O objetivo deste trabalho é apresentar uma aplicação do Lean Seis Sigma na Logística de uma empresa de autopeças visando à redução do nível do inventário por meio da eliminação dos desperdícios e redução da variabilidade nos processos de gestão de estoque. O estudo de caso usou como espinha dorsal para condução do projeto a metodologia DMAIC e foram aplicadas ferramentas de análise de dados e estatística tais como matriz de causa e efeito, mapa do processo, cartas de controle e análise da capacidade, juntamente com as ferramentas do Lean: Value Stream Mapping, JIT, Kanban, 5S e Padronização. O resultado alcançado teve uma redução no inventário de aproximadamente 30%, ou seja, no início do projeto o estoque estava em torno de R$ 4.700.000,00 e após implantação reduziu para R$3.350.000,00. Estes números foram alcançados mantendo-se o nível de serviço em torno da média histórica da empresa de 98% para um faturamento estável dentro do período analisado. Em outras palavras, se a empresa puder entender e controlar os desperdícios e as variações nos processos, do fornecedor ao cliente, então será capaz de reduzir a dependência dos estoques e melhorar a produtividade, portanto, desta forma a empresa pode alavancar competitividade em relação aos seus concorrentes. / The objective of this paper is to present an application of Lean Six Sigma Logistics in a auto parts company in order to reduce the inventory level by eliminating waste and reducing variability in the processes of inventory management. The case study used as the backbone of the project to conduct the DMAIC methodology and tools were applied for data analysis and statistics such as cause and effect matrix, map of the process control charts and capability analysis, along with tools Lean: Value Stream Mapping, JIT, Kanban, 5S and standardization. The result achieved was a reduction in inventory of approximately 30%, ie, early in the project the stock was around $2,500,000.00 and after implantation decreased to $1,900,000.00. These numbers were achieved while maintaining the level of service around the companys historical average of 98% for a stable revenue in the period analyzed. In other words, if the company can understand and control waste and variations in processes from supplier to customer, then you will be able to reduce dependence on inventory and improve productivity, so this way the company can leverage competitive with their competitors.

Previous issue date: 2016-11-21 / The aircraft supply is a major cause of flight delays since this process is slow and, if it does not start as soon as the aircraft is available for the maintenance team, the risk of being terminated after the last passenger has already boarded increases. The procedure usually only starts after the information of how much must be supplied get through the flight dispatcher and this information typically take some time to get to the maintenance team. However, it is intended to test a new scenario: start to supply with the minimum quantity of fuel and, if necessary, complete with the amount belatedly informed by dispatcher. Therefore, this paper aims to analyze the application of Six Sigma in this process through the Student's t-test and the Statistical Process Control (SPC). Sigma in this process through the Student t test and Statistical Process Control (SPC). The method adopted was multi cases study by interviews and organizational practices observations. The results showed that the new process is favorable in relation to the former, the Case 1 average time of flight delays has been reduced from 14 to 6 minutes (57%), Case 2 average time has been reduced from 10 to 6 minutes (53%), about flight delays quantity upper than 15 minutes, has reduced to 43% and 39% respectively. It concludes that the application of Six Sigma in aircraft refueling process is innovative to the scientific literature and was favorable for both companies, also helped to mitigate the risk of fines and penalties and, consequently, improved the level of quality of service offered by the airline company. / O abastecimento de aeronaves é uma das principais causas de atrasos de voo uma vez que este processo é lento e, caso este não seja iniciado assim que a aeronave esteja disponível para a equipe de manutenção, o risco de ser finalizado depois do último passageiro já ter embarcado aumenta. O processo de abastecimento usualmente só é iniciado após a informação de quanto se deve abastecer, esta informação costuma demorar a chegar para a equipe de manutenção. A melhoria no processo considera iniciar o abastecimento com a quantidade mínima de combustível e, se necessário, completar com a quantidade tardiamente informada. O objetivo desse trabalho é analisar a aplicação do Seis Sigma utilizando o teste t de Student e o Controle Estatístico do Processo (CEP) nas duas maiores companhias aéreas brasileiras. O método adotado foi estudo de múltiplos casos por meio de entrevista e observação na prática organizacional. O resultado demonstra que o tempo médio de atrasos de voo para o Caso 1 foi reduzido após a aplicação do Seis Sigma, de 14 para 6 minutos (57%). Além disso, a quantidade de atrasos acima de quinze minutos foi reduzida em 53%. Já no Caso 2, o tempo médio foi reduzido de 10 para 6 minutos, uma melhora de 43%, quanto a quantidade de atrasos acima de quinze minutos, houve uma redução de 39%. Conclui-se que a aplicação do Seis Sigma no processo de abastecimento de aeronaves é inovadora para a literatura científica e foi favorável para as duas empresas, também colaborou para mitigar os riscos de multas e penalidades e, consequentemente, melhorou o nível de qualidade do serviço oferecido pelas companhias aéreas.

