Modelos de regeneração hepática em animais em crescimento: estudos histológicos, moleculares e avaliação de efeitos de imunossupressores / Experimental models of liver regeneration in growing animals. Histological and molecular studies, and evaluation of the effects of immunosuppressants

INTRODUÇÃO: Transplantes parciais de fígado em crianças têm sido realizados com maior freqüência, enfatizando a importância do estudo da regeneração hepática, bem como dos efeitos de drogas imunossupressoras sobre a mesma. A regeneração do parênquima é o resultado do balanço entre multiplicação celular e apoptose, esta última definida como morte celular programada. Neste processo, estão envolvidas expressões de genes pró-apoptóticos (Bak e Bax) e anti-apoptóticos (Bcl-XL e Bcl-2). Dentre as proteínas relacionadas à proliferação hepatocitária, destaca-se a interleucina-6 (IL-6). Embora o modelo de ressecção de 70% da massa hepática de ratos adultos seja amplamente utilizado para estudos de regeneração, não há trabalhos com animais em crescimento. MÉTODOS: Na presente pesquisa, foi padronizado o modelo de hepatectomia parcial em ratos recém-nascidos e em recém-desmamados: realizou-se ligadura com fio de algodão do pedículo dos lobos esquerdo lateral, esquerdo medial e direito medial, seguida da ressecção do parênquima desses lobos. Os fígados remanescentes foram imediatamente pesados e comparados com os pesos dos fígados de animais controles. Para caracterização dos modelos de regeneração, 40 ratos recém-nascidos e 30 recém-desmamados foram submetidos à hepatectomia descrita e mortos nos dias subseqüentes (1, 2, 3, 4 e 7), e o fígado residual submetido a análises de peso e histologia convencional. A seguir, 36 animais (18 recém-nascidos e 18 recém-desmamados) foram divididos nos seguintes grupos: controle, cirurgia simulada e hepatectomia. Um dia após, utilizando métodos moleculares (técnica do RT-PCR), estudou-se a expressão do gene da IL-6, dos genes pró-apoptóticos e anti-apoptóticos nos fígados desses animais e, por meio de métodos imunoistoquímicos, analisou-se a presença de antígenos relacionados à proliferação celular (PCNA) e apoptose (TUNEL) em lâminas preparadas pela técnica do \"tissue microarray\". Em outros 36 ratos (18 recém-nascidos e 18 recém-desmamados), foram administradas drogas imunossupressoras (metilprednisolona, ciclosporina A ou tacrolimus, separadamente) no ato da hepatectomia, e o parênquima hepático remanescente submetido às mesmas análises moleculares e imunoistoquímicas, um dia após. RESULTADOS: A ressecção do parênquima hepático correspondeu a 70% da massa total do fígado. A mortalidade relacionada à hepatectomia nos animais recém-nascidos e recém-desmamados foi 30% e 0% respectivamente. Na análise histológica observou-se maior quantidade de mitoses em hepatócitos no terceiro dia nos recém-nascidos e no segundo dia nos recém-desmamados, com normalização da arquitetura do parênquima até o 7º dia e recuperação total do peso de ambos. No animal recém-nascido, notou-se intensa esteatose associada ao processo regenerativo. A hepatectomia provocou aumento na expressão do gene da IL-6 no fígado residual e diminuição da expressão dos genes pró-apoptóticos em ambos os modelos, além de aumento do anti-apoptótico Bcl-2 nos animais recém-desmamados. O estudo realizado sobre o efeito das drogas imunossupressoras mostrou resultados diferentes daqueles descritos em animais adultos, não havendo alteração no número de células em proliferação (PCNA positivas) ou apoptose (TUNEL positivas). As drogas não tiveram efeito sobre a expressão do gene da IL-6. Metilprednisolona e tacrolimus ocasionaram aumento da expressão do gene anti-apoptótico Bcl-2 em ambos os modelos; metilprednisolona e ciclosporina provocaram aumento na expressão do gene pró-apoptótico Bak nos ratos recém-nascidos. CONCLUSÕES: