Development of a multifocal confocal fluorescence lifetime imaging microscope for high-content screening applications

Fluorescence lifetime imaging microscopy (FLIM) is an imaging modality that is able to provide key insights into subcellular processes. When used to measure Förster resonance energy transfer (FRET), for instance, it can discern protein-protein interactions and conformational changes. This kind of information is highly useful in the drug screening process in order to determine the effectiveness of drug leads and their mechanisms of action. FLIM has yet to be successfully translated to high-content screening (HCS) platforms due to the high throughput and fine temporal and spatial resolution requirements of HCS. Our prototype HCS FLIM system uses a time-resolving instrument called a streak camera to multiplex the FLIM scanning process, allowing for 100 confocal spots to be simultaneously scanned across a sample. There have been a few major advancements to the prototype. First the fiber array used to connect the fluorescence channels to the streak camera was characterized. Its alternating fiber delay scheme was successful in greatly reducing optical crosstalk between adjacent channels. Next, an optical beam scanner for parallel excitation beams was designed and implemented, greatly improving the possible scan speeds of the system. The streak camera was upgraded to a higher repetition rate sweep, and modifications to system components and reconstruction procedures were made to accommodate the new sweep unit. A single-photon avalanche diode array was also tested as a possible replacement for the streak camera, and was found to offer photon detection efficiency advantages. Finally, improvements were made to the excitation power and optical throughput of the system in order to reduce the required exposure time. These advances to the prototype system bring it closer to realizing the requirements of HCS FLIM, and provide a clear picture for future improvements and research directions. / Thesis / Doctor of Philosophy (PhD) / Fluorescent proteins are commonly used to tag subcellular targets so that they can easily be distinguished with a fluorescence microscope. While this can help visualize where different organelles and proteins are located in the cell, a great deal more information can be gained by measuring the fluorescence lifetime at each point in the sample, which is highly sensitive to the microenvironment. Fluorescence lifetime imaging microscopy (FLIM) has the potential to be a powerful technique for testing drug leads in the drug discovery process, although current FLIM systems are not able to provide the high throughput speeds and high temporal resolution required for drug screening. This thesis project has succeeded in improving a highly parallel FLIM microscope by reducing inter-channel crosstalk, implementing an optical scanner, improving power and optical throughput, and investigating future time-resolving instruments. This progress has brought the prototype setup closer to being used in a drug screening environment.

Degradation Mechanisms in Small-Molecule Organic Electronic Devices / Alterungsmechanismen in organischen Halbleiterbauelementen basierend auf kleinen Molekülen

Over the last decades organic light-emitting diodes (OLEDs) and organic solar cells (OSCs) have gained considerable attention as efficient, flexible, lightweight, and potentially low-cost technology for lighting and display applications or as a renewable energy source, respectively. However, achieving long-term stability remains challenging. Revealing and understanding aging processes is therefore of great interest. This work presents fundamental investigations to understand and circumvent organic device degradation. In the first part, single materials used in organic devices were investigated. By tailoring an attenuated total reflection infrared (ATR-IR) spectrometer to the specific needs and subsequent measurements, it is shown that the tris(8-hydroxyquinoline)aluminum (Alq3) molecule, a well known fluorescent green emitter, degrades during air exposure by the formation of carbonyl groups. By using a laser desorption/ionization time of flight mass spectrometer (LDI-TOF-MS) it was shown that a,w-bis-(dicyanovinylen)-sexithiophen (DCV6T-Bu4), a well known small-molecule material which is used as part of the active layer, reacts with oxygen during ultraviolet (UV) irradiation. By using climate boxes and a sun simulator the impact of dry and humid air as well as sunlight on C60, a widely-used acceptor molecule in organic solar cells, was investigated. The breaking of the C60 cage to C58 and C56 and the further reaction of these components with oxygen as well as the dimerization of C58 and C56 molecules were found. The degradation products such as C58O increase with air exposure time but they are independent of the humidity level of the ambient air as well as sunlight irradiation. Subsequent annealing leads to a decrease of the C58O concentration. Many efficient n-dopants are prone to degradation in air, due to the low ionization potentials, thereby limiting the processing conditions. It was found that the air exposure of the highly efficient n-dopant tetrakis(1,3,4,6,7,8-hexahydro-2H-pyrimido[1,2-a]pyrimidinato)ditungsten(II) (W2(hpp)4) leads to oxidation reactions of the molecule to [W(hpp)2 + O] and other degradation products. The decay constant of W2(hpp)4 and the matching mean growth time of the [W(hpp)2 + O] degradation as well as a second very quick degradation of the dopant could be determined. The two decay constants can be explained by the assumption that W2(hpp)4 molecules, which are involved in the charge transfer, do degrade slower due to the fact that the charge transfer leads to a downshift of the energy levels of the W2(hpp)4 molecule. Apart from the properties of the organic materials, other effects such as the impact of different purification systems on the material purity as well as the dependence of material purity on the OLED lifetime has been investigated. No correlations between the purification grade and the amount of impurities were found. OLEDs which contain N,N\'-di(naphthalen-1-yl)-N,N\'-diphenyl-benzidine (alpha-NPD) purified in a vertically interlaced stainless steel sublimation systems shows slightly higher external quantum efficiencies compared to tube-based vacuum sublimation systems. The devices which contain alpha-NPD purified by a sublimation system have an extended lifetime. Finally, the impact of residual gases during device fabrication on OLED lifetime and electrical characteristics was investigated. It was found that water vapor introduces an additional series resistance to the OLED, while the other gases do not influence the electric characteristics. The presence of nitrogen or oxygen impacts the lifetime of the OLEDs by the same amount. Nitrogen is non-reactive, this leads to the conclusion that the influence of nitrogen and oxygen on the OLED lifetime is of non-chemical nature, such as changes in the morphology of the organic layers. Water vapor introduces an additional, even faster degradation process within the first hours of OLED operation. As major sources of device degradation, the dimerization of 4,7-diphenyl-1,10-phenanthroline (BPhen) as well as the complexation reaction of alpha-NPD with a bis(1-phenylisoquinoline)iridium(III) (Ir(piq)2) fragment was identified.

