The aim of the presented Diploma Thesis was to develop a new method for the indirect determination of formaldehyde in wood-based products using gas-diffusion microextraction coupled with electrochemical detection on unmodified screen-printed electrodes (MLEM-SPCE). Formaldehyde released from the sample is derivatized using an acetylacetone reagent present in an acceptor solution. The product of derivatization of formaldehyde with acetylacetone is 3,5-diacetyl-1,4-dihydrolutidine (DDL) which forms a selective oxidation voltammetric peak at a potential of 0.4 V. Detection and quantification limits of 0.57 mg kg−1 and 1.89 mg kg−1 , respectively, were obtained, together with intra- and inter-day precision below 10% (as relative standard deviation, RSD). The developed methodology was applied to determine formaldehyde content in seven samples. Similar results were obtained from the European standard method EN 717-3 with a significant reduction of total analysis time. The developed method MLEM-SPCE, which combines the use of a new sample preparation procedure for volatile compounds with the firstly introduced determination of formaldehyde (as the derivative product DDL) on unmodified SPCEs, proves to be a promising alternative for the determination of formaldehyde in wood-based products and other samples.

Presented Diploma Thesis is focused on the development, characterization, and utilization of simple and inexpensive electrochemical DNA biosensor for the detection of DNA damage caused by oxidative stress. The initial part of the work is devoted to preparation and characterization of a large-surface carbon film electrode (ls-CFE) modified with carbon nanotubes (CNT/ls-CFE). Carbon nanotubes improve electrochemical properties of the transducer and increase the amount of adsorbed DNA on the electrode surface. Testing of the electrode surface modified with multiwalled carbon nanotubes (MWCNT) was performed using cyclic voltammetry (CV) and electrochemical impedance spectroscopy (EIS) using a redox system [Fe(CN)6]4-/3- and using square wave voltammetry without any redox indicator. Carbon nanotubes have proved to be unsuitable material for our type of biosensor, but it can be used inanalytical chemistry for the determination of electroactive substances. The second part of this Thesis deals with the application of the prepared DNA biosensor for the detection of DNA damage by oxidative stress. This Diploma Thesis is focused on study of electrochemical behavior of 2,4,6-trinitrotoluene (TNT) and 2,4,6-trinitrophenol (picric acid) on finding the optimum conditions for their determination using direct current voltammetry (DCV) and differential pulse voltammetry (DPV) at a mercury meniscus modified silver solid amalgam electrode (m-AgSAE) in the solution of Britton-Robinson (BR) and on finding of the limit of quantification (LQ) for these substances. Practical applicability of the newly developed methods was verified on direct determination of TNT and picric acid in model samples of drinking and river water. Moreover, the electrochemical behaviors of TNT and picric acid was studied using cyclic voltammetry (CV). Optimum medium for the determination of TNT at m-AgSAE was: methanol-BR buffer pH 4.0 (1:9). Upon the DCV it is proper to apply regeneration potentials Ereg,1= 0 mV and Ereg,2= -1100 mV and upon the DPV was apply regeneration potentials Ereg,1= 0 mV and Ereg,2= -600 mV were applied. The concentration dependence of the peak current was found to be linear for both techniques over the concentration range of 1·10-6 -1·10-4 mol/l with LQ of 0.54 µmol/l (for DCV) and 0.46 µmol/l (for DPV). The practical part is the production of biomodified nanostructured electrodes by anodi- zation W/Al layers with galvanic deposition of gold or deposition of mercury, a modifi- cation of the electrodes by 11-mercaptoundecanoic acid and by bovine serum albumin (BSA). The thesis includes SEM images of nanostructured electrodes contact angle mea- surements of these electrodes and form an electrical circuit with subsequent simulation waveforms.

