Determinação de corantes sintéticos em bebidas isotônicas empregando extração em fase sólida dispersiva e HPLC-UV/Vis / Determination of synthetic dyes in sports drinks by dispersive solid-phase extraction and HPLC-UV/Vis

Coordenação de Aperfeiçoamento de Pessoal de Nível Superior / Sports drinks are specially formulated for people who are undertaking physical activity, being mainly composed by carbohydrates, electrolytes and vitamins. Synthetic dyes are food additives commonly used in sports drinks to improve their consumer characteristics and appearance. Besides the lower cost of production in relation to dyes of natural origin, synthetic colorants have several advantages such as high stability to light, oxygen and pH, color uniformity and low microbiological contamination. However some of these compounds cause adverse health effects (allergy, asthma, hyperactivity, thyroid tumors, etc.), especially when consumed in excess. Thus, their use is strictly controlled around the world. The present study aimed to develop a simple, fast and efficient method for the determination of the synthetic dyes tartrazine, sunset yellow, amaranth, ponceau 4R, indigo carmine and brilliant blue in sports drinks employing dispersive solid-phase extraction (d-SPE) and high eficiência liquid chromatography with ultraviolet/visible detection (HPLC-UV/Vis) In the extraction step, it was used 25 mg of the polymeric sorbent Oasis® HLB and the elution step was performed with metanol:ammonium hydroxide (95:5, v/v). Recovery results, evaluated at 2.0; 4.0 and 6.0 mg L-1, were between 76 and 108% with relative standard deviation (RSD) <18%. Method limits of detection and quantification were 0.6 and 2 mg L-1, respectively. The method was applied on 8 commercial samples of sports drinks and the results indicated that all studied samples presented dye levels in conformity with the Brazilian legislation. The proposed d-SPE method for the determinatios of 6 synthetic dyes in sports drinks using HPLC-UV/Vis shown to be rapid, effective and cheap, and can be applied in routine analysis. / As bebidas isotônicas são especialmente formuladas para pessoas que praticam atividade física, sendo compostas principalmente por carboidratos, eletrólitos e vitaminas. Os corantes sintéticos são aditivos alimentares comumente utilizados em bebidas isotônicas para aprimorar suas características de consumo e aparência. Além do menor custo de produção em relação aos corantes de origem natural, os corantes sintéticos apresentam diversas vantagens, tais como alta estabilidade à luz, oxigênio e pH, uniformidade de cor e baixa contaminação microbiológica. No entanto, alguns destes compostos podem causar efeitos adversos à saúde (alergia, asma, hiperatividade, tumores de tireoide, etc.), especialmente quando consumidos em excesso. Assim, seu uso é estritamente controlado em todo o mundo. O presente estudo teve como objetivo desenvolver um método simples, rápido e eficiente para a determinação dos corantes sintéticos tartrazina, amarelo crepúsculo, amaranto, ponceau 4R, azul de indigotina e azul brilhante em bebidas isotônicas empregando extração em fase sólida dispersiva (d-SPE) e cromatografia líquida de alta eficiência com detecção por ultravioleta/visível (HPLC-UV/Vis). Na etapa de extração utilizou-se 25 mg do sorvente polimérico Oasis® HLB e a etapa de eluição foi realizada com seringa, empregando uma solução de metanol:hidróxido de amônio (95:5, v/v). Os resultados dos ensaios de recuperação, avaliados nos níveis 2,0, 4,0 e 6,0 mg L-1, foram entre 76 e 108% com desvio padrão relativo (RSD) <18%. Os limites de detecção e de quantificação do método foram 0,6 e 2 mg L-1, respectivamente. O método foi aplicado em 8 amostras de bebidas isotônicas comerciais e os resultados indicaram que todas as amostras estudadas apresentaram corantes dentro dos limites máximos permitidos pela legislação brasileira. O método d-SPE proposto para a determinação de 6 corantes sintéticos em bebidas isotônicas empregando HPLC-UV/Vis demonstrou ser rápido, barato e eficaz, podendo ser aplicado em análises de rotina.

Study on bioaccumulation and integrated biorefinery of vegetable oil and essential oil of Coriander (Coriandrum sativum L.) / Etude de la bioaccumulation et du bioraffinage des huiles végétale et essentielle de coriandre (Cordiandrum Sativum L.)

