The role of auxin in cell differentiation in meristems / Rôle de l'auxine dans la différenciation des cellules au sein des méristèmes

L'auxine régule la croissance et le développement des plantes grâce aux facteurs de transcription de la famille des "AUXIN RESPONSE FACTOR" (ARF). Chez Arabidopsis thaliana en particulier, ARF5, 6, 7, 8 et 19 activent l'expression de gènes cibles en réponse à l'auxine. Ces cinq ARF activateurs contrôlent de façon plus ou moins redondante des processus divers au cours du développement de la plante, notamment la régulation des croissances au niveau des méristèmes racinaires et aériens ainsi que la formation des racines latérales ou des méristèmes axillaires.Chacun de ces cinq ARF activateurs présente des patrons d'expression uniques dans chacun des tissus racinaires et aériens, en association avec leurs fonctions particulières. Il est probable que cette expression tissu-spécifique trouve son origine dans un contrôle différencié de leur transcription. Dans cette étude, des régulateurs amonts de la transcription de ARF5, 6, 7, 8 et 19 ont été identifiés par une méthode haut-débit de crible simple hybride en levure (Y1H). Une procédure d'expression transitoire en protoplastes a permis de confirmer que l'expression de chaque ARF activateur est contrôlée par des régulateurs spécifiques, dont la majorité se comportent comme des répresseurs de la transcription des ARF in planta. Parmi les régulateurs identifiés, les facteurs de transcription ont été étudiés grâce à des mutants pour préciser les interactions in planta. Ces mutants montrent des phénotypes développementaux typiques de perturbations de l'auxine dans les racines et les tiges : altérations des cinétiques de croissance, de l'émergence des organes latéraux ou de réponses à l'auxine et modification de l'expression des ARF activateurs.Par ailleurs, ce travail aborde également les dialogues entre les voies de signalisation de l'auxine et des cytokinines, et en particulier le rôle de ces interactions dans le développement des racines et des tiges. Une des interactions identifiées dans le crible Y1H est la répression de ARF7 par CRF10, un gène membre de la famille des "Cytokinin Response Factors". Nous avons mis en évidence l'importance de cette interaction pour le maintien de l'architecture du méristème apical racinaire, pour la sénescence des feuille et pour la réponse phototropique à la lumière bleue dans les hypocotyles. / Auxin regulates plant growth and development through the transcription factors of the AUXIN RESPONSE FACTOR (ARF) gene family. Most notably in Arabidopsis thaliana ARF5, 6, 7, 8 and 19 activate expression of target genes in response to auxin. These five ARF activators control both variable and overlapping processes during plant development including regulation of growth at the root and the shoot apical meristems, lateral root and axillary shoot formation. Each of the five ARF activators shows unique tissue-specific expression patterns in the root and the shoot associated with their distinct functions. This tissue-specific expression is likely derived from the differences in the control of ARF activator transcription. In this study the upstream regulators of ARF5, 6, 7, 8 and 19 transcription were identified. This was achieved by utilizing a high-throughput yeast one-hybrid (Y1H) method. The transient protoplast assay revealed that each ARF activator is controlled by specific transcriptional regulators and that the majority of these regulators are repressors of ARF transcription in planta. Mutants of the regulatory transcription factors were utilized to additionally investigate the interactions in planta. These mutants display auxin-related developmental phenotypes in the root and the shoot including alternations in growth kinetics, emergence of lateral organs, responses to auxin and altered expression of ARF activators. Furthermore, this study additionally focuses on cross-talk between the auxin and cytokinin signaling pathways and its role in root and shoot development. One of the interactions identified in the Y1H screen is a repression of ARF7 by CRF10, a member of the Cytokinin Response Factors gene family. The importance of this interaction in maintaining architecture of the root apical meristem, in leaf senescence and in the phototropic response to blue light in hypocotyls is studied.