os modelos de regeneração hepática em ratos recém-nascidos e recém-desmamados foram exeqüíveis e adequados para a pesquisa; a hepatectomia promoveu estímulo da proliferação de hepatócitos com inibição da apoptose; as drogas imunossupressoras utilizadas não exerceram efeito sobre a proliferação de hepatócitos porém provocaram aumento da expressão de genes relacionados a apoptose. / INTRODUCTION: Partial liver transplantation has been performed in children with increasing frequency, and this emphasizes the importance of the studies of hepatic regeneration and the effects of immunosuppressive drugs on this phenomenon. Liver regeneration is controlled by the balance between cell proliferation and apoptosis (defined as a programmed cell death that results from the expression of pro-apoptotic genes - Bax and Bak - and anti-apoptotic genes - Bcl-2 and Bcl-XL). Among proteins related to hepatocyte proliferation, interleukin-6 is an important one. Although the adult rat model of 70% hepatectomy has been widely utilized for studies of liver regeneration, there are no studies using growing animals. METHODS: In the current paper, two experimental models were created utilizing newborn and weaning rats: using a cotton thread, the vascular hilum and the hepatic vein were ligated and the left lateral, left medial and right medial lobes were resected. The remaining liver was immediately harvested and weighted to be compared to control livers. The animals were sacrificed on days 1, 2, 3, 4, and 7 after the operation, the remnant livers were weighted and harvested for histological examinations. Then, 36 animals (18 newborn and 18 weaning animals) were divided into the following groups: control, sham and hepatectomy. One day after, the expressions of IL-6 gene, pro-apoptotic and anti-apoptotic genes were studied in the remnant livers. Immunohistochemical stainings for cell antigens related to cell proliferation (PCNA) and apoptosis (TUNEL) were also performed utilizing tissue microarray sections. In another group of 36 animals (18 newborn and 18 weaning animals), immunosuppressive drugs were administered just after the hepatectomy (methylprednisolone, cyclosporine A or tacrolimus separately), and the remnant liver submitted to the same molecular and immunohistochemical studies. RESULTS: The resected liver corresponded to 70% of the total liver weight. The mortality rates after hepatectomy were 30% and 0% for newborn and weaning rats, respectively. The histological examinations showed a great number of mitoses of hepatocytes on the third day in newborns and on the second day in weaning rats, and normalization of histological aspects by 7 days after hepatectomy and weight recuperation. In the newborn group liver regeneration was related to an intense steatosis. Hepatectomy promoted an increase in the expression of IL-6 gene of the remnant liver, a decreased expression of pro-apoptotic genes in both models, and an increased expression of anti-apoptotic Bcl-2 gene in weaning rats. The study of the effects of immunosuppressants showed different results from those described in adult animals, with no alterations in the number of cells in proliferation (PCNA positive) and apoptosis (TUNEL positive). Drugs had no effect in expression of IL-6 gene. Methylprednisolone and tacrolimus promoted an increased expression of anti-apoptotic gene Bcl-2. In addition, methylprednisolone and cyclosporine promoted an increase in the expression of the pro-apoptotic gene Bak in newborn rats. CONCLUSIONS: The experimental models were feasible and adequate for the current investigations; hepatectomy stimulated hepatocyte proliferation and inhibited hepatic cells apoptosis; the utilized immunosuppressant drugs did not affect hepatocyte proliferation although an increased expression of apoptosis-related genes was verified.

