Meiosis is a specialized form of cell division in which haploid gametes are produced from diploid progenitors. This reduction in ploidy results from proper meiotic chromosome segregation and is ensured by crossover recombination events. Given their importance, it is no surprise that crossover formation is regulated in most eukaryotes. Crossover assurance is a regulatory mechanism that helps to ensure that each pair of chromosomes gets at least one crossover during meiosis.
We seek to better understand how crossover assurance works. To do so, we have developed a system in which crossover formation between a pair of chromosomes is restricted to a defined region. If crossover assurance functions in this context, then crossovers should frequently form in this defined region.
Our experiments involve three yeast strains: Homolog: diploid Saccharomyces cerevisiae. Homeolog: Diploid S. cerevisiae, but with one copy of III derived from S. paradoxus and one from S. cerevisiae. Homo-meolog: The homeolog strain, but with the HIS4 region of the S. paradoxus III replaced with the corresponding S. cerevisiae sequence.
S. cerevisiae and S. paradoxus are largely syntenic and have 80-90% sequence homology. This level of sequence divergence greatly reduces the incidence of meiotic crossing over. Thus, in the Homeolog strain chromosomes III will frequently fail to form crossovers. In the Homo-meolog strain, a defined region of homology surrounding HIS4 (a hotspot for meiotic recombination) exists in a chromosomal context of homeology. In the Homo-meolog strain, crossover assurance should result in a high incidence of crossover formation in the HIS4 region. By comparing the spectrum of meiotic recombination events in the HIS4 region in the three strains, we will gain insight into the means through which crossover assurance is enforced.
These experiments are in the preliminary stage. Strain construction and data collection are ongoing, but our preliminary results demonstrate an elevated incidence of crossing over in the HIS4 region in the homo-meolog strain relative to both the homolog and homeolog strains. Spore viability patterns in the homo-meolog strain are not statistically distinguishable from that of the homolog strain, but are different from that of the homeolog strain. Taken together, these results suggest that the crossovers are targeted to the HIS4 region in the homo-meolog strain, possible through the action of a crossover assurance mechanism. Further analysis of the patterns of recombination in these strains may provide insight into the means through which this regulation is exerted.

In this work, the Traveling Salesman Problem (TSP) is discussed. The Hybrid Genetic Algorithm for solving the TSP is presented. The traveling salesman problem is formulated as follows: given matrix D=(dij)nxn of distances between n objects and the set P of permutations of the integers from 1 to n, find a permutation p=(p(1), p(2), ..., p(n)) P that minimizes. Many heuristic algorithms can be applied for the TSP. Recently, genetic algorithms (GAs) are among the advanced heuristic techniques for the combinatorial problems, like the TSP. genetic algorithms are based on the biological process of natural selection. The original concepts of GAs were developed in 1970s. Many simulations have demonstrated the efficiency of GAs on different optimization problems, among them, bin–packing, generalized assignment problem, graph partitioning, job–shop scheduling problem, set covering problem, vehicle routing. One of the main operators in GAs is the crossover (i.e. solution recombination). This operator plays a very important role by constructing competitive GAs. In this work, we investigate several crossover operators for the TSP, among them, CX (cycle crossover), PMX (partialy mapped crossover), POS (position based crossover), ER (edge recombination crossover), edge-NN (edge recombination crossover, nearest neighbour) and AP (alternating-positions crossover). Comparison of these crossover operators was performed. The results show high efficiency of the edge-NN, ER and PMX crossovers.