Caracterização da superexpressão do fator sigma ECF σx em Pseudomonas aeruginosa PA14 / Characterization ofthe ECF sigma fator σx overexpression in Pseudomonas aeruginosa PA14.Ana Laura Boechat Borges 05 July 2013 (has links)
Pseudomonas aeruginosa é uma proteobactéria do grupo gama muito versátil, capaz de colonizar ambientes variados e infectar hospedeiros filogeneticamente distintos, incluindo humanos imunocomprometidos. Os fatores sigma de função extracitoplasmática (ECF) são membros de sistemas de sinalização de superfície celular (CSS), abundantes em P. aeruginosa. Vinte genes codificando fatores sigma ECF estão presentes nos genomas sequenciados de P. aeruginosa, a maioria fazendo parte de sistemas TonB relacionados à captação de ferro. Neste trabalho, seis fatores sigma pobremente caracterizados foram superexpressos na linhagem PA14 a partir de um promotor induzível por arabinose para investigar seu papel na expressão dos sistemas de dois componentes PvrSR e RcsCB, que atuam na regulação da fímbria CupD, além de sua influência no crescimento de culturas de P. aeruginosa. Não foi observado efeito positivo de nenhum dos fatores sigma testados na expressão dos sistemas de dois componentes e a superexpressão de cinco deles tampouco levou a qualquer alteração no crescimento, porém a produção de piocianina foi alterada na superexpressão de PA14_55550 e a superexpressão de PA14_26600 e PA14_46810 levou a um discreto aumento no início da formação de biofilme em PA14. Por outro lado, culturas superexpressando σx (ALB04) apresentaram um perfil alterado de lipopolissacarídeo e uma curva de crescimento bifásica, alcançando precocemente uma fase estacionária seguida de uma recuperação do crescimento até uma segunda fase estacionária. Durante a primeira fase estacionária, a maior parte das células aumenta de tamanho e morre, mas as células remanescentes retornam à morfologia selvagem e seguem para a segunda fase de crescimento exponencial. Isso não acontece devido a mutações compensatórias, uma vez que células coletadas de pontos tardios da curva e diluídas em meio novo repetem este comportamento. Apesar de trabalhos com a linhagem PAO1 associarem σx à transcrição de oprF, que codifica a principal porina não específica de Pseudomonas, nas condições dos nossos ensaios em PA14 a expressão dessa porina não foi induzida pela superexpressão de σx. Assim, os efeitos observados nessa superexpressão também não podem ser atribuídos a OprF. A transcrição de oprF em PA14 mostrou-se majoritariamente dependente da região promotora a que se atribui a ligação de σ70, ao contrário dos relatos na literatura da dependência da região de ligação a σx. Análises proteômicas foram realizadas para investigar os elementos envolvidos nesses efeitos de superexpressão de σx, o que revelou a indução de diversas enzimas envolvidas na via de biossíntese de ácidos graxos. As células superexpressando σx apresentam uma maior proporção de ácidos hexadecanoico (C16) e hexadecenoico (C16:1) e dados de anisotropia mostram uma maior fluidez da(s) membrana(s). Este trabalho é o primeiro relato de um fator sigma ECF envolvido em biossíntese de lipídeos em P. aeruginosa. / Pseudomonas aeruginosa is a very versatile gammaproteobacteria, able to colonize different environments and to infect phylogenetically distinct hosts, including immunocompromised humans. The extracytoplasmic function sigma factors (ECFs) are members of cell signaling systems (CSS), abundant in P. aeruginosa. Twenty genes coding for ECF sigma factors are present in the sequenced genomes of P. aeruginosa, most of them being part of TonB systems related to iron uptake. In this work, six poorly characterized sigma factors were overexpressed in strain PA14 from an arabinose inducible promoter to investigate their role in the expression of the two-component systems PvrSR and RcsCB, which regulates CupD fimbria, and their influence in P. aeruginosa cultures growth. None of