A Comparison of Two Immunoturbidimetric Assay Methods for Serum Amyloid A in Cats.

The analysis of acute phase protein serum amyloid A (SAA) has recently been brought into clinical use in veterinary medicine. Some of the difficulties with incorporating the SAA method in clinical practice have been the expensive and rather large equipment required for the method. Due to these difficulties only larger clinics can afford to use the SAA analysis. The company Equinostic has recently developed a smaller instrument that costs one-tenth of a larger instrument. The instrument is named EVA1 and has so far only been used to analyze SAA in horses. The aim of this study was to investigate if the EVA1 instrument could be used to analyze SAA in cats. This study included 24 serum samples from cat, which were first analyzed twice on the EVA1 instrument and then sent to the Strömsholm Referral Animal Hospital in Sweden where they reanalyzed the samples using a validated reference method. Both instruments are based on an immunoturbidimetric assay. The correlation between the two instruments was good (r=0.97) but the EVA1 instrument showed constantly lower results than the reference method. The difference between the duplicates when analyzed on the EVA1 instrument was larger than expected. The conclusion is that EVA1 could be used to analyze SAA in cats. However, before it could be used clinically in veterinary practice an extended study is recommended.

Introduction: Available data suggest an association between presence of secondary (AA) amyloidosis and MCP-1 (monocyte chemoatracttant protein-1) and MIP-1alpha (macrophage inflammatory protein-1 alpha) genes polymorphisms. Some studies have also shown an impact of polymorphisms in exon 3 of SAA 1 (serum amyloid A 1) gene on the incidence of AA amyloidosis in different populations. Methods: The incidence of single genotypes MCP-1, MIP-1alpha and SAA 1 genes was investigated. Serum levels of SAA, MCP-1 and MIP-1alpha were measured and potential relation between serum levels and genotypes were analyzed. All examinations were performed in patients with AA amyloidosis (43), rheumatoid arthritis (RA) without amyloidosis and healthy control group (100). Results: Significantly more frequent occurrence of 1.1/1.1 genotype in SAA 1 was recorded in AA amyloidosis group compared to RA group as well as in control group (p<0,001). No statistically significant differences in distribution of another genotypes were found. Distribution of neither 1.1/1.1 genotype nor another ones did not vary among RA group and control group. No significant difference in distribution of another examined genotypes was recorded among all three groups. Nos últimos anos, nosso grupo de pesquisa vem descrevendo vários efeitos da SAA em células do sistema imune no que diz respeito à expressão e liberação de citocinas pró-inflamatórias. Neste estudo centramos nossa atenção na verificação dos efeitos da SAA sobre células mononucleares. Para isto, usamos três modelos experimentais. Em murinos, descrevemos a habilidade da SAA induzir a produção de NO por macrófagos peritoneais e, com uso de animais knockout para TLR4, sugerimos que SAA seja um ligante endógeno do TLR4. Em células mononucleares de sangue periférico humano, a SAA induz a expressão e liberação de CCL20, uma quimiocina importante na transição da resposta imune inata para adaptativa, bem como a expressão dos fatores M-CSF e VEGF. Em células THP-1, mostramos a cinética de fosforilação de proteínas tirosina quinases promovida pela SAA e comparamos com LPS, um estímulo pró-inflamatório clássico. Ainda em células THP-1 mostramos que a SAA induz a fosforilação de duas proteínas importantes no processo inflamatório por induzirem a ativação de NF&#954;B; a p38 e a ERK1/2. Com este estudo contribuímos com o conhecimento a respeito do papel regulatório da SAA em células mononucleares. A ação da SAA sobre estas células torna-se importante, pois estas são cruciais na resposta imune inata e também atuam como células acessórias na resposta imune adaptativa. Desta forma, evidencia-se que, no processo de fase aguda, a expressão e a síntese de SAA resultam na modulação de etapas que controlam este processo e sua progressão. / In the past few years, our research group has described various effects of serum amyloid A (SAA) on cells of the immune system regarding the expression and release of pro-inflammatory cytokines. In this study we have focused on the effects of SAA on mononuclear cells. In order to do this, we have used three experimental models. In the murine experimental model, we described SAA\'s ability to induce the production of NO through peritoneal macrophages and, by using knockout animals for TLR4, we suggested that SAA is an endogenous agonist of TLR4. In mononuclear cells of peripheral human blood, SAA induced the expression and release of CCL20, an important chemokine in the transition from the innate to the adaptive immune response, as well as the expression of M-CSF and VEGF-factors. In THP-1 cells, we showed the phosporylation kinetics of tyrosine protein kinases induced by SAA, and we compared it to LPS, a classic pro-inflammatory stimulus. We also demonstrated, in THP-1 cells, that SAA induced the phosphorylation of two proteins, namely p38 and ERK1/2, that are crucial in the inflammatory process because they induce the activation of transcription factors. With this study, we contributed to the knowledge of the regulatory role of SAA in mononuclear cells. Activity of SAA on these cells is highly important, for they are crucial in the innate immune response and act as accessory cells in the adaptive immune response. Hence it is evident that, in the acute phase process, the expression and synthesis of SAA result in the modulation of the phases that control this process and its progression.

