The diploma thesis deals with the suggestopedical English language teaching in the context of leisure education. Characterizes the interest adult learning as a part of lifelong education, defines its objectives, functions, motivation and language barriers within the adult education. More specifically, it focuses on the alternative method suggestopedia in detail; it examines its origin, development, features and principles. It describes the different stages of learning that forms the characteristic profile of this unconventional method. The empirical part is focused on the suggestopedic course itself. The use of case study helped to understand the features of suggestopedic education, processes and conditions from the point of experiences and attitudes of the course participants, i.e. the advantages and disadvantages of this method.

Vybrané charakteristiky hybného aparátu u dětí ve věku 13 a 15 let / Selected characteristics of the mobility for children aged 13 and 15 years

The aim of the diploma thesis is to evaluate the mobility of the spine, the condition of flaton and the basic anthropological characteristics of boys and girls aged 13 and 15 years. Testing will be performed with non-invasive, standard tests and tests used in anthropological and medical practice. The data will be compared with the results of previous research. From the measured values, basic somatic dimensions, such as body weight, body height, circumference of right arm and head, were selected. The body mass index was calculated from body weight and height values. The diploma thesis deals with the assessment of the mobility of spine and flats. The Stibor test, Otto's test, Schober's test, the Cepoj test, the lateroflex test, and the modified Thomayer test were selected from the tests that evaluated the spine. In spine functional tests, the most interesting results of the Čepoj symptom are that the average values of cervical spine mobility deteriorate with the increasing age of boys. In the lateroflex tests, 13-year-old girls and 15-year-old boys beat the best average values. Additionally, these boys had the worst results in Stibor and Schrober's flag when deflected, but paradoxically achieved the best average values. The findings were compared with the results of previous research.

The main aim of this thesis was to study the genetic variation of several performance traits under pond management conditions and to estimate future perspective of sustainable selective breeding program in common carp. In the present study, Amur mirror carp (AM) and Hungarian synthetic mirror carp (HSM) were used. Most of the studied performance traits showed sufficient genetic variation (h2 = 0.12 1.0), suggesting a good potential for genetic improvement of the traits through selective breeding. It was shown that selection of common carp under pond management conditions should be optimally applied after the second overwintering. Winter survival is often a bottleneck for common carp production. As a result, the genetic background of winter survival and traits that might be correlated to the survival was studied. Main focus was given to the muscle fat and body weight and their dynamics through winter period and successive third growing period. It was found that selection for i) lower fat content before and after winter, ii) lower decrease in muscle fat content and/or body weight during winter, iii) higher condition factor may lead to better survival during both winter and third growing period and growth of fish during the third growing season. Unfortunately, direct selection for slaughter yields is impossible on live breeding candidates. Therefore, morphological predictors that can be measured in vivo are considered as an interesting alternative. As a result, external and internal measures were combined on 1553 fish by linear regression to predict log-log residuals (Logr) of slaughter yields. It was found that the accuracy of the prediction of slaughter yields may be solid. From the genetic point of view, model-predicted (h2 = 0.48 0.63) and even individual predictors (h2 = 0.34 0.72) of slaughter yields were highly heritable and favourably genetically correlated to the Logr yields. Hence, selection on trait predictors has an interesting perspective for genetic improvement of slaughter yields in common carp. Furthermore, genetic parameters of fillet fatty acid content and performance traits in market size common carp cultured under semi-intensive pond conditions were estimated. For flesh FA composition analysis 158 individuals were processed. Heritability estimates of total muscle fat, some FA groups and most performance traits were moderate to high (0.23 0.62). Interestingly, genetic correlations showed that genetic improvement of growth via selective breeding under Central European pond conditions without changing the production technology would very likely negatively affect carp flesh quality with respect to FA composition. Finally, restriction-site-associated DNA sequencing (Rad-seq) was used to identify and genotype single nucleotide polymorphism (SNP) markers for subsequent parentage assignment, construction of a medium density genetic map (12,311 SNPs), and testing of efficiency of marker-assisted (MAS) and genomic selection (GS) for growth and Koi herpes virus (KHV) resistance. No genome-wide significant QTL was identified for growth. However, genomic prediction of its breeding value may outperform the traditional pedigree-based prediction, resulting in an 18% improvement in prediction accuracy. On the other hand, genome-wide significant QTL affecting resistance to KHV was identified on linkage group (LG) 44 explaining approximately 7% of the additive genetic variation. The presented results add further evidence supporting the application of selective breeding in common carp cultured even under traditional pond management conditions. However, it is evident that the rearing technology will need suitable modifications. In addition, real economic impact of selective breeding on carp culture should be verified by calculation of realized heritabilities, real genetic gains and the total fish biomass yield of genetically improved stocks from a pond area unit.

