Crescimento, acúmulo de macronutrientes e produção de chicória em função do período de proteção com polipropileno /

Orientador: Arthur Bernardes Cecílio Filho / Banca: Pedro Luis da Costa Aguiar Alves / Banca: Rovilson José de Souza / Resumo: O experimento foi realizado na UNESP - Campus de Jaboticabal, SP, com o objetivo de avaliar diferentes períodos de cobertura de plantas de chicória com tecido de polipropileno (TP), branco, de 20 g m-2 sobre o crescimento, acúmulo de macronutrientes e produção de chicória crespa. O delineamento experimental foi o de blocos casualizados, em esquema fatorial 2x4, com quatro repetições. Os tratamentos constituíram-se de duas cultivares de chicória (Chicória Crespa e AF-218) e sistemas de cultivo, os quais distinguiram-se quanto a cobertura ou não com polipropileno e o tempo de permanência da cobertura, ou seja: 1- não cobertura da chicória com polipropileno; 2- 15 dias após o transplantio (DAT) coberto, 3- 30 DAT coberto e 4- 42 DAT coberto, o que correspondeu a todo o período de crescimento da chicória no campo. Para a avaliação do crescimento e acúmulo de macronutrientes, o delineamento experimental adotado foi o de parcelas subdivididas, com 4 repetições. Na parcela, os quatro tratamentos constituíram-se duas cultivares e dois sistemas de cultivo (com e sem polipropileno) e na subparcela a idade da planta (7, 14, 21, 28, 35 e 42 dias após o transplantio). A semeadura foi realizada em 02-06-2003 e o transplantio em 27-06-2003. A colocação do polipropileno foi feita diretamente sobre as plantas, fixando-se as laterais na parte externa do canteiro. Observou-se que o aumento do período de cobertura das plantas com polipropileno aumentou a altura, número de folhas e a massa fresca da parte aérea (MFPA) das duas cultivares. O emprego do TP durante todo o período pós-transplantio promoveu precocidade da colheita em oito dias, além de qualidade superior com folhas mais tenras e limpas. Somente para acúmulo de nitrogênio e cálcio não houve diferença entre os tratamentos no final do ciclo. A cobertura com TP proporcionou maior acúmulo de P, K, Mg e S para ambas... (Resumo completo, clicar acesso eletrônico abaixo). / Abstract: The aim of this work was to evaluate the growth, nutrient uptake and production of endive curly subjected to periods of plant covering with white polypropylene cloth (PC) (20 g m2). The experiment was carried out at Universidade Estadual Paulista (UNESP)- Campus of Jaboticabal, SP, Brazil. The experimental design was a randomized block design with a factorial 4 x 2 and four replications. The treatments consisted of two cultivate of endive (Chicória Crespa and AF-218), and different cultivation systems, with and without the polypropylene covering, and it’s permanence, that is: 1- without polypropylene covering; 2- 15 days after the transplanting (DAT), covering 3- 30 DAT and covering 4 - 42 DAT, what corresponded the whole period of growth of the endive in the field. For the growth and nutrient uptake evaluation the experiment was set in a randomized block design with divided-plots and four replications. In the plot, the four treatments consisted of two cultivate and two cultivation systems (with and without polypropylene) and in the subplot the age of the plant (7, 14, 21, 28. 35 and 42 days after transplant). The sowing was done in June 2nd , 2003 and the transplanting in June 27th. The placement of the polypropylene was made directly over the plants, fixing the lateral parts in the external side of the plot. Observed that the increasing of the covering period with polypropylene increased the aerial plant part, number of the leaves and fresh matter of the aerial part (FMAP) of both cultivate. The use of the PC was associated higher accumulation of P, K, Mg and S in both genotypes. Plants of AF-218 covered with PC during the whole for all period after of transplant, and with higher FMAP, accumulated 836, 515, 205, 144, 90 and 65 mg plant, respectively of N, K, Ca, Mg, S, and P. / Mestre