This work documents the first case of idiopathic AA amyloidosis in humans caused by mutation in the promoter region of SAA1 gene. Knowledge of the mechanism of the disease may be an indication for targeted treatment in the future. Mutations in the SAA1 promoter should be considered in all cases of idiopathic forms of AA amyloidosis in which neither the immune nor the inflammatory component of the disease are clearly present.

Stochastic Optimization of Energy Prosumption Systems : A case study on energy balancing for self-sustainable societies

To achieve a sustainable future fossil electricity is being replaced with renewable, leading to higher uncertainties in electricity production. This has resulted in an incentive for consumers to produce, sell, and store their own electricity, hence becoming prosumers. Austerland Skags is a Swedish project that explores the possibility to convert a small society into a prosumption system. The system includes solar and wind power as electricity producers and hydrogen-fueled vehicles for commodity transport. To capitalize the most on their produced electricity they want to store excess electricity. This master thesis uses Austerland Skags as a case study to develop a stochastic linear optimization model to determine the optimal energy storage solution for an energy prosumption system with both electricity and hydrogen demand. The method used in this thesis was the sample average approximation (SAA) algorithm. The results from the SAA were compared to the expected results from the expected value problem (EEV) to show the difference between a stochastic and deterministic solution. The results from the SAA turned out to consistently outperform the EEV for the samples created. Since hydrogen demand could only be sourced in-house, the model was forced to use an electrolyzer and hydrogen tank. The final result from the SAA showed that both a battery and fuel cell was used in addition to the electrolyzer and hydrogen tank in the optimal solution. All capacities stayed within reasonable levels showing the possibility of realizing a cost-effective prosumption system. / För att uppnå en hållbar framtid byts fossila bränslen ut mot förnyelsebara energikällor, vilket leder till högre osäkerheter i elproduktionen. Detta har skapat ett initiativ för konsumenter att börja producera, sälja, och lagra sin egen elektricitet och därav bli prosumenter. Austerland Skags är ett svenskt projektsom undersöker möjligheten att konvertera ett litet samhälle till ett prosumentsystem. Systemet är uppbyggt med sol och vindkraft för produktion av elektricitet och planerar att använda vätgasdrivna fordon för transport av råvror. För att utnyttja så mycket som möjligt av den producerade elektriciteten vill de kunna lagra överskotts elektricitet. Den här masteruppsatsen använder Austerland Skag som en fallstudie för att utveckla en stokastisk linjär optimeringsmodell för att avgöra den optimala energilagringslösningen för ett energiprosumentsystem med både el och vätgasbehov. Metodvalet i denna uppsats var sample average approximation (SAA) algoritmen. Resultatet från SAA jämfördes med det förväntade resultatet från förväntade värdeproblemet (EEV) för att visa skillnaden mellan stokastiska och deterministiska lösningar. Resultatet från SAA visade sig ständigt ge bättre resultat än EEV för undersökta stickprov. Eftersom vätgasbehovet endast kunde förses in-house i modellen var den tvingad att dimensionera upp ett elektrolysör och vätgaslager. Slutresultatet av SAA visade att både batterier och bränsleceller var aktuellt tillsammans med elektrolysör och vätgaslager i den optimala lösningen. Alla kapaciteter förhöll sig inom rimliga nivåer vilket påvisar möjligheten att realisera ett kostnadseffektivt prosumentsystem. / Austerland Skags

