Pharmacokinetics and physiologic effects of xylazine-ketamine-butorphanol administered intramuscularly or in combination with orally administered sodium salicylate on biomarkers of pain in Holstein calves following concurrent castration and dehorning

Coetzee / Objective—To determine the pharmacokinetic parameters of xylazine, ketamine, and butorphanol administered IM and sodium salicylate administered PO and to compare their effect on biomarkers of pain and distress following sham (Period 1) and actual (Period 2) castration and dehorning. Animals—40 Holstein bull calves Procedures—Calves weighing 108 to 235 kg received the following treatments prior to sham castration and dehorning (Period 1) and castration and dehorning (Period 2) (n=10 calves/group): (i) 0.9% saline solution IM (PLACEBO) (ii) sodium salicylate (SAL) supplied free-choice in water to provide concentrations from 2.5 to 5 mg/mL beginning 24 hours prior to Period 1 to 48 hours after Period 2; (iii) 0.025 mg/kg butorphanol, 0.05 mg/kg xylazine, 0.1 mg/kg ketamine co-administered IM immediately prior to both periods (XKB); and (iv) a combination of treatments (ii) and (iii) (SAL + XKB). Plasma drug concentrations, average daily gain (ADG), chute exit speed, serum cortisol concentrations and electrodermal activity (EDA) were evaluated. Results—ADG (0-13d) was significantly greater in the SAL and SAL + XKB groups. Calves receiving XKB had significantly slower chute exit speed in both periods. Serum cortisol concentrations were significantly increased in all groups during Period 2 compared to Period 1. However, XKB attenuated serum cortisol response for the first hour after castration and dehorning while oral salicylate significantly reduced cortisol from 1-6 hours. XKB administration significantly decreased EDA scores in both periods. Conclusions and Clinical Relevance—Free-choice sodium salicylate decreases cortisol concentrations and reduced weight loss associated with castration and dehorning in calves.

Relationship between castration and morbidity and their effects on performance and carcass quality

Most would agree that a discount is warranted but determining the appropriate discount to apply is considerably more difficult. Being able to calculate this discount under varying conditions would help stock operators maintain a certain level of profitability or recognize opportunities to make more profit when excessive discounts are being applied. The goals of this study were to determine how castration timing affects performance (as measured by average daily gain), morbidity, and carcass quality and how morbidity affects performance and carcass quality. Ordinary Least Squares regression and logit models were estimated to quantify the effects of various management and environmental factors on performance, morbidity, and carcass quality. These model estimates of production variables along with price and cost assumptions were used to calculate breakeven purchase prices and price discounts for bulls relative to steers, accounting for the possibility of contracting bovine respiratory disease, if owned for a short background period or if ownership is retained through slaughter. Model results confirm that late-castrated steers do indeed exhibit diminished performance and increased morbidity probabilities relative to early-castrated steers. Increased morbidity also decreases average daily gain. However, this study found that castration timing and morbidity during the backgrounding period have minimal effects on carcass quality, with morbidity only impacting hot carcass weight and castration timing significantly affecting days to market and only tending to impact hot carcass weight. Ultimately, based on 2009 market conditions, bulls should be discounted at feeder calf sales compared to steers. The average calf arrived at 459 pounds, and at this weight bulls should be discounted $4.69/cwt relative to the same weight steers. The discount increases to $5.37/cwt for 400 pound calves and drops to $4.20/cwt for 500 pound calves. If ownership is retained through slaughter, required discounts will change to $6.77/cwt, $4.91/cwt, and $7.55/cwt, respectively.

Effect of age and castration method on neurohormonal, and electroencephalographic stress indicators in Holstein calves

The aims of this study were (i) to evaluate the physiological response to pain induced by castration in calves and (ii) to elucidate age-related differences in pain response of calves subjected to different castration methods. Seventy six Holstein bull calves were blocked by age (≤ 6 weeks and ≥ 6 months) and randomly assigned to one of four treatment groups: control (n = 20), castration by banding (n = 18), cut and clamp surgical castration (n = 20), and cut and pull surgical castration (n = 18). Measurements included electroencephalogram, heart rate variability, infrared thermography, electrodermal activity, and concentrations of serum cortisol, and plasma substance P prior to, during, and following castration. Electroencephalogram recordings showed desynchronization for all treatments, consistent with increased arousal; yet the magnitude of desynchronization was greatest for 6-month-old calves castrated by cut and clamp. Additionally, older calves in the cut and pull group showed greater desynchronization than younger calves in the same group. Based on the heart rate variability analysis, 6-month-old calves in the control or cut and pull castration groups showed greater sympathetic tone than younger calves in the same treatment groups. Overall, younger calves showed lower electrodermal activity than older calves. Regardless of treatment, concentrations of cortisol and plasma substance P were greater in 6-month-old calves relative to their younger counterparts. In summary, neurohormonal and electroencephalographic stress responses of calves to castration were age-specific. Castration by cut and clamp showed the most pronounced stress response in 6-month-old calves.

