Total Syntheses of (+)-Geldanamycin, (-)-Ragaglitazar, and (+)-Kurasoin A and Phase-Transfer-Catalyzed Asymmetric Alkylation

Geldanamycin possesses various biological activities as seen in the NCI 60 cell line panel (13 nM avg., 70 nM SKBr-3 cells). The predominant mode of action providing these unique results arises from the ability of geldanamycin (GA) to bind to the chaperone heat shock protein 90 (Hsp90). Despite its complicated functionality, the first total synthesis of GA was accomplished, which included two new reactions developed specifically to address the stereochemical features. The final step in the synthesis of GA was a demethylation-oxidation sequence to generate the desired para-quinone. This step could only be accomplished with HNO3/AcOH, producing GA in 5% yield. A GA model study, which closely resembled the aromatic core, was extensively investigated to solve this critical oxidation issue. A protected hydroquinone model compound was determined to be the optimum choice. Using Pd in the presence of air with a 1,4-hydroquinone provided the desired para-quinone quickly and nearly quantitatively in 98% yield. This study formulated the recipe of success for para-quinone formation of GA and future synthetic analogs. Asymmetric glycolate alkylation has been developed using phase-transfer-catalysis (PTC). Diphenylmethoxy-2,5-dimethoxyacetophenone with trifluorobenzyl cinchonidinium catalyst and cesium hydroxide provided alkylation products at —35 °C in high yield (80-99%) and with excellent enantioselectivity (up to 90% ee). Useful α-hydroxy products were obtained using bis-TMS peroxide Baeyer—Villiger conditions and selective transesterification. The intermediate aryl esters can be obtained with >99% ee after a single recrystallization. The newly developed PTC glycolate alkylation was applied to the asymmetric syntheses of ragaglitazar and kurasoin A. Ragaglitazar is a potent antihyperglycemic and lipid modulator, currently in phase II clinical trials. Kurasoin A is a relatively potent protein farnesyltransferase (PFTase) inhibitor with an IC50 value of 59.0 micromolar. PTC glycolate alkylation was optimized to provide 4-benzyloxy glycolate intermediates in excellent overall yield and with 96% ee after recrystallization. Ragaglitazar was then synthesized after considerable experimentation to provide the potent lipid modulator with yields and enantiopurity rivaling the best-known routes produced by industry standards. Kurasoin A was produced through an α-triethylsiloxy Weinreb amide to provide the highest overall yielding route to this PFTase inhibitor currently disclosed.

Målet med detta examensarbete är att ersätta den tidigare lösningen av sensorfingret med andra komponenter, eller hitta en annan sensor med motsvarande funktion. Arbetet har begränsats av att ESAB vill ha ett mekaniskt finger för användarvänlighetens skull. Efter en förundersökning av olika typer av sensorer har vi valt att använda oss av trådtöjningsgivare som sensorer till det mekaniska sensorfingret. I och med detta val behövde vi även komma med ett teoretiskt förslag på en mekanisk upphängning av sensorfingret. Vi har byggt upp två separata Wheatstonebryggor med varsin instrumentförstärkare (INA125). Utsignalen från bryggorna är relativt liten och behövde förstärkas cirka 2000 gånger. Nollbalansering av bryggorna har skett manuellt med potentiometrar men vi har även gett ett förslag på en lösning där nollbalansering kan ske med ett enkelt knapptryck. Knappen är placerad på upphängningen på ett sådant sätt att man inte kan trycka oavsiktligt på knappen. Resultatet av arbetet är en uppkoppling på en experimentplatta samt förslag på hur upphängningen och sensorfingret designmässigt kan se ut. För vidare arbetsgång behövs en prototyp för att testa livslängden och ta reda på vilken känslighet som är mest optimal. / The goal with this degree project was to find a solution with different components to the existing design, or find a sensor with similar function that could replace the previous design of the sensor finger. Our work has been limited by ESAB due to the user friendly mechanical design of the previous sensor finger. After examining different sensors we choose to use strain gages as sensors for the mechanical sensor finger. With this choice we also needed to suggest a theoretical design for a mechanical attachment of the sensor finger. We have designed two separate Wheatstone bridges with separate instrumental amplifiers (INA125). The output signals from the bridges are relatively small and need to be amplified around 2000 times. The zero balancing of the bridges has been done manually with potentiometers but we have also suggested a solution where the zero balancing can be performed by a push-button. The button is placed where you cannot push it unintentionally. The result of this degree project is a circuit on a veroboard, a designing proposal for the attachment and the sensor finger. In order to further develop this design it would be necessary to build a prototype to evaluate its life span and find the optimal sensitivity.