Dans la plupart des organismes, les crossovers se formant au cours de la méiose sont interférérents : deux crossovers sont rarement à proximité. Nous avons étudié ce phénomène en détail dans la plante modèle Arabidopsis thaliana en utilisant une grande population de rétro-croisements, en méiose mâle et en méiose femelle. Nous avons utilisé le modèle Gamma à deux voies, superposant à la voie interférente une proportion p de crossovers d'une deuxième voie non interférente. La méiose femelle montre une interférence plus élevée et une proportion p plus faible que la méiose mâle. Par ailleurs nos comparaisons intra-chromosomiques concluent qu'il existe des variations d'interférence entre le bras gauche et le bras droit d'une part et entre la partie centrale et les régions distales d'autre part. Nous avons ensuite développé des tests statistiques qui ont révélé des hétérogénéités dans la proportion p le long des chromosomes. De telles variations ont ensuite été trouvées dans un jeu de données de tomate où notre analyse statistique nous a permis de montrer pour la première fois sur un organisme «sauvage» que la deuxième voie était très peu interférente et qu'en fait les deux voies avaient un peu de « cross-talk ». Vu que la limitation la plus sévère des modèles utilisés jusqu'à présent est l'hypothèse de constance du paramètre p, nous avons développé un modèle généralisé où les deux voies peuvent avoir des paysages de recombinaison différents. Ce modèle nommé PSL-GS a été implémenté en logiciel et testé sur des données simulées et sur trois jeux de données de plantes. L'utilisation du critère BIC suggère que tout comme chez la tomate, les paysages de recombinaison d'Arabidopsis et du maïs sont différents entre les 2 voies. / For most organisms, crossovers forming during meiosis exhibit crossover interference – nearby crossovers are rare. This phenomenon was studied in great detail in Arabidopsis thaliana using a large backcross population for male and female meiosis. We used the gamma two-pathway model by superposing proportion (1- p) of interfering crossovers with p of non-interfering crossovers. It was observed that female meiosis shows higher interference but lower proportion of non-interfering crossovers than male meiosis. Further intra-chromosomal interference comparisons conclude that there are variations between (a) left and right arms, and (b) the central versus distal regions of a chromosome. Then statistical tests revealed heterogeneities in the non-interfering crossover proportion along chromosomes. Thereafter various statistical tools developed to examine a very novel wild-type tomato data annotating crossover positions from both pathways provided evidence for ‘cross-talk’ between the two pathways of crossover formation as opposed to being independent. Finally a model named Pathway-Specific Landscapes Gamma-Sprinkling (PSL-GS) incorporating chromosomal non-uniformity in the individual pathways has been proposed to extend present state-of-the-art interference modeling which consider both pathways to be uniform along chromosomes.

Caractérisation d’un point chaud de recombinaison méiotique chez Arabidopsis thaliana / Characterization of a meiotic recombination hotspot in Arabidopsis thalianaKhademian, Hossein 13 March 2012 (has links)
La recombinaison méiotique initiée en prophase I de méiose génère soit des crossing-over (COs), qui sont des échanges réciproques entre segments chromosomiques, ou des conversions géniques non associées aux COs (NCOs). Les deux types d'événements se produisent dans de petites régions (moins de 10 kilobases) appelées points chauds, qui sont distribuées de manière non homogène le long des chromosomes. L'objectif de ma thèse était la caractérisation d'un point chaud de recombinaison méiotique (nommée 14a) chez Arabidopsis thaliana (i) dans différentes accessions (ii) dans le mutant msh4, un gène impliqué dans la formation des COs. Dans les deux hybrides ColxLer et ColxWs (i) 14a a un taux très élevé de COs 0,85% et 0,49%, respectivement (ii) Les COs sont regroupés dans deux petites régions de quelques kilobases, 14a1 et 14a2 avec une distribution de type gaussienne observée aux points chauds décrits dans d'autres espèces (iii) 14a1 est aussi un point chaud de NCO avec un taux aussi élevé que celui des COs (0,5%) dans ColxLer (iv) un biais de l'initiation de recombinaison a été trouvé dans 14a1 aussi bien pour les COs que les NCOs dans le fond génétique ColxLer.Une réduction de la fréquence de CO a été observée dans le mutant msh4 dans le fond génétique ColxLer à 14a1 et 14a2 (6,4% et 18,7% par rapport au sauvage). Cela confirme le rôle précédemment connu de la protéine MSH4 impliqué dans la formation de CO. La fréquence de NCO à 14a1 est similaire à celle observéedans le fond sauvage. Le rôle des H3K4 histones trimethyltransferase d’Arabidopsis dans la recombinaison méiotique (comme précédemment observé comme Set1 chez S. cerevisiae ou PRDM9 chez les mammifères) a également été étudiée. Aucun des dix gènes d’histones méthyltransférase étudié n'a montré de rôle dans la méiose. Cela pourrait être dû à (i) une forte redondance de la fonction entre les protéines (ii) une autre histone méthyltransférase en charge de l'étiquetage des points chauds de recombinaison méiotique (plus de 29 putatif histone méthyltransférase ont été identifiés dans le génome d'Arabidopsis!) (iii) contrairement à S. cerevisiae, les souris et l'homme, un autre mécanisme de contrôle épigénétique de la recombinaison méiotique. / Meiotic recombination initiated in prophase I of meiosis generates either crossovers (COs), which are reciprocal exchanges between chromosome segments, or gene conversion not associated to crossovers (NCOs). Both kinds of events occur in narrow regions (less than 10 kilobases) called hotspots, which are distributed non-homogenously along chromosomes. The aim of my PhD was the characterization of a hotspot of meiotic recombination (named 14a) in Arabidopsis thaliana (i) across different accessions (ii) in msh4 mutant, a gene involved in CO formation. In both ColxLer and ColxWs hybrids (i) 14a had a very high rate of COs 0.85% and 0.49%, respectively (ii) COs clustered in two small regions of a few kilobases, 14a1 and 14a2 with typical Gaussian curve distribution observed in other organisms (iii) 14a1 was also a hotspot of NCO with high rate (0.5%) in ColxLer (iv) a bias of recombination initiation at 14a1 CO and NCO hotspot was found in ColxLer. A reduction of CO frequency was observed in msh4 mutant in ColxLer background at 14a1 (6.4%) and 14a2 (18.7%) compared to wild type. This confirmed previously known role of MSH4 protein in CO formation. Frequency of NCO at 14a1 was similar to wild type. The role of putative Arabidopsis histone H3K4 trimethyltransferase in meiotic recombination as previously observed like Set1 in S.cerevisiae or PRDM9 in mammals (mice and human) was also studied. None of ten putative histone methyltransferase genes was involved in meiosis. This could be due to (i) a strong redundancy of function between gene products (ii) another histone methyltransferase in charge of labeling meiotic recombination hotspots (more than 29 putative histone methyltransferase have been identified in the Arabidopsis genome!) (iii) contrary to S. cerevisiae, mice and humans, another mechanism for epigenetic control of meiotic recombination