This diploma thesis focuses on the implementation of waterproofing of underground constructions using an LLDPE membrane hinged on anchors. In the future, this method will replace the commonly used waterproofing system in Scandinavia with extruded polyethylene foam. This method of tunnel construction and waterproofing is compared with the NRTM method, often used in Central Europe. The thesis describes the installation of a waterproofing system, especially materials, accessories, welding devices and auxiliary installation equipment. The main focus of the thesis is to verify the reliability of the individual types of welds and to assess the waterproofing system in terms of implementation and environmental demands.

Contrôle de l'organisation moléculaire en 2D et 3D par l’utilisation de liaisons hydrogène, de coordination métallique et d'autres interactions

La stratégie de la tectonique moléculaire a montré durant ces dernières années son utilité dans la construction de nouveaux matériaux. Elle repose sur l’auto-assemblage spontané de molécule dite intelligente appelée tecton. Ces molécules possèdent l’habilité de se reconnaitre entre elles en utilisant diverses interactions intermoléculaires. L'assemblage résultant peut donner lieu à des matériaux moléculaires avec une organisation prévisible. Cette stratégie exige la création de nouveaux tectons, qui sont parfois difficiles à synthétiser et nécessitent dans la plupart des cas de nombreuses étapes de synthèse, ce qui empêche ou limite leur mise en application pratique. De plus, une fois formées, les liaisons unissant le corps central du tecton avec ces groupements de reconnaissance moléculaire ne peuvent plus être rompues, ce qui ne permet pas de remodeler le tecton par une procédure synthétique simple. Afin de contourner ces obstacles, nous proposons d’utiliser une stratégie hybride qui se sert de la coordination métallique pour construire le corps central du tecton, combinée avec l'utilisation des interactions plus faibles pour contrôler l'association. Nous appelons une telle entité métallotecton du fait de la présence du métal. Pour explorer cette stratégie, nous avons construit une série de ligands ditopiques comportant soit une pyridine, une bipyridine ou une phénantroline pour favoriser la coordination métallique, substitués avec des groupements diaminotriazinyles (DAT) pour permettre aux complexes de s'associer par la formation de ponts hydrogène. En plus de la possibilité de créer des métallotectons par coordination, ces ligands ditopiques ont un intérêt intrinsèque en chimie supramoléculaire en tant qu'entités pouvant s'associer en 3D et en 2D. En parallèle à notre étude de la chimie de coordination, nous avons ii examiné l'association des ligands, ainsi que celle des analogues, par la diffraction des rayons-X (XRD) et par la microscopie de balayage à effet tunnel (STM). L'adsorption de ces molécules sur la surface de graphite à l’interface liquide-solide donne lieu à la formation de différents réseaux 2D par un phénomène de nanopatterning. Pour comprendre les détails de l'adsorption moléculaire, nous avons systématiquement comparé l’organisation observée en 2D par STM avec celle favorisée dans les structures 3D déterminées par XRD. Nous avons également simulé l'adsorption par des calculs théoriques. Cette approche intégrée est indispensable pour bien caractériser l’organisation moléculaire en 2D et pour bien comprendre l'origine des préférences observées. Ces études des ligands eux-mêmes pourront donc servir de référence lorsque nous étudierons l'association des métallotectons dérivés des ligands par coordination. Notre travail a démontré que la stratégie combinant la chimie de coordination et la reconnaissance moléculaire est une méthode de construction rapide et efficace pour créer des réseaux supramoléculaires. It relies on the spontaneous self-assembly of molecules called tectons. These molecules have the ability to recognize themselves using various intermolecular interactions. The resulting assembly can produce molecular materials with predictable organization. This strategy requires the creation of new tectons, which are sometimes difficult to synthesize and require in most cases many synthetic steps, which prevents or limits their practical application. Moreover, once formed, the bonds joining the central core of the tecton with the groups used for molecular recognition cannot be broken, which means that it is not possible to recycle or reform the tecton by simple synthetic procedures. To avoid these obstacles, we propose to use a hybrid strategy that uses metal coordination to build the central core of the tecton, combined with the use of weaker interactions to control the association. We call such entities metallotectons due to the presence of metal. To explore this strategy, we constructed a series of ditopic ligands containing either pyridine, bipyridine or phenanthroline to promote metal coordination, substituted with diaminotriazinyl groups (DAT) to allow inter-complex association by the formation of hydrogen bonds. In addition to the possibility of creating metallotectons by coordination, these ditopic ligands have an intrinsic interest in supramolecular chemistry as entities that can associate in 3D and 2D. In parallel to our study of coordination chemistry, we examined the association of ligands by X-ray diffraction (XRD) and scanning tunneling microscopy (STM). The adsorption of these molecules on the graphite surface at the liquid-solid interface results in the formation of different networks through a process of 2D nanopatterning. To understand the details of iv molecular adsorption, we systematically compared the 2D organization observed STM with the 3D structures determined by XRD. We also simulated the adsorption by theoretical calculations. This integrated approach is essential to characterize the molecular organization in 2D and to understand the origin of the observed preferences. These studies of the ligands themselves may therefore serve as a reference when we study the association of metallotectons derived by ligands coordination. Our work demonstrates that the strategy combining coordination chemistry and molecular recognition is a rapid and an efficient method to create supramolecular networks. We verified that the strategy of molecular tectonics is also effective in leading the organization in 3D and 2D, which often shows a significant homology. We found that our heterocyclic ligands have unexpected ability to adsorb strongly on the graphite surface, creating networks organize in nanoscale. Together, these results provide promising applications in several fields, including heterogeneous catalysis and nanotechnology. The Blanka tunnel has two boring sections: Brusnice tunnel and Královská obora tunnel. Both tunnels were boring in difficult geologycal conditions, but the geology conditions of these tunnels are not same. Also they had not the same hazards during boring. There was used a spectrum of different technical works with hazard reduction purpose and safety process of boring. There were three serious accidents, wnich everyone made a creater on the surface, but they have not the same causes. This text has a task to describe a geological survey before and during the boring of both tunnels. How other reasons affected the process of boring? How the tunnels were bored? What events preceded the everyone of three major failure? This text does not deal with economical things of boring and effects of specific personal influence on the process of boring. V této diplomové práci bylo zkoumáno téma kalibrace zařízení a sekcí ve vztahu k aerodynamickému tunelu na brněnském institutu leteckého inženýrství. Přístroje používané v aerodynamickém tunelu jsou kalibrovány a jsou popsány a provedeny kalibrace sekcí na testovací sekci. Kalibrační listy a šablony sestav jsou vytvořeny za účelem zjednodušení budoucích procesů. Nově kalibrovaná sestava se používá k provádění měření síly a probuzení na modelovém voze, která jsou porovnána s předkalibrací měření, aby dále ilustrovala užitečnost kalibrací.

