Adolescent stress and social experiences : developmental antecedents of adult behavioural responses to unfamiliar stimuli and the underlying neuroendocrine mechanisms

During adolescence, animals leave the natal home and interact with potentially threatening stimuli (i.e. stressors), e.g. unfamiliar environments and conspecifics. Adolescent stressors can result in fewer interactions with unfamiliar stimuli in adulthood, plausibly due to sustained effects of glucocorticoid exposure on stress physiology (e.g. glucocorticoid secretion and receptor expression). The current thesis tested the hypothesis that adolescent glucocorticoid exposure and social experiences act as stressors by quantifying the effects of the adolescent experiences on behavioural responses to unfamiliar stimuli and the underlying neuroendocrine mechanisms when in adulthood using two captive species, zebra finches and rats. In study one, adolescent zebra finches were dosed with the glucocorticoid corticosterone. In adulthood, birds dosed with corticosterone in early adolescence took longer to enter an unfamiliar environment when tested individually and had lower expression of the glucocorticoid receptor GR in the hippocampus and hypothalamus, brain regions that regulate stress responses. Glucocorticoids therefore appear to be an endocrine mechanism behind the long-term effects of adolescent stress. Subsequent studies explored whether higher social density and more unfamiliar social interactions during adolescence act as stressors. In study two, early adolescent zebra finches were housed in groups varying in conspecific number and density. In adulthood, females raised in larger groups secreted a higher stressor-induced corticosterone concentration and, if raised at lower density, spent more time in an unfamiliar environment when group housed. In study three, adolescent female rats were housed in familiar pairs or exposed to unfamiliar conspecifics. Unfamiliar adolescent interactions had no effects on responses to unfamiliar environments or stress physiology in adulthood, but heightened ultrasonic call rates. In this thesis, adolescent social experiences do not act like stressors, but modulate (especially female) social behaviour. Adolescent stressors and social experiences therefore have distinct effects on responses to unfamiliar stimuli and stress physiology that are maintained into adulthood.

The effect of nutritional Nucleotides and parenteral Glucocorticoids on improving immunoglobulin absorption and growth by neonate calves : reducing the carbon footprint of dairy calves

Antibiotics are routinely used in modern livestock production systems to treat and prevent disease as well as to enhance livestock production and/or outputs. This widespread use of antibiotics has led to a growing worldwide interest in antibioticfree animal production. The addition of feed supplements such as nucleotides to improve intestinal health as well as the early development of the immune system needs to be investigated as an environmentally-friendly nutritional management activity. In this study four (4) groups of newborn Holstein calves (n=24) were evaluated after being treated as follows: Group 1 (Negative control), Group 2 (Investigational Veterinary Product (IVP) 1 – Oral supplement containing nucleotides, vitamins, essential fatty acids, essential amino acids, pre-biotics and trace-minerals), Group 3 (IVP 2 – Oral supplement containing nucleotides only at concentration and dosage equal to IVP1) and Group 4 (IVP 3 - Parenteral glucocorticoids). Results of the study indicated that: Pre-colostral Serum IgG titres – After titration of the serum the anti-BVDV results showed no difference between the study groups. All calves recorded a SP ratio of zero value (0.00 ± 0.2) indicating that they had not been nursed by their mothers prior to the start of the study. This was an entrance requirement for the calves to be enrolled into the study. Apparent Efficiency of Absorption % (AEA%) – Literature shows that an AEA% result of between 20%-30% is good and 35% is excellent. The AEA% recorded for this study fell within this range although there was no statistically significant difference shown between the groups. In spite of a strictly controlled study protocol, 7 of the 24 calves (29%) still suffered from FPT in this study. The result falls within the range of 19% - 37% reported on United States of America (USA) farms by Doepel and Bartier in 2014. Serum cortisol - When compared to the control group, there were no statistical significant difference evident for group 2 and group 3. However, the difference between the Glucocorticoid – Group 4 and the control and other groups was statistically significant (P = 0.0001; table 12) at the various time intervals. It was also evident that the inclusion of parenteral glucocorticoids (group 4) may have assisted in prolonging the natural “gut closure”. Gut closure – a positive 2 point linear regression forecast line indicated an increasing trend in IgG absorption post-24 hours for group number 4 whilst groups 1, 2 and 3 each had a negative 