The starting point of this study is the crossover phenomenon and how the needs of adult readers of young adult fiction can be met in the library services. To investigate this, the main focus is on how public librarians perceive cross-reading adults, and in what ways literature promotion can facilitate or restrict adults' reading of young adult fiction. Six librarians were interviewed for this study, and the results were analyzed from a theoretical perspective that emphasizes literature promotion as an activity that can affect the perceptions, and actions of the recipients. The librarians express an open attitude to cross-reading in general, which creates favorable conditions for anyone who wishes to read across age boundaries. Furthermore, there are several situations where the librarians sometimes choose to recommend young adult literature to adults, or when fiction for adults and young adults is shelved together. However, the main part of the collection is still organized and shelved based on age categories. The study suggests that when literature is directed at a specific age group, there may be a restricting effect on those who do not fit into that age category, unlike in cases where literature is contextualized based on content. Literature may also be seen as more appropriate for certain age groups by its proximity or distance to other literature departments in the library. One conclusion drawn is that if the boundaries between different age groups are loosened, this may be advantageous for more than one group of library users.

An Exploration of the Promises and Limitations of Passive Direct Methanol Fuel CellsRosenthal, Neal Stephen 01 September 2011 (has links)
"While Direct Methanol Fuel Cells (DMFC) have a promising future as a long-lasting and environmentally friendly energy source, the use of balance of plant (BOP) equipment, such as pumps, fans, and compressors, create a complex system that can significantly reduce plant efficiency and increase cost. As an alternative, passive DMFCs have been designed and studied due to their ability to run under ambient conditions without any BOP equipment. However, before they become a feasible energy source, more must be understood about their promise and limitations. In this thesis, performance of a self-designed and constructed passive DMFC was investigated. In addition, an analytical mathematical model was developed in order to gain a better understanding of the limitations of the passive DMFC. The model was compared with literature's data to ensure reliability. Passive DMFCs, consisting of one to twelve Membrane Electrode Assemblies (MEAs) were designed, constructed and tested. The smaller scale fuel cell was optimized using different setups and elaborately tested using a variety of fuels, most notably methanol chafing gel, to determine an optimal performance curve. The larger fuel cells were further used to test for long-term performance and practical feasibility. The compact four-cell units could run for at least 24 hours and can provide performance akin to an AA battery. A larger 12-cell fuel cell was also designed and built to test feasibility as a convenient power supply for camping equipment and other portable electronics, and was tested with neat methanol and methanol gel. In all fuel cell prototypes, polarization plots were obtained, along with open circuit voltage (OCV) plots and long-term performance plots. While it is currently not possible to differentiate which methanol fuel source is the best option without a more thorough investigation, methanol gel has shown great potential as a readily available commercial fuel. The three largest restrictions in passive DMFC performance are 1) slow mass transfer of fuel to the anode, 2) slow kinetics of methanol and oxygen electrodes, and 3) methanol crossover. The developed model correctly predicts the effect of methanol crossover and the resulting crossover current on OCV as well as on performance of the fuel cell over the entire voltage-current range. Further, the model correctly predicts the effect of increasing methanol feed concentration on reduced OCV but increased limiting current density. The effect of the proton exchange membrane thickness is also well explained. Finally, the model describes the significant power losses from larger overpotentials, as well as crossover current, and the resulting significant heat generated and low efficiency. Overall, PDMFCs show great promise for potential application provided the cost can be reduced significantly."