Avaliação do consumo de oxigênio em jogadores de polo aquático : comparação e concordância entre protocolos executados dentro e fora da água / Oxygen consumption assessment in water polo players : comparison and agreement between protocols executed in and out-water

O polo aquático (PA) é um esporte coletivo e de invasão praticado no meio líquido. Dentre as técnicas utilizadas pelos jogadores de PA, destaca-se o eggbeater (EB), movimento propulsivo de membros inferiores que permite ao jogador a permanência na posição vertical e deslocamentos. Devido às especificidades do gesto de EB e à falta de informações a respeito das características fisiológicas de jogadores de PA, o objetivo deste estudo foi comparar e verificar a concordância de resultados obtidos de testes de esforço máximo realizados dentro (com EB) e fora da água (em cicloergômetro). Para tal, doze jogadores de PA (30,5 ± 7,7 anos de idade; 79,2 ± 7,2 kg de massa corporal; 179,1 ± 5,9 cm de estatura) foram avaliados em duas etapas: (1) em laboratório com teste máximo em ciclo-ergômetro e (2) em piscina com teste máximo em EB. Em ambas as etapas foram obtidos: consumo máximo de oxigênio (VO2max), taxa de troca gasosa (RER), frequência cardíaca máxima (FCmax) e esforço percebido (EP). Entre os dados obtidos de ambos os testes foram realizadas análises descritivas, análises de comparação, correlação intra-classe e concordância. Não foram encontradas diferenças entre os valores máximos obtidos nos dois testes (ciclo-ergômetro: VO2max = 40,2 ± 2,7 ml.kg-1.min-1, RER = 1,17 ± 0,08, FCmax = 181,4 ± 11,7 bpm; EP = 20; eggbeater: VO2max = 38,4 ± 5,7 ml.kg- 1.min-1, RER = 1,19 ± ,12, FCmax = 179 ± 11,7 bpm; EP = 20). De modo geral, os valores encontrados no teste em EB apresentaram maior variabilidade, porém análises de Bland-Altmann indicam limites de concordância aceitáveis para VO2max, RER e FCmax obtidos de diferentes métodos (dentro e fora da água) entre jogadores de PA. / Water polo (PA) is a team and invasion sport performed in water. Among the techniques used by the players, the eggbeater kick (EB) is a propulsive movement of the lower limbs, which allows the player to remain in the upright position and to displace. Due to the specifics of the EB and the lack of information about the physiological characteristics of PA players, the aim of this study was to compare and to verify the agreement of maximal exercise tests in (EB) and out (cycling) water. Twelve PA players (30.5 ± 7.7 years, 79.2 ± 7.2 kg body mass; 179.1 ± 5.9 cm height) were evaluated in two steps: (1) in laboratory with maximal test on a cycle ergometer and (2) in pool with maximum test in EB. In both phases, maximal oxygen uptake (VO2max), gas exchange ratio (RER), maximal heart rate (HRmax) and perceived exertion (EP) were obtained. The descriptive analysis, comparison analysis, intra-class correlation and level of agreement were performed. No differences were found between the maximum values obtained in the two tests (cycle ergometer: VO2max = 40.2 ± 2.7 ml.kg-1.min-1, RER = 1.17 ± 0.08, FCmax = 181.4 ± 11.7 bpm; EP = 20; eggbeater: VO2max = 38.4 ± 5.7 ml.kg-1.min-1, RER = 1.19 ± 12 water, FCmax = 179 ± 11.7 bpm; EP = 20). In general, the values in EB test showed greater variability than in cycle ergometer, but the Bland-Altmann analyzes indicate acceptable limits of agreement for VO2max, RER and HRmax obtained from different methods (in and out water) in PA players.

