Establishing associations for the evaluation of mobility screen (EMS) in an adult South African population

Background: Muscle, joint and bone injuries affect mobility and stability, which in turn limits physical activity. Screening tests such as the Functional Movement Screen (FMS) are used to assess an individual’s mobility and stability to determine whether any movement dysfunctions exist. Screening tests aim to establish an individual’s injury risk with the goal of guiding an intervention program. The Evaluation of Mobility Screen (EMS) is a screening test that has been developed at the Sports Science Institute of South Africa. The EMS has been adapted from the FMS by exchanging the Rotatory Stability test for the Seated Rotation test. The current use of screening tools is limited because of the lack of normative data sets that represent the diversity of age, gender and physical activity levels in the general population. Most current published data represent athletes or younger populations. By establishing the relationship between screening outcomes and variables such as age, gender and physical activity level, the effectiveness of screening tests may be improved. Aim: To describe associations between EMS scores for males and females across different age groups and levels of physical activity. Objective: To evaluate and compare differences in EMS scores relating to age, gender and physical activity levels. Methods: This was a quantitative study, with a descriptive, correlational design. The sample consisted of 135 males and 127 females between the ages of 18 and 60. The EMS data were collected at the HighPerformance Centre, in the Sports Science Institute of South Africa, Cape Town. Results: There was no difference between the total scores of males and females (median = 17). The two youngest groups (20-30 and 31-40 years) scored the highest (median = 17), while the oldest group (51-60 years) scored the lowest (median = 15). Gender had a significant effect (p < 0.05) on five subtests (Single Leg Hurdle, Shoulder Mobility, Asymmetric Leg Raise, Stability Push Up and Seated Rotation). Age had a significant effect (p < 0.05) on three subtests (Overhead Squat, Single Leg, Hurdle Split Squat). Physical activity level had a significant effect (p < 0.05) with two subtests (Single leg Hurdle and Stability Push Up). Conclusion: Gender, Age and Physical Activity are associated with changes in EMS scores. EMS total scores declined as age increased. While the total scores remain similar between genders, there were clear variations within the different subtests. The oldest participants (51-60 year) scored the lowest throughout all subtests. Males scored higher in the strength components, while females scored higher in the flexibility components. Physical activity levels did not have a clear pattern as expected but still demonstrated association with two subtests. The results add to the sentiment that the focus should move away from the composite scoring system, and towards analyzing individual subtest scores. Future studies should also investigate if subtest scores can be improved by targeted intervention programs.

Internal and external load measures as predictors of overuse injury risk in professional football players

Background Football is the most popular sport worldwide. Football has grown into a faster, intensive and more competitive game with a substantial increase in technical and physical demands. To reach the peak demands of match play, extensive training is necessary to improve performance and to reach the top level in professional football. Inadequate training loads prevent optimal performance adaptions, place the player at higher risk of being underprepared and may increase the risk of overuse injuries. Determining an optimal training load that improves performance and decreases the risk of overuse injuries is important. Therefore, monitoring and understanding individual responses to training loads are necessary. To date there is limited research regarding prediction of risk of overuse injuries with respect to optimal TL in professional football players. Aim To describe the pattern of injuries and determine the influence of load metrics and injury risk in South African professional football. The total GPS distance covered, the number of GPS measured high-intensity sprints and session Rating Perceived Exertion load and the effects on the risk of developing an overuse injury in professional football players. Objectives (1) To determine the relationship between total GPS distance (m) covered, ACWLR and overuse injuries in a full competitive season. (2) To determine the relationship between GPS measured high-intensity sprints, ACWLR and overuse injuries in a full competitive season. (3) To determine the relationship between session rating of perceived exertion, ACWLR and overuse injuries in a full competitive season. (4) To determine the overuse injury risk per playing position (defenders, midfielders and attackers). (5) To determine the patterns of injury during a full competitive season. (6) To determine the effect of the internal load (sRPE) and external load (GPS) in a congestion week compared to a normal week on overuse injury risk. Methods Data was collected from 32 professional football players in the first and reserve team over one full competitive Premier Soccer League season (2016/17). Training load metrics were assessed using the acute:chronic workload ratio (ACWLR) to predict overuse injury risk within the team. The relationship between total GPS distance (m) covered (TDC), GPS measured high-intensity sprints (HIS), session rating of perceived exertion (sRPE) and ACWLR and overuse injuries was determined. Overuse injuries were described based on frequency, anatomical position and injury type as well as with regards to playing position (defenders, midfielders and attackers). The effect of a congestion week on overuse injury risk was also determined. Results No significant outcomes were recorded when predicting overuse injuries for the whole team, with regards to average TDC, HIS and sRPE ACWLR. Overuse injuries may be predicted when monitoring the individual player loads, thereby taking into account the peak demands of match play per playing position. Large difference between TDC and HIS and large increases or decreases (20%) within weeks may increase the risk of overuse injuries. Hamstrings and groins injuries are the most common injuries sustained and defenders sustained the most overuse injuries within the team relative to exposure time. Congestion weeks did not predict overuse injury risk.

