A three-phase study was conducted. In the first phase, individual interviews were conducted with 15 secondary school teachers to develop indigenous items for assessing teacher burnout and lack of reciprocity. In the second phase, 187 secondary school teachers were included to determine the items of newly-developed instruments and to examine their psychometric properties. In the third phase, the aims were to examine the validity and reliability of each measurement, to analysis the relations between teacher burnout and demographic variables, and to explore the relationship among lack of reciprocity, ERI, meaning of life and teacher burnout in a sample of 509 teachers from 25 secondary schools in Changchun, China. / According to the relationship between teacher burnout and demographic variables, results indicated that younger teachers reported more EE and DP. Teachers of junior rank reported more burnout. Teachers who taught Chinese, mathematics and English reported more experience of EE than teachers who taught other subjects. / Based on the social exchange theory of burnout, the effort-reward imbalance (ERI) model, and the existential theory of burnout, the purpose of this research was to investigate the relationship among investment-reward imbalance, meaning of life, and burnout among Chinese secondary school teachers. As one of the key variables in this study, investment-reward imbalance included two operational definitions which were named lack of reciprocity and ERI. / Limitations, theoretical and practical implications of the findings were also discussed. / Results showed that, based on the Maslach Burnout Inventory, the newly-modified Burnout Measurement consisted of three subscales: emotional exhaustion (EE), depersonalization (DP), and reduced personal accomplishment (RPA). The self-developed Reciprocity Scale included three dimensions, namely, reciprocity in the relationship with students, intangible reciprocity in the relationship with the school, and tangible reciprocity in the relationship with the school. All measurements in this study were found to possess good validity and reliability. / Structural equation modeling was used to test a set of hypothetic models of the relationship among lack of reciprocity, ERI, meaning of life and teacher burnout. Results showed that, lack of reciprocity in the relationship with students positively influenced FE and RPA. Lack of intangible reciprocity in the relationship with the school was positively associated with RPA. Lack of tangible reciprocity in the relationship with the school was positively associated with EE. Among the three dimensions of lack of reciprocity, lack of reciprocity in the relationship with students is a more important predictor to teacher burnout than other components. ERI was positively associated with EE. Considering the two operational definitions of investment-reward imbalance in the prediction of teacher burnout, lack of reciprocity predicted RPA better than ERI, while ERI predicted EE better than lack of reciprocity. Meaning of life was negatively associated with EE, DP and RPA. In addition, results supported the hypothesis that EE influenced DP on the structure of teacher burnout. / 李歆瑤. / Advisers: Patrick S. Genome-wide identification of novel candidate tumor suppressor genes in Hong Kong common tumors through integrative cancer epigenetics and genomics. / CUHK electronic theses & dissertations collection

Cancer is the leading cause of death in Hong Kong (21,300 new cases and 11,500 deaths in 2003), with nasopharyngeal carcinoma (NPC), esophageal cancer (ESCC), and colorectal cancer (CRC) among the common ones. For these tumors, most patients present with advanced stage disease and poor treatment outcome, with an urge of early detection. Epigenetic inactivation of tumor suppressor genes (TSG) by CpG methylation represents an important mechanism of tumorigenesis, in addition to genetic abnormalities. Tumor-specific methylation can also be used as biomarkers for the identification of novel TSGs and for cancer early diagnosis and prognosis prediction. / Finally, for the purpose of development of epigenetic biomarker for cancer molecular diagnosis, I screened gene methylation in the serum samples. Aberrant methylation of PCDH10 and DLC1 was detected in serum samples (2/14 (14%) and 4/14 (29%) respectively) from tumor patients but not in normal controls. It suggests that screening for PCDH10 and DLC1 methylation in sera could be a tumor-specific and non-invasive epigenetic biomarker for molecular diagnosis and prognostics. (Abstract shortened by UMI.) / In the second approach, 1-Mb array-based comparative genomic hybridization (aCGH) was carried out to detect DNA copy number aberrations, which contain potential TSG loci, in a panel of NPC and ESCC cell lines. Frequent deletions include: 1p36.3, 3p14-11, 4p16-15, 5p13-q12, 6p21-12, 8p22-cent, 9p, 9q22-31, 10p, 13q12, 14q32, 16q23-24, 17q11.2, 18q in NPC, and 1p21, 4q21, 7p21, 7q35, 8p22-23, 8q11, 10p11, 11q22, 13q31, 14q32, 18q11-23 in ESCC. Several deletions (3p14-11 and 16q23) were further investigated in detail in this study. More than 12 genes were identified to be frequently silenced by methylation in tumors, including FHIT (3p14), WNT5A (3p14), ADAMTS9 (3p14), FEZF2 (3p14), ROBO (3p12), CADM2 (3p12), EPHA3 (3p11), RAB (11q22), ADAMTS18 (16q23), and TUSC8 (16q23), while