Déterminismes génétiques et environnementaux des composantes architecturales à l'origine de l'élaboration de la forme du rosier buisson / Genetic and environmental determinisms of the architectural components that gave rise to the design of the bush rose shape

La forme, et donc l’architecture d’une plante ornementale en potcomme le rosier buisson détermine sa qualité visuelle et peut êtrecontrôlée via la création variétale et/ou l’utilisation de techniquesculturales comme l’application de facteurs environnementaux.Cependant, ces méthodes sont appliquées empiriquement etprennent rarement en compte l’interaction Génotype x Environnement(GxE). Dans un premier temps, cette étude a montré que, parmiles trois techniques culturales testées sur cinq à huit cultivarsde rosier buisson de formes contrastées, la restriction hydrique modifiait fortement l’architecture avec un effet compactant. Une forteinteraction GxE a également été montrée avec trois réponses architecturales,de faible à forte, pouvant s’expliquer en partie par desconcentrations différentes en cytokinines et acide salicylique entregénotypes.Dans un deuxième temps, l’analyse génétique par cartographiede l’architecture du rosier à partir de deux populationsconnectées en ségrégation et remontantes a montré que la plupartdes caractères architecturaux étudiés étaient contrôlés par un ouplusieurs QTLs à effets faibles dont certains étaient infl uencés parl’environnement et/ou par le fond génétique. L’introgression detels caractères dans d’autres fonds génétiques par exemple seradonc vraisemblablement diffi cile. Cette étude a souligné l’importancede prendre en compte les interactions GxE et QTLxE pourmieux raisonner les programmes de création/sélection, mais aussipour l’utilisation des techniques culturales dans le but de contrôler l / The shape and therefore the architecture of a potted ornamentalplant such as bush rose determines its visual quality. Firstly, among three cultivationtechniques evaluated on fi ve to eight rose bush cultivars withcontrasted shapes, water restriction highly modifi ed the architectureleading to more compact plants. A high GxE interaction was revealedwith three architectural responses ranging from low to high. Theywere partly explained by different cytokinins and salicylic acidconcentrations between genotypes.Secondly, the genetic analysisby mapping of the bush rose architecture based on two connectedrepeat-blooming populations in segregation showed that most ofthe architectural traits were controlled by one or several QTLs withlow effects. Some of them were infl uenced by the environmentand/or by the genetic background. The introgression of such traitsin different genetic backgrounds for example will likely be diffi cult.This study underlined the importance of taking into considerationthe GxE and QTLxE interactions to better orientate breedingprograms but also to better use cultivation techniques in order to control plant architecture.

This master thesis deals with the mapping and analysis of Russian-Georgian relations in the period between 2004-2011. It focuses primarily on the development of relations after the onset of the new Georgian President Mikhail Saakashvili and after his transformation of Georgian Policy. The work is divided into chronological historical axis of events, from the history of Georgia and Georgian people, the author gets to the problems of August war in 2008. The main objective of this work is to find the causes of the war between Russia and Georgia, to map out its long-standing hostile relations and try to find its solutions. Starting point of this work is the assumption that Georgia and Russia have always been nations culturally very similar and therefore its friendly relations are essential for the regulation of conflicts on Georgia's borders. Russia as a powerful neighbor, has always had a superiority over Georgia. Therefore, in subsequent chapters is enhanced the need for intervention by international organizations such as NATO, OSCE, EU, etc. issues in Russian-Georgian conflict, in order to offset the forces of power on both sides. Syftet med uppsatsen är att undersöka vilken didaktiska potential som kan finnas i Umberto Ecos roman Rosens namn (1980) i gymnasieskolans svenskundervisning. Ansatsen är litteraturvetenskaplig och rör dels hur elever kan tänkas lära sig hantera litterära texters egenart, dels elevers användning av genreteori för att förstå romanen. För att utforska det första ämnet används Viktor Sklovskijs teori om automatisering, främmandegöring och medvetet försvårad form. Det andra ämnet utgår i huvudsak från Tzvetan Todorovs artikel ”Kriminalromanens typologi”. Det visar sig att romanens didaktiska potential är stor om eleverna ges god vägledning. Romanens omfattande användning av latin och andra främmande språk är en tillgång vid tillämpning av Sklovskijs teorier om främmandegöring, då det finns gott om utrymme för tolkning och reflektion. Även genreanalysens didaktiska potential är hög eftersom romanen på både typiska och otypiska vis anknyter till spänningslitteraturen. / The aim of the paper is to examine what didactic potential may exist in Umberto Eco’s novel The Name of the Rose (1980) in the education of the Swedish language for the upper secondary school. It is approached from a literary scientific angle and is in part about how pupils can learn how to manage the uniqueness of the literary texts and in part how pupils can use genre theory to understand the novel. To explore the first subject Viktor Shklovsky’s theories about automatism, defamiliarisation and roughened form are used. The other subject is mainly studied with Tzvetan Todorov’s article “The Typology of Detective Fiction” as a starting point. It is deduced that the didactic potential of the novel is great if the pupils are given good guidance. The novel’s extensive use of Latin and other foreign languages is an asset when using Shklovsky’s theories, because there is a lot of room for interpretation and reflection. The genre analysis too has great didactic potential because the The Name of the Rose relates to suspense novels in both typical and atypical ways.

