Závislost velikosti proudu IKs kanálu srdce na stimulaci / Cardiac IKs channel: rate-dependence of the current magnitude

This diploma thesis deals with study of the rate-dependence of the magnitude of a current through the heart channel that conducts slowly activating component of delayed rectifier outward current (IKs). This property is very important for the IKs channel function. When other repolarizing currents are insufficient, but also when the heart rate accelerates, especially during elevated sympathetic tone, IKs provides so-called repolarization reserve, which prevents excessive lengthening of cardiac action potential repolarization. The IKs channel structure is encoded by the KCNQ1 (pore-forming -subunit) and KCNE1 (modulatory -subunit) genes. Mutations in these genes disrupt the physiological function of the IKs channel and cause inherited arrhythmogenic syndromes, especially long QT syndrome (LQTS). Such mutations include the c.926C>T (p.T309I) mutation in the KCNQ1 gene, which results in LQTS type 1 in heterozygous carriers. The theoretical part of the thesis provides basic information about the IKs channel and the patch clamp technique, this knowledge is necessary for the practical part. The experimental part is focused on cultivation of the CHO cell line and its transient transfection for subsequent electrophysiological measurements by whole-cell patch clamp technique to study the dependence of the IKs magnitude on stimulation frequency, both in the wild type channels (i.e. without mutation) and in those with cotransfected wild type and T309I subunits.

The main aim of this thesis is to propose changes to the current motivational program in a selected business corporation and other areas closely related to employee motivation. To achieve this goal, the relevant theoretical background has been studied. Subsequently, key terms such as motivation, stimulation and others were defined. Furthermore, an analysis of the current situation in the company was made. The research consists of a statistical test that examines the relationship of pre-specified qualitative factors. The calculations are based on data from the questionnaire survey. The results can, for example, indicate whether factors in the form of work environment, age or level of education affect employee preferences in motivation. Based on the acquired knowledge and data, specific changes in relation to employee motivation were proposed. For example, a proposal in the area of financial or non-financial bonuses, a program for young people, leisure activities, competition employee of the month and others.

This thesis deals with questions of scanning electric activity of brain,¬ so-called electroencephalograph (undermentioned EEG), methods of audiovisual stimulation (undermentioned AVS) and a data-measurement processing. Theoretical part of the thesis is engaged in a theory of EEG signal creation, history and even in current methods of purchasing and processing of the EEG signal, theory of AVS and a theory of biofeedback. For measuring EEG signal with or without an application of AVS methods has been used EEG diagnostic device by Alien company. Its attributes are described in the thesis as well. For elaboration and analysis has been created a programme aplication EEG_xhroze00.fig which realizes frequency spectrum analysis using Fast Fourier Transform algorithm (FFT) and another programme aplication brain_mapping.fig for mapping activity of brain using designed algorithm.

This master´s thesis deals with the problem of heart stimulation by using external pacemaker. Such external pacemaker is used for a short – time stimulation of patient to overcome the time before the implantation of such apparatus. The master´s thesis informs about the basic function of heart and possible heart disorders which are treated by pacemaker. Great deal of master´s thesis is devoted to the design of external pacemaker wiring. This section describes the possibilities of controlling the pacemaker, circuits for detection R-R interval, a design of output circuit eith regard to required of stimulation and pacemaker feeding. The program Pspice was used to check the functional appropriateness of designed circuits and it was successively simulated in a non - soldering field.

Cílem této diplomové práce je najít a popsat souvislost mezi fixací očí v emočně zabarveném stimulu, kterým je obrázek či video, a EEG signálu. K tomuto studiu je třeba vyvinout softwarové nástroje v prostředí Matlab k úpravě a zpracování dat získaných z eye trackeru a propojení s EEG signály pomocí nově vytvořených markerů. Na základě získaných znalostí o fixacích, jsou v prostředí BrainVision Analyzeru EEG data zpracovány a následně jsou segmentovány a průměrovány jako evokované potenciály pro jednotlivé stimuly (ERP a EfRP). Tato práce je vypracována ve spolupráci s Gipsa-lab v rámci výzkumného projektu.