Identifying and alleviating pain associated with routine husbandry procedures performed on pre and post weaning dairy calves

Edwards-Callaway / This thesis includes two studies that assessed pain responses to husbandry procedures in order develop mitigation tools. The objective of the first trial was to identify method-related differences in behavioral pain responses in calves of two ages (6 week and 6 months) subjected to castration: surgical cut (CP; n=18), surgical cut and emasculator (CC; n=20), rubber banding (BAND; n=18), or control manipulation of the scrotum (CONT; n=20). Behavior was evaluated pre- and post-castration to record foot stamps, tail flicks, kicks, elimination, in addition to standing and lying post-castration. For 6 week calves, the probability of kicking and eliminations was greater for surgical castrates and all castrated animals, respectively. The probability of kicking was greatest for all 6 month castrates while BAND and CONT had greater probability of elimination. Both age groups increased standing post-castration. Lying decreased in 6 week calves but was unchanged for 6 month calves. Six week calves displayed more tail flicks and tended to display less foot stamps than 6 month calves. Six week calves showed a decrease in tail flicks and foot stamps in response to castration while 6 month calves showed an increase in both behaviors. CP and CC, but not BAND, resulted in less tail flicks than CONT post castration. The second trial compared the effects of preemptive analgesics administered to calves subjected to dehorning with local anesthesia. Six month Holstein steers were randomly assigned to receive one of the following treatments (n= 8/group): meloxicam (1 mg/kg PO), gabapentin (15mg/kg PO), meloxicam (1 mg/kg) and gabapentin (15 mg/kg) PO, flunixin (2.2 mg/kg IV), or a placebo. Drug, cortisol, ex-vivo prostaglandin, haptoglobin, and substance P concentrations, ocular thermography, algometry, and average daily gain were evaluated. Analgesic-treated calves had lower plasma SP concentrations and improved ADG compared with controls. Flunixin calves had reduced cortisol and ex-vivo prostaglandin concentrations for 24h compared to controls. Meloxicam treated calves showed an increase in MNT at two horn bud sites compared with the other treatments. Overall, the results provide validation of responses to noxious stimuli that can be used to develop pain alleviation for livestock.

Orientador: Celso Antônio Rodrigues / Resumo: O comportamento das mulas, durante o estro, influencia negativamente sua função zootécnica. A ovariectomia tem se apresentado como forma de torná-las mais cooperativas. Procedimentos realizados por videocirurgia se apresentam com uma possibilidade para reduzir as complicações trans e pós-operatórias. Então, foram utilizadas 9 mulas, com idade de 7,9 ± 3,2 anos, pesando 352 ± 76 kg. Realizou-se estudo do comportamento reprodutivo dos animais durante a rufiação com garanhão equino, nas diferentes fases do ciclo estral. A duração do estro foi de 6,4 ± 1,4 dias. As ovulações ocorreram 5,3 ± 2,6 dias após o início dos sinais de estro. Os sinais que apresentaram maior correlação foram micção frequente (MF) e levantamento da cauda (LC) (0,79) e imobilidade (IM) e LC (0,76) enquanto que os que apresentaram menor correlação foram aproximação do macho (AM) e LC (0,26) e AM e MF (0,29). Posteriormente foi realizada a ovariectomia por videolaparoscopia e no pós-operatório foi realizado o acompanhamento clínico-laboratorial para caracterizar possíveis complicações da técnica. Para hemostasia dos vasos presentes no pedículo ovariano utilizou-se no grupo A um bisturi ultrassônico e no grupo B um aplicador de clips de laqueação de titânio. Observou-se que o grau de dificuldade da ovariectomia foi inversamente proporcional ao tamanho do ovário. Ovariectomy is an interesting way to make these animals more cooperative. Videosurgeric procedures has become a possibility to reduce operative and postoperative complications. Nine mules underwent the procedure and were followed from January to May 2017. The average age of the animals was 7,9 ± 3,2 years old and the weight approximately 352 ± 76 kg. A prospective evaluation of reproductive behavior was set during the ruffling of the stallion equine, along different phases of the estrus. Estrus length was 6,4 ± 1,4 days. Ovulation occurred 5.3 ± 2.6 days after the onset of estrus signs. The signs that presented the highest correlation were frequent micturition (MF) with tail lifting (LC) (0,79) and immobility with just LC (0,76). The least correlations were male approximation (AM) with LC and AM with MF. Subsequently, video-laparoscopic ovariectomy was performed. In post-operative days, clinical follow up and laboratory assay were performed to detect possible complications. Two groups were randomly selected for ultrasonic scalpel (group A) and titanium clips (group B) aiming the hemostasis of the vascular pedicle on the ovaries. Estudo do programa de esterilização das populações canina e felina no Município de São Paulo, período 2001 a 2003 / A study of the sterilization programme of canine and feline populations in São Paulo, from 2001 to 2003