homozygous deletion of these genes was infrequently detected. Aberrant methylation of these genes was also frequently detected in primary tumors in a tumor-specific manner. The tumor suppressor functions of TUSC8, WNT5A, CADM2 and ROBO were further investigated and validated. Further experiment indicated that induction of tumor cell apoptosis may contribute to the tumor suppressor function of TUSC8. / Modified genomic methylation subtractive approaches using uracil-DNA glycosylase or combined with pharmacological demethylation were developed. GADD45G, PCDH10, ROR2, DLC1L1 were among a series of novel methylated targets identified by these approaches. Methylation-associated silencing of these genes was frequently detected in various types of tumor cell lines and primary tumors including NPC, ESCC and CRC, in a tumor-specific manner. Ectopic expression of these genes strongly suppressed tumor cell growth and colony formation of silenced tumor cells. Epigenetic inactivation of GADD45G is the major mechanism for the loss of its response to environmental stresses. Reintroduction of PCDH10 strongly suppressed tumor cell migration and invasion. Ectopic expression of DLC1L1 in silenced tumor cells resulted in a remarkable suppression of tumor cell clonogenicity, which depends on its GAP activity. Furthermore, DLC1L1, but not its inactivating mutants, inhibited Ras mediated oncogenic transformation. Development and 6-month validation of a typology of Chinese women experiencing miscarriage based on pregnancy, peraonality and cultural factors. / CUHK electronic theses & dissertations collection

In the first phase of this study, the cluster analysis results of 208 WEM provided initial empirical support for this typology. Three subtypes of WEM were identified. They were "Adjusted Women" (AW, N = 54, 26%), "Dysphoric/Mixed Type Women" (DW, N = 57, 27%), and "Gender-Bound Women" (GW, N = 97, 47%). Partially supporting the hypothesis, GW experienced the worst adjustment as reflected in their high level of perinatal grief symptoms compared to AW and DW. Further examination of the demographic characteristics of each WEM subtype showed that while DW reported relatively stronger personality (i.e., trait anxiety, trait depression, and neurotic personality) and cultural vulnerabilities (i.e., traditional ideal personhood and self-sacrifice) compared to AW, GW's reports of personality and cultural vulnerabilities faked the worst among the 3 WEM subtypes. Cluster-constrained hierarchical regression analyses revealed a distinct set of predictors for immediate postloss adjustment of AW, DW, and GW. Instead of a complete nested model, the present data fitted a partially nested model where AW were nested within GW, and DW represented a mixed type of WEM. Specifically, AW's perinatal grief was affected by pregnancy factors whereas GW's was affected by pregnancy, personality, and cultural factors. DW's perinatal grief was not affected by pregnancy factors but by personality and cultural factors. / In the fourth phase of this study, pregnancy, personality, and cultural factors as well as spousal emotional social support were reexamined for their possible implications for WEM's and the pregnant controls' psychological distress, state anxiety symptoms, and state depressive symptoms at 6 months following the initial assessment. The results showed that spousal emotional support at a 6-month follow up was a salient predictor of psychological distress state anxiety symptoms, and state depressive symptoms at 6 months after the initial assessment for both WEM and the pregnant controls While spousal emotional support at the initial assessment did not have the same effect, this result suggested that to mitigate the longer term poor psychological adjustment of WEM and pregnant women, sustained spousal emotional support is needed. Trait anxiety at the initial assessment was also a strong predictor of WEM's psychological distress, state anxiety symptoms, and state depressive symptoms at 6 months post miscarriage. Pregnancy factors at the initial assessment were only moderately related to the psychological adjustment of WEM and the pregnant controls, and cultural factors at the initial assessment were not related to any of the adjustment indicators at the 6-month follow up. (Abstract shortened by UMI.) / In the second phase of this study, comparisons were made between the 3 WEM subtypes and women with healthy uncomplicated pregnancy (pregnant controls, N = 258). The results showed that the 3 WEM subtypes experienced varying levels of adjustment problems---that is, psychological distress, state anxiety symptoms, and depressive symptoms---compared to the pregnant controls. GW, in particular, were 8 times more likely to be classified as psychological distress caseness and 4 times more likely to be classified as state anxiety caseness and state depression caseness, even after controlling for pregnancy factors and spousal emotional social support. / In the third phase of this study, the author attempted to establish predictive validity of the proposed WEM typology using 6-month 2-wave longitudinal data A subsample from Phase One and Phase Two of this study, including 103 WEM (AW = 33, 32%; DW = 27, 26%; GW = 43, 42%) and 139 pregnant controls, provided information on their psychological distress, state anxiety symptoms and depressive symptoms, as well as