5 Abstract This Diploma Thesis is targeted on the determination of 5-nitrobenzimidazole (5-NBIA) using following techniques: DC voltammetry (DCV) and differential pulse voltammetry (DPV). As working electrodes, a silver amalgam paste electrode (AgA-PE), which was constructed for this determination in the form of a new prototype, a bismuth film electrode (BiFE), at which the optimum conditions for the deposition of the bismuth film at a suitable substrate (glassy carbon electrode and gold electrode were tested for this purpose) were initially optimized, and a glassy carbon electrode (GCE) were used. The optimum conditions for the voltammetric determination of 5-NBIA at the AgA-PE (in a medium of Britton-Robinson buffer (BR-buffer) of pH 7.0 for both DCV and DPV), at the BiFE with gold substrate (BR-buffer of pH 9.0 for both DCV and DPV; the film was deposited "ex situ" in a stirred plating solution (1000 mg L-1 Bi(III) solution in 0.1 mol L-1 acetate buffer of pH 4.5) for 300 s), and at the GCE (BR-buffer of pH 5,0 for both DCV and DPV). This Diploma Thesis is focused on investigation of the voltammetric behavior of the explosive penthrite (pentaerythritol tetranitrate) (PETN) and on searching for optimum conditions for its determination using differential pulse voltammetry (DPV) at a hanging mercury drop electrode (HMDE) (this part of the Diploma Thesis follows the topic investigated in the author's Bachelor Thesis; in this Diploma Thesis, the results of the Bachelor thesis are applied to the determination of PETN in environmental matrices) and at a mercury meniscus modified silver solid amalgam electrode (m-AgSAE) and on investigation of voltammetric behavior in real samples (deionized, drinking, and river water). For investigating the behavior of PETN in aqueous-methanolic media on the m-AgSAE, mixtures of Britton-Robinson buffers (BR-buffers) and methanol in various volume ratios were used. The aminoacid sequence of papain (EC 3.4.22.2) consists of 212 aminoacids. It has only one free sulfhydryl group, which is located in the active site of the protein. Some organometallic complexes could be bonded only to this free -SH group due to their structure. The artificial metalloproteins synthesised by this way may have different electrochemical properties. In this work, we have studied the electrochemical properties of papain and its derivatives. We compared the ability of papain and its three artificial derivatives to catalyse the hydrogen evolution by the chronopotenciometry. The work was completed by the study of the electrochemical properties of the organometallic complexes of ruthenium, which were used for the artificial metalloprotein preparation. The electrochemical properties of the compounds were never studied before. The process of the hydrogen evolution catalysed by the proteins is held in the adsorbed state of the catalyst. Due to this fact we have also studied the adsorption properties of papain on the substrates with different level of hydrofobicity. (In Czech)

Differential pulse voltammetry (DPV) was used for developing a method for the determination of phenothiazine derivatives, namely chlorpromazine (CHP) and levomepromazine (LMP), using a glassy carbon electrode (GCE) and a graphite electrode, both with a 2 mm disc diameter. Comparison of quantification limits (LQ) for both substances on both electrodes was performed. After optimizing and processing the results, the optimal conditions for measuring calibrations were as follows: Britton- Robinson (BR) buffer at pH 4.0. Calibration dependences were measured inthe concentration range from 1·10-7 to 1·10-4 mol·dm-3 . There was no need for electrochemical electrode regeneration or matrix pretreatment. The results showed higher measurement sensitivity for CHP, and the graphite electrode also showed higher sensitivity. Although there were measured lower concentrations with the graphite electrode in comparison to the GCE, because of the low repeatability of the measurements in the lower concentration range 1-10·10-7 mol·dm-3 , the LQ is comparable to the results reached on the GCE. Besides to the BR buffer, measurements in other matrices (drinking water and river water) were also made. This master‘s thesis concerns gel polymer electrolytes formed on a methyl methacrylate base with selected types of nanoparticles. In the thesis are also analyzed the methods for measuring electrochemical properties. The practical portion deals with sample preparations of gel polymer electrolytes with different contents of alkaline salt in a solvent, creating gels with different nanoparticle content and comparing gel polymer electrolytes polymerized with heat and UV radiation. The thesis deals with the evaluation of these samples from the viewpoint of electrical conductivity and potential windows as well as thermal analysis of selected samples.
