Functional characterization of R2R3-MYB activators and repressors as flavonoid transcriptional regulators in poplar

Flavonoids are important and ubiquitous secondary metabolites and are known to participate in various developmental and stress response processes in plants. Common flavonoids include anthocyanins, proanthocyanidins and flavonols. This thesis aims to determine, at the molecular level, how the biosynthesis of flavonoids, in particular the proanthocyanidins, is regulated in poplar. Poplars accumulate large amount of flavonoids and the major flavonoid biosynthetic genes in poplar have been identified. Flavonoid biosynthesis is known to be regulated by MYB transcription factors. Previous work had identified MYB134 as a key regulator of proanthocyanidin synthesis in poplar. Here I describe experiments on five additional genes encoding MYB activators (MYB115 and MYB117), MYB repressors (MYB165 and MYB194), and one bHLH cofactor (bHLH131) as possible flavonoid regulators in poplar. The objective of this work is to determine the in planta functions of these new flavonoid regulators using reverse genetic methods, phytochemical and transcriptome analysis, to identify their target genes and to determine how these transcriptional regulators interact using promoter transactivation and yeast two- hybrid assays. MYB115 was identified as a second proanthocyanidin regulator. Similar to the effects of MYB134, overexpression of MYB115 in poplar led to increased proanthocyanidin content and upregulated flavonoid biosynthesis genes, but reduced the accumulation of salicinoids. Overexpression of repressor type MYBs, MYB165 or MYB194 led to reduced anthocyanin, salicinoid and hydroxycinnamic ester accumulation in leaves, while reducing proanthocyanidin content in roots. Transcriptome analysis demonstrated the downregulation of most flavonoid genes in these transgenics, as well as some shikimate pathway genes, confirming the broad repression function on the phenylpropanoid pathway. By contrast, MYB117 encodes an anthocyanin activator, and was shown to be specific to this branch of the flavonoid pathway. Overexpression of MYB117 in transgenic poplar increased accumulation of anthocyanin in all tissues, resulting in red poplar plants. One bHLH cofactor, bHLH131 was shown to interact with both MYB activators and repressors and required by MYB activators to activate flavonoid gene promoters. This indicate an important role of bHLH131 in the flavonoid biosynthesis. Proanthocyanidin MYB activators, MYB134 and MYB115 could activate each other. This indicates a positive feedback loop of proanthocyanidin MYB activators. Interestingly, repressor MYB165 suppressed expression of other flavonoid MYB repressors including MYB194 and MYB182, which shows a negative feedback loop of MYB repressors. The expression of bHLH131 was also regulated by MYB activators and repressors. These results reveal the complex interaction between these regulators. iii iv Unexpectedly, overexpression MYB134, MYB115 or MYB117 poplars upregulated flavonoid 3’5’-hydroxylase and cytochrome b5 genes, and lead to enhanced flavonoid B- ring hydroxylation and an increased proportion of delphinidin, myricetin Overexpression of flavonoid 3’5’-hydroxylase in poplar confirmed its function in enhancing B-ring hydroxylation. However, overexpression of cytochrome b5 in flavonoid 3’5’-hydroxylase- overexpressing plants did not further increase flavonoid B-ring hydroxylation. Thus its role in flavonoid B-ring hydroxylation remained unclear. These results show that flavonoid MYBs could also alter flavonoid structure. Together, these studies outline the complex regulatory network formed by flavonoid MYB activators and repressors, and bHLH cofactors controlling both flavonoid accumulation and structure. / Graduate / 2020-12-04

Preparação e caracterização de hidrogéis neutros de colágeno aniônico:gelatina:extrato de semente de uva / Preparation and characterization of anionic collagen:gelatin:grape seed extract neutral hydrogels

O desenvolvimento de uma matriz de colágeno associado à gelatina tem potencial como biomaterial devido sua alta biocompatibilidade, capacidade de alterar suas propriedades físico-químicas e estruturais por modificações químicas e a habilidade de formar géis estáveis. Porém, um ponto negativo de sua aplicação está na biodegradabilidade. Desta forma, para reduzir esta degradação, agentes de reticulação, como a proantocianidina (PA), que age formando ligações de hidrogênio as quais estabilizam o complexo proteína-PA, podem ser empregados. Este trabalho teve como objetivo a obtenção de hidrogéis neutros de colágeno aniônico:gelatina:extrato de semente de uva. Para tanto, foram utilizadas duas proporções de extrato, 