on their motivation to reproduce at 6 months after the initial interview. Although GW were significantly more likely than AW and DW to report being pregnant or having the intention to conceive at 6 months post miscarriage, no significant differences were observed between the 3 WEM subtypes in their psychological distress, state anxiety symptoms, and state depressive symptoms. / The present 4-phase study was an attempt to propose an integrated conceptual model to advance understanding of Chinese women's adjustment to miscarriage; that is the perinatal grief symptoms, psychological distress, state anxiety symptoms, and depressive symptoms they experience in response to miscarriage. Through a comprehensive review of sociobiological theory, attachment theory, psychoanalytic theory and the feminist perspective, the author proposes a conceptual model involving 3 major pathways, namely pregnancy, cultural, and personality factors. It was hypothesized that 3 subtypes of women who experienced miscarriage (WEM) (Adjusted Women, AW; Dysphoric Women, DW; Gender Bound Women, GW) could be identified with each subtype being affected by a combination of different factors. A rent-gap-theory-based cellular automata approach to urban land use simulation. / CUHK electronic theses & dissertations collection

Theoretical modeling and simulation are two common approaches to the study of land use change. While theoretical models are often limited by the simplicity in their assumptions, simulations are mostly low in generalization due to their specificity in applications. The integration of these two approaches will thus help us to model more closely the reality by having a firm theoretical foundation for real-life simulation. In the present research, a theory-based cellular automata (CA) simulation is proposed to simulate the changes of land uses. The land use change generally involves urban development and urban redevelopment processes. Rent gap theory is selected as the theoretical model to describe the mechanism of urban redevelopment. On the other hand, relevant conditions related to urban development are incorporated into the simulation of new development because rent gap theory is inadequate in accounting for all mechanisms that are working together to shape the pattern of land use change. In order to perform CA simulation under random fluctuation in time, stochastic formulation of the rent gap theory is first made in the form of a build up value equation to seek for the time of appearance of rent gap. The time of urban redevelopment depends on the appearance of rent gap while the time of urban development depends on the conditions of local characteristics and government policy. To substantiate the theoretical arguments, the land use change of Hong Kong in the period of 1966--2001 is selected as a case study. For the pattern of future development of Hong Kong, the simulation is carried out up to 2016. It also serves as a basis for the evaluation of the integrative utilization of a theoretical model and a simulation method. Delivery planning under uncertainty in logistics and express industry. / CUHK electronic theses & dissertations collection

In this dissertation, we aim to investigate some delivery planning problems under different aspects of uncertain conditions in order to present some insights on how to develop more efficient and effective logistics solutions in the challenging, dynamic and competitive industry of express delivery nowadays. / Lastly, we introduce a travel time estimation method which can be considered as an upstream extension on the first delivery planning problem or any routing problems that require travel time in a road network as an input parameter. We realize that different trips may traverse on some links that are transited by other trips as well; and therefore linear and quadratic programming models can be developed to infer the travel time of any road segments which are traversed by more than one trip from the consideration of the common road segments of various trips. Bus routes information is proposed as the data source because of their coverage on the road network and their data available. Robustness of the proposed models is evaluated according to the deviation between the resultant inferences and the means of the segment travel times. It is found that the basic models suffer from a fundamental problem of underspecification issue if the number of observed trips (i.e. constraints) is fewer than the number of road segments (i.e. variables) to be estimated. Therefore, two additional types of constraints are introduced to address these issues. / The focus of the second delivery planning problem addressed in this dissertation is switched to the randomness of the demands at the delivery locations. We consider a single-depot problem in which a large volume of packages have to be delivered in a dense and small area with a lot of high-rise buildings within an extremely tight delivery commitment time window. At each vertex, the package volume (i.e. demand) is a highly stochastic variable and its service time is also dependent on the demand. This version of problem is typically faced by express service providers which offer premium express services to customers that are protected by the money back guarantee (MBG) condition. We name this version of problem as a time-constrained vehicle routing problem