Energy-efficient and lifetime aware routing in WSNs

Network lifetime is an important performance metric in Wireless Sensor Networks (WSNs). Transmission Power Control (TPC) is a well-established method to minimise energy consumption in transmission in order to extend node lifetime and, consequently, lead to solutions that help extend network lifetime. The accurate lifetime estimation of sensor nodes is useful for routing to make more energy-efficient decisions and prolong lifetime. This research proposes an Energy-Efficient TPC (EETPC) mechanism using the measured Received Signal Strength (RSS) to calculate the ideal transmission power. This includes the investigation of the impact factors on RSS, such as distance, height above ground, multipath environment, the capability of node, noise and interference, and temperature. Furthermore, a Dynamic Node Lifetime Estimation (DNLE) technique for WSNs is also presented, including the impact factors on node lifetime, such as battery type, model, brand, self-discharge, discharge rate, age, charge cycles, and temperature. In addition, an Energy-Efficient and Lifetime Aware Routing (EELAR) algorithm is designed and developed for prolonging network lifetime in multihop WSNs. The proposed routing algorithm includes transmission power and lifetime metrics for path selection in addition to the Expected Transmission Count (ETX) metric. Both simulation and real hardware testbed experiments are used to verify the effectiveness of the proposed schemes. The simulation experiments run on the AVRORA simulator for two hardware platforms: Mica2 and MicaZ. The testbed experiments run on two real hardware platforms: the N740 NanoSensor and Mica2. The corresponding implementations are on two operating systems: Contiki and TinyOS. The proposed TPC mechanism covers those investigated factors and gives an overall performance better than the existing techniques, i.e. it gives lower packet loss and power consumption rates, while delays do not significantly increase. It can be applied for single-hop with multihoming and multihop networks. Using the DNLE technique, node lifetime can be predicted more accurately, which can be applied for both static and dynamic loads. EELAR gives the best performance on packet loss rate, average node lifetime and network lifetime compared to the other algorithms and no significant difference is found between each algorithm with the packet delay.

Essays on intergenerational income mobility, geographical mobility, and education

