Influência de agentes suspensores e edulcorantes na suspensão de estearato de eritromicina / Influence of suspending agents and sweeteners in erithromycin stearate suspension

As suspensões de estearato de eritromicina, de procedência nacional, apresentam a inconveniência que reside na elevada viscosidade ou flutuação do fármaco disperso. Foram estudados diferentes adjuvantes farmacotécnicos, a fim de verificar fatores interferentes que poderiam melhorar a estabilidade da suspensão de estearato de eritromicina. Como agentes suspensores foram utilizados BoniasolR2600 (carboximetilcelulose sódica), veegumR HV (silicato de alumínio e magnésio) e ViscosolR RV (glicolato de amido sódico) e como edulcorantes a glicerina, sorbitol a 70% e sacarose. As determinações analíticas para avaliar a estabilidade das suspensões foram: pH, densidade, viscosidade, aspecto da suspensão, volume do fármaco disperso, características de ressuspensão e doseamento do antibiótico. As suspensões contendo BoniasolR 260 e glicerina ou sorbitol não apresentaram o fenômeno da flutuação. As preparações contendo VeegumRRV e glicerina ou sorbitol na concentração de 30% (p/v) apresentaram estabilidade adequada. / Suspensions of erithromycin stearate of national origin present the inconvenience of the high viscosity or of the flotation of the dispersed drug. Different pharmaceutical helpers were studied in order to verify of the erythromycin stearate suspension. BoniasolR2600 (sodium carboxymethyl cellulose), VeegumR HV (aluminum magnesium silicate) and ViscosolR RV (sodium starch glycollate) were used as suspending agents and glycerin, sorbitol at 70% and sucrose were used as sweeteners. Analytic determination to evaluate the suspensions stability were: pH, density, viscosity, aspecto of suspension, volume of the dispersed drug, resuspension characteristics and determination of the antibiotic dose. Suspensions containing BoniasolR 2600 and glycerin or sorbitol didn\'t present floatation. Preparation with veegumR HV and glycerin or sorbitol in a concentration of 30% (p/v) presented adequate stability.

Synthèse et caractérisation de nouveaux synthons et mousses biosourcés, à partir de sorbitol / Synthesis and characterization of new building blocks and biobased foams from sorbitol

Dans un contexte de valorisation de molécules issues de la biomasse, de nouvelles architectures moléculaires et polyols ont été développé à partir du sorbitol et divers synthons biosourcés en se basant autant que possible sur les principes de la chimie verte. A partir de réactions d’estérifications contrôlées plusieurs polyols polyesters ont été synthétisés, en l’absence de solvant. Grace à l’utilisation de diols de différentes tailles (C2 à C12) comme monomères, la viscosité et la teneur en fonctions hydroxyles des polyols finaux ont pu être adaptée jusqu’à obtention de propriétés satisfaisantes à l’élaboration de mousses polyuréthanes. Ainsi, des mousses polyuréthanes semi-flexible et des mousses polyisocyanurate rigides ont pu être formulés avec des profils cinétiques de moussage rapide (inférieur à 3 min). Les mousses polyisocyanurates rigides présentent d’excellente propriétés mécaniques et thermiques pouvant pleinement satisfaire à la l’isolation thermique de bâtiment. D’autre part, une voie de transestérification entre le sorbitol et diméthyle carbonate a été étudié afin d’élaborer une nouvelle molécule plateforme bi-fonctionnelle : un bis-cyclocarbonate. A partir de cette molécule plateforme des réactions de polymérisation par ouverture de cycles et d’aminolyses ont été mise en place pour la synthèse de diols, polyéthers réticulés, et de polyuréthanes sans isocyanate (NIPU). Les synthèses de NIPU réalisées à partir de diamines courtes ou longues (issus de dimer d’acide gras) a permis d’étudier les relations existantes entre le choix des monomères et les températures de transitions vitreuses des matériaux polymères résultant. / In a context of renewable molecules valorization, new molecular architectures and polyols have been developed from sorbitol and various biobased building-blocks with respect to the green chemistry principles. Several polyesters polyol have been synthesized from controlled esterification reactions in bulk conditions. Thanks to the used of variable size diols (C2 to C12) monomers, polyols final viscosity and hydroxyls values were tuned until the obtaining of suitable properties for polyurethanes foams elaborations. Thus, semi-flexible polyurethane foams and rigid polyisocyanurate foams were formulated with fast foaming kinetic profiles (less than 3 min). Rigids polyisocyanurates foams exhibit excellent mechanical and thermal properties, in great agreement with building insulating application requirement. Otherwise, transesterifications reaction involving sorbitol and dimethyl carbonate were studied in order to develop a new bi-functional chemical platform, a bis-cyclocarbonate. Ring opening polymerization and aminolysis reactions were investigated from this chemical platform to the elaboration of cross-linked polyether and non-isocyanate polyurethanes (NIPU). NIPU syntheses were performed with short and long diamines in order to study the relationships between monomers choice and the resulting polymer material temperature of glass transition.

