The house is located in a quiet part of Strachotín. The plot itself is now used as agricultural land. The bio corridor belt creates a natural dominance around the area and ensures safe drainage of water during torrential rains. The concept is based on a wide range of services and care. The home for seniors will become part of the life of all seniors in the village and nearby. Supporting home-based autonomy allows seniors to live in their own home and only use the services that are available. Visits and social activities are a matter of course. Accommodation in the house itself will preferably be provided to seniors who require constant care. The operating standard of the home is designed to a very good standard. The home offers a barrier-free environment, single or double rooms. Meeting rooms are an important part of the house, along with a garden and bathing biotope. Separate living from operation guarantees a high level of privacy and allows direct contact with the garden.

Design, Syntheses, and Bioactivities of Conformationally Locked Pin1 Ground State Inhibitors

Pin1 (protein interacting with NIMA 1) is a peptidyl-prolyl isomerase involved in mitosis. As a potential anti-cancer drug target, Pin1 interacts and regulates the activity of an increasing number of cell cycle enzymes by an unknown mechanism. These cell cycle enzymes include Cdc25, Cdc27, Cyclin D1, Myt1, Wee1, NIMA, Cdc2, Plk1 and c-Myc. Recent research has revealed that Pin1 is overexpressed in a variety of cancer cell lines and Pin1 inhibitors inhibit proliferation activity of several cancer cells overexpressing Pin1. The most potent Pin1 inhibitors identified so far are in the micromolar range and no pharmacophore has been identified. In order to assist the understanding of the biological function of Pin1 using molecular probes, two amide isosteres of Ser-<i>trans</i>-Pro and Ser-<i>cis</i>-Pro dipeptides were designed and stereoselectively synthesized. The conformationally locked Ser–<i>trans</i>–Pro mimic, Boc-SerΨ[(<i>E</i>)CH=C]Pro–OH, was synthesized through the use of an Ireland-Claisen [3,3]-sigmatropic rearrangement in nine steps with 13% overall yield from a serine derivative. The Ser-<i>cis</i>-Pro mimic, Boc-SerΨ[(<i>Z</i>)CH=C]Pro–OH, was synthesized through the use of a Still-Wittig [2,3]-sigmatropic rearrangement in 11 steps with an overall yield of 20% from the same starting material. Conformationally locked peptidomimetics, including two exactly matched peptidomimetics, Ac–Phe–Phe–pSer–Ψ(<i>E</i>)CH=C]Pro–Arg–NH2 and Ac–Phe–Phe–pSer–Ψ[(<i>Z</i>)CH=C]Pro–Arg–NH2, were synthesized from these Ser-Pro isosteres using Fmoc SPPS. A protocol for in vitro Pin1 inhibition assay was established for measuring the inhibition constant for these peptidomimetics. A conformationally locked cis peptidomimetic inhibits Pin1 with a <i>K</i><sub>i</sub> of 1.7 <i>μ</i>M, 23-fold more potent than its trans counterpart, illustrating the preference of Pin1 for a cis amide bond in its PPIase domain. The A2780 ovarian cancer cell antiproliferation activity of these peptidomimetics parallels their respective Pin1 inhibition data. This research provides a start toward more drug-like Pin1 inhibitor design. Gly–<i>trans</i>–Pro isosteres were synthesized using the Ireland-Claisen route. The construction of a non-peptidic (Z)-alkene library for Pin1 inhibition was attempted using the Ser-<i>cis</i>-Pro mimic, Boc—SerΨ[(Z)CH=C]Pro–OH as the core. / Ph. D.

Undersökning av rumslig tillgänglighet av livsmedelsbutiker inom Karlstads kommun : En analys av multimodal transport / Analysis of the Spatial Accessibility of Grocery Stores within Karlstad Municipality

Examensarbete undersöker rumslig tillgänglighet av livsmedelsbutiker inom Karlstads kommun. Studien fokuserar på butikskedjor som exempelvis Ica och Coop, som spelar en central roll i att tillhandahålla mat och andra nödvändigheter för befolkningen. Syftet med studien är att analysera tillgängligheten till livsmedelsbutiker genom gång- och cykeltrafik, med målet att förbättra folkhälsanoch värna grönare trafikalternativ.För att uppnå detta mål användes geografiska informationssystem (GIS) för att utföra rumsliga analyser. ArcGIS Pro användes för att skapa en nätverksdataset som möjliggjorde nätverksanalyser, inklusive OD-kostnadsmatrisen för att beräkna antalet tillgängliga butiker per stadsdelsområde ochtjänsteområden från livsmedelsbutikerna. Befolkning- och inkomstdata integrerades för att inkludera socioekonomiska aspekter i analysen.Resultaten visar att det finns betydande skillnader i tillgänglighet mellan olika stadsdelar och transportmedel. Tingvallastaden framstår som ett område med mycket god tillgång till livsmedelsbutiker inom både gång- och cykelavstånd, medan andra områden har lite till ingen tillgång. Studien finner att äldre personer oftare (5 %) bor närmare butiker inom gång- och cykelavstånd jämfört med den totala befolkningen.Arbetet belyser vikten av att använda GIS för att förstå och förbättra tillgängligheten tilllivsmedelsbutiker. GIS baserade nätverksanalyser har potentialen att integrera relevant geodata för en effektiv, informativ och mångpräglad analys som kan användas för att förbättra både folkhälsa och miljövänliga transportmedel. / The thesis investigates the spatial accessibility of grocery stores within the Karlstad municipality. The study focuses on retail chains such as Ica and Coop, which play a central role in providing food and other necessities for the population. The purpose of the study is to analyze the accessibility of grocery stores through walking and cycling traffic, with the goal of improving public health and promoting greener traffic alternatives.To achieve this goal, geographic information systems (GIS) were used to perform spatial analyses. ArcGIS Pro was utilized to create a network dataset that enabled network analyses, including the OD cost matrix to calculate the number of accessible stores per district and service areas from the grocery stores. Population and income data were integrated to include socioeconomic aspects in the analysis.The results show significant differences in accessibility between different districts and modes of transport. Tingvallastaden stands out as an area with very good access to grocery stores within both walking and cycling distance, while other areas have little to no access. Treatment with delgocitinib cream improves itch, pain and other signs and symptoms of chronic hand eczema: Results from the Hand Eczema Symptom Diary in a phase IIb randomized clinical trial