Les apiaceae peuvent être définies en tant qu' Aroma-Tincto-Oleo-Crop (ATOC), plantes qui contiennent à la fois une huile végétale et une huile essentielle. Appliquer le concept d'agroraffinage aux ATOC revient à proposer un procédé séquentiel alliant une co-extraction huile végétale et huile essentielle à une valorisation des résidus en tant que source de molécules biosourcées et de susbtrat pour la formulation d'agromatériaux . Les objectifs de cette thèse seront donc d'étudier la faisabilité biologique et technologique d'application du concept d'ATOC-raffinage à la coriandre (Coriandrum sativum L.). Le chapitre I présente l'état de l'art bibliographique sur l'extraction et l'analyse des huiles végétales et huiles essentielles de coriandre tandis que dans le chapitre II sont décrits les matériels et méthodes mis en oeuvre au cours de la thèse tant au niveau échantillonnage, extraction, analyse que traitement des données. Le chapitre III est centré sur l'étude des différents paramètres biologiques pouvant influencer la bioaccumulation des huiles végétales et huiles essentielles dans la coriandre (différentes variétés, différents organes de la plante, différents stades de développement biologique) et leur impact sur l'activité anti-oxydante des extraits obtenus à partir des résidus d'extraction. Dans le chapitre IV, la technologie d'extrusion (mono-vis et bi-vis) a été appliquée aux graines de coriandre dans le but d’évaluer la faisabilité du pressage mécanique du fruit de la coriandre pour l’extraction d'une huile végétale aromatisée. L’influence des conditions expérimentales sur le rendement d'extraction en huile végétale (diamètre de buse et distance buse-vis (extrudeur mono-vis) ou configuration de vis, coefficient de remplissage et température de pressage (extrudeur bi-vis)) a été étudiée tandis que la faisabilité de la valorisation du résidu solide d'extraction en agromatériaux (thermo-pressage) a été montrée. / Apiaceae could be defined as Aroma Tincto Oleo Crops (ATOC), e.g. plants containing both vegetable oil and essential oil. Applying agroreffinery concept to ATOC led to propose a sequential fractionation process coupling co-extraction of vegetal oil and essential oil to a valorization of by-product residues as biosourced active molecules and substrates for designing agromaterials. The aim of this thesis is to determine the biological and technological feasability of application of the ATOC-refinery concept to coriander (Coriandrum sativum L.) Chapter I reports a bibliographic state of the art study on extraction and characterisation of coriander vegetal oil and essential oil while chapter II describes materials and methods setting up during the thesis for sampling, extraction, analysis and data processing. Chapter III focus on the study of major various biological parameters influencing bioaccumulation of vegetal oil and essential oil in coriander (different plant varieties, different plant organs, different biological stages) and their impact on anti-oxidant activity of extracts obtained from extraction residues. In chapter IV, coriander fruits are processed by extrusion technology (mono screw and twin-screw extruder) in order to evaluate the feasability of mechanical pressing for extracting a flavored vegetal oil. Influence of operating parameters on vegetal oil extraction yields (nozzle diameter and nozzle/screw distance (single-screw extruder) or screw configuration, device’s filling coefficient and pressing temperature (twin-screw extruder)) is studied while the feasability of valorization of extraction cake as agromaterial (thermopressing) was stated.

L'encapsulation de protéines pose de nombreuses difficultés liées à leurs propriétés physicochimiques, leur sensibilité aux conditions environnementales et opératoires. La structure 3D spécifique de chaque protéine étant directement liée à son activité biologique, un contrôle de l'intégrité protéique est indispensable pour assurer l'efficacité et la sécurité des systèmes formulés. Dans cet optique, l'encapsulation par émulsion multiple a été appliquée à une protéine modèle : la myoglobine (Mb). Le but de cette thèse a été l'étude du procédé d'encapsulation à travers 3 objectifs. Le premier fut d'étudier l'influence de paramètres de formulation sur les propriétés des particules obtenues et sur la conformation de la Mb. Les mesures en spectrométrie UV/Vis. et le calcul des principaux rapports d'absorbance ont constitué une méthode de contrôle fiable et rapide. Le second fut de valider la pertinence et de montrer les limites de cette méthode en la comparant à une seconde méthode utilisant des mesures conductimétriques. Enfin, l'étape de solidification des particules par élimination du solvant ainsi que le changement de solvant ont été étudiés.. Des microsphères creuses de 15μm avec un rendement d'encapsulation de 36% ont pu être obtenues tout en maintenant la conformation native de la Mb. L'emploi de polymère de masse moléculaire élevée et un taux d'élimination de solvant trop rapide sont 2 paramètres altérants significativement la protéine. Ces travaux ouvrent la voie au développement de transporteurs d'O2 utilisables par exemple dans le cas de pathologies musculaires / Protein encapsulation results in several problems related to their physicochemical properties, their sensitivity to environmental conditions and operating procedures. The specific 3D structure being directly linked to their biological activity, monitoring of protein integrity is crucial to ensure the efficacy and security of formulations. In this context, the multiple emulsion method was applied to a model protein: myoglobin (Mb). The aim of this thesis was to study the encapsulation process through 3 objectives. The first was to study the influence of formulation parameters on the particle properties and the conformation of Mb. Measurements by UV/Vis. spectrometry and calculation of key absorbance ratios established a reliable and rapid method for protein monitoring. The second was to validate the pertinence and show the limits of this method by comparing it to a second one using conductimetry measurements. Finally, the particle solidification by solvent removal and the solvent exchange were studied. The process maintains the Mb native conformation in Hollow microspheres of 15μm in diameter and an encapsulation efficiency of 36% were obtained, while keeping intact the Mb native conformation. The use of high molecular weight polymer and a fast solvent removal rate are 2 parameters inducing significant protein alteration. This work paves the way for the development of O2 carriers for muscular diseases for example