The oceans are essential for humanity, yet these systems continue to be degraded and suffer from pollution, habitat destruction and overexploitation. The Costa Rican Gulf of Nicoya is a productive gulf that is fished by both large-scale industrial fisheries and small-scale artisanal fishermen. However, the gulf is profoundly overfished and there has been a shift in both the type of species and size of fishes being targeted, which has had devastating economic and ecological effects in the region. To improve their livelihoods, artisanal fishermen have together with governmental agencies instituted partially protected marine protected areas, known as AMPRs which are intended to give artisanal fishermen exclusive fishing rights and to better the health of the ecosystems. However, these AMPRs can vary greatly within and between each other in aspects such as what types of fishing gear is allowed. To investigate how fishing techniques impact the health of the fish stocks and the wellbeing of fishermen within an AMPR, the size of a target fish, Spotted Rose Snapper (Lutjanus guttatus) and the number of undesired fish caught, was compared based on the number of approved fishing methods such as nets, longlines and commercial scuba diving, between different zones. This was conducted by identifying and measuring catch in collaboration with the National University of Costa Rica, who contributed with databases and field assistance, and artisanal dead bait longline fishermen (bottom long line) in the Paquera-Tambor AMPR. The results showed that in areas where fishing gear is more restricted, the mean weight of Spotted Rose Snapper was higher, but the relative proportion bycatch was not different. Furthermore, there was no difference in legal capture or breeding lengths between areas with high and low restriction of fishing gear. This indicates that the effects of overfishing are less predominant in zones where fewer fishing methods are permitted. The restriction of fishing methods could increase biodiversity and size in fish by exploiting less cohorts of populations. The preservation of size in fish species is very important regarding reproductive success, and it is therefore paramount that larger individuals are protected. This could be accomplished by for example, restricting the amount of allowed fishing gear or introducing maximum catch sizes. However, an improvement of the fish stocks, and therefore human wellbeing cannot be achieved in the Gulf of Nicoya without a revision of the strategies of two influential governmental bodies, the Costa Rican coast guard and the Costa Rican Institute of Fisheries and Aquaculture.