2 point linear regression forecast line (figure 23). Weight / ADG (D42) - Group 2 showed a statistically significant increase in mean D42 weight (P = 0.0042) of 59.167 kg ± 3.545 kg when compared to the other study groups and compared to the control group (P = 0.0227). A comparison of the relative increase in mass of groups indicated that Group 2 efficiently achieved this result with a statistically significant ADG of 0.536 kg (P = 0.014) compared to the other study groups and compared to the control group (P = 0.022). In addition, weight / FCR – FCR was calculated for all groups as follows: G1 – 4.000 kg, G2 - 2.593 kg, G3 – 2.703 kg and G4 - 3.012 kg feed required for the production of 1 kg meat. The results indicated that the group 2 feed input was the most economical followed by groups 3, 4 and control respectively. In conclusion, it is evident that a nutritional supplement containing nucleotides, vitamins, trace- minerals, essential fatty acids, essential amino acids and pre-biotics may contribute significantly to important economic indicators such as weight gain, ADG and FCR on a commercial dairy farm. / Environmental Sciences / Ph. Efeito da dexametasona na proteômica do fluido endometrial de éguas suscetíveis a endometrite / Effect of dexamethasone on proteomics of endometrial fluid from mares susceptible to endometritis

A corticoterapia tem sido utilizada frequentemente nas éguas suscetíveis. O uso de isuflupredona melhora a taxa de prenhez e altera o perfil proteico do líquido endometrial em relação a éguas não tratadas. A utilização de dexametasona diminui o acúmulo de líquido pós-cobertura, reduz o edema do útero, porém, desconhecem-se seus efeitos no perfil proteico do líquido endometrial. O objetivo do presente estudo foi analisar o efeito da dexametasona em éguas suscetíveis à endometrite persistente póscobertura sobre perfil proteico do líquido endometrial na presença ou ausência de infecção. Nove éguas suscetíveis foram utilizados, com idade entre 7 a 30 anos. Após a verificação dos sinais de estro as éguas foram submetidas a quatro tratamentos: (C) éguas não receberam nenhum tipo de tratamento e serviram como controle; (D) éguas receberam 40mg de dexametasona (IV), no momento da cobertura, com coleta da amostras após 6 horas, (I-6 e I-24) infusão intra-uterina de 1 x 109 de S. zooepidemicus/mL, com coleta da amostra após 6 e 24 horas; (I/D-6 e I/D-24) infusão intra-uterina de 1 x 109 S. zooepidemicus/mL e administração de 40mg de dexametasona (IV), com coleta da amostra após 6 e 24 horas. Todas as éguas foram submetidas a todos os tratamentos. As amostras foram coletadas e submetidas à eletroforese bidimensional para separação proteica e espectrometria de massa para a identificação das bandas proteicas relevantes. A corticoterapia provocou alteração na proteômica do líquido endometrial de éguas suscetíveis, caracterizada pelo aumento (TTR) e/ou diminuição (ApoA1) na densidade óptica de proteínas da fase aguda da inflamação. Conclui-se que a utilização da dexametasona em éguas com e sem presença de infecção altera a proteômica do fluido endometrial de éguas suscetíveis. Sugere-se que a dosagem ou a frequência de aplicação da dexametasona deva ser aumentada. / Corticotherapy has often been used in susceptible mares. The use of isuflupredona improves pregnancy rate and alters the protein profile of the endometrial fluid in relation to untreated mares. The use of dexamethasone decreases the post breeding fluid accumulation, reduces the uterine edema, however is unaware of its effects on the protein profile of endometrial fluid. The aim of the present study was analyze the effect of dexamethasone in mares susceptible to post-breeding persistent endometritis on the protein profile of endometrial fluid in the presence or absence of infection. Nine susceptible mares were used, aged 7-30 years old. After checking the signs of estrus, mares were subjected to four treatments: (C) mares received no treatment and served as controls; (D) mares received 40 mg of dexamethasone at breeding time, with collection of samples after 6 hours; (I-6 and I-24) intrauterine infusion of 1 x 109 S. zooepidemicus/ml and the sample was collected after 6 and 24 hours; (I/D-6 and I/D-24) intrauterine infusion of 1 x 109 S. zooepidemicus/ml and 40 mg of dexamethasone administration, collecting the sample after 6 and 24 hours. All of the mares were subjected to all treatments. Samples were collected and subjected to two-dimensional electrophoresis for protein separation and mass spectrometry for the identification of relevant protein bands. Corticotherapy resulted in alteration of the protein profile of the endometrial fluid of susceptible mares, characterized by an increase (TTR) and/or decrease (ApoA1) in optical density of the acute phase of proteins of inflammation. We conclude that the use of dexamethasone in mares with and without the presence of infection alters the protein profile of endometrial fluid of susceptible mares. It is suggested that the dosage or frequency of application of dexamethasone should be increased.