Uso de paclobutrazol em tomateiro cultivado em dois ambientes /

O trabalho foi desenvolvido de abril a outubro de 2007, na UNESP - Campus de Ilha Solteira, Estado de São Paulo. O PBZ foi aplicado às mudas, aos 15 dias após a germinação. Para avaliação das mudas, foi utilizado delineamento inteiramente casualizado, em esquema fatorial, com 4 repetições e 10 plântulas por parcela. Para avaliação do desenvolvimento e produtividade dos híbridos após transplante, adotou-se o delineamento em blocos casualizados, em esquema fatorial, com análise para grupo de experimentos, modelo fixo, com 4 blocos por ambiente e 9 plantas por parcela. O uso de concentrações crescentes de paclobutrazol reduziu a taxa de crescimento e a altura de plantas, aumentou o diâmetro da haste, reduziu a brotação lateral e reduziu a produtividade da cultura. O PBZ também influenciou significativamente na massa de seca de parte aérea, com aumento linear na massa seca da parte radicular. O híbrido Jennifer apresentou maiores taxas de crescimento absoluto, resultando em plantas com maior altura que o AF 7631, sem que houvesse diferenças entre eles, no que se refere ao diâmetro da haste, brotação lateral e produtividade. Em ambiente protegido, foram obtidas plantas mais vigorosas, com maior altura, brotação lateral e maior produtividade que em campo aberto. O ambiente de cultivo não interferiu em nenhuma das características qualitativas avaliadas, para o híbrido AF 7631, enquanto que, para o Jennifer houve efeito significativo do ambientes de cultivo sobre o teor de sólidos solúveis e acidez titulável... The work was conducted from April to October of 2007, at UNESP - Campus of Ilha Solteira, Sao Paulo State. The PBZ was applied to seedlings, at 15 days after germination. For evaluation of the seedlings, the used statistical experimental design was completely randomized, in factorial outline, with 4 repetitions and 10 seedlings per plot. For evaluation of the development and productivity of two hybrids after transplant it was used a randomized block design, in a factorial array, with analysis for group of experiments, in a fixed model, with 4 replications for environment and 9 plants per plot. The use of increasing concentrations of paclobutrazol reduced the height of plants and their growth rate, increased the stem diameter, decreased the side shooting and reduced the crop yield. PBZ also influenced significantly in the dry shoot, with lineal increase in the dry roots. The hybrid Jennifer showed higher rates of absolute growth, resulting in higher plants than AF 7631, but, with no differences between them, regarding to the stem diameter, to side shooting and crop yield. In the greenhouse, it was obtained higher plants with higher side shooting and higher yield than that observed in the open field. The cultivation environment didn't interfere in none of the appraised qualitative characteristics, for the hybrid AF 7631, while, for Jennifer there was significant effect of the cultivation environment on the contents of soluble solids and titratable acidity, not affecting, however, the maturation index. La prédiction du transfert de cadmium du sol à la plante passe par l'élaboration d'un modèle décrivant la dynamique du métal dans le végétal. Cette thèse analyse les cinétiques d'accumulation du Cd dans Noccaea caerulescens, à l'échelle de la plante entière, de ses organes et au cours de son cycle de végétation. Elle cherche également à établir un modèle prédictif simple, fondé sur les relations entre concentrations d'exposition et en Cd dans la plante. Lorsque N. caerulescens est exposée à une concentration constante en conditions contrôlées, l'allocation de biomasse et la translocation du Cd aux parties aériennes sont constantes dans le temps. Une relation linéaire étroite existe entre la quantité de Cd prélevé, la biomasse et la concentration d'exposition, représentée par le facteur de bioconcentration (BCF). Le développement de la plante n'a pas d'effet sur l'influx racinaire en Cd, qui reste constant dans le temps et proportionnel à l'exposition. Ces résultats suggèrent que le Cd est alloué aux différents tissus aériens de la plante sans prédilection. A contrario, lors des cultures extérieures en terre, les concentrations en Cd, Ni et Zn diminuent au cours du temps après la vernalisation. Les trois métaux ont des comportements similaires en termes d'accumulation dans les différents tissus. La quantité maximale de Cd dans les organes aériens est observée à 2100°Cj. Lorsque la plante est en fleur, elle ne semble pas présenter d'organe aérien privilégié pour l'hyperaccumulation du Cd, les BCF étant constants dans le temps. L'utilisation du BCF permet une prédiction correcte des concentrations dans les parties aériennes, contrairement au modèle de Barber-Cushman / Prediction of cadmium transfer from soil to plant can be achieved by the development of a model describing metal dynamics in the plant. This thesis analyzes Cd accumulation and distribution kinetics in Noccaea caerulescens, in the whole plant and its organs during a growth cycle. A simple predictive model, based on the relationship between Cd exposure concentration and plant Cd concentration, is also developped. This work is based on long-term experiments in controlled conditions with a constant exposure and on outside soil experiments. When N. caerulescens is exposed to a constant concentration under controlled conditions, biomass allocation and Cd translocation to the shoots are constant over time. A strong linear relationship exists between the amount of Cd taken up, biomass and exposure concentration, represented by the bioconcentration factor (BCF). The plant development has no effect on Cd root influx, which remains constant during time and proportional to Cd exposure concentration. These results suggest that Cd is allocated to the various shoots? tissues without predilection. On the other hand, in the field, Cd, Ni, end Zn concentrations in shoots decrease over time after vernalization. The three metals have similar behaviors in terms of accumulation in the different tissues. The maximum amount of Cd in shoots is observed at 2100°Cdays. When the plant is in flower, no privileged shoots parts appear in Cd hyperaccumulation, the BCF is constant over time. The use of the BCF, measured in controlled conditions, allows a correct prediction of shoots concentrations, unlike the Barber-Cushman model

