Studium hydroxidů a oxidů kovů ve vodných roztocích / Study of Metal Oxides and Hydroxides in Aqueous Solutions

This dissertation work deals with analysis of nickel hydroxide phases, their oxidation compounds, their stability and degradation mechanisms of electrochemically more active alpha phase on standard beta phase. The active material was prepared by both methods, electrodeposition and chemical precipitation. Main analysis method was Electrochemical Quartz Crystal Microbalance in combination with common analytical methods (cyclic voltammetry, potenciometry) can resolute between alpha and beta phases and quantitatively describe differences in main reaction by monitoring mass changes in the active material. Poor stability of the ?-Ni(OH)2 were improved by adding cations with valence two three and four into the structure instead of Ni atoms. The most important role plays cobalt and its hydroxide. Totally new way is to use titanium in combination with other cations. In electrolyte the most significant addition is LiOH, which has beneficial influent on cycle ability, stability in strong alkaline medium and cycle life.

Techniky přípravy elektrod s nanostrukturovaným povrchem a jejich charakterizace / Preparation Techniques and Characterization of Electrodes with Nanostructured Surface

Nowadays, nanostructures fixed on solid substrates and colloidal nanoparticles permeate through all areas of human life, in area of sensors and detection as well. This dissertation thesis deals with the fabrication of nanostructures on the surface of planar electrodes via self-ordered nanoporous template of aluminum trioxide. The nanofabrication, as one of many possible techniques, is used to increase the active surface area of electrodes by creating unique surface types with specific properties. These electrodes are very perspective in the applications, such as biomolecules electrochemical detection and measurement. The transformation of aluminum layer into non-conductive nanoporous template in the process of anodic oxidation is a fundamental technique employed to obtain the array of nanostructures in this thesis. The fabrication of high quality nanoporous membranes with narrow pore size distribution on various types of metallic multilayers is one of the key experimental parts in this work. Several problems associated with the production of the thin-film systems, including the dissolving the barrier oxide layer, are discussed and solved. Another part of this work deals with the use of nanoporous membrane as a template for the production of metallic nanostructures via electrochemical metal ions deposition directly into the pores. The obtained nanostructures as nanowires, nanorods or nanodots are characterized by the scanning electron microscopy and energy-dispersive or wavelength X-ray spectroscopy. The electrode surface, modified by gold nanostructures suitable for the detection of biomolecules, has been chosen for the electrochemical measurements, due to the gold biocompatibility. The nanostructured electrodes were characterized by electrochemical impedance spectroscopy and cyclic voltammetry. The effect of nanostructured surface geometrical parameters, including the size of the electrochemically active area, on the results of electrochemical measurements has been observed and compared to flat gold electrodes. Two model biomolecules, namely guanine and glutathione, have been chosen for the study of potential application of these nanostructures in biosensors.

Hlavním cílem této práce je charakterizace vzorků titanátů, jako materiálů pro sodíko-iontové (Na-ion) akumulátory. Syntéza některých vzorků je součástí této práce. Charakterizace je zaměřena na elektrochemické vlastnosti, složení a morfologii použitých materiálů. První část se zabývá lithium-iontovými (Li-ion) akumulátory. Byly vybrány, protože jsou ve vědeckých článcích dobře popsány a základní funkční princip je aplikovatelný také na Na-ion akumulátory. Materiály používáné pro katody, anody a elektrolyty následují po krátké části shrnující parametry a konstrukci Li-ion akumulátorů. Následující kapitola je zaměřena na sodíko-iontové akumulátory. Srovnání sodíku a lithia je následováno materiály používanými pro elektrody a elektrolyty (s důrazem na anodové materiály). Třetí část popisuje analytické metody použité pro charakterizaci elektrod a materiálů. Jedná se o elektrochemickou charakterizaci (cyklická voltametrie a galvanostatické cyklování s potenciálovým omezením), morfologii (rastrovací elektronová mikroskopie) a složení (X-ray difrakční spektroskopie). Poslední dvě kapitoly obsahují syntézu a charakterizaci sodného titanátu a charakterizaci dvou vzorků TiO2. Výsledky této práce jsou shrnuty v závěru.

This work focuses on optimization and application of voltammetric methods for determination of 5-nitroquinoline in model samples of drinking and river water using carbon film electrode (CFE). The advantages of carbon film electrode are primarily its wide potential window in both cathodic and anodic regions and also low environmental stress compared to mercury electrodes. In this contribution, CV and AdSV were used to observe electrochemical processes. For determination of 5-nitroquinoline DPV and FIA were used. Solid phase extraction was investigated as a method for preliminary separation and preconcentration for DPV. Determination of 5-nitroquinoline at CFE is based on cathodic reduction of nitrogroup. This work demonstrates the application of carbon film electrode for determination of 5-nitroquinoline in submicromolar concentrations in model samples of water.