Background: Measuring patient-reported outcomes is crucial to fully capture the burden of chronic hand eczema (CHE). - Objectives: To assess the effect of delgocitinib cream on itch, pain and nine additional key signs and symptoms reported by patients with CHE using the Hand Eczema Symptom Diary (HESD). - Methods: In a double-blind, phase IIb dose-ranging trial (NCT03683719), 258 adults with mild to severe CHE were randomized to delgocitinib cream 1, 3, 8 or 20 mg/g or cream vehicle twice daily for 16 weeks. Patients assessed 11 signs and symptoms of CHE daily through the HESD using an 11-point numeric rating scale; this was an exploratory endpoint. - Results: Delgocitinib cream 20 mg/g was associated with an early and sustained reduction in itch and pain, along with clinically relevant reductions of ≥4 points from baseline to Week 16 in 48.4% and 63.6% of patients, respectively (17.9% and 5.9% with cream vehicle). There were improvements versus cream vehicle in all assessed CHE signs and symptoms (20 mg/g, p < 0.05). - Conclusions: Delgocitinib cream reduced itch, pain and other signs and symptoms in patients with CHE. This data correlated with clinician-reported outcomes, indicating that the HESD may be a useful assessment tool for CHE management.

Thesis: Abortion in the light of case law of European Court of Human Rights This thesis focuses on the legal regulation of abortion in selected European countries in order to find single European standard. It concentrates primarily on issues of assessing violations of particular articles of the Convention on Human Rights and Fundamental Freedoms by the European Court of Human Rights or former European Commission of Human Rights in relation to specific cases associated with abortion. Abortion means the deliberate termination of pregnancy by means of surgery or administration of drugs to induce abortion. Abortion is a major ethical issue that raises number of discussions and disputes. The complexity of the problem is due to the fact that on the one hand there is the mother's right to freely decide to terminate or continue the pregnancy and on the other hand, there is considerable right of the embryo/foetus to life. The whole text of the thesis is interwoven with finding a balance between these competing interests of women and the unborn child based on my own opinion or the opinion of the international judicial body. The aim of this master's thesis is to examine the topic of using style guides in institutional translation, especially in the context of translation in the EU institutions. The theoretical part of the thesis first defines institutional translation and then describes the topic of translation in the EU institutions and of translation manuals and style guides in institutional translation in general and specifically in the EU. The empirical part is focused on the analysis of the Interinstitutional Style Guide as a set of rules and guidelines used in institutional translation in the EU institutions; the IISG is also compared with the grammar and spelling rules of the Czech language in the Czech Internet Language Reference Book by the Czech Language Institute of Czech Academy of Sciences. The thesis examines the observance rate of those rules in the Czech language versions of legal documents published in the Official Journal of the EU from 2004 to 2020. The analysis is conducted in a corpus compiled of EU legislative texts through ApSic Xbench. The results of the analysis are then compared to the occurrence of the examined language phenomena in a corpus of original Czech legal documents. This diploma thesis deals with the issues of numerals in the context of teaching Czech as a foreign language. The theoretical part describes the reasons why numerals are so problematic in the process of teaching and the common methodological recommendations for the teaching of numerals are described here as well. The goal of this thesis is to suggest the most suitable way to teach selected numerals along with the related grammatical cases. Numerals in the chosen textbooks of Czech for foreigners and the analysis of the chosen forms in the CzeSL-SGT learner corpora were analysed for this purpose. The analysis of the corpus helps to define the difficult cases in more detail. Based on the analyses, the worksheets were created for the chosen types of numerals that can be used in the teaching process. These materials also contain recommendations for working with the individual activities.

Diploma thesis "Content issues of teaching civic education through the changes of time (with focus on professional journals dealing with civic education)" is dealing with an evolution of a content of civic education in the 20th and at the beginning of the 21st century on pages of professional-oriented journals. The first part of the diploma thesis deals with the history of civics. It describes the evolution since the establishment of civics in Czechoslovakia until the evolution after the Velvet Revolution in the Czech Republic. The second part of the diploma thesis introduces five journals devoted to a subject of civic education. These are the magazines Časopis československé obce učitelské, Časopis pro občanskou nauku a výchovu, Společenské vědy ve škole, Občanská výchova: časopis pro občanskou výchovu, filosofii, psychologii a logiku a Společenskovědní předměty: čtvrtletník pro pedagogy základních a středních škol.