O gênero Passiflora é um excelente modelo para estudos de transição de fase, pois há diferenças morfológicas evidentes entre as plantas nas fases juvenil, adulta vegetativa e adulta reprodutiva. Há relatos de que quando se introduzem ápices de plantas adultas de Passiflora edulis, quer estejam na fase vegetativa ou reprodutiva, em condições de cultivo in vitro, observa-se a regressão à fase juvenil. O objetivo geral deste trabalho foi caracterizar as modificações morfológicas envolvidas na reversão à juvenilidade de ápices caulinares adultos de P. edulis quando cultivados em meio de cultura sem adição de reguladores de crescimento além de avaliar o efeito de reguladores de crescimento no intuito de evitar a reversão à juvenilidade no cultivo in vitro de P. edulis. O trabalho foi desenvolvido a partir da combinação da técnica da micropropagação in vitro e microscopia eletrônica de varredura, sendo dividido em três experimentos: o cultivo in vitro de ápices caulinares em meio MSM sem reguladores de crescimento e análise de amostras em microscópio eletrônico de varredura, cultivo in vitro de ápices caulinares em meio MSM com reguladores de crescimento e cultivo in vitro de ápices caulinares em meio MSM adicionados com BAP na concentração de 10?M com análises de amostras em microscópio estereoscópico. Através de micrografias eletrônicas e imagens estereoscópicas, foi possível caracterizar as fases do desenvolvimento de P. edulis, demonstrando os caracteres morfológicos assim como a manutenção das estruturas cultivadas in vitro. Não foi possível observar o rejuvenescimento in vitro quando ápices caulinares foram cultivados em meio ½ MS ou MSM sem adição de reguladores de crescimento. O rejuvenescimento de ápices caulinares adultos de P. edulis foi obtido a partir do cultivo em meio MSM com adição de 10?M de BAP. Entretanto, o rejuvenescimento foi parcial, pois nem todas as características foram revertidas. A formação de folhas lanceoladas in vitro e o desenvolvimento de brotos adventícios in vitro caracterizaram a reversão à juvenilidade em P. edulis. O meio MSM suplementado com 10 ?M de BAP foi favorável ao desenvolvimento de gemas axilares, além de facilitar a indução do rejuvenescimento dos ápices caulinares. Logo, a reversão à juvenilidade foi alcançada sob as condições in vitro adotadas / The genus Passiflora is an excellent model for phase transition studies because there are obvious morphological differences between plants in the juvenile, adult vegetative and adult reproductive phases. It has been reported that when introducing shoot tips of adult plants of Passiflora edulis, whether in the vegetative or reproductive phase, under in vitro culture conditions, regression to the juvenile phase is observed. The general objective of this work was to characterize the morphological changes involved in the reversion to juvenility of adult shoot tips of P. edulis when grown in culture medium without addition of growth regulators in addition to evaluating the effect of growth regulators in order to avoid reversion to juvenility in the in vitro cultivation of P. edulis. The work was developed from the combination of in vitro micropropagation and scanning electron microscopy, and was divided in three experiments: in vitro culture of shoot tips in MSM medium without growth regulators and analysis of samples in scanning electron microscope; in vitro culture of shoot tips in MSM medium with growth regulators and in vitro culture of shoot tips in MSM medium added with BAP at the concentration of 10?M and analysis of samples under a stereoscopic microscope. Through electronic micrographs and stereoscopic images, it was possible characterize the phases of the development of P. edulis, demonstrating the morphological traits as well as the maintenance of the structures cultivated in vitro. In vitro rejuvenation could not be observed when shoot tips were grown in ½ MS or MSM medium without addition of growth regulators. The rejuvenation of P. edulis adult shoot tips was obtained from the cultivation in MSM medium with the addition of 10?M BA. However, the rejuvenation was partial, since not all the characteristics were reversed. The formation of in vitro lanceolated leaves and the development of in vitro adventitious shoots characterized the reversion to juvenility in P. edulis. The MSM medium supplemented with 10?M BA was favorable to the development of axillary buds, in addition to facilitating the induction of the rejuvenation of the shoot tips. The fate of 13C labelled root and shoot litter in soil and earthworm casts : a multidisciplinary approach based on a mesocosm experiment / Devenir de litières de parties racinaires et aériennes marquées au 13C dans le sol et les turricules de vers de terre : une approche pluridisciplinaire basée sur une expérimentation en mésocosmes

Le sol représente un puits de carbone important, dont les entrées et les sorties sont gouvernées par de nombreux facteurs biotiques et abiotiques. Nous nous intéressons à deux facteurs biotiques essentiels : la qualité de la litière, principale source des MOS; et les vers de terre, ingénieurs clés des sols tempérés. Nous étudions le devenir de parties racinaires et aériennes, marquées au carbone 13, dans le sol et des turricules de Lumbricus terrestris, au cours d'une expérimentation en mésocosmes d'une année. Nous utilisons une approche pluridisciplinaire à différentes échelles spatio-temporelles pour quantifier, localiser et caractériser le carbone organique incorporé dans le sol et les turricules, pendant les 54 semaines d'expérimentation. La composition chimique des parties racinaires et aériennes a un impact significatif sur le devenir du carbone dans le sol et semble influencer la palatabilité de la litière pour les vers de terre. Après une année d'expérience, la contribution des racines, par rapport aux parties aériennes est plus importante dans le sol et les turricules. Les interactions physiques semblent également jouer un rôle important dans la formation et l'évolution des turricules. Les vers de terre minimisent l'effet contrasté des parties racinaires et aériennes sur le carbone du sol et des turricules. Les invertébrés du sol doivent être pris en compte lors de l'étude des processus d'incorporation, de décomposition et de stabilisation du carbone dans le sol. Les microorganismes jouent un rôle essentiel