Chlortoluron is a herbicide from the group of substituted ureas and it is used for weed control in production of cereals and poppy. Substituted ureas are widely used but these substances are known for their high mobility in soil, persistence and contamination of underground water. Therefore, the development of analytical methods suitable for residual concentrations determination is necessary. This thesis deals with the determination of chlortoluron using carbon paste electrode (CPE) by the methods of differential pulse voltammetry and high-performance liquid chromatography with electrochemical (HPLC-ED) and spectrophotometric (HPLC-UV) detection. The aim of the work was to find optimal conditions suitable for chlortoluron determination. Further, the possibility of using miniaturized carbon paste electrode (mCPE) and usability of tested methods for determination of chlortoluron in model samples of river water and soil were studied. Concentration dependences in deionized water were measured by the method of DPV in the Britton-Robinson (BR) buffer (pH = 3) and methanol (10 %, v/v). The limit of detection was 3,7∙10-7 mol∙dm-3 in the case of CPE and 8,7∙10-8 mol∙dm-3 in the case of mCPE. The mediator is deposited on electrode surface formed by basal plane of highly ordered pyrolytic graphite. Využití tuhých elektrod na bázi uhlíku k elektrochemické oxidaci acikloviru / Utilization of carbon-based solid electrodes to electrochemical oxidation of acyclovir

Electrochemical oxidation of acyclovir at glassy-carbone electrode and boron-doped diamond electrode gives one pH-dependent oxidation signal; the corresponding electrode reaction is controlled by diffusion. It is followed by a second indistinctive signal in the pH range 2,0 - 9,0. Further, optimization of conditions was carried out for determination of acyclovir using DC and DP voltammetry. The calibration dependence on GC electrode in B−R buffer (pH = 6,0) is linear only in the concentration range from 2 to 10 μmol∙l−1 for DCV with limit of detection 0,38 μmol∙l−1; for DPV it is not linear. BDD electrode has to be activated to prevent its passivation. Anodic activation at +2,4 V for 15 s with simultaneous stirring of solution results in relative standard deviation of 0,9% for acyclovir concentration of 1∙10−4 mol∙l−1. Using DCV and DPV methods linear concentration dependences were obtained in 0,1 mol∙l-1 nitric acid and in pH 6,0 B−R buffer. The lowest limit of detection and limit of quantification (LOD = 0,47 μmol∙l−1 a LOQ = 1,55 μmol∙l−1) and linear range from 0,6 to 100 μmol∙l−1 was reached in pH 6,0 B−R buffer. This work is devoted to the study of boron doped diamond as electrode material, its properties and use in electroanalytical methods - in voltammetric and subsequently amperometric methods in combination with high performance liquid chromatography. The series of boron doped diamond films was tested with respect to the effect of boron concentration on their morphology, quality, electrochemical and spectral properties using scanning electron microscopy, atomic force microscopy, Raman spectroscopy, and cyclic voltammetry and differential pulse voltammetry. Further, the effect of boron concentration on the determination of selected substances was investigated, both for their oxidation (2-aminobiphenyl, benzophenone-3) and for their reduction (5-nitroquinoline). Furthermore, a voltammetric and amperometric method was developed for the determination of a mixture of aminobiphenyls and aminonaphthalenes using a boron doped diamond electrode. In this doctoral thesis, a novel method for the determination of primary bile acids cholic acid and chenodeoxycholic acid is presented. Bile acids play various vital roles in the mammalian body. Moreover, their determination is extremely helpful in liver and biliary disease diagnosis and management. These saturated organic compounds lack strong chromophores and fluorophores in their structure, and thus are usually hard to detect in spectroscopy. For this reason, either instrumentally advanced but expensive methods, such as mass spectrometry, or less reliable enzymatic methods are commonly employed in bile acids quantitation. Hence, the demand for simple and reliable methods for their determination is strong. Bile acids are also known to be virtually inert for direct electrochemical oxidation. Herein, a simple method for their chemical activation for electrochemical oxidation on bare electrode materials was developed, optimized and applied to cholic acid and chenodeoxycholic acid determination. The activation is based on a dehydration reaction of a primary bile acid with 0.1 mol L-1 HClO4 in acetonitrile (water content 0.55%) that introduces double bond(s) into the originally fully saturated steroid core. Vývoj voltametrických metod pro detekci fytosterolů na elektrodách na bázi uhlíku / Development of voltammetric methods for detection of phytosterols on carbon-based electrodes

The aim of this diploma thesis was to develop an electrochemical method for three phytosterols, namely stigmasterol β-sitosterol and ergosterol using voltammetry techniques on glassy carbon electrode and boron doped diamond electrode. The work builds on previous results, where it was shown that the electrochemical oxidation of phytosterols requires perchloric acid in acetonitrile. When this acid is used, a dehydration reaction occurs. Calibration dependences were measured here for all phytosterols in an environment with a water content of 0.43%. The stability of the signal with increasing distilled water content was further investigated. Then the calibration dependences in the increased water content were measured again. If we want to use HPLC-type separation techniques, we must first find out how the phytosterols will behave in this environment, and therefore the signal stability on the buffer content was investigated. The buffers used were phosphate and acetate buffer. Subsequently, the concentration dependence was measured in an environment with a higher content of phosphate buffer. Furthermore, the possibility of detecting two phytosterols at once was investigated.