Does Foam Rolling have a Positive Effect on Performance and Recovery from Post Exercise Induced Muscle Damage: A Systematic Review of the Literature to Guide Practitioners on the use of Foam Rolling

Foam rolling is a form of myofascial release performed by the individual using their body weight, as opposed to the pressure being applied by the clinician. Foam rolling is currently used by athletes at all levels, from recreational to elite athletes. Foam rolling is used as a warm up to aid performance and/or recovery. This study aims to review the literature to determine if foam rolling enhances performance, positively affects recovery from exercise induced muscle damage, and whether there is a consensus on the protocol to achieve performance and recovery enhancement. Seven electronic databases, Google Scholar, Science Direct, Pubmed Central, Pubmed, ISI Web of Science, Medline and Scopus, were searched using terms related to foam rolling between January 2006 and April 2017. Published articles that included foam rolling as a recovery intervention and/or a performance enhancing tool were included in the study and assessed using the PEDro scale for methodological quality ratings. This dissertation will consolidate and add to the knowledge on the use of foam rolling. It will highlight when foam rolling should be used and recommend a specific protocol that should be used for performance and/or recovery purposes. This may address the confusion around when foam rolling should be used and show foam rolling to be effective as a recovery tool or for enhancing performance. This may help with better management of athletes by sports personnel, and in turn improve their performance and enhance their recovery. A total of 33 articles met the inclusion criterion and were systematically reviewed. There is evidence supporting the use of foam rolling in a warm-up consisting of dynamic stretching and an active warm-up to enhance performance; mainly through its effects on flexibility while maintaining muscle contractility. There is also evidence supporting the use of foam rolling to enhance recovery from exercise induced muscle damage and delayed onset of muscle soreness, with its main effects being the shortening of time to return to baseline performance, flexibility and pressure pain threshold. The mechanism by which foam rolling acts seems to be a neural response to pressure exerted. Sixty to ninety seconds of foam rolling may suffice to achieve the above desired effects. Further research is needed to determine the exact mechanisms of action of foam rolling, as well as the risks that may be associated with foam rolling.