dans la décomposition de la litière et semblent contribuer au carbone stable du sol, et ce particulièrement dans les turricules. / Soil represents an important carbon sink, which inputs and outputs are governed by numerous biotic and abiotic factors. We focused on two essential biotic factors: litter inputs, the main source of SOM; and earthworms, key soil engineers in temperate regions. We investigated the fate of 13C-labelled Ryegrass root and shoot litter, in soil and epi-anecic earthworm casts (Lumbricus terrestris), based on a one year mesocosm experiment. Soil samples were collected at two depths, as well as surface casts, regularly during the 54 weeks of experiment. SOM complexity and heterogeneous composition was considered, using a multidisciplinary approach at different spatio-temporal scales to quantify, localize and characterize the organic carbon incorporated in soil and earthworm casts. The chemical composition of roots and shoots had a significant impact on the fate of carbon in soil, with an increased contribution from root-derived carbon in soil and casts after one year. The chemical composition tended to influence the litter palatability for earthworms. As organo-mineral associations tended to increase during drying and ageing of casts, physical interactions might have also played an important role in cast formation and evolution. Earthworms tended to minimize the diverging fate of root and shoot residues on both soil and cast carbon, after the year of experiment. Thus, while studying soil carbon incorporation, decomposition and stabilization, the role of soil invertebrates should be particularly considered. We also observed the crucial role of microorganisms in decomposing litter and their potentially high participation to stable carbon pool in soil and particularly in casts.

Substâncias húmicas extraídas de turfa associadas com N-ureia: influência no crescimento da cana-de-açúcar e nas transformações do N no solo / Humic substances extracted from peat associated with N-urea: influence on sugar cane growth and N transformations in soil

As substâncias húmicas (SH) podem alterar o crescimento das plantas bem como estimular a absorção de nutrientes. A ureia é o fertilizante nitrogenado mais utilizado na agricultura, porém, apresenta baixa eficiência de uso. Sabe-se que as substâncias húmicas podem aumentar a eficiência de uso da ureia por meio de efeito bioestimulante no crescimento radicular. Somado a isso, as SH atuam nas bombas de próton das raízes estimulando a absorção de nitrogênio e também alteram a dinâmica de hidrólise e mineralização do N-ureia. O objetivo deste trabalho foi verificar se as SH extraídas de turfa, associadas ou não ao N-ureia, alteram as transformações do N-ureia no solo, assim como o crescimento radicular e da parte aérea no desenvolvimento inicial da cana-planta. Nesse sentido, foram instalados dois experimentos em casa de vegetação, um com a presença de plantas para aferir parâmetros morfológicos e nutricionais e outro, sem plantas, para verificar a dinâmica de hidrólise e mineralização do N-ureia. Os experimentos foram planejados em delineamento experimental inteiramente casualizado com 16 tratamentos e cinco repetições, correspondendo a um esquema fatorial completo com quatro doses de nitrogênio na forma de ureia: 0; 100; 200 e 300 mg dm-3 e quatro doses de SH: 0; 13,8; 27,6; 41,7 mg dm-3 de C, e o experimento sem plantas utilizando o mesmo delineamento porém com 3 repetições e análises feitas em duplicatas. As substâncias húmicas aumentaram a hidrólise da ureia elevando a concentração de NH4+ no solo. O processo de nitrificação da ureia foi reduzido na presença das substâncias húmicas. As aplicações isoladas de N e SHs estimularam o crescimento radicular e da parte aérea da cana-de-açúcar, bem como modificaram as características morfológicas das raízes e parte aérea da cana-planta. A associação dos produtos resultou em decréscimos no crescimento vegetal devido às altas concentrações de amônio causadas pela maior hidrólise da ureia e também atuaram na redução do acúmulo e teor de nitrogênio na planta. Tanto a ureia quanto as SHs se mostraram eficientes no estímulo do crescimento inicial da cana-de-açúcar, no entanto, devido às altas doses de N-ureia utilizadas, o possível efeito sinérgico dos produtos não foi verificado. / Humic substances (HS) can alter plant growth and stimulate the absorption of nutrients. Urea is the most widely used nitrogen fertilizer in agriculture, however, has low use efficiency. It is known that humic substances can increase the efficiency of urea through bio-stimulant effect on root growth. Added to this, the SH proton pumps operate in the roots stimulating the uptake of nitrogen and also alter the dynamics of urea\'s hydrolysis and mineralization. The aim of this study was to verify whether the HS extracted from peat, associated or not with N-urea, alter the dynamics of transformation of urea-N in the soil and the growth of roots and shoots in the cane-plant cropcycle. In this direction, two experiments were carried out in a greenhouse, with the presence of plants to assess morphological and nutritional parameters and one without plants, to verify the dynamics of hydrolysis and mineralization of N-urea. The experiments were designed in a completely randomized design with 16 treatments and five replications, corresponding to a full factorial design with four rates of nitrogen as urea: 0, 100, 200 and 300 mg dm-3 and four rates of HS: 0, 13.8, 27.6, 41.7 mg dm-3 C, and experiment without plants using the same design but with three replications and analyzes performed in duplicates. The humic substances increased hydrolysis of urea by raising the concentration of NH4+ in soil. The nitrification of urea was reduced in the presence of humic substances. Individual applications of N and SHs stimulated root and shoot of sugar cane growth and modified the morphological characteristics of roots and shoots in the crop cycle of the cane-plant. The association of the product resulted in decreases in plant growth due to high ammonium concentrations caused by increased hydrolysis of urea and also reduced accumulation and nitrogen content in the plant. Both urea and the SHs were effective in stimulating early growth of sugar cane , however , due to high doses of N-urea used, the synergistic effect of the products has not been verified.