0,25 e 0,50% e três diferentes tempos de hidrólise alcalina do colágeno, 24, 72 e 120 horas, gerando nove diferentes biomateriais, incluindo os hidrogéis sem tratamento com o extrato. O colágeno foi extraído de tendão bovino e o agente reticulante foi o extrato de semente de uva cujo componente majoritário é a proantocianidina. A caracterização foi feita por termogravimetria (TG), calorimetria exploratória diferencial (DSC), espectroscopia de absorção no infravermelho (FTIR), microscopia eletrônica de varredura (MEV), cinética de absorção de água e ensaios de citotoxicidade in vitro, pelos métodos de difusão em ágar e difusão de extrato em solução (MTT). Estudos de TG mostraram a perda de água em um processo único, representando em média 95% do hidrogel. As curvas DSC mostraram que quanto maior a concentração de extrato, maior é a temperatura de desnaturação, aumentando em média 6,8°C com a adição de 0,50% de extrato, o que indica a eficácia da proantocianidina na reticulação do colágeno. Além disso, notaram-se temperaturas menores para maiores tempos de hidrólise alcalina do colágeno, sendo de 58,4°C, 49,7°C e 46,5°C para hidrogéis preparados com colágeno 24, 72 e 120 horas, respectivamente. A presença do extrato não causou alterações significativas nos espectros FTIR, apenas surgimento das bandas em 1118 e 1288 cm-1 referentes ao anel aromático da proantocianidina, mas gerou mudanças nas estruturas internas dos hidrogéis, visualizadas por MEV, como aumento do número de poros e interconectividade entre eles. A cinética de absorção de água mostrou que o equilíbrio é atingido em aproximadamente 10 minutos, indicando vantagem para a aplicação do hidrogel, já que é obtido rapidamente a partir de sua forma liofilizada, adequada para o armazenamento. Também foi observado que quanto menor é o tempo de hidrólise, maiores são as absorções, variando de 540 até 1360%. Com os ensaios de citotoxicidade foi possível concluir que o hidrogel C24GE50 mostrou-se mais adequado para uma aplicação como biomaterial, com um índice de 92,7% de sobrevivência celular. / The development of a collagen matrix associated with gelatin has potential as biomaterial due to its high compatibility, ability to change its physical-chemical and structural properties by chemical modifications and also the ability to form stable gels. However, a negative point of its application is the biodegradability. Thus, to reduce this degradation, crosslinking agents, such as proanthocyanidin (PA), which acts forming hydrogen bonds which stabilize the PA protein complex may be employed. This project aimed to obtain neutral hydrogels prepared by mixture of anionic collagen: gelatin: grape seed extract. It was used two extract proportions (0.25 and 0.50%) and three different periods of time for collagen alkaline hydrolysis (24, 72, 120 hours), giving nine different biomaterials, including hydrogels without treatment with the extract. The collagen was extracted from bovine tendon and the crosslinking agent was grape seed extract whose major component is the proanthocyanidin. The characterization was done by thermogravimetry (TG), differential scanning calorimetry (DSC), Fourier transform infrared spectroscopy, scanning electron microscopy (SEM), water absorption kinetics and in vitro cytotoxicity assays, by agar diffusion method and diffusion of extract in solution (MTT). TG results showed loss of water in a single process, representing about 95% of the hydrogel.DSC curves showed that the higher the concentration of the extract, the higher the melting temperature, increasing on average 6.8 °C with the addition of 0.50% extract, which indicates the effectiveness of the proanthocyanidin crosslinking of the collagen. Moreover, lower temperatures were observed for longer periods of alkaline hydrolysis of collagen, being 58.4°C, 49.7°C and 46.5°C for preparations with 24, 72 and 120 hours, respectively. The presence of the extract did not cause significant changes in the FTIR spectra, only appearance of bands at 1118 and 1288 cm-1 related to the aromatic ring of proanthocyanidin but led to changes in internal structures of the hydrogels viewed by SEM, showing increased number of pores and interconnectivity between them. The water absorption kinetics showed that the equilibrium is achieved in approximately 10 minutes, indicating advantages in using this hydrogel, which is quickly obtained from its lyophilized form, a suitable form for storage. It was also observed that the shorter the time of hydrolysis, the greater the absorptions, ranging from 540 to 1360%. It was possible to conclude from the cytotoxicity analysis that the C24GE50 is more suitable for application as a biomaterial, with an index of 92.7% cell survival.