Um aspecto importante relacionado à operação de sistemas de distribuição é o referente à segurança nas ações de restabelecimento pós-perturbações, em manobras emergenciais e programadas, onde necessite de transferência de cargas entre alimentadores aéreos primários de uma mesma subestação ou para uma subestação adjacente, de forma segura e confiável. Devem-se observar os requisitos de carregamento, tensão a nível sistêmico, proteção, entre outros. Atualmente as decisões e tomadas de ações referentes às manobras necessárias para os remanejamentos de cargas são de competência dos operadores que atuam nas salas de controle de operações e, em muitos casos, respaldadas somente na experiência de cada operador, acumulada ao longo do tempo. Este trabalho visa o estudo dos fenômenos físicos envolvidos em situações que coloquem alimentadores aéreos primários de distribuição em paralelo ou mesmo quando se forme uma configuração em anel fechado. Estas manobras podem ser executadas através de alimentadores e transformadores distintos, de mesma potência ou de potências diferentes, estabelecendo diretrizes e respaldando os operadores das salas de controle em situações de emergência, urgência ou programadas no sistema aéreo de distribuição. O objetivo principal é oferecer informações técnicas, possibilitando as tomadas de decisões mais adequadas na coordenação de manobras no sistema aéreo de distribuição, em tempo real. Cada uma delas tem condições que se pode considerar como a mais adequada para a execução dos procedimentos estabelecidos, porém sem os principais subsídios técnicos. Desta forma, algumas variáveis como tensão mínima, limites de carregamento de equipamentos, ângulo de fase dos transformadores, restrições operativas do sistema e de consumidores, devem ser aplicadas em tempo real para a tomada de decisão por parte dos operadores do sistema elétrico. / An important aspect related to the distribution systems operation concerns the safety required in the post-disturbances restorement actions or in the emergency and scheduled maneuvers, which involve the need for load transfer between primary overhead feeders of a same sub-station or for an adjacent sub-station, in a safe and reliable way, complying with the loading requirements, voltage at systemic level and protection, among others. Currently the decisions and taking actions related to the necessary maneuvers to carry out the load transfers are responsibility of the operators who work in the operations control rooms, and in many cases, only endorsed by each operator´s own experience, gathered along the time. This paper aims at the study of the involved physical phenomena in situations that place distribution primary overhead feeders in parallel or even when a configuration in closed ring is formed, via distinct transformers and feeders of same or different powers, establishing guidelines and endorsing the control rooms operators in emergency, urgency or scheduled situations in the distribution overhead system. The main objective is to provide technical information, enabling more adequate taking decisions in the distribution overhead system maneuvers coordination, in real time. Each of them has conditions that can be considered the most adequate to carry out the established procedures, however without the main technical information. Thus, some variables such as minimum voltage, equipment loading limits, transformers´ phase angle, the operational restrictions of the system and customers must be applied in real time for the taking decision by the electrical system operators.