with stochastic demands and service times (SVRP-D). Since both the demand and service time of a vertex are stochastic, the vehicle capacity and the commitment-time (i.e. deadline) constraints may not be satisfied after the demands are realized, and recourse action is thus necessary to "rescue" the "infeasible" delivery plan through additional effort and cost. Three typical recourse actions are independently considered: (1) restocking; (2) outsourcing; and (3) reassignment. A two-stage stochastic integer programming is proposed such that the first-stage problem is to determine a set of a priori routes which minimize the vehicle travel cost and the cost for any known recourse actions. Dependent on a first-stage solution, the second-stage problem is to decide the optimal recourse plan with least cost according to the pre-determined recourse action. / Two delivery planning problems and one travel time estimation model under various aspects of randomness are addressed in this dissertation. The first delivery planning problem considers an extension on a well-known problem---the vehicle routing problem with time windows (VRPTW), where the travel time required between each delivery location is a stochastic variable, instead of a fixed value. We name this version of the VRPTW as a vehicle routing problem with time windows and stochastic travel times (VRPTWST). A two-stage stochastic integer programming with recourse model is developed and its objective is to minimize the vehicle travel cost together with the expected loss due to the violation of the delivery time windows. An exact branch-and-cut (B&C) algorithm is proposed. This algorithm is effective for small-size problems, e.g. instances with only 8 vertices. Impairment mitigation for high-speed optical communication systems. / CUHK electronic theses & dissertations collection

Electronic equalization has recently attracted considerable interest for impairment compensation for its significant cost saving and adaptive compensation capability. In this thesis, we propose novel maximum-likelihood sequence estimation (MLSE) structures for various advanced modulation formats. Electronic equalization of advanced modulation formats further extends the transmission reach and relaxes the speed limitation of electronic devices. We also propose novel application of MLSE for mitigation of timing misalignment between the pulse carver and data modulator in return-to-zero (RZ) systems. / In access networks, we focus on the achievement of centralized light source (CLS) wavelength-division-multiplexing passive optical networks (WDM-PON) with data rate of 10 Gbit/s for both downstream and upstream signals. The previous CLS WDM-PON schemes at 10 Gbit/s suffer from chromatic dispersion (CD) and/or asynchronous upstream modulation. We propose two solutions to mitigate these impairments. By eliminating the modulation synchronization module and all-optical CD compensation module, the proposed methods greatly reduce the cost and operation complexity of high-speed WDM-PON. / In the monitoring for impairment compensation, we propose a polarization-insensitive monitoring scheme for synchronized phase re-modulation by using a narrowband optical-passband filter (OBPF). With the optimal central wavelength of the OBPF, high monitoring sensitivity is achieved. / The increasing bandwidth demands have aroused a myriad of industry and academic activities to develop cost-effective optical communication systems with data rates of 10 Gbit/s and beyond. However, as the capacity grows, many signal degradation effects become prominent and seriously limit the data rate and the transmission distance. The mitigation of the impairments inevitably increases the operation complexity and implementation cost. The focus of this thesis is to develop new impairment mitigation approaches to improve the impairment compensation performance and/or to reduce the operation complexity and cost. As a result, cost-effective high-speed optical communication systems are enabled. / To freely enable the employment of advanced modulation formats for optical communications, we propose all-optical conversion from 40-Gbit/s RZ signal to 40-Gbit/s inverse-RZ/10-Gbit/s differential-phase-shift-keying orthogonal modulation signal to interface high-speed transmission systems using RZ format with networks using orthogonal modulation format. Shi Hua is an important genre in the traditional Chinese literary criticism. The Qing dynasty (1644-1911) is the prime period of the composition of Shi Hua, the number of which amounts to a total of more than 1,000 works. Compared with other dynasties, this figure is unsurpassable. The study of Shi Hua usually focuses on one author, one school or one dynasty, and a large-scale research from the vision of area is rare. This dissertation aims to study the works of Shi Hua of the Guangdong Province in the Qing Dynasty. Starting to appear in the Ming dynasty (1368-1644), the works of Shi Hua of Guangdong became abundant in the Qing dynasty. Even after the founding of the Republic of China in 1911, the composition of the works of Shi Hua was still occasionally observed. During the Qing dynasty, about 100 works of Shi Hua were composed by the authors of Guangdong nativity, while only 30-odd works are still extant. The rich content of these works demonstrates the intimate relationships between the