the tested sigma factors led to two-component systems upregulation and overexpression of five of them caused no change in the growth profile, but pyocyanin production was altered in PA14_55550 overexpression and PA14_26600 and PA14_46810 overexpression led to a slight increase in biofilm initiation in PA14. By the other side, cultures overexpressing σx (ALB04) presented an altered lipopolysaccharide profile and a biphasic growth curve, reaching an early stationary phase followed by a growth resuming untill a second stationary phase. During the early stationary phase, most cells swells and dies, but the remaining cells return to wild type morphology and proceed to the second exponential phase of growth. This is not due to compensatory mutations, since cells collected from late points of the curve and diluted in fresh medium repeat this behavior. Although studies with strain PAO1 associate σx with transcription of oprF, encoding the major nonspecific porin of Pseudomonas, under our experiments conditions with PA14, this porin expression is not induced by σx overexpression. Thus, the effects observed in this overexpression cannot be attributed to OprF. Transcription of oprF in PA14 proved to be mainly controlled by the σ70-dependent promoter region instead of the σx-dependent promoter region reported in the literature. Proteomic analyses were performed to investigate the elements involved in these effects of σx overexpression, which revealed the induction of several enzymes involved in fatty acids biosynthesis. Cells overexpressing σx exhibit a greater proportion of hexadecanoic (C16) and hexadecenoic (C16: 1) acids and anisotropy data show higher fluidity of the membrane (s). This work is the first report of an ECF sigma factor involved in lipid biosynthesis in P. aeruginosa.

Tem como objetivo geral propor um mÃtodo que avalia o alinhamento da estratÃgia corporativa ao nÃvel de serviÃo desejado pelos clientes e apresenta possÃveis soluÃÃes para a sua melhoria. O mÃtodo de raciocÃnio utilizado neste trabalho foi o hipotÃtico-dedutivo. Quanto à metodologia para o alcance dos objetivos propostos, fez-se uso de uma pesquisa classificada como aplicada, qualitativa, exploratÃria, bibliogrÃfica, documental e estudo de caso. Os conhecimentos pesquisados acerca de estratÃgia, alinhamento estratÃgico, BSC, Seis Sigma e nÃvel de serviÃo logÃstico serviram para fundamentar o mÃtodo proposto, que prioriza o alinhamento com base nos indicadores do BSC e realiza as melhorias com a aplicaÃÃo do Seis Sigma. A aplicaÃÃo do mÃtodo foi realizada em uma lavanderia hospitalar que serviu como o caso em estudo. A principal conclusÃo do trabalho realizado foi a constataÃÃo da utilidade do mÃtodo proposto na avaliaÃÃo e melhoria do alinhamento da estratÃgia corporativa com o nÃvel de serviÃo desejado pelos clientes. / The focus of this dissertation is to study the alignment between corporate strategy and the service level desired by customers. Its main purpose is to develop a method that analyses the alignment between corporate strategy and the service level desired by customers and presents possible solutions to its improvement. The method of reasoning used in this dissertation was the hypothetical-deductive. Regarding the methodology for achieving the proposed objectives, it was used an applied research classified as qualitative, exploratory, bibliographical, documentary and case study. Strategy, strategic alignment, BSC, Six Sigma and logistics service level were the knowledge researched to substantiate the proposed method, which prioritizes the alignment based on BSC indicators and promotes improvements by the use of Six Sigma. The application of the method was performed in a hospital laundry that served as case study. The main conclusion of the study was the assessment of the usefulness of the proposed method in evaluating and improving the alignment of corporate strategy with service level desired by customers.

