 adult cattle in 55 herds, in the municipalities of Itirapuã and Patrocínio Paulista, a voluntary sample of 813 animals was obtained in 37 herds whose owners agreed in participating in the study. A questionnaire about risk factors that could be associated to the prevalence rates was applied. Serum samples of all the 813 animals were collected and tested by the rose Bengal plate test (RBPT); positive samples were then tested by the complement fixation test (CFT). Milk samples were taken from each can at the industry and tested by the milk ring test (MRT). Of the 813 serum samples, 26 (3.2) tested positive in the serial application of RBPT and CFT, and of the 37 herds, 12 (32.4%) had at least one positive animal. Factors that showed association with the prevalence rate were number of animals in the herd, lack of vaccination with strain 19 and frequent introduction of animals. The agreement between individual serological tests and MRT to detect infected herds was not good, since MRT showed positive results in only four (10.8%) of the 37 herds

Insertion de la morphogenèse racinaire dans L grass, un modèle structure-fonction de graminées fourragères / Insertion of root morphogenesis in L-grass, a functional-structural plant model of forage grassesMigault, Vincent 24 February 2015 (has links)
Un modèle de morphogenèse aérienne de graminées fourragères, nommé L grass, a été développé lors de travaux précédents. Le but de la thèse est de proposer une méthode d'insertion de la morphogenèse racinaire dans ce modèle afin de tenir compte de l'impact des ressources telluriques sur la structure et le fonctionnement de la plante.</br>Un simulateur de plante entière de ray-grass anglais (Lolium perenne L.) incorporant (i) des règles d'autorégulation de l'architecture aérienne, (ii) le développement architecturé du système racinaire, (iii) des processus de coordination entre les parties aérienne et racinaire et (iv) un système d'allocation du carbone a été développé. La nature autorégulée du modèle lui permet de reproduire les réponses à l'intensité d'une défoliation et à la compétition pour la lumière sans nécessiter de contrôle central. Le couplage du modèle avec un modèle de sol permet de simuler le développement de la plante subissant différents scénarios d'apport en eau sans nécessiter de consignes morphogénétiques données a priori.</br>La représentation architecturée du système racinaire présente quelques limites, principalement en termes de ressources informatiques, pour les simulations de peuplements. Afin de pallier ces limites, un modèle continu de dynamique de densités racinaires a aussi été incorporé pour simuler l'enracinement. Les comparaisons entre les deux modèles de système racinaire ont montré l'intérêt indéniable des modèles de densités racinaires notamment pour les études à l'échelle d'un couvert végétal. / A model of the shoot morphogenesis of forage grasses, called L-grass, was developed in previous studies. The aim of this thesis is to propose a method for introducing root morphogenesis in the current model so the effect of telluric resources availability can be taken into account for shaping plant form and functioning.</br>A simulator of perennial ryegrass (Lolium perenne L.) whole plant, which includes (i) the rules of shoot architecture self-regulation,(ii) the architectural development of the root system, (iii) the coordination processes between shoot and root parts and (iv) a system of allocation of carbon, was developed. The self-adaptable character of the model allows the reproduction of the responses to the intensity of defoliation and the competition for sunlight without the need of any central control. The coupling of the model with a soil model enables to recreate the development of the plant undergoing different scenarios of water supply with no requirement of previously established morphogenetic guidelines.</br>The architectural representation of the root system presents some shortcomings, mainly regarding the need in computing power when simulating plant populations. In order to overcome these limitations, a continuous model of the dynamics of root densities has been also incorporated to simulate plant rooting. The comparison between both models of root systems has shown the undeniable interest of modeling root density, especially for the studies at the vegetation cover level.