Specifically it is a part of tunnel complex Blanka. The name of this part is Kralovská obora. If the all tunnel parts are built this tunnel complex Blanka can be complemented to already opened tunnels Zlíchovský, Mrázovka and Strahovský which are the part of city traffic circle. This technology preparation includes schedules and financial plans of the building, technological prescription for the primary tunnel lining, safety plan, control and trial/test plans, environmental plan, project of construction site, design of the main working machines, items budget of the north tunnel tube, coordination situation plan with wider relationships transport routes and technical report of building technological project.

This work describes the visualization design of the flight related quantities in a cockpit of a modern light sport aircraft. It focuses on the utilization of the state of the art trends in flight data displays and introduces the innovative implementation of the aircraft's energy state smart clues that reduce pilots' workload. The initial part of the work presents a research into the flight, engine and navigation data presentation on analog instruments, followed by their illustrative depiction in glass cockpits. Within the framework of this thesis, Microsoft Flight Simulator has been used as a source of the flight related data. Final advances in the display design were introduced through the implementation of the synthetic vision system and a visualization of the virtual tunnel in the sky.

Per tant, la via d'extensió de la vida és capaç de prevenir l'apoptosi a través d'un mecanisme desconegut. / Sustained Hog1 activation leads to an inhibition of cell growth. In this work, we have observed that the lethal phenotype caused by sustained Hog1 activation is prevented by SCFCDC4 mutants. The prevention of Hog1-induced cell death by SCFCDC4 mutation depends on the lifespan extension pathway. Upon sustained Hog1 activation, SCFCDC4 mutation increases Msn2 and Msn4 dependent gene expression that leads to a PNC1 overexpression and a Sir2 deacetylase hyperactivation. Then, hyperactivation of Sir2 is able to prevent cell death caused by sustained Hog1 activation. We have also observed that cell death upon sustained Hog1 activation is due to an induction of apoptosis. The apoptosis induced by Hog1 is decreased by SCFCDC4 mutation. Therefore, lifespan extension pathway is able to prevent apoptosis by an unknown mechanism.