Previous issue date: 2014-10-10 / The Plastisol is a fluid bulk based on PVC, mineral fillers, plasticizers, stabilizers and depending of its final application, a pigment and resins for adhesion providing. It can be produced by using two different types of mixers: The sigma and the cowles. This work studied what was the best mixer for producing the Plastisol and also evaluated the adhesion, viscosity and thixotropic properties gotten using these two producing processes. For a better understanding of adhesion, viscosity and thixotropic phenomenon, it was produced in the laboratory four different samples, each one with different producing process. It was prepared specimens of Plastisol for each producing process used, where the tensile strength was measured in a dynamometer for taking the force intensity and a rheometer for viscosity and thixotropic studies. This way was possible to evaluate what was the best producing process adopted. / O Plastisol é uma pasta fluida composta por resinas de PVC, cargas minerais, plastificantes, estabilizantes e dependendo da aplicação final, pigmentos e uma resina que promova adesão. Ele pode ser produzido a partir da utilização de dois tipos de homogeneizadores: sigma e cowles. Neste trabalho estudou-se qual é o melhor tipo de homogeneizador para a produção do Plastisol e avaliou as propriedades de aderência, viscosidade e tixotropia obtida com a utilização desses dois processos. Para melhor observar o fenômeno da aderência, viscosidade e tixotropia, foram produzidas em laboratório quatro amostras em situações processuais diferentes. Foram confeccionados corpos-de-prova para cada tipo de processo utilizado, aos quais se aplicaram o teste de tensão de cisalhamento em um dinamômetro obtendo os valores da intensidade da força e um reômetro para o estudo de viscosidade e tixotropia determinando desta forma qual foi o melhor processo utilizado.

A dor neuropática atinge cerca de 6 a 10% da população global e estima-se o seu aumento nos próximos anos. Essa síndrome não tem cura e afeta consideravelmente a qualidade de vida das pessoas por ela acometidas. Os medicamentos utilizados atualmente para o seu tratamento, como antidepressivos, anticonvulsivantes, opióides, dentre outros, não proporcionam um resultado satisfatório pelo fato de não reduzirem consideravelmente os sintomas e/ou por terem muitos efeitos colaterais. Pesquisas recentes mostram que o receptor sigma-1 pode ser utilizado no tratamento da dor neuropática. Verificou-se na literatura uma nova série de pirimidinas que são capazes de se ligar ao receptor sigma-1, atuando como seus antagonistas, sendo potenciais alvos para a produção de fármacos que podem ser utilizados no tratamento da dor neuropática. Então, estudos de Relações Quantitativas Estrutura-Atividade (QSAR) foram realizados utilizando os métodos de Mínimos Quadrados Parciais (PLS) e Redes Neurais Artificiais (ANN) para prever a atividade biológica dessa série de pirimidinas. Os resultados obtidos se mostraram satisfatórios tanto para o método de PLS (r2 = 0,877, q2 = 0,800 e r2teste = 0,738), quanto para o método de ANN (r2trein = 0,734, r2val = 0,753 e r2teste = 0,676), mostrando que o conjunto de compostos antagonistas do receptor Sigma-1 pode ser descrito tanto de forma linear quanto de forma não-linear. / Neuropathic pain affects about 6 to 10% of the global population and it is estimated to increase in the coming years. This syndrome has no cure and considerably affects the life quality of people affected by it. Medications currently used for its treatment, such as antidepressants, anticonvulsants, opioids, among others, do not provide a satisfactory result because they do not significantly reduce the symptoms and/or have many side effects. Recent research shows that the sigma-1 receptor can be used in the treatment of the neuropathic pain. A new series of pyrimidines have been found in the literature, which are capable of binding to the sigma-1 receptor, acting as its antagonists, and have been synthesized as potential targets that can be used in the treatment of the neuropathic pain. Therefore, Quantitative Structure-Activity Relationships (QSAR) were performed using Partial Least Squares (PLS) and Artificial Neural Networks (ANN) methods to predict the biological activity of this series of pyrimidines. Through the mathematical models obtained by PLS (r2 = 0.877, q2 = 0.800 and r2test = 0.738) and ANN (r2trein = 0.734, r2val = 0.753 and r2test = 0.676) methods, it was showed that they were able to predict the biological activity of the studied pyrimidines.