Relationships between carbohydrate supply and reserves and the reproductive growth of grapevines (Vitis vinifera L.)

Viticultural practices such as trunk girdling and shoot topping along with defoliation, shading and node number per vine treatments were used to alter the carbohydrate physiology of mature Chardonnay grapevines growing in the cool climate of Canterbury, New Zealand. The timing of vine defoliation in the season previous to fruiting decreased concentrations of over-wintering carbohydrate reserves (mostly starch) in both the trunks and roots of grapevines. Roots were particularly sensitive, with defoliation as early as 4 weeks after bloom in the previous season reducing starch concentrations to 1.5%Dwt at bud burst compared with 17%Dwt in non-defoliated vines. In contrast, partial vine defoliation as early as bloom in the previous season reduced root starch concentrations to 4-7%Dwt at bud burst compared with 15%Dwt in non-defoliated vines. Vine shading and trunk girdling treatments at bloom in the previous season, resulted in small reductions in root starch concentrations (16%Dwt) compared with non-shaded and non-girdled vines (19%Dwt), but shoot topping did not. Study across three growing seasons established that higher concentrations of over-wintering trunk and root carbohydrate reserves were associated with warmer and sunnier weather in the previous growing season. Individual shoot leaf removal at either the beginning or towards the end of the inflorescence initiation period, reduced shoot starch concentrations to 3-6%Dwt compared with 11 %Dwt for no leaf removal, such reductions persisted through to the following season. Shoot topping at the start of the initiation period had no effect on shoot carbohydrate accumulation, but trunk girdling temporarily increased shoot starch concentrations during the first 31 days after treatment. Reductions in over-wintering trunk and root carbohydrate reserves were associated with a reduction in inflorescences per shoot and flowers per inflorescence in the following season, the reduction as much as 50% compared with non carbohydrate stressed vines. While there were strong linear or curvilinear relationships between the concentration of starch in trunks and roots at bud burst and inflorescences per shoot and flowers per inflorescence, in case the of inflorescences per shoot, there was not an immediate cause and effect because inflorescences were initiated in the previous season. Individual shoot leaf removal during the inflorescence initiation period illustrated that leaf removal directly inhibited the initiation of inflorescences in latent buds. Shoot carbohydrate measurements showed a strong curvilinear relationship to the number of inflorescences per shoot, with a threshold starch concentration of 10-12%Dwt during the inflorescence initiation period required for a maximum number of inflorescences per shoot. Furthermore, examination of individual node positions emphasised the importance of the subtending leaf on the initiation of inflorescences within the latent bud. The number of inflorescences per shoot post bud burst was reduced on vines that were both carbohydrate reserve stressed (by previous season's defoliation) and had a high node (108) number retained per vine after winter pruning compared with little or no reduction in inflorescences per shoot on carbohydrate reserve stressed vines that had a low (20) node number per vine. The reduction in inflorescences per shoot on high node vines was associated with reduced carbohydrate reserves and reduced shoot vigour (thinner and lighter shoots). Flowers per inflorescence were reduced by as much 50% in response to lower overwintering carbohydrate reserves. Fewer flowers per inflorescence were attributed to a reduction in primary branching of the inflorescence and also a reduction in flowers per branch. Strong linear relationships between the concentrations of starch in trunks and roots and flowers per inflorescence indicate that the determination of flowers per inflorescence, unlike inflorescences per shoot, may be dependent on the level of overwintering carbohydrate reserves. This is most likely due to changes in branching of the inflorescence and individual flower formation occurring during the bud burst period. Per cent fruitset was not affected by reductions in carbohydrate reserves, so fewer inflorescences per shoot and flowers per inflorescence resulted in reduced vine yield. The findings of this thesis indicate that changes in the level of carbohydrate production and partitioning in response to a range of viticultural management practices and seasonal weather contribute to seasonal variation in grapevine flowering and yields in New Zealand's cool climate environment. The relationships between carbohydrate reserves and flowering illustrate the potential to use this information to predict grapevine flowering and forecast yields. The practical implications of this research illustrate that the viticulturist must manage grapevines not only for the current crop, but also for subsequent crops by maintaining sufficient carbohydrate reserves for balanced growth flowering and fruiting from season to season.