This thesis consists of an introductory part and the following four self-contained papers: In Paper [I] we analyze the implications of social identity and self-categorization for optimal redistributive income taxation. A two-type model is supplemented by an assumption that individuals select themselves into social categories, in which norms are formed and education effort choices partly depend on these norms. The results show, among other things, that externality correction by a welfarist government leads to an element of tax progression that serves to reduce the discrepancy between the effort norm and the actual effort chosen by low-productivity individuals in the high-effort group. Furthermore, if the preference for social identity is sufficiently strong, increased wage-inequality leads to higher social welfare through a relaxation of the selection constraint. It may thus be desirable to use publicly provided education to induce more wage-inequality, even if higher wage-inequality increases the intrinsic utility of a potential mimicker. In Paper [II] I employ high quality register data to present new facts about income mobility in Sweden. The focus of the paper is regional differences in mobility, using a novel approach based on a multilevel model. This method is well-suited when regions differ greatly in population size as is the case in Sweden. The maximum likelihood estimates are substantially more precise than those obtained by running separate OLS regressions. I find small regional differences in income mobility when measured in relative terms. Regional differences are large when adopting an absolute measure and focusing on children with below-median parent income. On the national level I find that the association between parent and child income ranks has decreased over time, implying increased mobility. In Paper [III] I study the long term effects of inter-municipal moving during childhood on income using Swedish register data. Due to the richness of the data I am able to control for important sources of selection into moving, such as parent separation, parents' unemployment, education, long run income, and immigration background. I find that children's long run incomes are significantly negatively affected by moving during childhood, and the effect is larger for those who move more often. For children who move once, I also estimate the effect of the timing and the quality of the move. I measure the quality of each neighborhood based on the adult outcomes for individuals who never move. The quality of a move is defined as the difference in quality between the origin and the destination. Given that a family moves, I find that the negative effect of childhood moving on adult income is increasing in age at move. Children benefit economically from the quality of the region they move to only if they move before age 12 (sons) and age 16 (daughters). In Paper [IV] I study the bias of IGE estimates for different missing-data scenarios based on simulated income processes. Using an income process from the income dynamics and risks literature to generate two linked generations’ complete income histories, I use Monte Carlo methods to study the relationship between available data patterns and the bias of the IGE. I find that the traditional approach using the average of the typically available log income observations leads to IGE estimates that are around 40 percent too small. Moreover, I show that the attenuation bias is not reduced by averaging over many father income observations. Using just one income observation for each generation at the optimal age (as discussed in the paper) or using weighted instead of unweighted averages can reduce the bias. In addition, the rank-rank slope is found to be clearly less sensitive to missing data.

Untersuchung der Fluoreszenzlebensdauer von BODIPY-Farbstoffen in Polymerlösungen und Polymerschmelzen

Die vorliegende Arbeit befasst sich mit dem Fluoreszenzverhalten, speziell der Fluoreszenzlebensdauer, von BODIPY-Farbstoffen in Polymerlösungsmittelgemischen mit unterschiedlicher Polymerkonzentration sowie in Polymerfilmen bei unterschiedlichen Temperaturen. Dazu werden zunächst die Synthesen von vier verschiedenen BODIPY-Fluorophoren mit einem Phenylsubstituent in meso-Position aufgezeigt. Dahingehend wurde eine Synthesestrategie entwickelt, um eine einzelne Polypropylenkette an diese Farbstoffsysteme anzubinden. Dabei soll aufgezeigt werden, dass die Länge des Substituenten am Phenylsubstituenten am chromophoren Kern maßgeblich das Fluoreszenzverhalten der Sonde beeinflusst. BODIPY-Farbstoffe mit makromolekularen Substituenten zeigen im Vergleich zu Derivaten mit kürzeren Substituenten eine deutlich größere Fluoreszenzlebensdauer und eine nicht so stark ausgeprägte Temperaturabhängigkeit. Mehrere Zeitkomponenten der Fluoreszenzlebensdauer der Fluorophore in reinem Polypropylen bzw. deren Mehrkomponentensystemen (Polyethylenpropylen Copolymer oder Kraton) im Vergleich zu reinen Lösungsmitteln (Toluol oder Dodecen) deuten dabei auf lokale Heterogenitäten im Material hin. Außerdem wird der Einfluss der Viskosität auf die Fluoreszenzlebensdauer in Polymer/Lösungsmittelgemischen mit unterschiedlicher Polymerkonzentration untersucht und die Rolle des Wasserstoffbrückennetzwerkes zwischen den Polymer- und Lösungsmittelmolekülen diskutiert.

Dans ce mémoire, nous proposons une méthodologie statistique permettant d’obtenir un estimateur de l’espérance de vie des clients en assurance. Les prédictions effectuées tiennent compte des caractéristiques individuelles des clients, notamment du fait qu’ils peuvent détenir différents types de produits d’assurance (automobile, résidentielle ou les deux). Trois approches sont comparées. La première approche est le modèle de Markov simple, qui suppose à la fois l’homogénéité et la stationnarité des probabilités de transition. L’autre modèle – qui a été implémenté par deux approches, soit une approche directe et une approche par simulations – tient compte de l’hétérogénéité des probabilités de transition, ce qui permet d’effectuer des prédictions qui évoluent avec les caractéristiques des individus dans le temps. Les probabilités de transition de ce modèle sont estimées par des régressions logistiques multinomiales. / In this master’s thesis, we develop a statistical method to estimate the lifetime expectancy of clients in the insurance domain. The forecasts are personnalized according to the clients’ own features, the most notable being the fact that they can have any combination of automobile and residential insurance products. Three approaches are compared. The first approach is the simple Markov model which assume homogeneity and stationnarity of the transition probabilities. The other model suggested – which is implemented both by direct computation and by simulation – allows for heterogeneity of the transition probabilities, thus providing forecasts which evolve in time along with the characteristics of the clients. The transitions probabilities are estimated using multinomial logistic regressions.