Com o intuito de avaliar os potenciais efeitos fisiológicos de radiotransmissores do tipo mochila em falcões quiri-quiri (Falco sparverius) de vida livre, oito machos desta espécie foram capturados e divididos em dois grupos: indivíduos marcados com radiotransmissores (aproximadamente 5% do peso vivo, n = 6) e indivíduos controle (anilhados, n = 2). Em seguida, amostras fecais foram coletadas duas horas após a captura (dia −1) e durante os dias 0 (definido como o dia de soltura dos animais), 4, 7, 15, 30, 40 e 55. Previamente às análises laboratoriais, o ensaio imunoenzimático para dosagem de glucocorticóides fecais foi testado empregando procedimentos padrões de validação hormonal (paralelismo, curva de dose-resposta, validação fisiológica e coeficientes de variação inter- e intra-ensaio). Em relação aos machos monitorados, ambos os grupos apresentaram um aumento significativo nos níveis de glucocorticóides fecais durante o dia 0 (P < 0,001), mas as concentrações retornaram aos níveis pré-equipagem após 4 dias da soltura. Além disso, os níveis de glucocorticóides fecais não diferiram significativamente entre os falcões radiomarcados e os anilhados durante os 55 dias de monitoramento (P > 0,05). Embora o número de indivíduos neste estudo seja reduzido, estes resultados sugerem que radiotransmissores do tipo mochila não induzem efeitos persistentes sobre a atividade adrenal de machos de falcões quiri-quiri de vida livre. Considerando a literatura consultada, esta é a primeira pesquisa a analisar os efeitos de radiotransmissores sobre a resposta adrecortical de aves de vida livre. / Although radiotelemetry is considered a valuable technique for ornithological field studies, several assumptions have been made about the impact that transmitters may cause on the estimation of behavioral, ecological and reproductive parameters. To assess potential effects of backpack radiotransmitters, we captured and assigned 8 male American kestrels (Falco sparverius) in 2 groups: radio-tagged (n = 6) and control individuals (leg-banded, n = 2). Thereafter, we collected feces two hr after capture (day −1), and subsequently during days 0 (releasing day), 1, 4, 7, 15, 30, 40 and 55. Prior to fecal analysis, EIA corticosterone assay was validated using standard procedures (e.g. parallelism, dose-response curve), and physiological significance of fecal glucocorticoid metabolites was confirmed through adrenocorticotropin (ACTH) challenge, which induce an increase of 4-fold (446.10 ± 60.73 ng/ g) above baseline (114.27 ± 15.23 ng/ g) within 4 hr (P < 0.001). Both groups exhibited a significant increase in fecal glucocorticoids during day 0 (P < 0.001), but concentrations returned to pre-attachment values within 4 days. Fecal glucocorticoid concentrations did not differ between samples of radio-tagged and leg-banded kestrels (P > 0.05). In spite of the small number of monitored subjects, these findings suggested that radiotransmitters did not affect adrenocortical activity in male American kestrel. To our knowledge, this is the first research to evaluate the effects of transmitters on stress responses in free-ranging birds.