Οι ομάδες των ορμονών που επηρεάζουν σημαντικά την αύξηση και την ανάπτυξη των παιδιών και των εφήβων, ανήκουν κυρίως στον άξονα της αύξησης και της αναπαραγωγής, στο σύστημα του stress και στο ανοσολογικό σύστημα. Η κορτιλόλη, η λεπτίνη και η IL-6 αποτελούν στρεσογόνους παράγοντες οι οποίοι συνδέονται με τη δυναμική της ανάπτυξης και ενηβωσης του ανθρώπινου οργανισμού. Εκτός από την επίδραση τον ορμονών, η ενήβωση και η αναπαραγωγή του φύλου μπορεί να επηρεαστεί από φαρμακολογικούς παράγοντες, όπως είναι η υδροκορτιζόνη, τα αναβολικά στεροειδή και τα οιστρογόνα. Η φυσική άσκηση είναι ένας τύπος φυσικού stress που προκαλεί πλήθος ορμονικών, μεταβολικών και ανοσολογικών αλλαγών ενώ οι αθλητές και οι αθλήτριες ρυθμικής και ενόργανης γυμναστικής εκτίθονται σε έντονους φυσικούς και ψυχολογικούς στρεσογόνους παράγοντες. Τα τελευταία χρόνια οι προσδιορισμοί στο σίελο προσελκύουν τεράστιο ενδιαφέρον (λόγω του ανώδυνου τρόπου συλλογής του σιέλου) και κυρίως χρησιμοποιούνται στην Ενδοκρινολογία για την παρακολούθηση των επιπέδων των στεροειδών ορμονών. Υλικά και Μέθοδοι: Μελετήθηκαν 142 αθλήτριες και 97 αθηλτές της ενόργανης γυμναστικής υψηλού αγωνιστικού επιπέδου καθώς και 107 αθλήτριες της ρυθμικής γυμναστικής. Ως ομάδα ελέγχου μελετήθηκαν 73 κορίτσια και 68 αγόρια, τα οποία δεν αθλούνταν. Η συλλογή των δειγμάτων έγινε χρησιμοποιώντας μια ειδικά σχεδιασμένη συσκευή συλλογής σιέλου (Salivette). Για τον προσδιορισμό της λεπτίνης στο σίελο χρησιμοποιήθηκε ραδιοανοσολογική μέθοδος ενώ για τον προσδιορισμό της κορτιζόλης η μέθοδος ηλεκτροχημειοφωταύγειας. Για την μέτρηση της IL-6 στο σίελο αναπτύχθηκε και επικυρώθηκε μέθοδος Elisa υψηλής ευαισθησίας. Αποτελέσματα: Οι αθήτριες της ρυθμικής και της ενόργανης γυμναστικής χαρακτηρίζονται από υψηλότερα επίπεδα κορτιζόλης στο σίελο (p<0,01) συγκριτικά με τους μάρτυρες και τους αθλητές της ενόργανης. Τόσο στους αθλητές όσο και στις αθλήτριες της ενόργανης γυμναστικής, οι απογευματινές τιμές κορτιζόλης (μετά από άσκηση) παραμένουν στα ίδια επίπεδα με τις πρωινές τιμές. Στις αθλήτριες της ενόργανης τα επίπεδα της IL-6 παρουσιάζουν τάση αύξησης και της λεπτίνης τάση μείωσης μετά από άσκηση, αν και οι μεταβολές αυτές δεν είναι στατιστικά σημαντικές. Η συλλογή του σιέλου είναι μια μη επεμβατική, ανώδυνη τεχνική που επιτρέπει την επαναλαμβανόμενη δειγματοληψία. Η ραδιοανοσολογικής μέθοδος είναι απλή και εύκολη για τον προσδιορισμό της λεπτίνης στο σίελο, ενώ η μέθοδος της ηλεκτροχημειοφωταύγειας αποτελεί αξιόπιστη επιλογή για τη μέτρηση της κορτιζόλης στο σίελο. Για τον προσδιορισμό της IL-6 στο σίελο απαιτείται η ανάπτυξη και η επικύρωση μεθόδου Elisa, λόγω του ότι ο σίελος επηρεάζει τη μέτρηση της συγκεκριμένης κυτοκίνης. Τόσο στις αθλήτριες όσο και στους αθλητές της ενόργανης γυμναστικής υψηλού αγωνιστικού επιπέδου, ο νυχθημερήσιος ρυθμός έκκρισης της κορτιζόλης καταργείται. Επίσης οι αθλήτριες της ενόργανης γυμναστικής χαρακτηρίζονται από υψηλότερες τιμές δεικτών stress συγκριτικά με τους αθλητές. / The aim of this study was to evaluate the effects of intensive physical exercise and acute psychological stress during high level athletic competition as reflected on the levels of salivary cortisol, leptin and IL-6 in elite artistic and rytmic gymnasts. Design: The study included two hundred and forty-nine (249) Caucasian athletes (elite Gymnasts) -142 girls(aged 16.0 ± 1.6 years) and 97 boys (aged 16.8 ± 1.1 years) from 44 European countries, during theEuropean Championship of Gymnastics held in Volos-Greece on April 2006. One hundred and forty-one pubertal adolescents, who were recruited from public schools in Greece, not engaged in strenuous sports activities were used as controls (73 girls aged 16.0 ± 1.4 years and 68 boys aged 15.3 ±2.0 years). Methods: Saliva samples were collected, the morning before training and in the afternoon shortly after the competition. Cortisol concentrations were measured using a chemmiluminesce method. Leptin and IL-6 concentrations were measured using RIA and ELISA methods. Acute stress was assessed using a questionnaire designed for the study. Results: No difference was found between morning and afternoon salivary cortisol levels in both male and female AG. Female AG