Shi Hua of Guangdong and the poetry of the same area as well as the entire Qing dynasty. / The study of this dissertation is three-layered. Firstly the historical development of the works of Shi Hua of Guangdong in the Qing dynasty is discussed, and the information about the extant works and their authors is investigated. Secondly the development and evolution of the Guangdong poetry are analyzed on the basis of the content of the works of Shi Hua. Thirdly the theories and the value of several important works of Shi Hua in Guangdong are studied. The dissertation is divided into eight chapters: Chapter 1, the development of the works of Shi Hua of Guangdong in the Qing dynasty: in a chronological order, the development of the works of Shi Hua of Guangdong in the Qing dynasty is discussed in order to outline their literary thoughts. Chapter 2, the works of Shi Hua of Guangdong in the Qing dynasty and their authors: the authors' nativities and the background of composition are scrutinized. Chapter 3, the discussions about the Guangdong poetry in the works of Shi Hua of Guangdong in the Qing dynasty: the regional features of the Guangdong poetry discussed in the Shi Hua and their relations with poetry societies, clans, paintings, political background and the compilation of collectanea are studied, and the poetic thought of 'Tang-following' is emphasized. Chapter 4, the study in the works of Shi Hua of Huang Pei-fang: Huang's attitude towards the Qing poetry is studied, his poem creation theory is examined so as to reveal his poetic thought of 'Tang-following'. Chapter 5, the study in the works of Shi Hua of Zhang Wei-ping: Zhang's comments on the Qing poetry is studied, the principles and influence of his works of Shi Hua are deduced, and his poetic thoughts of 'spirit' and 'profoundness' are explored as well. Taking into account of the colonial nature of modern Hong Kong, this author is to examine how the TWGHs as a medical space gradually developed from one that used only Chinese medicine into one in which Chinese medicine and western medicine coexisted. However, it finally became a western style hospital using only western medicine in the inpatient services in the 1940s, along with the growing hegemony of western medicine that was underpinned by colonial power. The multidimensional relationships among different agents in the process of transformation of medical services in the TWGHs constitutes another important theme of this thesis. These relationships touched upon a series of significant interactions between colonial government and Chinese community, colonial authorities and the Tung Wah Board of Directors, Chinese and western medical practitioners, Chinese community and the Tung Wah authorities, and so on. / The implantation, dissemination and expansion of modern western medicine, as an important part of western learning that infiltrated into the Orient, exerted profound impacts on Chinese traditional medical patterns and Chinese medical ideas and practices. As the center for exchange between Chinese and Western Culture, Hong Kong became a significant space for the spread and practice of western medicine. A wide range of western medical services and activities were delivered and developed by the colonial government, western missionaries, benevolent societies, and private practitioners in order to promote the development and popularization of western medicine among the Chinese community, including the establishment of hospitals, dispensaries and clinics, the opening of medical schools and training of western doctors, and the promotion of public health education. / This thesis also points out that the early intense prejudice and resistance against western medicine is not necessarily and cannot be entirely attributed to the underlying difference in the concept and practice of healing and sickness in the two different medical systems. Instead, I argue that a number of technical and practical factors in the delivery of western medical services provided by different agencies greatly affected and determined the choices and uses of the Chinese population. At the same time, the gradual recognition and reception of western medicine among the Chinese was not only the passive result of the compulsory western medical system developed by the colonial government, but also an active realization of the real efficiency and value of western medicine among the indigenous population and their consent and acceptance of its ideology and cultural value, to a great extent. / This thesis examines the confrontation and interaction between Chinese medicine and Western medicine, and the diverse and complicated Chinese attitudes towards western medicine by studying the history of the introduction of western medicine into Hong Kong and the case of transformation of Chinese and western medical services in the Tung Wah Group of Hospitals (TWGHs) during the period between 1894 and 1941. The history of the TWGHs dates back to the opening of the Tung Wah Hospital in 1870. Originally intended for the accommodation and treatment of those Chinese who had strong fears and prejudices against western medicine, the Tung Wah Hospital was founded to provide treatment only by Chinese doctors using Chinese medicine. The bubonic plague of 1894 in Hong Kong marked an important turning point in the history of medical services of the Tung Wah Hospital.