Previous issue date: 2008 / Resumo: Este trabalho tem o objetivo de mostrar a eficiência da aplicação da metodologia Lean Six Sigma na Logística, isto é, mais especificamente na redução do tempo de atendimento a pedidos. Tanto o Lean quanto o Six Sigma, até então aplicados de maneira isolada, tinham reduzido significativamente o nível das perdas (estoques) ou melhorado a velocidade dos processos ou reduzido a variabilidade. Porém, se fazia necessária uma abordagem diferente e completa para se reduzir ainda mais o lead time dos pedidos. E, a partir daí, um método é proposto e aplicado neste trabalho para focar a atenção na redução destas perdas, melhorar o processo de atendimento aos pedidos e, conseqüentemente, diminuindo o tempo de atendimento. A metodologia LeanSigma proposta para processos da Logística consiste basicamente em melhorar o gerenciamento dos inventários, isto é:

As the competitive environment changes the way companies do business, companies are embracing both (Lean and Six Sigma) initiatives together to support cost reductions and quality improvements in several different business areas. This work has the objective of showing the efficiency application of the Lean Six Sigma
methodology applied in the Logistics context and more specifically in the order leads time reduction. As much Lean as Six Sigma, up to now applied in an isolated way, they had reduced the level of the losses (stocks) significantly or improved the speed of the processes or reduced the variability. A method is proposed and applied in this work to focus the attention in the reduction of these losses, to improve the service process to the requests and, consequently, reducing the time of

Service quality improvement through lean management at King Khalid International Airport in Saudi ArabiaAlmuharib, Tariq Mohammed January 2014 (has links)
This thesis aims at investigating the level of service at departure area of KKIA. The study further measures the perceived quality of service and passenger satisfaction in term of tangibles, reliability, responsiveness, assurance and empathy by using five steps of lean six sigma (Define, Measure, Analyse, Improve and Control). The study aims to implement and evaluate the effectiveness of using a lean six-sigma managerial approach to the departure area of KKIA to enhance the level of service (LOS) and customer satisfaction. The passenger flow through the airport is used to measure the quality of services. Existing studies that discuss lean six-sigma implementation in airports were evaluated. Data was collected about KKIA departure area and its LOS both before and after the lean six-sigma implementation. Additionally, a SERVQUAL questionnaire was used to evaluate pre- and post-implementation review. The questionnaire features easy-to-collect data to measuring the perceived quality of service and evaluate passenger satisfaction with the LOS in KKIA. Before the study concepts of LOS and Lean was unheard within the airport community. Initial evaluation show poor customer service and many delays (waste) within the departure are at KKIA. During the study a growing awareness of Lean and LOS showed improvements in the customers’ experience, e.g, removing the shop at the entrance, improving flow thought the airport. Concluding the study, there was a total reduction in waiting from 54.74 minutes to 34.87 minutes (35.7% reduction). Also the LOS on the IATA Performance Standard went from E (Inadequate) to C (Good). This is improvement, excellent but without continuous improvements the Airport’s LOS will reduce back as passenger numbers grow from 16 million passengers per year (up from 9.8 million in 2004), to an expected 25 million by 2025. Interesting different cultural approaches to lean management became evident during the study when compared to other implementations in Europe to the Middle East. Cultural difference will always affected the reliability of the results. However, by considering these cultural effects in answering questions and interviews, the lean implementation was still successful. As culture changes are ongoing, evaluation of the situation at KKIA is required for ongoing improvements. Best-practice for the continued improvement of ongoing lean management is suggested for the departure area at KKIA. These key strategies and best-practice approaches are useful to future implementations at other airports.