Nutrient alterations in Scots pines (<em>Pinus sylvestris</em> L.) under sulphur and heavy metal pollution

Abstract In this study, mineral nutrition and its relation to the vigour of Scots pines growing under the influence of sulphur, copper and nickel stress were investigated. This was done by analysing the nutrient status and needle damage of pines along a pollution transect extending from the proximity of a large S, Cu and Ni emitters on the Kola Peninsula to a background area in Finnish Lapland. Additionally, pine seedlings were exposed to Cu and Ni stress in order to gain more detailed information about the mechanisms behind metal stress. The total sulphur concentrations of the youngest needles in the vicinity of the largest point source were about double compared to those in the reference area (&lt; 800 mg in kg of dry needles vs. > 1900 mg kg-1). In the case of Cu and Ni, this difference was close to 100-fold (from 2-3 mg kg-1 to over 250 mg kg-1). While the elevated sulphur concentrations were not attributable to particle deposition, the particles on needle surfaces raised the total concentrations of Cu and Ni occasionally over 1.5-fold compared to the inside concentrations. The Mn and Zn concentrations showed a decreasing trend, whereas K and P slightly increased towards the Monchegorsk smelters. Enhanced needle senescence and tip necrosis were related to high total foliar levels of Cu, Ni and S and low levels of Zn and Mn. Stomatal chlorosis and other discolourations showed a distinct relation to high SO2 levels in the air and also to high foliar concentrations of Ca, Fe, Si and Cl. Particles deposited on needle surfaces did not cause directly visible injuries in pines. In seedlings, Cu and Ni were enriched in roots: the Ni concentration was up to 16-fold and the Cu concentration 6-fold compared to that added into the soil. While Cu was mostly retained in roots, Ni had much easier access to foliage. The effect of metal stress on other nutrients varied between nutrients, plant parts and metals added. Soil analyses from both the field study and the seedling experiment gave mostly a weak estimation about the plant availability of nutrients. Foliar nutrients remained above the deficiency limits in all studies and were hence not the primary cause for the decrease of pine vigour and the consequent growth retardation and forest decline.

The role of the exercise instructor in older adults' uptake and adherence to exercise classes

Encouraging older adults to become more active and maintain that activity is critical to the promotion of their health and well-being, social networks and independence. Leadership behaviour and quality of instruction is important in influencing engagement of older adults in exercise classes. Instructors' attitudes could influence older adults' uptake and adherence to classes, but little is known about the relationship between attitudes and characteristics of instructors and their delivery in relation to uptake and adherence of older adults to exercise classes. Methods: Underpinned by the Theory of Planned Behaviour and using a mixed methods approach this thesis uses three studies to:1) Survey 731 United Kingdom exercise instructors with Level 3 older adults exercise qualification to investigate instructors' characteristics and attitudes towards older-adults' participation in exercise. 2) Interview 19 instructors to further explore instructors' attitudes, experiences and beliefs in relation to their exercise classes for older adults and how their attitudes, experiences and beliefs are influenced by their training and characteristics.3) Follow up a cohort of 16 instructors and 193 of their class participants over 6 months to explore what characteristics of exercise instructors, the group and class participants influence adherence and also whether instructors influence uptake and adherence to home exercise. Results and Conclusion: The first study establishes that there is a relationship between instructors' training, experience, characteristics and their attitudes. The second study supports these findings and demonstrates how instructors' think that these factors and others not only influence how they deliver and promote their classes but also influence older adults' uptake and adherence to exercise classes. The final study enables us to look at the relationship between instructor variables and the participant within the exercise class setting. Class participants' mental well-being, education and housing were key factors related to their attendance. Having attended the class for more than six months at baseline was an important factor related to adherence. Individual factors such as participants' attitudes, beliefs about group cohesion and instructor variables such as personality traits and experience emerged in the final models both in relation to participant attendance and adherence. There are a series of complex interactions between the instructor, participant, the group and others which influences beliefs and attitudes. It is clear that the instructor can influence participants and they have an important role to play in creating an atmosphere and environment of which participants want to be a part of.