DINÂMICA DO CARBONO- 13C DE RAÍZES E RESÍDUOS DE PARTE AÉREA DE CULTURAS DE VERÃO EM DOIS SOLOS DE VÁRZEA / 13C-CARBON DYNAMICS OF ROOTS AND SHOOTS FROM SUMMER CROPS IN TWO LOWLAND SOILS

The use of isotopic techniques has been widely used in studies related to changes in the soil C. The chemical quality of crop residues and the type of management used are considered to be the main factors that control the dynamics of soil C. However, the stabilization of C derived from crop residues is also dependent on soil texture. Therefore, the objectives of these studies were: a) to evaluate the efficiency of repeat pulse labeling technique for the isotopic enrichment of leaves, stems and roots of rice, sorghum and soybeans plants with 13C under greenhouse conditions and b) to evaluate the dynamics of C during the decomposition of 13C enriched shoots and roots of rice, sorghum and soybean plants in two lowland soils with different texture. The studies were conducted in the greenhouse of the department of soils, Federal University of Santa Maria (UFSM), Brazil. In the first study, rice, sorghum and soybeans plants were exposed weekly to isotopic pulses with 33% 13CO2 atoms, for 1-1,5h in closed acrylic chambers till grain filling stage. Delta 13C enrichment, chemical composition and the recovery and distribution of 13C in plant parts and soil was determined. In the second study, enriched residues obtained in the first study were used to study the dynamics of C in two lowland soils The soils were Planossolo Hidromórfico Eutrófico gleissólico and Planossolo Hidromórfico Eutrófico arênico. The experimental design was completely randomized with three replications with 18 treatments, which were composed of a combination of 13C enriched shoots and roots with non-enriched shoots and roots (natural abundance). The soil was collected after 30, 90 and 180 days and the amounts of 13C remaining in residues and soil and the new C in soil was determined. The dry matter (DM) production and chemical composition of the residues was not affected by the 13C enrichment. The enrichment values were +377 for rice, + 466.5 for sorghum and + 453.7 for soybean. The isotopic enrichment of plants with 13C by repeated pulses was efficient for the homogeneous enrichment of rice, sorghum and soybean plants in greenhouse conditions. Regarding the dynamics of the soil C, the management of rice residues affected the decomposition and the addition of new C in the soil. The quality of the residues influenced the decomposition process. There was no difference in the decomposition of the roots of the three crop plants. The type of the soil did not affect the decomposition of shoot and root, residues of soybean, sorghum and rice plants. The amount of new C was influenced by the type of residues and soil. The roots contributed greater amounts of C in the soil in relation to the shoots. / Resumo O uso de técnicas isotópicas tem sido amplamente utilizado em estudos relacionados a transformações do C no solo. A qualidade química dos resíduos culturais e o tipo de manejo utilizado são considerados os principais fatores que controlam a dinâmica do C no solo. Contudo a estabilização do C oriundo dos resíduos culturais é dependente da textura do solo. Os objetivos desses estudos foram: a) avaliar a eficiência do enriquecimento isotópico com 13C de folhas, talos e raízes das plantas de arroz, sorgo e soja cultivadas em solo de várzea em condições de casa de vegetação; b) avaliar a dinâmica do C durante a decomposição da parte aérea e de raízes de arroz, sorgo e soja enriquecidos isotopicamente com 13C em dois solos de várzea com textura distinta. Os estudos foram conduzidos em casa de vegetação na Universidade Federal de Santa Maria utilizando solos de várzea. No primeiro estudo, plantas de arroz, sorgo e soja foram expostas semanalmente aos pulsos isotópicos com 33% átomos de 13CO2, durante 1-1,5h, em câmaras fechadas de acrílico até o estádio de enchimento de grãos. Foram determinados os valores de enriquecimento com 13C dos órgãos das plantas, a composição química dos resíduos e valores de recuperação e distribuição do 13C nos órgãos da planta e no solo. No segundo estudo, foi realizada uma incubação com duração de 180 dias, com a utilização dos resíduos culturais enriquecidos no estudo I. Foram utilizados resíduos de parte aérea e raízes de arroz (Oryza sativa L.), sorgo (Sorghum bicolor L.) e soja (Glycine max L.) em dois solos de várzea, um Planossolo Hidromórfico Eutrófico gleissólico e um Planossolo Hidromórfico Eutrófico arênico. O delineamento utilizado foi o inteiramente casualizado com três repetições e 18 tratamentos, os quais foram compostos por uma combinação de palha enriquecida com 13C e com raízes também enriquecidas com 13C ou com raízes não enriquecidas (condição natural). Foram realizadas três datas de coleta aos 30, 90 e 180 dias nas quais foram avaliadas as quantidades de 13C remanescente nos resíduos e no solo, e o C novo no solo. A produção de MS e composição química dos resíduos não foi afetada pelo enriquecimento com 13C. Os valores de enriquecimento foram de +377 em arroz, +466,5 em sorgo e +453,7 em soja. O enriquecimento isotópico das plantas com 13C por meio de pulsos repetidos foi eficiente para o enriquecimento homogêneo de plantas de arroz, sorgo e soja em condições de casa de vegetação. Em relação a dinâmica do C no solo, o manejo dos resíduos de arroz afetou a decomposição e a adição de C novo no solo. Houve influência da qualidade dos resíduos sobre o processo de decomposição. Não houve diferença na decomposição das raízes das três culturas estudadas. O tipo de solo não afetou a decomposição da palha e raízes de soja, sorgo e arroz. A quantidade de C novo foi influenciada pelo tipo de resíduo e pelo solo. As raízes adicionam maior quantidade de C ao solo em relação à parte aérea.