L'antagonisme entre les besoins croissants d'énergie au niveau mondial et l'obligation de ralentir le réchauffement climatique fait partie des défis auquel l'humanité fait face. Dans le but d'assurer un confort thermique suffisant, les domaines de l'habitat, de l'automobile ou encore des bâtiments agricoles consomment de grandes quantités d'énergie. Pour réduire ces besoins, une des solutions proposées vise à améliorer l'isolation thermique de ces bâtiments grâce à l'utilisation de matériaux innovants. Un des objectifs majeurs est d'améliorer l'isolation des fenêtres qui représentent une partie importante des pertes énergétiques. Les recherches décrites dans cette thèse ont permis de développer des matériaux transparents aux rayonnements solaires visibles tout en étant des boucliers efficaces contre les rayonnements ultra-violets et proche-infrarouges. De tels matériaux sont le fruit de la combinaison entre des motifs à clusters de métaux de transition présentant des propriétés d'absorption recherchées et une matrice hôte permettant la mise en forme de ces matériaux (processabilité). La synthèse, ainsi que la méthodologie de modulation des propriétés d'absorption et d'intégration des clusters dans différentes matrices de type sol-gel ou polymères sont présentées. Un intérêt tout particulier est porté sur les relations qui s'établissent entre la structure des motifs à clusters et leurs propriétés d'absorption grâce aux comparaisons faites entre des études expérimentales et des études de chimie quantique utilisant la théorie de la fonctionnelle de la densité (DFT). In fine, l'augmentation du niveau de compréhension des relations structures-propriétés d'absorption des motifs à clusters étudiés permettra d'envisager l'étude de nouvelles compositions pour des matériaux encore plus efficaces à l'avenir pour le contrôle solaire. / The antagonism between growing global energy needs and the obligation to slow the global warming is one of the challenges humanity faces. In order to ensure sufficient thermal comfort, the housing, automotive or agricultural buildings sectors are major energy consumers. To reduce these needs, one of the proposed solutions aims to improve the thermal insulation of these buildings through the use of innovative materials. One of the major objectives is to improve the insulation of the windows which represent a significant part of the energy losses. The research described in this thesis has made it possible to develop transparent materials for visible solar radiation while being effective shields against ultraviolet and near-infrared radiation. Such materials are the result of the combination between transition metal cluster patterns having desired absorption properties and a host matrix for shaping these materials (processability). The syntheses as well as the methodology for modulating the absorption and integration properties of clusters in different sol-gel or polymer matrices are presented. Of particular interest is the relationship between the structure of cluster pattern and their absorption properties through comparisons between experimental studies and quantum chemistry studies using density functional theory (DFT). In fine, the increase in the level of understanding of the structure-absorption properties of the clusters studied will make it possible to consider the study of new compositions for even more efficient materials in the future for solar control.

Inorganic ions are one of the most important group chemicals. Their importance is crucial for all living organisms on the planet and monitoring of their content is essential for this purpose in clinical or environmental analysis but also in the food industry, agriculture or many other industries. Many analytical methods for ion detection have been designed and used, depending on the area of their use. Recently, numerous methods have been adapted to areas where not primarily intended. One such methods may be, for example, laser ablation with inductively coupled plasma mass spectrometry, which has recently begun to affect the analysis of biological samples. As a complementary method, able to detect also non-metallic ions is capillary electrophoresis. In this master thesis, the possibility of applying capillary electrophoresis with indirect photometric detection for the separation of inorganic ions in cell lysates of HeLa cells by capillary electrophoresis was verified. Main attention was paid to chloride ions. The ability to separate inorganic compounds in cellular material using salicylic acid as an absorbing anion in the base electrolyte was tested.

Charakterizace rostlinné složky vybraných potravin pomocí technik molekulární biologie a instrumentálních metod / Characterization of plant-based component of selected foodstuffs using techniques of molecular biology and instrumental methods

The aim of this work was to compare authenticity analysis in selected food products with fruit component by using, instrumental and molekular methods. Particularly, the presence of blueberries in fruit–based foodstuffs was verified. The theoretical part is focused on the characterization, chemical composition and botanical classification of blueberries (European blueberry and Canadian blueberry). It also contains an overview of instrumental and molecular diagnostic methods that can be used for the analysis of these fruits. The experimental part focuses on the selection of a suitable method of DNA isolation, and primers for the detection of blueberries in commercial products. DNA analysis was performed by qPCR and HRM analysis. In the experimental part, DNA was isolated in sufficient quality for PCR and the presence of blueberries in foodstuffs was verified by qPCR. Using HRM analysis, we were able to differentiate between bilberry (Vaccinium myrtillus) and blueberry (Vaccinium corymbosum) in control samples and in some commercial products. Certain phenolic acids and some flavonoids specific for blueberries were detected by HPLC. The total content of polyphenols and flavonoids was determined by UV / VIS spectrophotometry.

