This diploma thesis is theoretically-empirical. The theoretical part provides us with information about stress and burnout syndrome obtained on the basis of study of relevant literature. It defines the basic concepts of stress and burnout syndrome and introduces us to their basic characteristics. In the second empirical part is the rate and frequency of burnout staff is determined on the basis of a questionnaire survey in the sample of pedagogical staff. As a partical part targeted interviews with selected teachers are used as a complementary method.

Successfull cardiopulmonary resuscitation is an essential life-saving tool; nevertheless, general ischemia during cardiac arrest may trigger different pathways that could turn even into a fatal damage; this condition is called post-cardiac arrest syndrome. It has been repeatedly shown that oxidative stress (OS) plays one of the key roles in the development of ischemia-reperfusion injury. However, current evidence on the possible participation of OS in the pathogenesis of post-cardiac arrest syndrome is insufficient. We tested following hypotheses: (i) ischaemia-reperfusion injury after cardiac arrest is accompanied by OS and (ii) mild therapeutical hypothermia decreases OS cardiac arrest. In the experimental part of our work we studied the effects of hypothermia and normothermia on hemodynamic parameters, markers of organ damage and on the OS burden in porcine model of cardiac arrest. Furthermore, we compared the effects of hypothermia with ischaemic postconditioning and nitric oxide administration in the porcine model of extracorporeal cardiopulmonary resuscitation. We found protective effects of hypothermia on all major endpoints including OS in comparison with normothermia; moreover, hypothermia improved also selected variables compared to ischemic postconditioning and nitric oxide. My diploma work deals with the ethical approach of workers who come into contact with the clients committed criminal offences. The work focus on ethical issues of the social workers who are in contact with the clients with social pathology, perpetrators, involuntary clients and with persons suffering from asocial behaviour. The ethical approaches are connected with various ethical paradoxes, problems and they are a part of ethical dilemma. These facts arise in ethical attitudes in connection with client and simultaneously in situation of burn-out syndrome. The target of this work is to focus on ethical issues and on the problems which are connected with the duties of the social workers. The main target of my diploma work was to find out ethical errors of workers in social services in access to the client and find out the range of burn-out syndrome by social workers. The aim of my research has been achieved. The method of quantitative analytic research was applied. The questionnaire survey and its assessment was used. It is the quantitative research. As respondents were chosen members of the Prison Service of the Czech Republic and workers of the Educational institution for Children and Youth and the Educational Centre for Children and Youth in Šindlovy Dvory. The second file of respondents was comprised by members of the Probation and Mediation Service of the Czech Republic.

In the first part of the thesis I address the theoretical basis of the issue of social work, lifelong learning, the burnout syndrome and the ethical code of a social worker. Of health social worker help the others who face a problematic situation. They help people in order to enable them to exist and function in interaction with their social environment at a socially appreciated level. In the practical part of the thesis, the objective of the research that I set myself was to ascertain whether the health social workers have the competences to exercise their profession and what techniques they use to prevent the burnout syndrome. I decided for a written anonymous questionnaire to collect the data. The research was conducted in health and social facilities in 8 regions of the Czech Republic. 160 questionnaires were distributed altogether. The total research set consisted of 106 respondents. I examined two hypotheses. The first one was: A majority of health social workers does not have sufficient education to exercise the profession. The second one was: Of health social workers realize the risks of the burnout syndrome.