Muscle atrophy occurs by an imbalance between catabolic and anabolic protein levels. Among catabolic proteins, FOXO3a, MuRF-1 and Atrogin-1 are directly related to muscle atrophy induced by dexamethasone. Furthermore, it is known that some inflammatory proteins (TNF-&#945; and IL-6) also participate in reduction of muscle weight. We have shown that aerobic exercise attenuates some of the side effects of dexamethasone, but nothing is known about interval training (IT) performed before and concomitant dexamethasone treatment. This study investigated if interval training is effective in attenuating muscle atrophy induced by dexamethasone and if the proteins FOXO3a, MuRF-1, Atrogina-1, TNF-&#945;and IL-6 are involved in this response. Rats were distributed into 4 groups: sedentary control (SC), sedentary + Dexa (SD), trained control (TC), and trained + Dexa (TD), and underwent an interval training period (50% and 80% of maximal capacity, 2 and 1 min, respectively, 1h/day, 5 days / week, 70 days) or remained sedentary. Dexamethasone was administered during the last 10 days (0.5mg/kg per day i.p.). The rats were weighed weekly during training and daily during the treatment. The tibialis anterior (TA), soleus (SOL) and flexor hallucis longus (FHL) were collected, weighed and stored for analysis of TNF-&#945;, IL-6, FOXO3a, MuRF-1 and Atrogin-1protein levels using electrophoresis method, Western Blotting. Administration of dexamethasone resulted in a significant decrease in body weight (-17%) followed by reduction in TA (-22%) and FHL (-19%) muscles weight. This reduction in muscle weight involved a significant increase in MuRF-1 protein levels in TA (+27%) and FHL (+18%) muscles, although TNF-&#945; (-37%FHL and -15% SOL) and IL-6 (-26% TA, - 24% FHL and -18% SOL) protein levels were reduced. Interval training was effective in blocking the increase of MuRF-1protein level in TA and FHL muscles, moreover interval training significantly reduced FOXO3a production level in TA muscle, in both groups, TC (-27%) and TD (-32%).This response was followed by an attenuation of TA muscle mass after training. Chronic treatment with dexamethasone, as well as training, did not change Atrogin-1protein level. The results of the present study allow us to conclude that MuRF-1 seems to be involved in TA and FHL muscle atrophy induced by dexamethasone treatment, independent of inflammatory proteins signaling. On the other side, interval training determined TA muscle atrophy attenuation by decreasing MuRF-1 and FOXO3a without changes in TNF-&#945; e IL-6 protein levels. / A dexametasona é amplamente utilizada no uso clínico, devido ao seu potente efeito antialérgico e anti-inflamátorio, entretanto o uso crônico pode induzir diversos efeitos deletérios, tais como hiperglicemia, hipertensão, hipercolesterolemia e atrofia muscular. A atrofia muscular se dá por um desbalanço entre proteínas catabólicas e anabólicas. Dentre as proteínas catabólicas, a FOXO3a, MuRF-1 e Atrogina-1 estão diretamente relacionadas com a atrofia muscular induzida pela dexametasona. Além disso, já se sabe que algumas proteínas inflamatórias (TNF-&#945; e IL-6) também participam na redução de peso muscular. Demonstramos recentemente que o exercício físico aeróbio atenua alguns dos efeitos deletérios da dexametasona, mas nada se sabe sobre os benefícios do exercício físico intervalado (TI) realizado antes e concomitantemente ao tratamento com a dexametasona. Portanto o objetivo deste trabalho foi investigar se o treinamento intervalado seria efetivo em prevenir a atrofia muscular induzida pela dexametasona (0,5mg/kg por 10 dias) e se as proteinas FOXO3a, MuRF-1, Atrogina-1, TNF-&#945; e IL-6 estão envolvidas nesta resposta. Foram utilizados 56 ratos Wistar, separados em 4 grupos: sedentário controle (SC), sedentário tratado com dexametasona (SD), treinado controle (TC) e treinado tratado com dexametasona (TD). Após adaptação na esteira, os animais foram