O objetivo deste trabalho foi elucidar a interação entre a proantocianidina (PA), um agente natural e não tóxico de ligação cruzada, e o colágeno dentinário tipo I. A hipótese proposta é que o modelamento de uma dentina desmineralizada mais resistente e com melhores propriedades propiciará um substrato mais estável e íntegro para restaurações adesivas. Para isto, o estudo foi dividido em 4 fases: 1) Validação da PA como agente exógeno de ligação cruzada, testando matrizes de dentina desmineralizadas tratadas com extratos a base de PA em teste microflexural para análise do módulo de elasticidade (ME), variando o tempo de exposição (10, 30, 60, 120, 240 min), o tipo de extrato usado (semente de uva-GSE, semente de cacau-CSE, oxicoco-CRE, canela-CNE e açaí-ACE) e a longevidade (imediato, 3, 6 e 12 meses); 2) Estudar parâmetros relacionados às soluções de PA, como solubilidade (água, etanol e acetona), fonte (GSE e CSE) e concentração (0,65%, 3,25%, 6,5%, 15% e 30%), através da mensuração do ME pelo teste micro-flexural de matrizes de dentina desmineralizada; 3) Caracterização da dentina tratada com PA (GSE e CSE), sorção de água, calorimetria e degradação enzimática; 4) Analisar a resistência de união entre a dentina tratada com PA (GSE/CSE) e sistemas de adesivos comerciais (Adapter Single Bond Plus e One Step Plus), variando a concentração (6,5% e 30%), tempo de exposição (1, 10 e 60 min) e longevidade (imediato, 3, 6 e 12 meses). Foram realizadas análises de variâncias e teste de contraste de médias, quando necessário, para todos os ensaios. Os resultados de ME foram influenciados pelo tratamento com PA, dependendo do tipo de extrato usado, exibindo os melhores resultados para o maior tempo de exposição (240 min) para os extratos GSE e CSE. Embora os resultados imediatos com os extratos CRE e CNE não tenham aumentado, estes se mantiveram constantes apos 1 ano de armazenamento, o que não aconteceu para o ACE e os grupos controles. A solubilidade das soluções é influenciada diretamente pelo modo de extração e manufaturação dos extratos, sendo que GSE é solúvel em água destilada e CSE em acetona-água. A efetividade do GSE mostrou ser concentração-dependente, exibindo o maior ME para a concentração de 30%. A dentina tratada com agentes a base de PA foi mais resistente contra a colagenase, obteve uma menor sorção de água e um aumento significativo na temperatura de desnaturação quando comparada ao grupo controle. O GSE foi capaz de aumentar a resistência de união (RU) imediata para ambos os sistemas adesivos, mantendo-se constante para o Adapter Single Bond Plus ao longo do tempo. Para o One Step Plus todos os grupos mostraram diminuição da RU após 1 ano, independente do tempo de exposição. Quando a dentina foi tratada por 1 minuto, os valores de RU para o grupo 30% de GSE foram maiores do que os demais e mantiveram-se constante pelo período de armazenamento. O efeito dos agentes ricos em PA mostrou ser concentração e tempo dependente, porém uma dentina mais resistente e estável ao longo do tempo é possível de ser obtida, levando-se em conta a estrutura química, conteúdo de PA, solubilidade e outros fatores inerentes ao extrato / The aim of this study was to elucidate the interaction between proanthocyanidin based extracts (PA), a natural and non-toxic cross-linker, and type I dentin collagen. The hypothesis is that if a stronger and more stable collagen layer is chemically engineered, the resultant hybrid layer will be stronger and less prone to degradation. So, this research was divided into four phases: 1) PA validation as an exogenous cross-linking agent, testing the elastic modulus (E) of demineralized dentin treated with various PA based extracts (grape seed-GSE, cocoa seed-CSE, cranberry-CRE, cinnamon-CNE, and açai berry-ACE), exposure times (10, 30, 60, 120, 240 min), and long term effectiveness (immediate, 3, 6 and 12 months); 2) Evaluation of different parameters related to PA solutions, like solubility (water, ethanol and acetone), manufacturer (GSE and CSE) and concentration (0.65%, 3.25% 6.5%, 15% and 30%), E was also obtained by micro-flexural strength measurements of demineralized dentin matrix, 3) Characterization of PA treated dentin (GSE and CSE): water sorption, calorimetry and enzymatic degradation and 4) Analyze the bond strength ( BS) of PA treated dentin (GSE / CSE) with commercial adhesives systems (Adapter Single Bond Plus-SB and One Step Plus-OS), varying concentration (6.5% and 30%), exposure time (1, 10 and 60 min) and long-term effectiveness (immediate, 3, 6 and 12 months). Data were statistically analyzed at a 95% confidence interval. GSE and CSE extracts showed a time-dependant effect and were able to improve and stabilize the E of the organic matrix. CRE and CNE extracts were able to maintain the E of collagen matrices constant after 12 months artifical saliva storage. ACE and controls groups showed no effect over dentin organic matrix, did not prevent degradation and consequently reduced the mechanical properties. Herbal extraction process and other pharmacognostic parameters have an important influence on extract solubility as well as constitution, therefore GSE is better dissolved in water and CSE in acetone-water. GSE effect on demineralized dentin is concentration-dependant, with highest E values at 30% GSE concentration. Dentin treated with PA-based agents was more resistant against enzymatic degradation, less susceptible to water sorption and showed a higher denaturation temperature. As compared to the controls, GSE stabilized the dentin-resin bond strength values over a period of one year for both adhesives, however better results were achieved with SB. CSE showed less predictable results depending on the adhesive system used. One minute treatment with 30% GSE was capable of increasing short-term BS and stabilizing it for at least 6 months. Increased mechanical properties and stability of dentin matrix can be achieved by the use of PA-rich collagen crosslinkers regarding inherent characteristics of the natural compounds as chemical structure, solubility and PA content.

So, this research was divided into four phases: 1) PA validation as an exogenous cross-linking agent, testing the elastic modulus (E) of demineralized dentin treated with various PA based extracts (grape seed-GSE, cocoa seed-CSE, cranberry-CRE, cinnamon-CNE, and açai berry-ACE), exposure times (10, 30, 60, 120, 240 min), and long term effectiveness (immediate, 3, 6 and 12 months); 2) Evaluation of different parameters related to PA solutions, like solubility (water, ethanol and acetone), manufacturer (GSE and CSE) and concentration (0.65%, 3.25% 6.5%, 15% and 30%), E was also obtained by micro-flexural strength measurements of demineralized dentin matrix, 3) Characterization of PA treated dentin (GSE and CSE): water sorption, calorimetry and enzymatic degradation and 4) Analyze the bond strength ( BS) of PA treated dentin (GSE / CSE) with commercial adhesives systems (Adapter Single Bond Plus-SB and One Step Plus-OS), varying concentration (6.5% and 30%), exposure time (1, 10 and 60 min) and long-term effectiveness (immediate, 3, 6 and 12 months). Data were statistically analyzed at a 95% confidence interval. GSE and CSE extracts showed a time-dependant effect and were able to improve and stabilize the E of the organic matrix. CRE and CNE extracts were able to maintain the E of collagen matrices constant after 12 months artifical saliva storage. ACE and controls groups showed no effect over dentin organic matrix, did not prevent degradation and consequently reduced the mechanical properties. Herbal extraction process and other pharmacognostic parameters have an important influence on extract solubility as well as constitution, therefore GSE is better dissolved in water and CSE in acetone-water. GSE effect on demineralized dentin is concentration-dependant, with highest E values at 30% GSE concentration. Dentin treated with PA-based agents was more resistant against enzymatic degradation, less susceptible to water sorption and showed a higher denaturation temperature. As compared to the controls, GSE stabilized the dentin-resin bond strength values over a period of one year for both adhesives, however better results were achieved with SB. CSE showed less predictable results depending on the adhesive system used. One minute treatment with 30% GSE was capable of increasing short-term BS and stabilizing it for at least 6 months. Increased mechanical properties and stability of dentin matrix can be achieved by the use of PA-rich collagen crosslinkers regarding inherent characteristics of the natural compounds as chemical structure, solubility and PA content.

