Ce mémoire, consacré au développement de méthodes physico-chimiques pour le contrôle de la médication par l'Harpagophytum et l'Eleutherococcus, principes actifs utilisés en phytothérapie équine, est constitué de cinq chapitres. Le premier chapitre situe les enjeux des recherches entreprises ; il détaille le principe d'un contrôle de la méditation et le déroulement de celui-ci. Les propriétés, tant botaniques que pharmacologiques, des plantes étudiées, l'Harpagophytum procumbens et l'Eleutherococcus senticosus y sont également présentées. Dans le second chapitre, les différentes techniques expérimentales et les méthodes théoriques employées au cours de cette thèse sont détaillées. Cela concerne la préparation d'échantillon avec les différentes métho! des d'extraction, l'analyse par couplage chromatographie – spectrométrie de masse, et la modélisation moléculaire. Le troisième chapitre, dédié à l'étude de l'Harpagophytum, est présenté sur publications. L'extraction des principes actifs à partir des fluides biologiques équins, leur analyse par LC/MS, une approche théorique de la cationisation de l'harpagide par le sodium ainsi qu'une approche métabolique de l'harpagoside y sont développées. Une application directe de ces résultats à l'étude des éleutherocoques est présentée dans le chapitre 4, complétée par la recherche des principes actifs dans les compléments alimentaires. Le cinquième et dernier chapitre est un bilan des applications, des résultats et des perspectives qui découlent du travail effectué au cours de ces trois anné! es.

An investigation into the medicinal properties of Tulbaghia alliacea phytotherapy.

<p>The reproductive health of individuals is severely compromised by HIV infection, with candidiasis being the most prevalent oral complication in patients. Although not usually associated with severe morbidity, oropharyngeal candidiasis can be clinically significant, as it can interfere with the administration of medications and adequate nutritional intake, and may spread to the esophagus. Azole antifungal agents are commonly prescribed for the treatment and prophylaxis of candidal infections. However, the emergence of drug resistant strains and dose limiting toxic effects have complicated the treatment of candidiasis. Consequently, safe and effective and affordable medicine is required to combat this fungus. Commercial garlic (Allium sativum) has been used time since immemorial as a natural antibiotic, however very little is known about the antifungal properties of two indigenous South African species of garlic, namely Tulbaghia alliacea and Tulbaghia violacea, that are used as folk medicines for a variety of infections. This study compares the in vitro anti-candidal activity of Tulbaghia alliacea, Tulbaghia violacea and Allium sativum extracts. It was found that the greatest concentrations of inhibitory components were extracted by chloroform or water. The IC50 concentrations of Tulbaghia alliacea were between 0.007 &ndash / 0.038% (w/v). Assays using S. cerevisiae revealed that the T. alliacea extract was fungicidal, with a killing half-life of approximately 2 hours. This inhibitory effect of the T. alliacea extracts was observed via TLC, and may be due to an active compound called Marasmicin, that was identified using NMR. This investigation confirms that extracts of T.alliacea exhibit anti-infective activity against candida species in vitro.</p>

A double-blind homoeopathic drug proving of Curcuma longa 30CH with the subsequent comparison to the Ayurvedic and phytotherapeutic indications thereof

Mini-dissertation submitted in partial compliance with the requirements for the Master’s Degree in Technology: Homoeopathy, Durban University of Technology, 2011. / Introduction The purpose of this study was to determine the therapeutic potential of Curcuma longa 30CH when administered to healthy individuals, thus revealing the materia medica of the substance. It was also the aim of this study to compare the existing therapeutic indications of the substance to the proving symptomatology. Methodology The proving took the form of a double-blind placebo controlled study and was conducted by two Master’s in Technology: Homoeopathy students using 30 healthy subjects. Twenty four provers were given the active medication and six provers were given the placebo. The remedy was manufactured according to the German Homoeopathic Pharmacopoeia in 30CH potency. The proving ran for a period of six weeks. Results The symptoms extracted from the proving were placed in different sections according to the repertory and was compared to the Ayurvedic and Phytotherapeutic indications of Curcuma longa. There were 202 symptoms produced as a result of the remedy, 141 rubrics were formulated using these symptoms. The largest number of rubrics i ii was allocated to the mind, head and dreams section of the repertory, other smaller sections of prominence included the eye, ear, nose and throat sections. A wealth of information was gained once the comparison was made between Curcuma longa 30CH and the Phytotherapeutic and Ayurvedic indications of use. Similarities between the materia medica of Curcuma longa and the Phytotherapeutic indications of use were found to exist with respect to sections such as eye, nose, face, stomach, stool, respiration, back, extremities, skin and generals. Conclusion The administration of Curcuma longa 30C to healthy provers according to the methodological protocol of this study resulted in the production of a variety of defined proving symptoms which comprise the materia medica thereof (first objective of the study). The subsequent comparison of the proving symptoms with the existing indications of Turmeric as an Ayurvedic and Phytotherapeutic medicine (second objective of the study) revealed clear correlations in a variety of defined areas.

L'objectif du travail effectué était la recherche de nouvelles substances actives d'origine végétale, présentant soit une activité antiplasmodiale soit une activité antifongique. Cette étude a été menée suivant deux stratégies différentes: l'étude de remèdes traditionnels antipaludiques identifiés suite à des enquêtes ethnopharmacologiques, et la mise en évidence des propriétés antifongiques d'huiles essentielles grâce à une stratégie bioinspirée. La première partie du travail a permis de mettre en évidence le rôle d'un quassinoïde connu, la simalikalactone D, dans l'activité antipaludique d'une tisane de jeunes feuilles fraîches de Quassia amara L. (Simaroubaceae). Dans le cas de la décoction de rameaux de Psidium acutangulum DC. (Myrtaceae), c'est cette fois un mélange de flavonoïdes glycosylés qui est responsable de l'activité du remède. Dans le cadre de la recherche de nouvelles substances antifongiques, le criblage effectué a permis d'identifier de nombreuses huiles essentielles présentant des activités intéressantes, validant ainsi la démarche bioinspirée retenue dans ce cas. L'huile essentielle d'Otacanthus azureus (Linden) Ronse a en particulier démontré une activité remarquable, à la fois seule et en combinaison avec des antifongiques azolés. Enfin, l'étude métabolomique de la composition des huiles essentielles a permis de mettre au point un outil pouvant orienter la sélection des huiles en fonction des données obtenues en GC/MS dans l'optique de la recherche de nouvelles substances antifongiques. Ce travail démontre donc la validité des stratégies retenues - ethnopharmacologie et bioinspiration - dans la recherche de nouvelles substances bioactives.

<p>In South Africa, the crude aqueous extract from Hypoxis hemerocallidea is used by AIDS patients to treat opportunistic infections, such as tuberculosis. The rapid emergence of multidrug-resistant tuberculosis, and extreme drug resistant tuberculosis, in recent years, is a major threat to human health. The treatment of TB, nosocomial bacterial infections, and fungal infections is now a clinical challenge, especially in the immuno-compromised individual. There is a dire need for novel antibiotic alternatives with phytotherapies and plant-derived compounds as potentially promising alternatives. The main objective of this study was to investigate the antimycobacterial activity of Hypoxis hemerocallidea, a South African medicinal plant, using Mycobacterium smegmatis.</p>