Selective C-O Bond Hydrogenolysis Of Polyols Over Supported Bi(Metallic) Catalysts In Aqueous Phase / Valorisation d'hémicelluloses en polyols pour la préparation de polyesters ou résines alkydes

L'étude a porté sur la conversion en presence de catalyseurs bimétaliques supportes de trois molécules modèles de polyols (érythritol, xylitol et sorbitol) en phase aqueuse à 150-240 ° C sous 30-120 bar de H2 pour obtenir sélectivement des produits linéaires C4, C5 et C6 désoxygénés qui sont des précurseurs de polymèrs. L'activité catalytique dépend fortement de la nature du support utilisé (TiO2 vs ZrO2) et la plus grande sélectivité pour les produits désoxygénés linéaires voulus C4, C5 et C6 à une conversion de 80% est de 71, 66 et 54%, respectivement, en présence de catalyseur mixte à base de rhodium et du rhenium à 200 ° C sous 80-120 bar. Les caractérisations des catalyseurs par chimisorption de CO, MET-EDX, TGA-MS et XPS suggèrent une distribution et une réductibilité différentes des espèces de Re sur les nanoparticules Rh supportées en fonction du support permettant d'expliquer ces différences / The aim of our research project reports a study of heterogeneously catalyzed conversion of three polyol model molecules (erythritol, xylitol, and sorbitol) in aqueous phase at 150-240 °C under 30-120 bar of H2 to obtain selectively linear deoxygenated C4, C5, and C6 products used as precursors for polymer applications. The activity was strongly dependent on the nature of the support (TiO2 vs ZrO2) and the highest selectivity to the desired linear deoxygenated C4, C5, and C6 products at 80% conversion reached 71, 66, and 54%, respectively, in the presence of Rh–ReOx bimetallic catalysts at 200°C under 80-120 bar. The characterizations of the catalysts by CO chemisorption, TEM-EDX, TGA-MS, and XPS suggest a different distribution and reducibility of Re species over the supported Rh nanoparticles depending on the support that can explain these differences

Récemment, les polymères obtenus à partir de ressources renouvelables ont connus un véritable engouement pour le remplacement de matériaux obtenus à partir de ressources pétrolières. L'objectif de notre étude a été de développer de nouveaux systèmes bio-sourcés réactifs, plus précisément des systèmes époxy, pour des applications dans le domaine des matériaux composites. Le monomère le plus souvent utilisé pour formuler des réseaux époxy est le diglycidyl éther de bisphenol A, DGEBA, obtenu à partir du bisphenol A et de l’épichlorhydrine. L’intérêt pour le remplacement du bisphénol A par un précurseur bio-sourcé est accentué par la toxicité de ce dernier. Dans cette étude, nous avons étudié différents systèmes obtenus à partir de prépolymères époxy bio-sourcés et les avons comparés à un système classique à base de DGEBA en utilisant l'isophorone diamine (IPD) comme agent durcisseur. Parmi les différents prépolymères époxy bio-sourcés étudiés, certains ont été obtenus à partir de dérivés polysaccharides tels que le sorbitol et l'isosorbide. Le sorbitol polyglycidyl éther est disponible commercialement, tandis que le diglycidyl éther d'isosorbide a été synthétisé soit par une voie classique faisant intervenir l’épichlorhydrine soit par l'intermédiaire du diallyle isosorbide. Un autre prépolymère époxy dérivé du cardanol a été étudié et est aussi un produit commercial. Les structures chimiques de ces prépolymères époxy bio-sourcés ont été analysées. Les phénomènes de gélification et de réticulation des systèmes obtenus à partir de ces prépolymères et de l’IPD ont été étudiés. L’influence de la structure du prépolymère bio-sourcé et de l'agent de réticulation (classique ou dérivé de ressources renouvelables) sur les propriétés des réseaux ainsi que l'absorption d'eau par ces réseaux ont également été discutés. / In recent years, bio-based polymers derived from renewable resources have become increasingly important as sustainable and eco-efficient products which can replace the products based on petrochemical-derived stocks. Bio-based epichlorohydrin is commercially available. So the challenge to obtain a fully bio-based epoxy prepolymer is to replace bisphenol A by a bio-based precursor. Another bio-based epoxy prepolymer was derived from cardanol and is also a commercial product. Properties of the cured networks were evaluated using dynamic mechanical analysis and thermo gravimetric analysis. Influence of the bio-based epoxy prepolymer structure on the system properties as well as the influence of the crosslinking agent structure (either derived from renewable resources or bio-based ones) was discussed. Water absorption of the bio-based networks was also studied.