As substâncias húmicas (SH) podem alterar o crescimento das plantas bem como estimular a absorção de nutrientes. A ureia é o fertilizante nitrogenado mais utilizado na agricultura, porém, apresenta baixa eficiência de uso. Sabe-se que as substâncias húmicas podem aumentar a eficiência de uso da ureia por meio de efeito bioestimulante no crescimento radicular. Somado a isso, as SH atuam nas bombas de próton das raízes estimulando a absorção de nitrogênio e também alteram a dinâmica de hidrólise e mineralização do N-ureia. O objetivo deste trabalho foi verificar se as SH extraídas de turfa, associadas ou não ao N-ureia, alteram as transformações do N-ureia no solo, assim como o crescimento radicular e da parte aérea no desenvolvimento inicial da cana-planta. Nesse sentido, foram instalados dois experimentos em casa de vegetação, um com a presença de plantas para aferir parâmetros morfológicos e nutricionais e outro, sem plantas, para verificar a dinâmica de hidrólise e mineralização do N-ureia. Os experimentos foram planejados em delineamento experimental inteiramente casualizado com 16 tratamentos e cinco repetições, correspondendo a um esquema fatorial completo com quatro doses de nitrogênio na forma de ureia: 0; 100; 200 e 300 mg dm-3 e quatro doses de SH: 0; 13,8; 27,6; 41,7 mg dm-3 de C, e o experimento sem plantas utilizando o mesmo delineamento porém com 3 repetições e análises feitas em duplicatas. As substâncias húmicas aumentaram a hidrólise da ureia elevando a concentração de NH4+ no solo. O processo de nitrificação da ureia foi reduzido na presença das substâncias húmicas. As aplicações isoladas de N e SHs estimularam o crescimento radicular e da parte aérea da cana-de-açúcar, bem como modificaram as características morfológicas das raízes e parte aérea da cana-planta. A associação dos produtos resultou em decréscimos no crescimento vegetal devido às altas concentrações de amônio causadas pela maior hidrólise da ureia e também atuaram na redução do acúmulo e teor de nitrogênio na planta. Tanto a ureia quanto as SHs se mostraram eficientes no estímulo do crescimento inicial da cana-de-açúcar, no entanto, devido às altas doses de N-ureia utilizadas, o possível efeito sinérgico dos produtos não foi verificado. / Humic substances (HS) can alter plant growth and stimulate the absorption of nutrients. Urea is the most widely used nitrogen fertilizer in agriculture, however, has low use efficiency. It is known that humic substances can increase the efficiency of urea through bio-stimulant effect on root growth. Added to this, the SH proton pumps operate in the roots stimulating the uptake of nitrogen and also alter the dynamics of urea\'s hydrolysis and mineralization. The aim of this study was to verify whether the HS extracted from peat, associated or not with N-urea, alter the dynamics of transformation of urea-N in the soil and the growth of roots and shoots in the cane-plant cropcycle. In this direction, two experiments were carried out in a greenhouse, with the presence of plants to assess morphological and nutritional parameters and one without plants, to verify the dynamics of hydrolysis and mineralization of N-urea. The experiments were designed in a completely randomized design with 16 treatments and five replications, corresponding to a full factorial design with four rates of nitrogen as urea: 0, 100, 200 and 300 mg dm-3 and four rates of HS: 0, 13.8, 27.6, 41.7 mg dm-3 C, and experiment without plants using the same design but with three replications and analyzes performed in duplicates. The humic substances increased hydrolysis of urea by raising the concentration of NH4+ in soil. The nitrification of urea was reduced in the presence of humic substances. Individual applications of N and SHs stimulated root and shoot of sugar cane growth and modified the morphological characteristics of roots and shoots in the crop cycle of the cane-plant. The association of the product resulted in decreases in plant growth due to high ammonium concentrations caused by increased hydrolysis of urea and also reduced accumulation and nitrogen content in the plant. Both urea and the SHs were effective in stimulating early growth of sugar cane , however , due to high doses of N-urea used, the synergistic effect of the products has not been verified.