Apesar da capacidade regenerativa da dentina já ser bem estabelecida, este processo pode ser insuficiente no caso de injúrias decorrentes de traumas e/ou lesões cariosas extensas, podendo levar à exposição pulpar e perda de sua vitalidade. Assim, pesquisas recentes no campo da engenharia tecidual têm identificado materiais e substâncias que poderiam ser utilizadas como biomodificadores da dentina e auxiliares na regeneração do complexo dentina-polpa. Entre eles estão os extratos ricos em proantocianidina, um composto fenólico bioativo presente no extrato de semente de uva (GSE). Assim, o objetivo deste trabalho foi avaliar o potencial estimulatório de quatro concentrações crescentes do GSE na atividade funcional de células odontoblastoides. Foram utilizadas células de camundongo da linhagem MDPC-23, cultivadas em garrafas de cultura até a subconfluência. Em seguida, as células foram cultivadas em placas de 24 poços em uma concentração de 104/poço e divididas em cinco grupos: células sem adição do GSE, células + 0.1 µg/mL de GSE; células + 1 µg/mL de GSE; células + 10 µg/mL de GSE; células + 20 µg/mL de GSE. Após 3, 7 e 10 dias, foram analisados os seguintes parâmetros: proliferação e viabilidade celular, detecção e atividade de fosfatase alcalina, quantidade de proteína total, detecção e quantificação de nódulos mineralizados, expressão quantitativa dos genes Alp, Col1a1 e Dmp1 por meio de PCR em tempo real e sua expressão proteica correspondente por meio de imunolocalização. Os dados obtidos foram analisados quanto à normalidade e submetidos aos testes estatísticos ANOVA e Kruskal-Wallis, com nível de significância estabelecido em 5%. Os resultados mostraram proliferação e viabilidade celular com as concentrações mais baixas de GSE (0,1 e 1 µg/mL), assim como a atividade de síntese de proteínas totais e da fosfatase alcalina. A deposição de nódulos mineralizados foi significativamente maior com a concentração de 1 µg/mL do extrato. Esta mesma concentração favoreceu a expressão quantitativa dos genes Alp, Col1a1 e Dmp1 e a secreção das proteínas correspondentes vistas por imunolocalização. Conclui-se que baixas concentrações do extrato de semente de uva podem influenciar e favorecer a atividade de células odontoblastoides, contribuindo para a regeneração dentinária, devido à suas características antioxidantes e biomineralizadoras / The process of dentinogenesis generally involves several steps of cell differentiation, culminating with an extracellular matrix very similar to bone tissue. Dentin regeneration may be necessary in cases of pulp exposure, root resorption and carious lesions. Studies on functional regeneration of lost tissues are being carried on recently, based on the presence of cells, scaffolds and/or substances that induce cell proliferation and differentiation. Among the several substances which can interact with cell metabolism are the extracts rich in proanthocyanidin, bioactive phenolic compounds present in fruits, vegetables and seeds. The purpose of this project is to evaluate the stimulatory potential of four different concentrations of grape seed extract (GSE) in the functional activity of odontoblast-like cells. MDPC-23 cell line was seeded in culture flasks until subconfluence followed by cell seeding in 24-well culture plates in the concentration of 104/well and divided in five groups: 1) cells without GSE, 2) cells + 0,1 µg/mL of GSE; 3) cells + 1 µg/mL of GSE; cells + 10 µg/mL of GSE and cells + 20 µg/mL of GSE. After 3, 7 and 10 days, there were evaluated the following parameters: cell proliferation and differentiation, ALP detection and activity, total protein content, mineralization detection and quantification, as well as the expression of genes Alp, Col1a1 and Dmp1 through Real Time PCR and their corresponding proteins by means of immunolocalization. Data were analyzed by ANOVA and Kruskal-Wallis statistical tests, with significance level set at 5%. The results demonstrated cell proliferation and viability with low GSE concentrations (0,1 and 1 µg/mL), as well as total protein content and ALP activity. Deposition of mineralized nodules was significantly increased with the GSE concentration of 1 µg/mL. The same concentration favoured the expression of genes Alp, Col1a1 and Dmp1 as well as the corresponding proteins. It is concluded that low concentrations of grape seed extract may influence functional activity of odontoblast- like cells, contributing to dentin regeneration by means of its antioxidant and biomineralizing properties