Le système de conduction ventriculaire (SCV) est responsable de la propagation rapide de l’activité électrique dans le cœur. Il est composé du faisceau de His, des branches droite et gauche, et des fibres de Purkinje. 1) Le développement du SCV a été étudié par l’utilisation de souris transgéniques exprimant la recombinase Cre inductible, insérée par recombinaison homologue au locus du gène de la connexine-40 (Cx40). Ce gène code pour une protéine exprimée dans les trabécules ventriculaires et le SCV. La recombinaison est observée uniquement dans les cellules exprimant la Cx40 et leur descendance. Mes résultats révèlent une restriction progressive du destin des trabécules Cx40-positives en cardiomyocytes conducteurs. Les progéniteurs Cx40-positifs participent à la formation du myocarde contractile et du SCV de E10.5 à E14.5 ; alors qu’ils ne donnent que des cardiomyocytes conducteurs après E16.5. 2) L’analyse du SCV a été étudiée dans un modèle de malformation cardiaque congénitale. Le mutant Tbx1-/-, modèle du syndrome de DiGeorge humain, présente une communication inter-ventriculaire. Des défauts morphologiques du SCV sont détectés chez ces mutants : absence de branche droite et un faisceau de His moins compacté. Ceux-ci sont corrélés avec des défauts de conduction. Le phénotype observé ne résulte pas d’un défaut d’activation du programme génique à l’origine de l’établissement du SCV ; mais semble dû à la présence de la communication inter-ventriculaire qui empêcherait les cellules progénitrices du SCV de rejoindre le sommet du septum inter-ventriculaire. / The ventricular conduction system (VCS) is responsible for the rapid propagation of electrical activity in the heart. The VCS is composed of the His bundle, the left and right bundle branches, and the peripheral Purkinje fibers. 1) The development of the VCS has been studied by using transgenic mice expressing the inducible Cre recombinase, introduced by homologous recombination at the locus of gene of connexin-40 (Cx40). Cx40 encodes for a protein expressing in the ventricular trabeculae and the VCS. The recombination is observed in the cells expressing Cx40 and in their descendants. My results suggest a progressive restriction of the fate of Cx40-positive trabeculae in conductive cardiomyocytes. Cx40-positive progenitors give rise to the formation of the compact myocardium and of the VCS when they are induced between E10.5-E14.5; while they participate only in the cardiomyocytes of VCS after E16.5. 2) The analysis of the VCS has been studied in a model of congenital heart malformation. The mutant Tbx1-/-, model of the DiGeorge syndrome, present a ventricular septal defect. Morphological defects of the VCS are found in Tbx1-/- hearts: an absence of right bundle branch and a non-compacted His bundle; which are correlated with functional defect. The phenotype observed in these mutants does not result from a defect of the activation in the genetic program being at the origin of the establishment in the VCS, but seems to be explain by the presence of a large ventricular septal defect, because it could block the progression of the progenitors of the VCS along the crest of the inter-ventricular septum.

There is no doubt that the human population is growing old worldwide. As a result we more often think about the ageing and the old age. One of the motives which is, however, connected with ageing is the rough handling, which may develop into maltreatment, abuse and neglect (it is known as the EAN syndrome - Elder Abuse and Neglect). In this graduation theses the authoress targets the violence on seniors in their own families. In this case the aggressor is their child, grandchild or another relative. The main aim of this study is to analyse the main cause which leads to EAN. This negative social phenomenon is described from the point of view of the employee of crisis phone line for seniors.

The thesis deals with problems of burnout among special educators working in special primary schools with students with multiple disabilities. There are defined basic concepts in the theoretical part of the work - burn-out syndrome, its symptoms, causes and stages of development. Separate chapters are devoted to the diagnosis of burn-out syndrome and prevention. Furthermore, the concept of a special educator and a multiple disability is explained in the theoretical section and a special primary school is described here. The practical part focuses on quantitative research into burn-out syndrome in two selected groups - special educators in special primary schools and teachers in ordinary primary schools. The results of the investigation are then compared with each other.

Background. Burnout is a syndrome which affects, particularly, employees of the so-called helping professions. It can influence quality of life and reduce job performance together with the quality of caring about clients. Thus it is not surprising that there is a tendency to prevent burnout and mitigate its symptoms. Developing mindfulness and self-compassion is one of the researched possibilities. Aims. This thesis aims to theoretically discuss and subsequently verify by research anticipated link between mindfulness, self-compassion and burnout affecting employees in Czech psychiatric hospitals and wards who work directly with their clients. Data and procedure. 160 adult respondents have filled in a demographic questionnaire, The Self-Compassion Scale (SCS-SZ), The Five Facet Mindfulness Questionnaire (FFMQ-15-CZ) and The Copenhagen Burnout Inventory (CBI) in an online survey. Statistical analysis. The figures of descriptive statistics and Pearson's correlation coefficients were calculated via Jamovi program. Results. The results have shown that there are significant links between mindfulness, self-compassion and the degree of burnout in case of the respondents. Limits. Author: Bc. Veronika Maršíčková Title: The Effect of Functional Electrical Stimulation Applied by the WalkAide Neurostimulator on Balance and Gait in Patients with Multiple Sclerosis. Objectives: The main aim of this diploma thesis is an assessment of therapeutic effect of functional electrical stimulation applied with using of the neurostimulator WalkAide on balance and gait of people with multiple sclerosis against standard outpatient physiotherapy. Partial aims are comparisons the immediate and persistent therapeutic effect of functional electrical stimulation. Methods: The thesis is a randomized controlled trial. For its realization it was necessary to create an intervention and a control group of probands. Both groups received 2 different rehabilitation programs. The intervention group received the WalkAide neurostimulator (for 2 months) for home use. The control group underwent a standard outpatient physiotherapeutic program with application of 20 therapies (at least) lasting 60 minutes (i.e. 2-3 times a week for 2 months). Data collection was performed by validated motor tests and questionnaires, which were focussed on endurance, speed and skilfulness of gait, static and dynamic balance and subjective perception of balance, overal mobility and pathological fatigue.