Assessment of the Occurrence and Potential Risks of Pharmaceuticals and their Metabolites in Fish and Water Using Liquid Chromatography Mass Spectrometry

A comprehensive method for the analysis of 11 target pharmaceuticals representing multiple therapeutic classes was developed for biological tissues (fish) and water. Water samples were extracted using solid phase extraction (SPE), while fish tissue homogenates were extracted using accelerated solvent extraction (ASE) followed by mixed-mode cation exchange SPE cleanup and analyzed by liquid chromatography tandem mass spectrometry (LC-MS/MS). Among the 11 target pharmaceuticals analyzed, trimethoprim, caffeine, sulfamethoxazole, diphenhydramine, diltiazem, carbamazepine, erythromycin and fluoxetine were consistently detected in reclaimed water. On the other hand, caffeine, diphenhydramine and carbamazepine were consistently detected in fish and surface water samples. In order to understand the uptake and depuration of pharmaceuticals as well as bioconcentration factors (BCFs) under the worst-case conditions, mosquito fish were exposed to reclaimed water under static-renewal for 7 days, followed by a 14-day depuration phase in clean water. Characterization of the exposure media revealed the presence of 26 pharmaceuticals while 5 pharmaceuticals including caffeine, diphenhydramine, diltiazem, carbamazepine, and ibuprofen were present in the organisms as early as 5 h from the start of the exposure. Liquid chromatography ultra-high resolution Orbitrap mass spectrometry was explored as a tool to identify and quantify phase II pharmaceutical metabolites in reclaimed water. The resulting data confirmed the presence of acetyl-sulfamethoxazole and sulfamethoxazole glucuronide in reclaimed water. To my knowledge, this is the first known report of sulfamethoxazole glucuronide surviving intact through wastewater treatment plants and occurring in environmental water samples. Finally, five bioaccumulative pharmaceuticals including caffeine, carbamazepine, diltiazem, diphenhydramine and ibuprofen detected in reclaimed water were investigated regarding the acute and chronic risks to aquatic organisms. The results indicated a low potential risk of carbamazepine even under the worst case exposure scenario. Given the dilution factors that affect environmental releases, the risk of exposure to carbamazepine will be even more reduced.

Platelet Transcriptome Heterogeneity: A Role for RNA Uptake in Vascular Health and Disease

As our understanding of the platelet’s systemic role continues to expand beyond hemostasis and thrombosis, interrogation of the platelet’s ability to affect diverse biological processes is required. Studies of the platelet’s non-traditional roles have focused on developing our understanding of the platelet’s relation to specific disease phenotypes as well as elucidation of platelet characteristics, content, and function. The generic content, traditional function and heterogeneity of platelets have long been accepted; more ambiguous and controversial has been how these factors are interrelated. Investigation of platelet content revealed the presence of biologically functional RNA in anucleated platelets, the correlation of platelet RNA to distinct phenotypes, and the ability of platelets to transfer RNA to other vascular cells; however how these processes occur is unclear. To further interrogate platelet RNA processes, we utilized sorting and RNA sequencing to develop platelet subpopulation transcriptome profiles. We found that platelet heterogeneity extends to the platelet transcriptome: distinct RNA profiles exist dependent on platelet size. We hypothesized that this RNA heterogeneity is the result of RNA transfer between platelets and vascular cells. Using in vitro and in vivo modeling, we were able to show the novel ability of platelets to take up RNA from vascular cells, correlating to the unique functional profile associated with small platelet transcriptomes. These findings reveal a role for platelet RNA transfer in platelet RNA heterogeneity, with potential correlation to platelet functional diversity previously proposed. The ability of the platelet to bidirectionally transfer RNA within circulation has implications for vascular health and beyond.