presented higher levels of morning salivary cortisol than female controls (p<0.05). Both male and female AG had higher degree of psychological stress in comparison with controls . Female AG had higher morning and afternoon salivary cortisol levels and higher degree of stress than males. Conclusions: In elite AG the diurnal rhythm of salivary cortisol has been abolished, probably due to the strenuous training and competition conditions. Female AG presented higher levels of morning salivary cortisol and psychological stress compared to both male AG and female controls. The long term consequences of these modifications of the HPA axis remain to be elucidated.

It is a well known that inadequate recovery will prolong the time it takes for the runner’s body to adequately adapt between training sessions and competitions. Anecdotal claims have been made about compression garments as a beneficial method to assist recovery after training sessions and competitions. Until now limited scientific research has addressed the influence that compression garments have on the recovery process after sporting activities. The benefits of compression garments, as a possible recovery modality, are that it is costeffective, practical and easily obtainable. This study endeavored to investigate the possible influence that compression garments may have on middle-aged long distance runners’ recovery rate after a prolonged run. This is the first study that has focused on compression garments as a post-exercise recovery modality for experienced middle-aged long distance runners. The other unique aspect of this study is the prolonged two-hour treadmill protocol that was used to induce muscle soreness. In addressing the aims, a randomized, crossover study design was used to investigate the possible benefits that the high pressure (CCL II 23-32 mmHg (mercury millimeter)) graduated compression garments may bring about. Seven competitive male long distance runners (height: 176.0 ± 8.6 cm; body mass: 92.5 ± 11.8 kg; VO2max: 45.7 ± 5.0 mL.kg-1.min-1) between the ages of 36 to 51 years volunteered for the study. The runners had to complete a two-hour treadmill run at 70 % of their predetermined maximum aerobic capacity, followed by a monitored 72-hour recovery period. The first part of the prolonged run was a 90–minute variant gradient run, followed by a 30-minute downhill run. Each subject acted as his own control and visited the Stellenbosch University’s Sport Physiology Laboratory (South Africa) on two occasions, separated by 7 to 28 days. One test was done with a compression garment (23 to 32 mmHg) and the other without. Testing included the measurement of lower limb circumferences (ankle, calf, mid- and proximal thigh), plasma lactate, lactate dehydrogenase and creatine kinase concentrations and the completion of subjective questionnaires on perceived muscle soreness (visual analog scale (VAS)). The lower extremities’ functional ability was determined with a time to exhaustion (TTE) step test, a vertical jump test (VJ) and modified sit-and-reach flexibility test. Preexercise measurements were taken and immediately after and during the 72 hour after the treadmill run and repeated for the second bout. The main outcomes of this study showed that the two-hour treadmill run induced delayed onset of muscle soreness, with and without the compression garment. Evidence of this was a significant rise in plasma creatine kinase (CKp) over the duration of both trials (P < 0.05). The compression garment significantly reduced swelling in the calf muscle (41.0 ± 0.2 vs. 41.5 ± 0.5 mm; P < 0.002). Runners showed a lower perceived muscular pain and discomfort while performing functional knee movements at 24 and 48-hours after the two-hour run with the compression garment (1.2 ± 1.6 vs. 3.8 ± 2.4 cm and 0.9 ± 1.8 vs. 3.0 ± 2.6 cm on VAS, respectively; P < 0.05). Significant differences in perceived muscle soreness between the WCG and WOCG trials were observed at 24-hours after the run during rest (0.1 ± 0.2 vs. 0.4 ± 0.8 cm; P = 0.02) and with stretching (1.9 ± 1.2 vs. 3.5 ± 2.5 cm on VAS P = 0.02). The perceived pain associated with pressure was significantly lower with the