submetidos ao protocolo de TI (50% e 80% da capacidade máxima, 2 e 1 minutos, respectivamente, 1h / dia, 8 semanas, 5 dias / semana) ou mantidos como sedentários. O peso corporal (PC) foi verificado semanalmente durante o treinamento e diariamente durante o tratamento. Os músculos tibial anterior (TA), sóleo (SOL) e flexor longo do hálux (FHL) foram coletados, pesados e armazenados para análises de produção proteica de TNF-&#945;, IL-6, FOXO3a, MuRF-1 e Atrogina-1 utilizando o método de eletroforese. O TI aumentou a capacidade física máxima dos animais treinados. A administração da dexametasona determinou diminuição significativa de PC (-17%) acompanhado de redução do peso muscular do TA (-22%) e FHL (-19%). Esta redução de peso muscular envolveu a proteína MuRF-1, que aumentou no TA (+27%) e FHL (+18%), embora tenha ocorrido diminuição significativa da produção proteica de TNF-&#945; (-37% FHL e -15% SOL) e IL-6 (-26% TA, -24% FHL e -18% SOL) no grupo SD. O TI foi efetivo em bloquear o aumento de MuRF-1 nos músculos TA e FHL, ademais diminuiu significativamente a produção proteica de FOXO3a no músculo TA, tanto para o grupo TC (-27%) como TD (-32%), resposta acompanhada de atenuação da redução de peso muscular no TA, mas não no FHL (TD, -15%). O tratamento crônico com dexametasona, assim como o TI, não determinou alteração na Atrogina-1. Os resultados do presente estudo nos permitem concluir que a MuRF-1 parece estar envolvida na atrofia nos músculos TA e FHL induzida por 10 dias de tratamento com dexametasona, independentemente das alterações das proteínas inflamatórias (TNF-&#945; e IL-6). O TI prévio, por sua vez, determinou atenuação da atrofia no TA por promover diminuição significativa na produção proteica de MuRF-1e de FOXO3a, sem alterar a produção proteica de TNF-&#945; e IL-6.

O uso de isuflupredona melhora a taxa de prenhez e altera o perfil proteico do líquido endometrial em relação a éguas não tratadas. A utilização de dexametasona diminui o acúmulo de líquido pós-cobertura, reduz o edema do útero, porém, desconhecem-se seus efeitos no perfil proteico do líquido endometrial. O objetivo do presente estudo foi analisar o efeito da dexametasona em éguas suscetíveis à endometrite persistente póscobertura sobre perfil proteico do líquido endometrial na presença ou ausência de infecção. Nove éguas suscetíveis foram utilizados, com idade entre 7 a 30 anos. Após a verificação dos sinais de estro as éguas foram submetidas a quatro tratamentos: (C) éguas não receberam nenhum tipo de tratamento e serviram como controle; (D) éguas receberam 40mg de dexametasona (IV), no momento da cobertura, com coleta da amostras após 6 horas, (I-6 e I-24) infusão intra-uterina de 1 x 109 de S. zooepidemicus/mL, com coleta da amostra após 6 e 24 horas; (I/D-6 e I/D-24) infusão intra-uterina de 1 x 109 S. zooepidemicus/mL e administração de 40mg de dexametasona (IV), com coleta da amostra após 6 e 24 horas. Todas as éguas foram submetidas a todos os tratamentos. As amostras foram coletadas e submetidas à eletroforese bidimensional para separação proteica e espectrometria de massa para a identificação das bandas proteicas relevantes. A corticoterapia provocou alteração na proteômica do líquido endometrial de éguas suscetíveis, caracterizada pelo aumento (TTR) e/ou diminuição (ApoA1) na densidade óptica de proteínas da fase aguda da inflamação. Conclui-se que a utilização da dexametasona em éguas com e sem presença de infecção altera a proteômica do fluido endometrial de éguas suscetíveis. Sugere-se que a dosagem ou a frequência de aplicação da dexametasona deva ser aumentada. / Corticotherapy has often been used in susceptible mares. The use of isuflupredona improves pregnancy rate and alters the protein profile of the endometrial fluid in relation to untreated mares. The use of dexamethasone decreases the post breeding fluid accumulation, reduces the uterine edema, however is unaware of its effects on the