O Greening ou Huanglongbing (HLB) é a doença mais devastadora dos citros na atualidade. Levantamentos mostram que a enfermidade está presente em cerca de 60% dos talhões do parque citrícola paulista, demostrando sua grande capacidade de disseminação. O psilídeo-asiático-dos-citros, Diaphorina citri Kuwayama (Hemiptera: Psylidae) é um importante vetor dos agentes causais associados ao HLB. Sua capacidade de dispersão está intimamente relacionada ao progresso da doença no campo. Não se tem conhecimento dos principais fatores que possam interferir no deslocamento do inseto até plantas cítricas. Sendo assim, a presente pesquisa teve como objetivo investigar a influência de alguns fatores ambientais sobre a dispersão do psilídeo D. citri, discutindo-os em relação à epidemiologia do HLB dos citros. Inicialmente foram realizados testes em laboratório para adaptação da metodologia de marcação, liberação e recaptura usando pó fluorescente. Grupos de psilídeos adultos foram marcados e sua sobrevivência avaliada, verificando a influência de quatro cores distintas de pó fluorescente (amarelo, azul, rosa e verde). Posteriormente realizaram-se estudos de campo com quatro liberações de psilídeos, com marcadores distintos, em quatro áreas experimentais circulares e adjacentes, sendo formadas por círculos concêntricos localizados a 18, 24 e 30 m de cada centro de liberação. Os insetos foram monitorados visualmente e por armadilhas adesivas amarelas. Nas 1ª e 2ª liberações, foi verificado o efeito das coberturas vegetais milho (Zea mays L.) e capim-braquiária (Brachiaria decumbens Stapf.) na dispersão de D. citri. Já nas 3ª e 4ª liberações, foi estudado o efeito de brotações no deslocamento do inseto. A influência dos ventos foi avaliada em ambas as situações. A análise de sobrevivência indicou que não houve interferência das quatro cores de pó fluorescente em D. citri, sendo a marcação visível no corpo dos insetos durante os 48 dias de avaliação. De acordo com as análises fatoriais, não foi observada qualquer interferência da cobertura vegetal milho, medindo 2 m de altura, na dispersão do inseto. Na presença de brotações, adultos de D. citri dispersaram menos em busca de novos hospedeiros, acumulando-se nas primeiras linhas de plantas cítricas. Após 6 horas da liberação, a capacidade de dispersão máxima de D. citri foi três vezes maior na condição de ausência de brotações. A dispersão média e a área total abrangida variaram assimptoticamente com o tempo, ou seja, o inseto possui inicialmente grande velocidade de dispersão, porém esta tende a se estabilizar com o incremento do tempo. Esses resultados deixam claro que D. citri realiza movimentos dependendo das condições fenológicas de seus hospedeiros, ressaltando a grande importância de se intensificar métodos de controle regionais em épocas em que o fluxo vegetativo das plantas cítricas não é tão intenso. / Greening or Huanglongbing (HLB) is the most devastating disease of citrus today. Surveys show that the disease is present in about 60% of the park stands citrus Sao Paulo, demonstrating its great ability to spread. The Asian citrus psyllid, Diaphorina citri Kuwayama (Hemiptera: Psylidae) is an important vector of the causative agents associated with HLB. Its ability to spread is closely related to disease progression in the field. There is no knowledge of the main factors that can affect the movement of the insect to citrus plants. Thus, the present study aimed to investigate the influence of some environmental factors on the dispersal of psyllid D. citri, discussing them in relation to the epidemiology of HLB of citrus. Initially tests were performed in the laboratory to adapt the methodology of marking, release and recapture using fluorescent powder. Groups of adult psyllids were marked and their survival assessed by checking the influence of four different colors of fluorescent powder (yellow, blue, pink and green). Subsequent studies were conducted with four field releases psyllids, with distinct markers on four experimental areas adjacent circular and being formed by concentric circles located at 18, 24 and 30 m for each center release. The insects were monitored visually and by yellow sticky traps. In the 1st and 2nd releases, was to determine the effect of cover crops maize (Zea mays L.) and grass (Brachiaria decumbens Stapf.) in the dispersal of D. citri. In the 3rd and 4th releases, were studied the effect of shootings in insect movement. The influence of the wind has been evaluated in both situations. Survival analysis indicated that there was no interference of the four colors of fluorescent powder on D. citri, and marking was visible on the body of insects during the 48 days of evaluation. According to the factor analysis, no interference of vegetation corn (measuring 2 m in height) was observed in the dispersal of the insect. In the presence of shoots, adult D. citri less dispersed looking for new hosts and it accumulate in the first lines of citrus plants. After 6 hours of release, the D. citri maximum dispersal was three times higher in the condition of absence of shoots. The dispersal medium and the total area covered varied asymptotically with time, ie the insect has initially high speed dispersal, but this tends to stabilize with increasing time. These results clearly show that D. citri performs movements depending on the phenology of its hosts, emphasizing the importance of intensifying regional control methods at times when the flow of plant citrus is not intense.