compression garment at 24 (307 %) and 48-hours (237 %) after the run (P < 0.05). Blood lactate levels were reduced during the acute phase of recovery at 10 (1.8 ± 0.5 vs. 2.2 ± 0.9 mmol.L-1; P = 0.05) and 30 minutes (1.8 ± 0.5 vs. 2.4 ± 0.4 mmol.L-1; P = 0.01) after the run, as well as plasma creatine kinase concentrations were statistically significantly lower at 24-hours (238.3 ± 81.3 vs. 413.3 ± 250.8 units.L-1; P = 0.005) after exercise with the compression garment. The two-hour treadmill run and the compression garment had no significant influence on the runners’ lower limb strength, power, endurance or flexibility (P > 0.05). Compression garments demonstrated the potential to enhance recovery after prolonged strenuous exercise in well trained middle-aged runners. In addition, runners did not experience additional fatigue from the moderate to high pressure garments. The effect of higher pressure compression garments on athletic performance and the psychological influence of the garment need further investigation. / AFRIKAANSE OPSOMMING: Verskillende tipes naoefening herstelstrategië, vorm deel van die moderne atleet se daaglikse routine. Dit is wel bekend dat onvoldoende herstel sal beteken dat die atleet se liggaam langer sal neen om aan te pas tussen inoefen sessies en kompetisies. Sekere bewerings word al gemaak omtrent die voordeligheid van kompressiesokkies tydens die herstelperiode na oefening sessies en kompetisies. Tot nou toe was daar beperkte wetenskaplike navorsing oor die invloed van kompressie sokkies of die herstel proses van sport aktiwiteite. Die voordeel van kompressie sokkies as ‘n moontlike herstelmetode, is dat dit koste-effektief, prakties en maklik verkrybaar is. Hierdie studie poog om ‘n ondersoek in te stel na die moontlike invloed wat kompressie sokkies op middeljarige lang-aftstandatlete se herstelperiode sal hê na ‘n verlengde hardloopsessie. Hierdie is die eerste studie wat konsentreer op kompressie sokkies as ‘n naoefenings hersteltegniek vir ervare middeljarige lang-afstandatlete. Die ander unieke aspek van die ondersoek is die langdurige tweeuur trapmeul protokol wat gebruik word om spierpyn te veroorsaak. Om die doel te bereik, is ’n lukrake oorkruis studie gebruik om ondersoek in te stel na die moontlike voordele van die hoë druk (CCL II 23-32 mmHg) kompressie sokkies. Die twee-ure trapmeul toets en die kompressie sokkies het geen betekenisvolle invloed gehad op die wedlopers se onderlyf ledemate se plofkrag, uithouvermoë of soepelheid (P > 0.05) nie. Kompressie sokkies het gewys dat dit potensiaal het om met herstel te help na lang en harde oefening in geoefende middeljarige atlete. Nietemin is daar verdere wetenskaplike navorsing nodig om dit te bevestig. Wedlopers het nie addisionele vermoeienis van die drukking van kompressie sokkies ervaar nie. Sterker drukkende kompressie sokkies sowel as die sielkundige invloed van die sokkies benodig verdere navorsing.spierpyn ervaaring (“visual analog scale” (VAS)). Kompressie sokkies het gewys dat dit potensiaal het om met herstel te help na lang en harde oefening in geoefende middeljarige atlete. Nietemin is daar verdere wetenskaplike navorsing nodig om dit te bevestig. Wedlopers het nie addisionele vermoeienis van die drukking van kompressie sokkies ervaar nie. The development of a fall risk assessment and exercise intervention programme for geriatric subjects

Thesis (M Sport Sc)--Stellenbosch University, 2012. / ENGLISH ABSTRACT: Falling is a common occurrence and one of the most serious problems in the elderly population (65 years and older). Falls account for 70% of accidental deaths in persons aged 75 years and older. Falls can be markers/indicators of poor health and declining function, and are often associated with significant morbidity. More than 90% of hip fractures occur as a result of falls, with most of these fractures occurring in persons over 70 years of age (Fuller, 2000). About one third of people aged 65 years and older fall each year, resulting in a substantial decrease in quality of life in addition to placing a huge burden on current health care systems. The purpose of this study was to determine whether a 12-week exercise intervention