Influence of SjGST on the growth and migration of human breast cancer cells

Glutathione S-transferase (GST) is an essential detoxification enzyme in eukaryotic cells, by catalysing the conjugation of electrophilic substrates to glutathione (GSH) and detoxification both in external and internal cellular enviornments. Previous studies of our team suggested that SjGST had profound on the growth of human breast cancer cells in vitro. In order to analyse the effect of SjGST on the proliferation and migration of human breast cancer cell line, MDA-MB 435s , we initially constructed plasmid containing gst fragment in pAcGFP-N1 and transfected into MDA-MB 435s cells. Assays for proliferation and migration of the transfected cells showed that no difference between transfected and non-transfected cells was found. Analysis of the transfected gene found a frameshift mutation occurred in the plasmid, and thus no expression was detedted. Thus, we carried out assays for proliferation and migration of the cancer cells using recombiation SjGST instead. Results from assays for proliferation and migration showed that SjGST enhance both proliferation and migration of MDA-MB 435s. Furthermore, the recombinant protein was a strong inhibitor to cell death, probably by detoxification pathway. But the mechanism of action of detoxification of shikonin by the recombinant protein, however remains unknow.

One of the most important aspects in MDA is meta-model, which is used to specify MDA style transformations. Theses present meta-model of PLA model and XSL transformations, used to retrieve PLA simulation model. Example of creating oil terminal simulation model is presented as well.

Dando continuidade às suas soluções de interoperabilidade, o Object Management Group (OMG) padronizou a Model Driven Architecture (MDA), uma arquitetura em camadas que, através de supressão dos detalhes de tecnologia, contorna os problemas de heterogeneidade e de adaptação das aplicações para adoção de novas tecnologias. A inclusão dos conceitos de arquitetura de software no processo de desenvolvimento auxilia este trabalho, uma vez que ela fornece uma visão de alto nível do sistema. Texten analyseras med hjälp av Systemisk funktionell grammatik (SFG) och de visuella elementen med Systemisk funktionell Multimodal Diskusanalys (SF-MDA). Det material som ingår i uppsatsen är litet och undersöks djupgående. Uppsatsen drar slutsatser om hur kampanjen låter bild och text samverka, både för att förhöja enade budskap och för att enhetligt presentera skilda budskap.

The role of the homeodomain protein Pitx3 in the development and survival of midbrain dopaminergic neurons

There is much interest in the study of midbrain dopaminergic (mDA) neurons as their functions include the regulation of motor function, emotion and reward pathways. Furthermore the dysfunction of these neurons is implicated in a number of human disorders such as Parkinson’s disease (PD), addiction and schizophrenia. PD is characterised by the degeneration of mDA neurons of the substantia nigra pars compacta (SNc), therefore, research into the specification and development of mDA neurons is of particular interest in relation to this disease. An understanding of the development of mDA neurons may lead to new methods of preventing their degeneration or potentially a human ES cell derived source of mDA neurons that could be used for transplantation in PD patients. Pitx3 is a bicoid-related homeodomain protein with an expression pattern restricted to the mDA neurons of the SNc and ventral tegmental area (VTA), within the central nervous system. To directly investigate a role for Pitx3 in mDA neuron development, I have analysed a line of transgenic mice with a green fluorescent protein (GFP) reporter under the control of the endogenous Pitx3 promoter. Use of the targeted GFP reporter as a midbrain dopaminergic lineage marker in the phenotypically normal heterozygous mice identified previously unrecognised ontogenetically distinct subpopulations of dopaminergic cells within the ventral midbrain. These subpopulations were detectable at E12.5 based on their temporal and topographical expression of Pitx3 and TH. Analysis of the Pitx3 null mice revealed that Pitx3 is required for the survival of a subset of nascent mDA neurons at the beginning of their terminal differentiation. The loss of mDA neurons via apoptosis continued throughout development resulting in a complete absence of SNc neurons whilst the VTA remained relatively intact in adult Pitx3 null mice. In addition, during embryonic development Pitx3 deficiency caused a loss of tyrosine hydroxylase (TH) expression specifically in the SNc dopaminergic neurons. Analysis of chimeric mice made with Pitx3 null and Pitx3 heterozygous ES cells revealed that Pitx3 acts in a cell autonomous manner. These findings point to two roles for Pitx3 in SNc mDA neurons, one in their survival and the other in regulation of TH expression. Taken together, these studies suggest that the ontogenetically distinct subpopulations may provide the molecular basis for the specific dependence of substantia nigra DA neurons on Pitx3. In addition, to establish whether the subpopulations identified at E12.5 do form the SNc and VTA, respectively, a strategy to track the fate of the earliest Pitx3- expressing cells has been initiated. In order to achieve this I have created transgenic mice in which a tamoxifen inducible form of Cre recombinase is under the control of the endogenous Pitx3 promoter. These mice can be crossed with existing mice which contain a ubiquitously expressed Cre-inducible reporter, such as LacZ or GFP, to give a temporally and spatially restricted reporter expression.

This paper supplies the reader with a broad overview of the basic concepts, the difference representation and application including the metamodel independent approach, and finally a narrow examination of the research topic, including constraints, examples and implementation details.</p>

Improving PIM Modeling with Design Pattern

Apply design pattern in software analysis and design can reduce time and cost by reuse proven and validated solution to avoid redesign. Design pattern provide general solution to specific problem domain, it is best to adopt design pattern in Platform Intendant Model (PIM). Model Driven Architecture (MDA) lack of methodology to judge problem domain from UML Diagram like sequential diagram, timing diagram or class diagram and how to apply design pattern in PIM mode. To alleviate the forgoing problem, this study proposed methodology to describe rule and step to apply design pattern, and present the difference between adopt design pattern by UML diagram. The research methodology is articulated using the design science research methodology. A usability study evaluation is performed to demonstrate its applicability with test case. With this proposed method, system analyst will improve software system flexibility and accuracy by design pattern after first phase PIM modeling, and object designed in software system will be low cohesion and high coupling .

This work analysis model transformation problems in Model Driven Architecture (MDA). MDA is an architecture created by Object Management Group (OMG). MDA separates business and application logic from underlying platform technology. Platform-independent models of an application or system can be realized through the MDA on any platform. The most important MDA problem is a transformation between plarform-independent models (PIM) and platform specific models (PSM). Currently, there is no standard language for these transformations. Purpose of this work is to analize possible transformations. Two technologies will be researched: XSLT and QVT XSLT (Extensible Stylesheet Language Transformations) is a language for transforming XML documents into other XML documents. QVT (Queries/Views/Transformations) is a language for transforming models in MDA architecture. Aggregation systems will be used as models for transformations. Platform-independent model of aggregation system will be created and transformed to Java platform models using XSLT and QVT transformations.