The aim of this thesis is to verify the effectiveness of children's listening activities in the preschool period from five to six years of age, to introduce the utilization of methodology connected with listening to the music by Jaroslav Herden in terms of preschool education, and to demonstrate the importance of motivation and integrative activities for the development of attention together with the overall harmonious development of children. In the theoretical part, I analyse the input of music into the consciousness of children in terms of basic psychological features and functions which are important for perception. I introduce basic musical abilities, particularly musical creativity and imagination. The theoretical part is concluded with the clarification of basic means of expression and shape features, which are important not only for the music perception as such, but also the music perception in an ontogenetic context. Moreover, I also present the philosophy of educational thinking concerning the listening activities proposed by Professor Jaroslav Herden. In the practical part, I introduce a comprehensive authorial project involving the methodology of Jaroslav Herden's music tailored to children who are aged from five to six years. Changes in chromosome number and structure may cause serious diseases. Cytogenetic tests leadin to set of karyotype are done for detecting these abnormalities. Chromosomes are visualised with proper methods and karyotype is made up most often. Manual karyotyping is time-consuming and expensive task. Because of this, researchers have been developing automated karyotyping systems. Karyotyping systems classify chromosomes into classes based on their characteristic features. Overlapping and bent chromosomes are limitations for automatic classification since they ocur at almost every mitosis. Accuracy and reliability of karyotyping systems still depend on the human intervention. Overcoming of these problems and development of fully automated system is the aim of modern approaches.

Obsahem této práce je návrh metody porovnání webových stránek na základě vizuálních rysů. Na začátku jsou popsány možné přístupy k porovnávání dokumentů s ohledem na jejich použití. Dále jsou prezentovány přístupy porovnávání zaměřené na vizuální vzhled dokumentů. Zde jsou nejdříve popsány metody pro porovnávání z vyrendrovaného obrázku dokumentu a pak přístup pomocí zdrojového kódu. Tato práce je dále detailně zaměřena na získání vizuální rysů ze zdrojového kódu dokumentu. Je zde popsán návrh nové metody pro porovnávání dokumentů na základě vizuálních rysů, která využívá strukturální popis dokumentu. Součástí je taky popis implementace aplikace a dosažené výsledky. V závěru jsou informace o možném rozšíření navržené metody a dalším pokračování.

Drought is one of the most important stress factors affecting plants. Increasing drought resistance via plant breeding is currently the main approach for improvement of plant productivity and reduction of water usage. Plant breeders and scientists search for reliable and easy methods of the determination of drought sensitivity in different genotypes. A precise knowledge about processes underlying plant stress response and defence against limited water availability is necessary for this. The aim of this study was a detailed analysis of photosynthetic processes and changes in leaf proteome (again aimed especially at proteins involved in photosynthesis) during drought stress. This analysis was a part of a complex project focused on the possible causes of genetic variability and drought resistance in agronomically important crops. Another goal was to analyse whether the measurement of selected photosynthetic parameters can be used for the determination of drought sensitivity per se and/or for the prediction of the response of hybrids based on known behaviour of their parents. In the case of time-consuming or arduous activity the individual bone, modifications of the individual skeleton may occur. Specific activities or movements are predominantly responsible for these modifications and are likely to be carried out for long periods from a young age of the individual. One of these modifications is considered to be some of the so- called non-metric traits on the postcranial skeleton. Special attention is being paid to changes of the articulation facets and changes in some muscular or ligaments' binding areas which are considered to be possibly related to excessive physical stress, occurring through the lifetime (squatting position). With respect to the frequency of 30 postcranial non-metric traits 293 skeletal remains in the Great Moravian population from the settlement agglomeration Mikulčice-Valy (9th - 10th century) have been compared. The primary goal of this research was to determine the frequency of non-metric traits and to verify the relationship with sex - age or the potential laterality differences in the case of bilateral traits.