Neste estudo, geramos ensaios de espectrometria de massa para detecção de 111 mutações nos genes RET, BRAF, NRAS, HRAS, KRAS, PIK3CA e AKT1 e avaliamos inúmeras linhagens celulares e tumores tiroidianos. Mostramos que as mutações dos genes BRAF e RAS refletem prognósticos distintos e que as mutações BRAF são altamente prevalentes em câncer metastático. Mutações dos genes PIK3CA e AKT1, esta última sendo reportada pela primeira vez no câncer de tiróide, são relativamente frequentes neste câncer. Avaliamos ainda a função do microRNA let-7 neste câncer. Mostramos que a ativação do rearranjo RET/PTC3 em células de tiróide PCCl3 reduz a expressão de let-7. Além disso, a transfecção deste microRNA em células TPC-1, que apresentam o rearranjo RET/PTC1, inibe a fosforilação de ERK, o crescimento celular e modula a expressão de genes do ciclo celular e diferenciação. Estes dados contribuem na aplicação de terapias dirigidas a efetores das vias PI3K e MAPK no câncer de tiróide, além de salientar o envolvimento do miRNA let-7 como um gene supressor tumoral nesta doença. / In this study, we designed an assay panel for genotyping 111 mutations by mass spectrometry in RET, BRAF, NRAS, HRAS, KRAS, PIK3CA, AKT1 and other related genes in many thyroid cancer cell lines and tumors. We show that patients with BRAF and RAS mutations have distinct prognosis and that BRAF mutations are highly prevalent in metastatic thyroid cancers. Mutations of PIK3CA and AKT1, the latter not previously described in this disease, are comparatively frequent in thyroid cancers. In addition, we evaluated the role of let-7 microRNA in this cancer. We show that RET/PTC3 activation in PCCl3 thyroid cells reduces let-7 expression. Moreover, transfection of let-7 in TPC-1 cells, which harbor RET/PTC1 rearrangement, inhibits MAPK activation, reduces cell growth and modulates the expression of cell cycle and differentiation genes. These findings may contribute to the design of therapies directed at MAPK and PI3K effectors and to highlight the function of let-7 as tumor suppressor gene in thyroid cancer.

Durante o preparo químico-cirúrgico são utilizados instrumentos e substâncias químicas, que constituem um binômio indivisível e necessário para alcançar a modelagem e a sanificação dos canais radiculares. Assim, propõe-se com este trabalho avaliar in vivo, por meio de método molecular de PCR quantitativo, baseado em DNA (qPCR), a eficiência do preparo químico-cirúrgico empregando como agente de irrigação o Hipoclorito de Sódio (NaOCL) a 2,5% ou o Gel de Endo PTC associado ao Hipoclorito de Sódio a 1,0% na redução bacteriana de canais radiculares de dentes portadores de periodontite apical primária. Foram selecionados 30 pacientes portadores de infecção endodôntica primária, totalizando 30 dentes, com rarefação óssea periapical visível na radiografia, sem tratamento endodôntico prévio. Os pacientes foram divididos de forma randomizada em dois grupos distintos, de acordo com a substância química auxiliar utilizada durante a instrumentação, NaOCL 1% + Endo-PTC leve ou NaOCL 2,5%. Em todos os casos empregou-se instrumentos Reciproc R40 ou R50 e as coletas foram realizadas antes (S1) e após o prepare químico-cirúrgico (S2). A análise de aderência foi realizada por meio do teste de Kolmogorov-Smirnov, as análises intragrupo foram realizadas com teste de Wilcoxon para amostras relacionadas e as comparações entre os dois grupos foram realizadas com o teste de Mann-Whitney, para a análise quantitativa de bactérias. Em ambos os grupos, houve diminuição significativa no número de bactérias entre S1 e S2 (p<0,05). No grupo NaOCL 1% + Endo-PTC leve, houve redução de 3,7x105(S1) para 5,7x104 (S2). No grupo NaOCl 2,5%, redução de 1,3x105 (S1) para 1,1x104(S2). Thus, the purpose of this study is to evaluate in vivo, the efficiency of the chemomechanical preparation using as the irrigant agent 2,5% sodium hypochlorite and Endo-PTC gel, associated to 1% sodium hypochlorite, to assess the bacterial reduction of root canals of teeth with primary apical periodontitis, using a molecular quantitative method DNA-based - polymerase chain reaction (qPCR). Were selected 30 patients with primary infection totaling 30 teeth, with visible periapical bone rarefaction on the radiography, without previous endodontic treatment. Patients were randomly divided into two distinct groups according to the auxiliary chemical substances used during the instrumentation, 1% sodium hypochlorite associated with Endo-PTC gel or 2,5% sodium hypochlorite.