Characterization by optical methods of the heat denaturation of bovine serum albumin (BSA) as affected by protein concentration, pH, ionic strength and sugar concentration

The thermal denaturation of proteins has been extensively studied using several methods including differential scanning calorimetry (DSC). A custom-built optical system was used to study thermal effects on protein as an alternative method to DSC measurements. It was used to investigate the thermal stability of bovine serum albumin (BSA) with a focus on comparisons with published DSC data. In the first study, the effect of protein concentration on the thermal denaturation (Td) of BSA was determined and validated using published DSC data for bovine serum albumin (BSA). The optical rotation (OR) and transmitted light (TL) signals indicating protein conformational changes and gel formation, respectively, were collected during the heating of BSA solutions at ~6��C/min from room temperature to ~85��C. The experiments were performed on 1, 2.5 and 5% (w/v) BSA in 0.01 M phosphate buffer at pH 7 and ionic strength (IS) 0.08. BSA���s Td values obtained from this investigation were consistent with published values and had low experimental variability (CV<2.5%). In agreement with some but not all published data, increasing BSA concentration did not affect its thermal stability. Protein gel formation, however, increased with protein concentration. In the second study, changes in the OR and TL signal of BSA in 0.01 M phosphate buffer at pH 6.1, 7 and 7.9 with IS maintained at 0.04, 0.08 and 0.16 were recorded during the heating of BSA solutions at ~6��C/min from room temperature to ~85��C. BSA showed a maximum and minimum thermostability at pH 7 and 7.9, respectively, consistent with published values determined by DSC. BSA formed opaque gel at pH 6.1 approaching the BSA���s pI values. Increasing IS level did not have a significant effect on BSA���s Td value but promoted gel formation. In the third study, the optical method was applied to investigate the heat stability of BSA as affected by low concentrations of sucrose, trehalose or sorbitol. BSA solutions (2.5% w/v) in the presence of 0 5% sucrose, trehalose and sorbitol were heated at ~6��C/min from ambient temperature to ~85��C. In contrast with published work on the thermal stability of BSA in the presence of higher sugar concentrations, this study showed that increasing sugar concentration did not enhance the thermal stability of this protein. Also, the ability to promote protein stability among sucrose, trehalose and sorbitol were not significantly different. The significance of these studies is that they demonstrate that the custom-built optical methods here developed can be used to study heat-induced protein denaturation and the effect of environmental conditions. Future studies will examine other proteins such as ��-lactoglobulin or ��-lacactalbumin. A further advantage of optical systems is their ability to conduct real-time measurements which could be used for food processing control. / Graduation date: 2007

After cooking (90°C/30min), instrumental texture and color of the surimi gels were evaluated. Then, the surimi was used for the elaboration of a surimi product and sensory analysis regarding to global acceptance. The protein and moisture contents in the surimi were not influenced either by washings or by cryoprotectors, while the ash content was higher when NaCl and sucrose were the cryoprotectors and the yield was higher when only distilled water was used for the washings. The highest lipid removal was achieved with the use of distilled water for the washings and NaCl and sucrose as the cryoprotectors. The highest NNP removals were achieved at both combinations: water for washings and NaCl and sucrose as the cryoprotectors and water with salts for washings and sorbitol and sodium tripolyphosphate as the cryoprotectors. This thesis deals with the sorting and use of sweeteners in the production of fruit spreads. The theoretical part focuses on the sorting of sweetening agents and their classification. Furthermore, it presents procedures of production of fruit spreads and their assortment. The main goal is to compare sensory attributes of jams that I produced according to an assigned recipe, which is described in the practical part of this thesis. These jams were made from regional fruits (strawberries, apricots, sour-cherries) and sweetened with both natural (sucrose, sorbitol, Steviol glycosides) and artificial (sucralose) sweetening agents. The control group of this research were the students and employees of the University of South Bohemia in České Budějovice in the age of 24-60 years. The evaluators filled in a sequence test protocol and a questionnaire for consumers. The results of the sensory analysis were processed into tables and graphs with the aid of statistical methods.

Resumo:O surimi consiste em um concentrado de proteínas miofibrilares obtido a partir do músculo do peixe triturado, lavado e adicionado de agentes crioprotetores. Este estudo teve o objetivo de caracterizar os surimis de tilápia obtidos com variações nos tipos de lavagens (água destilada ou água destilada + NaHCO3 + NaCl) e nos crioprotetores (NaCl + sacarose ou sorbitol + tripolifosfato de sódio), utilizando um esquema fatorial 2 x 2. Os tratamentos foram avaliados quanto a remoção dos compostos nitrogenados não proteicos (NNP), rendimento e composição centesimal dos produtos finais. Após o tratamento térmico (90°C / 30min), a textura e a cor dos géis de surimi foram avaliadas instrumentalmente. O surimi foi utilizado na elaboração de derivados, que foram analisados quanto à aceitação global. As concentrações de proteína e umidade dos surimis não sofreram influência dos processos de lavagens ou dos crioprotetores,enquanto que a concentração de cinzas foi maior quando NaCl e sacarose foram usados como crioprotetores e o rendimento foi maior quando apenas água destilada foi utilizada nas lavagens. A maior remoção de gordura foi alcançada quando as lavagens foram realizadas apenas com água destilada e os crioprotetores foram NaCl e sacarose. As maiores remoções de NNP foram alcançadas com ambas as combinações: apenas água destilada nas lavagens e NaCl e sacarose como crioprotetores e água com sais nas lavagens e sorbitol e tripolifosfato como crioprotetores. Os tratamentos utilizados produziram géis de cor e textura semelhantes.