Analýza rozkladu roztoků huminových kyselin diafragmovým výbojem / Analysis of humic acids solutions after their decomposition by diaphragm discharge

Preliminary results of research focused on the applications of DC diaphragm discharge in water solutions containing humic substances are presented in this diploma thesis. Electrical discharges in water produce various reactive species such as radicals (•OH, •O, •H), ions and molecules (H2O2, O3). These species have high oxidation potential and thus they easily react with order species and molecules. Such reactions could lead, for example, to destruction of organic pollutants dissolved in water. This work studies this effect on humic matters that can be contained in water coming from floods. Diaphragm discharge investigated by this work was created in the reactor using konstant DC high voltage up to 2 kV that gave the total input power from 100 to 200 W. Breakdown and discharge ignition started in the pin-hole in the dielectric barrier separating two electrode spaces (anode and cathode space). Presented work investigates decomposition of humic substances by the electric discharge in the dependence of solution properties and discharge conditions. Parameters such as initial solution conductivity, electrolyte kind or input power have been investigated. Moreover, substantial effect of pH on humic acid decomposition has been observed. Refraktometry and absorption spectroscopy in UV-VIS region together with fluorescence spectroscopy has been used for the detection of changes in humic solutions.

Solubilizační schopnosti polysacharidů / Solubilizattion properties of polysaccharides

In this diploma thesis were studied solubilization properties of polysaccharides by using hydrophobic solutes (Sudan Orange G, Sudan Red G, (±)-alpha-Tocopherol, Pyrene, Perylene, Nile red), which were represented by alkyl derivates of hyaluronan. At first, a behaviour of individual hydrophobic solutes was investigated in variously polar solvents (Methanol, 1 Propanol, Chloroforme, Cyklohexane, n Heptane) and in the environment of varying ionic strength (water, 0.1 M and 0.4 M NaCl). Afterwards, solubilization properties of Sodium Dodecyl Sulfate model solubilizated the hydrophobic solutes into a core of micelles was examinate. We were interested in the solubilization capacity as the mol of solubilized molecules per mol micelles of surfactant corresponding with a state of micelles saturation. In the case of the solubilization of (±)-alpha-Tocopherol into the core of micelles, it was not possible to determine the solubilization capacity. So we changed the determination of universally solubilization power. The solubilization power is defined as mol of molecules solubilized per mol surfactant relative to the quantity solubilizate at the micelles saturation. Model system of Sodium Dodecyl Sulfate as a simple surfactant carrying a negative charge as the alkyl derivates of hyaluronan was selected bacause of its characteristics.The surfactant forms unimolar micelles and its critical micelle concentrations and aggregation numbers are tabelated for the investigated microenvironment. The main aim of the study was investigating of hydrophobic domains of alkyl derivates of hyaluronan as free places for incorporation hydropbobic solutes in the microenvironment of varying ionic strength. The critical aggregation concentrations were determined by the Pyrene 1:3 ratio method. For the research of micropolarity of alkyl derivates hyaluronan’s domains were selected two concentrations of derivates for the next research of solubilization experiments - the first concentration near the critical aggregation concentration and the second concentration above it. The effect of concentration of Pyrene on a core polarity of derivates was investigated. We discovered the influence of the concentration and the other we found a stationary area of the concentration. In the end we investigated the influence of preparation of solutions of derivates of hyaluronan on the core polarity by the concentration of pyrene which corresponds to the stationary area. The study of solubilization properties of alkyl derivates of hyaluronan is not a simple case as we assumed. When we measured spectra of the absorbance, higher concentration of derivates of hyaluronan belittle absorbance of solubilizates. At the experiment of solubilization with Sudan Red G we found out that Sudan Red G is not able to solubilizate into the hydrophobic core of micelles of hyaluronan’s derivates because of lipophilic or steric effects. We had to change Perylene as a new solubilizate. From the measured emission spectra we found saturation micelles. We can express the solubilization power of hyaluronan’s derivates for the concentration of Perylene. The main aim of the diploma thesis was to determine optimal way of the preparation of hyaluronan’s derivates solutions with required degree of solubilization.

Stanovení karnitinu v potravních doplňcích / Determination of carnitine in food supplements

This work deals with the determination of L-carnitine in food supplements. L-carnitine is a substance naturally occurring in organism, essential for metabolism of fatty acids. In food supplements is used especially for reducing body weight as a "fat burner". L-carnitine is popular with athletes for improve athletic performance. While data are not available to support these positive effects of carnitine, the positive results of carnitine supplementation in the medicine were found, mainly on cardiovascular system. The literary part of this study describes the properties and the use of carnitine in the diet of human. Furthermore, a review of methods used for determination of L-carnitine in variety of samples is mentioned. In the experimental section the efficiency of SPE method for preparation matrix of real samples was investigated. A cation exchange solid phase extraction seems to be the most effective. The optimal conditions for isolation of L-carnitine by SPE extraction were not developed. L-carnitine was determined in six various food supplements by reverse phase chromatography with UV/VIS detection.