Foi realizada uma avaliação do Programa Saúde do Animal , com relação as esterilizações das populações canina e felina no Município de São Paulo no período de 2001 a 2003, na qual houve a integração de diversas entidades públicas e privadas. Utilizaram-se dados do Centro de Controle de Zoonoses do Município de São Paulo, com um total de 14.703 animais esterilizados. Os resultados foram quantificados e mapeados segundo sexo, espécie e faixas etárias. Também foram identificadas alternativas de controle populacional com seus respectivos custos. Foi observado que a grande parte dos animais esterilizados são cadelas adultas. A idade média de esterilizacão de 7 ± 3,6 anos, de cães foi de 6,8± 3,7 anos; de gatas foi de 5,9 ±3,4 anos, e de gatos foi de 5,8 ± 3,2 anos que existe uma maior atenção às zonas centro e sul do Município e há necessidade de implantar um adequado direcionamento dos animais atendidos, assim como estimular os proprietários a esterilizar animais antes dos dois anos de idade, fornecendo maior informação das técnicas existentes e seus benefícios, e fazendo com que a sociedade participe de forma ativa, garantindo o cumprimento dos objetivos do programa. / It was made an Animal Health Programme avaliation about cats and dogs sterilizations between 2001 and 2003 in São Paulo, Brazil. This work was made with collaboration of many public and private centers. Data of 14.703 sterilised animals from the Zonoses Cont rol Center in São Paulo were used. The results were quantified and maped consedring sex, species and ages. It was also identified alternatives of populationl control and their costs. Most part of these animals are female and adults. The male and female dogs had respectively 7,0±3,6 years old and 6,8 ±3,7 years old. And the male and female cats had 5,9±3,4 years old an 5,8 ± 3,2 years old., respectively. The sterilization control programme is more utilized in Center and South part of São Paulo. It`s necessary to implement an sterilization programme that enable an more adequate choice of the method to be used . It´s necessary to stimulate the animals owners to sterilize their cats and dogs before they complete 2 years old and to give more informations about the possible techiniques to be used and their benefits. The society must participate of this program to ensure the programnce objective performance.