This project investigas humeral torsion and activity-related change in the human upper limb. Increased humeral torsion angles have been identified in the professional throwing athlete and may be associated with strenuous activity. The nature of humeral torsion as an osteogenic response to the strain environment is investigated to identify its role in the behavioural morphology of the upper limb. These physical manifestations of strenuous physical activity provide an insight into the make-up of medieval armies prior to the establishment of standing armies. Populations analysed include two blade-injured samples, Towton and a subsample of blade-injured men from the Priory of St. Andrew, Fishergate, York. The men from the Mary Rose, a Tudor warship are also investigated. Other samples analysed include the rural sites of Wharram Percy and Hickleton, the urban cemeteries from the Priory of St. Andrew, Fishergate,York and the leprosarium of Sts. James and Mary Magdalene, Chichester, the modern cadaver-based Terry collection and non-human primates, Gorilla sp., Pan sp., Pongo sp., and Macaca sp.. Measurement of the humeral torsion angle and external measurements and indices of architecture, articulations and robusticity are employed. Cross-sectional geometric properties are investigated using CT imaging of the paired humeri from a sub-sample of blade-injured individuals and a comparative sample of those who were not. Bilateral asymmetry is investigated to identify the role of plasticity within the humerus and to reveal aspects of limb dominance. The results are compared with non-human primate species to obtain insight into inter-species differences. Results indicate the humeral torsion is not ontogenetically constrained, but is highly variable between and within populations, individuals and even between sides. Biomechanical analyses indicate that in the Towton population, humeral torsion may serve as part of a two-stage adaptation, in which the architecture is modified to enable greater biomechanical efficiency in distributing strain, reducing the need of increased cortical thickness. Changes in humeral torsion related to strenuous activity have been identified, although in the blade-injured samples it is decreased torsion angles, w hile in the comparative sample it is increased torsion angles that significantly correlate with limb hypertrophy. Humeral torsion appears to be influenced by other measurementd of humeral architecture, specifically, the amount of anterior bowing and anterior curvature to the distal humeral shaft. This work demonstrates the need for individual rather than population-based analyses, as the heterogeneity within population samples obscures individual variation in activity patterns. This analysis provides baseline data for typical populations of the Middle Ages. From this, it is then possible to investigate the individual within this baseline, to identify those who stand out from their samples through habitual, strenuous activity patterns. Movement patterns identified related to warfare include those consistent with the use of the longbow in the Towton sample and the use of a sword in the Fishergate blade-injured sample. <strong>Platforms for Molecular Mechanisms and Improvement in Subcutaneous Injection of Biotherapeutics</strong>

<p>Biotherapeutics, such as monoclonal antibodies (mAbs), represent a primary mechanism for treatment of human disease, and there has been a steady increase in Food and Drug Administration approvals since the first one in 1982. Subcutaneous (SC) injection of protein-based therapeutics is a convenient and clinically established drug delivery method that increases the convenience and reduces cost compared to other delivery methods. However, progress is needed to optimize bioavailability via this route. This dissertation describes the methods for evaluation of mass transport of high molecular weight proteins, such as mAbs, following SC injection using <em>in vitro</em> and <em>ex vivo</em> modeling developed to describe the factors relevant for optimal distribution prior to uptake into systemic circulation. The first chapter describes a novel collagen and hyaluronic acid (HA) based hydrogel for investigation of macromolecule transport based on the physiochemical properties of the diffusing molecule and the tissue matrix. This initial study demonstrated that, in collagen alone, collagen combined with HA, and HA alone, the molecules demonstrated different transport paradigms dependent primarily on molecule size, matrix viscosity, and electrostatic charge, respectively. This showed that the local tissue heterogeneity and therapeutic properties could be determining factors for molecule transport and bioavailability. The second, third, and fourth chapters describe two novel platforms for the investigation of injection plume formation in SC tissue utilizing three-dimensional X-ray tomography. Injection plume analysis has been studied comprehensively in the context of insulin transport using co-injection of radiopaque dyes to track the protein distribution. However high molecular weight therapeutics have vastly different physiochemical properties than insulin and are injected under different rates, concentrations, volumes, and viscosities due to dosing considerations. To address the gap mAb distribution, we first developed a novel protein conjugated to an x-ray contrast agent to directly track injection plume formation and investigated the effects of injection rate and tissue location through injections into ex vivo porcine tissue, described in chapters three and four. Ex vivo tissue analysis showed that the rate did not influence the distribution, however, plume volume was lower in porcine belly compared to neck tissue. Whereas porcine tissue is an excellent model to represent the structural features of human injection, the large heterogeneity between animal subjects and collected samples is a disadvantage. Therefore, the fourth chapter describes the fabrication of a gelatin hydrogel-based injection platform representing the dermal and subcutaneous tissue layers for controlled injection plume analysis. In summary, all three models represent useful platforms for the assessment of macromolecular mass transport, pharmaceutical autoinjector performance, as well as the potential impact of tissue properties and intersubject heterogeneity on plume formation. Overall, the findings in these studies might better inform drug designers and clinicians on how to most optimally engineer an injection to deliver the most efficient patient outcomes through better dosing and increased cost savings. </p>