programme, consisting of two 30 minute exercise sessions a week, could lower the risk of falling in a group of elderly women. Female subjects (n=22) with an average age of 79.5 years were selected from three retirement homes situated in Stellenbosch, Western Cape, South Africa, according to specific inclusion and exclusion criteria. The subjects included presented no major cardiovascular and pulmonary disease signs and symptoms as recognised by the American College of Sports Medicine (2011); had no serious illnesses or co-morbidities; were mobile with no significant musculoskeletal disorders; had no uncorrected visual or vestibular problems as well as no significant cognitive impairments or major psychological disturbances; were not taking any psychotropic medications or Benzodiazepines that could affect their progress. Subjects also had to be willing to follow the 12-week exercise intervention programme and sign an informed consent document. The selected subjects then underwent a pre- and post-intervention assessment consisting of a subjective rating of their fear of falling, the Fall Risk Assessment: Biodex Balance system, Balance Evaluations Systems Test (BESTest) and the 30-Second Chair Stand Test. Statistica 10 was used to analyse the data. Data was analysed to assess any significant improvements that the exercise intervention had on each fall risk variable tested. The main fall risk variables consisted of: fear of falling, muscular strength, balance, gait and getting up strategies. Statistically significant improvements (p<0.001) were seen in: Fear of falling, muscular strength, balance, gait and getting up strategies after the 12-week exercise intervention programme. This study suggests that exercise intervention has the potential to decrease the risk of falling among elderly women and should play an extremely important role in the prevention of falling amongst this population group. / AFRIKAANSE OPSOMMING: Om te val is ‘n alledaagse gebeurtenis en een van die mees ernstige probleme vir ons bejaarde bevolking (65 jaar en ouer). Insidente van val verklaar tot 70% van toevallige sterftes met betrekking tot persone van 75 jarige ouderdom en ouer. Om te val kan ‘n teken van swak gesondheid en/of ‘n afname in funksionele kapasiteit wees, en is gewoonlik met morbiditeitspatrone gekoppel. Meer as 90% van heupfrakture kom as gevolg van valle voor, waar die meeste van die frakture in persone bo 70 jarige ouderdom voorkom (Fuller, 2000). Minstens een derde van persone bo 65 jaar en ouer val elke jaar, so ‘n val het ‘n noemenswaardige afname in lewenskwaliteit tot gevolg asook ‘n enorme druk wat op huidige gesondheidsorg sisteme geplaas word. Die doel van die studie was om te bepaal of ‘n 12 week oefenintervensieprogram, wat uit twee oefen sessies van 30 minute elk bestaan, die risiko van val vir n groep bejaarde vroue kan verlaag. Vroulike individue (n=22) met ‘n gemiddelde ouderdom van 79.5 jaar uit drie ouetehuise/aftree oorde in Stellenbosch, Wes-Kaap, Suid-Afrika geleë; is volgens bepaalde insluitings- en uitsluitingskriteria geselekteer. Die geselekteerde individue het ‘n pre- en post-intervensie assessering ondergaan wat uit ‘n subjektiewe bepaling van hul vrees vir val bestaan het, die Val Risiko Assessering asook “Biodex Balans System Test, Balance Evaluations Systems Test (BESTest)” asook die 30 Sekonde Stoel-staan Toets. Statistica 10 is gebruik om die data te analiseer. Data was geanaliseer om enige merkwaardige verandering wat die oefenintervensie op elke val risiko veranderlike wat getoets was gehad het, te bepaal. Die belangrikste val risiko veranderlikes het uit: die vrees vir val, spier sterkte, balans, stappatroon en opstaan tegnieke bestaan. Betekenisvolle statistiese veranderinge (p<0.001) is gerapporteer in: die vrees vir val, spiersterkte, balans, stappatroon en opstaan tegnieke na die 12 week oefenintervensieprogram. The potential of exercise to reverse stress induced abnormalities in the rat brain