A privação dos hormônios sexuais em fêmeas contribui para o aparecimento de distúrbios metabólicos e endócrinos devido à ausência de estrógenos. Nesse estudo foi avaliado o efeito da castração e da suplementação com DHEA sobre aspectos metabólicos em ratas. A ovariectomia aumentou a adiposidade e o consumo de ração, diminuiu o consumo hídrico, além de promover resistência à insulina e intolerância à glicose, e o uso do DHEA reduziu a sensibilidade à insulina. Ainda, a castração levou a redução nos níveis plasmáticos de DHEA, estradiol, VLDL-c e triacilglicerol, albumina, uréia e creatinina, e o tratamento com DHEA restaurou parcialmente a concentração de estradiol no grupo OVX+DHEA. E por fim, a ooforectomia reduziu a expressão proteica do IRb, IRS1, PI3K e AKT no fígado, músculo gastrocnêmio ou coração, além diminuir o grau de fosforilação em tirosina da pp185 (IRS1/2). Deste modo, concluímos que a castração promoveu resistência à insulina e intolerância à glicose associada à obesidade, e neste modelo, o uso do DHEA piorou a sensibilidade à insulina nos animais. / Deprivation of sex hormones in females contributes to the onset of metabolic and endocrine disorders due loss of estrogen. In this study the effect of castration and supplementation with DHEA on metabolic aspects in rats was evaluated. Ovariectomy increased adiposity and food consumption, decreased water consumption, besides promoting insulin resistance and glucose intolerance, and the use of DHEA reduced insulin sensitivity. Furthermore, castration led to decreasing of DHEA plasma levels, estradiol, VLDL-c and triacylglycerol, albumin, urea and creatinine, and DHEA treatment partially restored the concentration of estradiol in OVX+DHEA group. Finally, ovariectomy reduced the protein expression IRb, IRS1, PI3K and AKT in the liver, gastrocnemius muscle or heart. Indeed, decrease tyrosine phosphorylation status of pp185 (IRS1/2) was observed. Thus, we conclude that castration promoted insulin resistance and glucose intolerance associated with obesity, and in this model, the use of DHEA impairs insulin sensitivity in animals.

Dentre os potenciais alvos utilizados em vacinas contraceptivas, o hormônio liberador de gonadotrofinas (GnRH) é considerado um dos mais atrativos. O GnRH é um decapeptídeo hipotalâmico que apresenta um papel central na reprodução de mamíferos. Entretanto, devido à sua baixa imunogenicidade, é necessário associar o GnRH com uma molécula carreadora capaz de estimular o sistema imune como, por exemplo, a subunidade B da enterotoxina termolábil de Escherichia coli (LTB). O presente estudo visou avaliar o potencial imunocontraceptivo de duas quimeras LTB/GnRH em camundongos machos da linhagem BALB/c. Os camundongos foram divididos aleatoriamente em oito grupos experimentais. O grupo LTB/GnRH-P recebeu a quimera expressa em P. pastoris. O grupo LTB/GnRH-P/A recebeu a quimera expressa em P. pastoris adsorvida em adjuvante oleoso. O grupo LTB/GnRH-E recebeu a quimera expressa em E. coli. O grupo LTB/GnRH-E/A recebeu a quimera expressa em E. coli adsorvida em adjuvante oleoso. O grupo LTB recebeu a proteína LTB expressa em E. coli. Os grupos Controle e Controle/A não receberam nenhum antígeno vacinal. O grupo Vacina comercial recebeu vacina comercial anti-GnRH utilizada em bovinos. A resposta imune humoral foi avaliada pela técnica de ELISA indireto e o potencial imunocontraceptivo foi avaliado por meio da quantificação de testosterona sérica e das análises histológicas das gônadas dos animais. Os grupos que receberam os antígenos LTB/GnRH (25 μg por vacina) apresentaram níveis de IgG total significativamente superiores quando comparados aos grupos controle, mostrando que ambos os antígenos são imunogênicos. Os grupos LTB/GnRH-P e LTB/GnRH-P/A apresentaram as maiores absorbâncias nos dias 28 (1,11 ± 0,09) e 42 (1,44 ± 0,04), respectivamente. Além disso, o grupo LTB/GnRH-P/A apresentou níveis de testosterona significativamente menores ao controle a partir do dia 28 (192 ng/dl ± 254,55). Os grupos LTB/GnRH-E e LTB/GnRH-E/A também obtiveram os maiores níveis de anticorpos nos dias 28 (0,33 ± 0,005) e 42 (1,44 ± 0,08), respectivamente. O grupo LTB/GnRH-E/A apresentou níveis de testosterona significativamente menores ao controle a partir do dia 28 (<12 ng/dl ± 0). Os resultados da histologia mostraram que ambos os antígenos causam alterações na espermatogênese, sendo que o antígeno expresso em E. coli foi relacionado com as maiores alterações nas gônadas. O presente estudo mostrou quimeras produzidas através da fusão da LTB com uma única molécula de GnRH é capaz de induzir a geração de resposta imune humoral, bloqueio das funções endócrinas relacionadas com a reprodução e alterações teciduais nas gônadas de camundongos machos. / Immunocontraception is recognized as one of the major new contraceptive methods for the control and management of fertility in different animal species. Among the potential targets used in contraceptive vaccines, gonadotropin-releasing hormone (GnRH) is considered one of the most attractive. GnRH is a hypothalamic decapeptide that presents a central role in mammalian reproduction. However, due to its low immunogenicity, it is necessary to associate the GnRH with a carrier molecule capable of stimulating the immune system such as, for example, the B subunit of Escherichia coli heat-labile enterotoxin (LTB). The present study aimed to evaluate the immunocontraceptive potential of two LTB/GnRH chimeras in male BALB/c mice. Mice were randomly divided into eight experimental groups. The LTB/GnRH-P group received the chimera expressed in P. pastoris. The LTB/GnRH-P/A group received the chimera expressed in P. pastoris adsorbed in oil adjuvant. The LTB/GnRH-E group received the chimera expressed in E. coli. The LTB/GnRH-E/A group received the chimera expressed in E. coli adsorbed in oil adjuvant. The Control and Control/A groups did not receive vaccine antigen. The Commercial vaccine group received an anti-GnRH commercial vaccine used in cattle. The humoral immune response was evaluated by indirect ELISA and the immunocontraceptive potential was assessed through the quantification of serum testosterone and the histological analysis of animal’s gonads. The groups that received LTB/GnRH antigens (25 μg per vaccine) presented total IgG levels significantly higher when compared to the control groups, showing that both antigens are immunogenic. The LTB/GnRH-P and LTB/GnRH-P/A groups showed higher absorbances at days 28 (1,11 ± 0,09) and 42 (1,44 ± 0,04), respectively. Furthermore, the LTB/GnRH-P/A group presented testosterone levels significantly lower to the control from day 28 (192 ng/dl ± 254,55). The LTB/GnRH-E and LTB/GnRH-E/A groups also obtained the highest antibody levels at days 28 (0,33 ± 0,005) and 42 (1,44 ± 0,08), respectively. The LTB/GnRH-E/A group presented testosterone levels significantly lower to the control from day 28 (<12 ng/dl ± 0). The results from histology showed that both antigens cause changes in spermatogenesis, wherein the antigen expressed in E. coli was associated with major changes in gonads. The present study showed that chimeras produced through the merge of LTB with a single GnRH molecule is capable to induce the generation of humoral immune response, block of the endocrine functions related to reproduction, and tissue alterations in the gonads of male mice.