Utilisation d'Opuntia ficus-indica en tant que biomoniteur de la pollution atmosphérique industrielle et urbaine / Investigation of opuntia ficus-indica as a biomonitor species of atmospheric pollution

L'objectif de cette étude est d'étudier le potentiel d'Opuntia ficus-indica (Ofi) en tant que biomoniteur dans un environnement contaminé par des dépôts atmosphériques enrichis en métaux lourds. Trois sites d'étude pollués ont été sélectionnés: industriel, minier et routier. Le phénomène de bioaccumulation a été validé en analysant la concentration en métaux lourds par SAA et ICP-MS dans plusieurs raquettes d'âge différent, dans les fruits et les racines. L'identification de l'origine du Pb a été étudiée à partir de la signature isotopique du Pb (TIMS) dans les raquettes, les racines et les sols, avec une caractérisation microscopique (MEB-EDX) des particules déposées à la surface des raquettes. Le taux de bioaccumulation du Pb suite à une exposition racinaire ou foliaire à des particules de fluorapatite synthétisées et enrichies en Pb à permis de préciser les voies de transfert vers la plante. Ainsi, la distribution particulaire ou dispersée du Pb dans les raquettes contaminées a été identifiée au moyen de cartographies MEB-EDX et µXRF. Les résultats obtenus montrent qu'Ofi est un bioaccumulateur du Pb, Cd et Cr. Les signatures isotopiques dans les raquettes révèlent la nature des diverses sources polluantes tout en donnant une indication sur l'évolution de la pollution atmosphérique au cours du temps. Le taux le plus élevé de Pb a été mesuré dans les raquettes contaminées à leur surface par de la fluorapatite. Les analyses microscopiques ont démontré pour les deux approches suivies l'intégration des particules déposées à la surface des raquettes au niveau de la cuticule, des trichomes et des stomates. Cette obsevation est cohérente avec la cartographie obtenue en µXRF. Notre étude démontre le potentiel de cette plante cosmopolite qu'est Ofi en tant que biomoniteur. / This study reports the potential of Opuntia ficus-indica (Ofi) as a biomonitor of atmospheric pollution in three heavily polluted sites, i.e., industrial, mining and road sites. The bioaccumulation phenomenon by Ofi was investigated by studying the heavy metal concentrations in different cladodes of various ages, roots and fruits, using AAS and ICP-MS. To identify the origin of Pb in cladodes, two types of methods were used: TIMS analysis which provided the lead isotopes ratios in cladodes, roots and soil samples, and SEM-EDX analysis at the cladode surface which provided the elemental distribution of Pb and airborne particles and a characterization of their chemical structure at the microscopic scale. A second study was also carried out by testing Pb bioaccumulation after foliar and roots exposure to synthesized fluorapatite particles enriched with Pb. SEM-EDS and µXRF analyses were used to follow the Pb concentration from the contaminated surface to the internal tissues. The results show that Ofi is indeed a bioaccumulator of Pb, Cd and Cr. The Pb isotopic signatures indicate that Ofi is a sensitive species to local aerial pollution and their successive cladodes obviously identify the evolution of air pollution with time. In addition, Ofi cladodes were found more contaminated with lead after direct foliar contamination. SEM-EDS measurements underlined in the two studies the retention of deposited particles on specific zones of the cladode surface (cuticle, trichome and stomata), which were also identified by µXRF cartography. These results represent the first demonstration of the potential of using Ofi, a cosmopolitan plant, as a biomonitor.