protein profile of endometrial fluid. The aim of the present study was analyze the effect of dexamethasone in mares susceptible to post-breeding persistent endometritis on the protein profile of endometrial fluid in the presence or absence of infection. Nine susceptible mares were used, aged 7-30 years old. After checking the signs of estrus, mares were subjected to four treatments: (C) mares received no treatment and served as controls; (D) mares received 40 mg of dexamethasone at breeding time, with collection of samples after 6 hours; (I-6 and I-24) intrauterine infusion of 1 x 109 S. zooepidemicus/ml and the sample was collected after 6 and 24 hours; (I/D-6 and I/D-24) intrauterine infusion of 1 x 109 S. zooepidemicus/ml and 40 mg of dexamethasone administration, collecting the sample after 6 and 24 hours. All of the mares were subjected to all treatments. Samples were collected and subjected to two-dimensional electrophoresis for protein separation and mass spectrometry for the identification of relevant protein bands. Corticotherapy resulted in alteration of the protein profile of the endometrial fluid of susceptible mares, characterized by an increase (TTR) and/or decrease (ApoA1) in optical density of the acute phase of proteins of inflammation. We conclude that the use of dexamethasone in mares with and without the presence of infection alters the protein profile of endometrial fluid of susceptible mares. It is suggested that the dosage or frequency of application of dexamethasone should be increased.

Estudo da expressão das proteínas envolvidas no estresse de retículo endoplasmático durante o remodelamento das ilhotas pancreáticas maternas no período perinatal / Study of endoplasmatic reticulum stress-related proteins in the maternal pancreatic islets remodeling during the peripartum

Na gestação há aumento da proliferação e redução da apoptose das células b pancreáticas. Prolactina (PRL) desencadeia estas mudanças, que são revertidas após o parto mesmo na presença de PRL. In vitro, dexametasona (DEX) se contrapõe a PRL. Avaliamos se o estresse do retículo endoplasmático (ERE) está envolvido na apoptose do pós-parto e se os glicocorticóides (GC) participam deste mecanismo. A fragmentação do DNA aumenta no 3° dia pós-parto (L3), em paralelo com a diminuição de pAKT e aumento do TRB3, indutor da apoptose por ERE. BiP, ATF4, CHOP, e a ligação de CHOP e CHOP-ATF4 no promotor do TRB3 aumentam em L3. O inibidor do ERE PBA restaurou os níveis de pAKT e CHOP e inibiu a apoptose. Células RINm5F tratadas com DEX (24h) têm aumento de BiP e ATF4, de p-eIF2 e do XBP-1 ativo. DEX também induz TRB3, mas inibe a ligação de CHOP ao TRB3. O tratamento por 72h não altera p-eIF2a, diminui XBP-1 ativo e promove apoptose, único evento revertido pela PRL. Concluímos que a apoptose das ilhotas em L3 é desencadeada por ERE, mas os GC não induzem este mecanismo. / During gestation occurs increase on the proliferation and apoptosis reduction of pancreatic b cells. Prolactin (PRL) promotes these changes which are reverted after delivery. Dexametasone (DEX) in vitro opposed to PRL. We evaluate whether endoplasmatic reticulum stress (ERS) was involved on post-delivery apoptosis and glycocorticoids (GC) participate on this mechanism. DNA fragmentation increased on the 3rd day post-delivery (L3), in parallel with pAKT diminution and inductor of apoptosis-TRB3 augment by ERS. BiP, ATF4, CHOP along with binding of CHOP and CHOP-ATF4 to the TRB3 promoter increased in L3. ERS inhibitor-PBA restored pAKT, CHOP levels and inhibited apoptosis. RINm5F cells with DEX (24h) showed increase in BiP, ATF4, p-eIF2 and in active XBP-1. DEX induced TRB3, but inhibited the binding of CHOP to TRB3. The 72h treatment did not alter p-eIF2a, diminished active XBP-1 and promoted apoptosis; the unique event reverted by PRL. We concluded that apoptosis of islets in L3 is generated by ERS; nevertheless this mechanism is not induced by GC.