Efeitos comparativos de diferentes géis à base de cranberry e proantocianidina sobre a dentina submetida à erosão: estudo in vitro / In vitro effect of Cranberry and proanthocyanidin on dentin erosion

Considerando que a Proantocianidina e outros componentes do Cranberry possam inibir as metaloproteinases da dentina quando esta é exposta a ação de agentes erosivos e tal fato pode minimizar o desgaste dentinário, o presente trabalho tem como objetivo avaliar o efeito protetor in vitro de géis à base de extrato de Cranberry e Proantocianidina em diferentes concentrações, aplicados sobre a dentina posteriormente submetida à erosão. Para tal, 150 espécimes de dentina bovina (4 x 4 mm) foram confeccionados e igualmente divididos em 10 grupos: G1- gel de clorexidina a 0,012%; G2- gel placebo; G3- gel a base de extrato de Cranberry a 0,05%; G4- gel a base de extrato de Cranberry a 1%; G5- gel a base de extrato de Cranberry a 5%; G6- gel a base de extrato de Cranberry a 10%; G7- gel a base de Proantocianidina 0,05%; G8- gel a base de Proantocianidina 1%, G9- gel a base de Proantocianidina 5%; G10- gel a base de Proantocianidina 10%. Os diferentes géis foram aplicados uma única vez sobre os espécimes antes do primeiro desafio erosivo durante 5 minutos. Em seguida, os espécimes foram submetidos a 3 ciclagens erosivas seguidas (imersão em bebida à base de Cola por 5 minutos seguido de imersão em saliva artificial por 3 horas) por dia, durante 5 dias. Após as 3 cilclagens erosivas diárias, os espécimes foram mantidos em saliva artificial por 15 horas. A perfilometria foi utilizada para quantificar o desgaste dentinário (&#x3BC;m). Os dados foram analisados pelo teste ANOVA seguido do teste de Fisher (p<0,05). Os resultados (G1: 15,6 ± 2,4b; G2: 23,1 ± 1,4e; G3: 15,1 ± 2,3b; G4: 20,4 ± 1,9cd; G5: 19,0 ± 2,1ac; G6: 18,7 ± 1,4a; G7: 18,2 ± 1,6a; G8: 21,1 ± 1,2d; G9: 19,2 ± 2,6ac; G10: 15,2 ± 2,5b) mostraram que o gel de Cranberry a 0,05% (G3) e gel de Proantocianidina 10% (G10) apresentaram eficácia estatisticamente significante na redução da erosão dentinária em comparação aos outros grupos analisados. / Considering that Proanthocyanidin and other Cranberrys components might inhibit dentin metalloproteinases exposed to erosive agents and it can prevent dental wear, the aim of this in vitro study was to evaluate the effect of Cranberry and Proanthocyanidin gels used in different concentrations on dentin before an erosive challenge. For this purpose, 150 bovine root dentin blocks (4x4mm) were treated by different gels and randomly divided into 10 groups: G1- 0.012% Chlorhexidine Gel (positive control), G2- Placebo gel with no active principle (negative control), G3- 0.05% Cranberry gel, G4- 1% Cranberry gel, G5- 5% Cranberry gel, G6- 10% Cranberry gel, G7- 0.05% Proanthocyanidin gel, G8- 1% Proanthocyanidin gel, G9- 5% Proanthocyanidin gel and G10- 10% Proanthocyanidin gel. The gels were applied over specimens once before the first erosive challenge for 5 minutes. After that, the blocks were subjected to 3 sequential erosive cycles (cola drink for 5 minutes and artificial saliva for 3 hours, each) per day, during 5 days. After 3 daily erosive cycles, the blocks were maintained in artificial saliva for 15 h. Profilometry was used to quantify the dentin wear (&#x3BC;m). Data were analyzed by ANOVA and Fisher\'s test (p<0.05).