A l’image des grands groupes, les PME souhaitent, elles aussi, profiter des méthodes modernes de management pour améliorer leurs processus, leurs performances et leur compétitivité. L’intérêt du secteur des PME se porte sur les éléments du concept Lean Six Sigma qui leur semblent appropriés pour réaliser ces objectifs. Après avoir constaté une lacune dans les recherches sur ce sujet et une lacune méthodologique concernant l’implémentation du LSS dans les PME, nous proposons une méthodologie de conception, d’application et de contrôle du LSS pour ces PME, que nous avons appelée LSS Plutus. Elle est dédiée aux PME de la production et elle prend en compte leur spécificité. Dans ce but, nous avons restreint la boîte à outils traditionnelle du LSS utilisée dans les grands groupes. Nous avons également créé deux variantes pour mettre en œuvre notre méthodologie qui correspondent aux besoins, capacités et maturités différents des PME. Nous avons élaboré des indicateurs, des outils, ainsi qu’un guide d’application de la méthode LSS pour les PME de la production et sa version informatique en TIBCO. Nous avons décidé de renforcer l’efficience des améliorations, en déployant les actions de planification, de standardisation et de pérennisation des changements. Dans ce but, nous proposons de modifier les critères du Modèle d’Excellence E.F.Q.M. pour qu’elles puissent prendre en compte les conditions de fonctionnement de la méthode LSS dans les PME. La méthodologie proposée a été vérifiée dans deux PME de la production en Pologne et en France, ce qui a confirmé son utilité et son efficacité en termes d’amélioration des performances et de développement des organisations / As well as the large companies, also the small and medium-sized enterprises desire to take advantage of modern management methods to improve their process, their performances and competitiveness. The interest of the SMEs is focused on the elements of the Lean Six Sigma concept that seem to be appropriate to achieve these goals. As we have identified a gap in the research on this topic and a methodological gap about the LSS implementation in the small and medium companies, we propose a methodology that we called LSS Plutus to design, apply and control Lean Six Sigma in the SMEs. It is dedicated to the small and medium production enterprises and it takes into account their specificity. For this purpose, we have limited the traditional LSS toolbox used by the large companies. We have also created two variants to apply our methodology according to the different needs, capacities and maturities of the SMEs. We have developed indicators, tools, as well as a practical guide for applying the LSS for the production SMEs and its computerized version in TIBCO. To enhance the efficiency of the improvements, we have deployed the planning, standardization and sustainability of the changes. For this purpose, we propose to modify the criteria of the EFQM Excellence Model in order to adapt it to the operating conditions of the Lean Six Sigma method in the SMEs. In a theoretical part of this work there is introduced a general view on logistics and its development, than a view on creating value for customer, basics of lean production, lean thinking and following methods as Six Sigma and Value Stream Mapping. At the end of the theoretical part there is mentioned outsourcing of logistics services and view on relationship between logistics provider and a client. In a practical part there is introduced a project of in-plant logistics optimalization in a factory producing steel pannels for MINI Cooper production. At the beginning there is introduced the factory itself, logistics services provider and a background of the project birth. Than there is a step by step description of a work on the project respecting a DMAIC methodology, it means that it's a classical example of a Six Sigma project using a Value stream mapping tool.