Cony, famoso autor e jornalista, com histórico de prisões políticas na época da ditadura, publicou Pilatos obra de linguagem considerada pornográfica ainda na vigência da censura. Rubem Fonseca, perseguido pelos censores desde Feliz ano novo (1975), devido às suas narrativas também consideradas contrárias à moral e aos bons costumes, publicou Secreções, excreções e desatinos livre de censura. Este estudo, a partir das análises dos textos referidos, questiona o que é o crime da literatura? E, se não há mais a conjunção de censura política e repressão moral, se está esgotado o embate entre argumentos moralistas e liberais, como podem ser lidos os novos códigos de controle desse imaginário? O objetivo desta tese é, sobretudo, discutir a possibilidade da permanência ou não destes livros como transgressores em relação ao tratamento dado ao corpo no panorama contemporâneo. Para tanto, serão analisadas as diferentes formas de representação do corpo nesses livros, a partir de conceitos, como castração, pornografia e obscenidade, investigados em campos de estudo sobre as representações do corpo nas áreas de psicologia, sociologia e da crítica literária / This paper investigates what we call of crimes of Literature in Carlos Heitor Conys book, Pilatos (1974) in a comparative perspective with the story telling book Secreções, excreções e desatinos (2001), written by Rubem Fonseca. Cony, the famous author and journalist, with a history of political prisons during the Brazilian dictatorial period, has published Pilatos a work of language that was considered pornographic even in the presence of censorship. Rubem Fonseca was perused by the censors since the release of Feliz Ano Novo (1975), narrative also considered contradictory to the moral and good manners, he published Secreções, excreções e desatinos, free from the censorship. From these analyses, this paper questions what is the crime of literature? And, if there are no more links between political censorship and moral repressions, if the arguments between the moralistic and the liberals are vanishing, how the new codes of the imaginary control can be read?The objective of this thesis is to mainly discuss the possibility of the permanence or not, of these books like transgressors, related to the treatment given of the body in a contemporary perspective. Therefore this paper will analyze the different forms of body representations on these books, considering concepts like castration, 'pornography', 'obscenity', investigated in study fields about body representations in the psychology, sociology and literary criticism area