Effet d'un rapport de Sillicium sur la senescence foliaire et les performances agronomiques du colza / Effect of silicon supply on foliar senescence and agronomic performances of rapeseed

Le colza (Brassica napus L.) est une plante de grande culture nécessitant de forts intrants azotés et présentant une faible Efficience d’Utilisation de l’Azote (EUA) principalement due à une faible efficience de remobilisation cet élément durant la sénescence foliaire. Actuellement, une amélioration du bilan agroenvironnemental de la culture du colza, nécessite la mise au point de nouvelles pratiques culturales permettant de maintenir (voire d’améliorer) son rendement tout en limitant les doses d’intrants azotés. Dans ce contexte, l’utilisation d’éléments bénéfiques tels que le silicium (Si) dont les effets ont déjà été démontrés dans la tolérance à certains stress nutritionnels (phosphore et potassium) constitue une alternative intéressante. Les objectifs de cette étude visent à étudier l’effet du Si (i) sur la progression de la sénescence foliaire, l’absorption et la remobilisation de l’azote et les performances agronomiques du colza cultivé en hydroponie ou en conditions de plein champ et soumis ou non à une limitation ou une privation en azote. Nos travaux montrent, qu’en dépit de son caractère faiblement accumulateur en Si, le colza absorbe le Si puis le stocke essentiellement au niveau racinaire. Par ailleurs, un apport de Si de courte durée (7 jours ; 1,7 mM) à des jeunes plantes privées en azote provoque un retard de sénescence des feuilles matures associé un maintien de leur teneur en chlorophylles et de leur activité photosynthétique. De plus, au champ, un apport de Si (12 kg ha-1) à des plantes cultivées avec 160 kg N ha-1 s’accompagne d’une augmentation du rendement grainier (+4,2 quintaux ha-1). L'utilisation de l’indice agronomique «Agronomic Nitrogen Recovery (ANR)» combinée à l’étude de l’expression de gènes codant des transporteurs d’azote (BnaNRT1.1, BnaNRT2.1, BnaAMT1.1) montrent que le Si agit notamment en stimulant l’absorption de l’azote chez le colza. Enfin, une analyse différentielle par RNAseq des transcriptomes racinaires de jeunes plantes traitées ou non avec du Si pendant une courte durée (7 jours ; 1,7 mM), montre que le Si module l’expression de 1079 gènes, 334 étant induits et 745 réprimés. Ce résultat indique qu’en plus de son rôle mécanique, le Si exerce également un rôle signalétique. / Rapeseed (Brassica napus L.) is a crop that requires high nitrogen inputs and has a low Nitrogen Utilization Efficiency (NUE) mainly due to a low remobilization efficiency during leaf senescence. Currently, an improvement of the agro-environmental balance of rapeseed, requires the development of new cultural practices to maintain (or even improve) its yield in response to low nitrogen inputs. In this context, the use of beneficial element such as silicon (Si) with previously demonstrated effects on tolerance to some nutritional stress (phosphorus and potassium), constitutes an interesting alternative. The objectives of this study are to investigate the effect of Si on the progression of leaf senescence, nitrogen uptake and remobilization but also on agronomic performances of rapeseed grown in hydroponics or in field conditions and submitted or not to a limitation or deprivation of nitrogen inputs. Our work shows that despite being considered as a low Si accumulator, rapeseed takes up and stores Si mainly in its roots. In addition, a Si supply for a short time (7 days; 1.7 mM) to young plants deprived of nitrogen leads to a delay of senescence of mature leaves associated with a maintenance of their chlorophyll content and their photosynthetic activity. In addition, in field conditions, a supply of Si (12 kg ha-1) to plants grown with 160 kg N ha-1 leads to an increase of yield (+4.2 quintals ha-1). Furthermore, calculation of the "Agronomic Nitrogen Recovery (ANR)" combined with the expression of genes encoding nitrogen transporters (BnaNRT1.1, BnaNRT2.1, and BnaAMT1.1) shows that Si increases the nitrogen uptake. Finally, a differential analysis by RNAseq of roots transcriptomes from young plants treated or not with Si for a short time (7 days; 1.7 mM), shows that Si modulates the expression of 1079 genes, 334 being induced and 745 repressed. This result indicates that in addition to its mechanical role, Si acts also a signal, a role very poorly demonstrated in the literature.