Medical Physiology.))--University of Stellenbosch, 2010. / ENGLISH ABSTRACT: Adverse experiences during early life causes alterations in the development of the central nervous system structures that may result in abnormal functioning of the brain. It is well known that, in humans, adverse early-life experiences such as social separation, deprivation, maternal neglect and abuse increase the risk of developing psychiatric disorders, such as depression, later in life. We used maternal separation in the rat as a model for early life stress to firstly determine how different brain systems are dysregulated by this stressful experience and additional chronic or acute stress during adulthood. Rat pups were separated from their mothers on postnatal day 2-14 for 3 hours per day while control rats were normally reared. The behavior, stress response, neurotrophin, apoptotic marker and serotonin levels in the ventral hippocampus, striatum and frontal cortex were measured during adulthood. A different group of maternally separated rats were allowed chronic voluntary exercise and similar measurements were done to determine whether exercise was able to normalize the deficits caused by early life stress. Differentially expressed cytosolic proteins of the ventral hippocampus of maternally separated rats versus normally reared rats were also identified. Protein expression levels of maternally separated rats that received chronic voluntary exercise or escitalopram treatment were subsequently determined to unravel the mechanism of therapeutic action for these two interventions. We found that maternal separation increased the baseline corticosterone response of rats and induced a blunted adrenocorticotropin hormone after acute restraint stress. Baseline neurotrophin levels were significantly decreased in the ventral hippocampus. Maternal separation followed by chronic restraint stress during adulthood resulted in increased depressive-like behavior compared to control rats. Maternal separation alone or followed by acute restraint stress during adulthood induced changes in apoptotic marker expression in the striatum and frontal cortex. In rats subjected to maternal separation and chronic restraint stress during adulthood, we found that chronic voluntary exercise decreased their depressive-like behavior and increased brain derived neurotrophin levels in the striatum. Serotonin levels were not affected by maternal separation, but chronic voluntary exercise increased serotonin in the ventral hippocampus of normally reared rats. Maternal separation induced a number of changes in the expression of cytosolic proteins and these stress-induced changes were identified in proteins relating to cytoskeletal structure, neuroplasticity, oxidative stress, energy metabolism, protein metabolism, and cell signaling. Chronic voluntary exercise was able to restore the expression levels of a number of proteins affected by maternal separation that increased the risk for neuronal death. When comparing the efficacy of exercise to that of escitalopram treatment it was evident that, in contrast to exercise, escitalopram targets a different subset of proteins affected by maternal separation, except for a few involved in energy metabolism pathways and neuroprotection. In this study we have shown that chronic voluntary exercise has therapeutic effects in maternally separated rats, decreasing depressive-like behavior, increasing neurotrophin expression and restoring cytosolic protein expression that were dysregulated by early life stress. / AFRIKAANSE OPSOMMING: Negatiewe stresvolle ervarings gedurende die vroeë stadium van ‘n mens se lewe veroorsaak veranderinge in die ontwikkeling van breinstrukture en het ‘n nadelige uitwerking op die funksionering van die brein. Dit is bekend dat stresvolle ervarings in kinders, byvoorbeeld sosiale afsondering, verwaarlosing en mishandeling, die risiko vir die ontwikkeling van psigiatriese steurings soos depressie gedurende volwassenheid kan verhoog. In hierdie studie gebruik ons moederlike skeiding van neonatale rotte as ‘n model vir vroeë lewensstres om te bepaal hoe dit verskillende sisteme in die brein negatief beinvloed, en dan ook die effek van addisionele kroniese of akute stres gedurende volwassenheid. Die neonatale rotte is weggeneem van hulle moeders af vanaf dag 2 tot 14 vir 3 ure elke dag terwyl kontrole rotte by hulle moeders gebly het.
