[pt] Ftalatos são os plastificantes mais utilizados industrialmente, estando disseminados em todos os ecossistemas do mundo. Como são compostos exclusivamente sintéticos, sua presença é indicativa da industrialização de uma região. Sua toxicidade é baixa, porém existem indícios de possíveis efeitos como interferentes endócrinos, especialmente no sistema reprodutor masculino. No Brasil, existem poucos estudos sobre a ocorrência e comportamento dos ftalatos no meio ambiente. Nesta tese foi estudada sua ocorrência em amostras de águas potáveis de diversos bairros das cidades do Rio de Janeiro e Niterói e do ecossistema da Baía de Guanabara (águas, sedimentos e mexilhões), a qual recebe inúmeros aportes industriais. Todas as amostras foram coletadas, extraídas em fase sólida e analisadas por cromatografia em fase gasosa, utilizando detecção por captura de elétrons. As da Baía foram amostradas nas estações seca e chuvosa, em diferentes anos. As extrações de todas as amostras obtiveram excelente repetibilidade e recuperações superiores a 76%. As águas potáveis do Rio de Janeiro e Niterói apresentaram teores de ftalatos em concentrações muito menores do que as informadas para cidades da Europa e Estados Unidos. Tais teores puderam ser correlacionados com as extensões das redes dos sistemas de abastecimento de ambas as cidades. Todas as amostras da Baía mostraram contaminações qualiquantitativas muito inferiores, quando comparadas a outros ambientes costeiros. O estudo do biomonitoramento ativo evidenciou a eficiência e rapidez da depuração dos ftalatos nos mexilhões mais contaminados do ecossistema da Baía de Guanabara. / [en] Phthalates are the most used plasticizers in industrial products, worldwide distributed in environments. Their presence is often used to indicate a region industrialization degree. Although their reported toxicity is low, phthalates are indicated for possible endocrine disrupting effects, specially in male reproductive system. In Brazil, there are few studies about the environmental occurrence and distribution of phthalates. This thesis studied the occurrence of those compounds in potable waters samples from residential areas of Rio de Janeiro and Niterói cities, and in samples from the ecosystem of Guanabara Bay (waters, sediments and mussels), which receives several industrial inputs. All the samples were collected, solid-phase extracted and the phthalate contents analyzed by gas chromatography with electron capture detection. The ones from the Bay were sampled during the dry and rainy seasons, in different years. All the extractions presented excellent repeatability and recoveries above 76%. The potable water samples from both cities presented phthalate values considerably lower than those reported for European and American cities. The results could be correlated with the water net distribution lengths for both cities. All results from the Bay ecosystem showed lower contamination when compared to other coastal environments. The controlled transplant proved quick and efficient phthalate depuration of contaminated organisms from Guanabara Bay.

La miniaturisation et la complexification croissante des composants microélectroniques induit une sensibilisation et une fragilisation accrue des composants vis-à-vis des contaminations présentes dans les zones de productions appelées “salles blanches”. Dans ces espaces, le contrôle actuel de la contamination organique n’est pas suffisant puisqu’il ne permet pas d’éviter la contamination de surface des plaquettes de silicium et des optiques des robots de production utilisés pour la photolithographie. Un contrôle accru des concentrations des contaminants organiques dans les atmosphères des salles blanches devient donc nécessaire et de nouvelles méthodes analytiques doivent être développées et validées. Dans le cadre de ce travail, des méthodes d’analyse ont été développées et validées afin de disposer d’une gamme d’outils permettant un suivi rigoureux des contaminations. Ces outils permettent d’identifier et de quantifier les contaminations surfaciques des plaquettes de silicium par des composés organiques semi-volatils (phtalates et organophosphorés) mais aussi de déterminer les concentrations de composés organiques volatils présents dans l’atmosphère des salles blanches. Ces méthodes font appel aux technologies du WOS/ATD-GC-MS « Wafer Outgassing System/Automated Thermal Desorber–Gas Chromatography–Mass Spectrometry » et de la DART-ToF-MS « Direct Analysis in Real Time-Time of Flight–Mass Spectrometry » pour les analyses de surfaces et au PTR-ToF-MS « Proton Transfer Reaction – Time of Flight - Mass Spectrometry » pour l’analyse de l’atmosphère. / The recent advances in the miniaturization and complexification of microelectronic components induce an increase in the sensitivity of these components regarding the organic contamination present in the production zone called “clean room”. Although, the control of organic contamination in the clean room is very rigorous it does not avoid the contamination of silicon wafer surfaces and robot lenses used in the photolithography process. The later implies that new analytical methodologies should be developed and validated. In this work, analytical methods were developed and validated in order to have a panel of tools which allows careful monitoring of organic contaminants. These tools allow the identification and quantitation of the contamination of silicon wafer surface by semi-volatiles organic compounds (phthalates and organophosphates) and the determination of volatile organic compounds concentrations in the clean room atmosphere. These methods uses the WOS/ATD-GC-MS « Wafer Outgassing System/Automated Thermal Desorber–Gas Chromatography–Mass Spectrometry » technology and the DART-ToF-MS « Direct Analysis in Real Time-Time of Flight–Mass Spectrometry » technology for wafer surface analysis and the PTR-ToF-MS « Proton Transfer Reaction – Time of Flight - Mass Spectrometry » technology for gas-phase analysis.

Étude des interactions PET - Eau minérale dans les eaux embouteillées au Liban et approches analytiques des risques sanitaires / Study of PET interaction with mineral water in lebanese bottled water and analytical approaches of health risks

L'évaluation de l'inertie des matières plastiques au contact de denrées alimentaires est régie par le règlement européen N°10/2011. Au Liban, l'absence de législations relatives à l'usage des matières en plastiques a conduit à des emballages non contrôlés au niveau de la production et du stockage, ce qui peut induire un risque sanitaire pour le consommateur. L'objectif de ce travail a été d'apprécier les phénomènes de migration des contaminants du PET vers l'eau. Pour cela, l'analyse de certains migrants potentiels a été initiée tels que des COV et des SV afin de vérifier la conformité de leur teneur par rapport aux LMA dans les règlementations internationales. Le dosage des aldéhydes dans l'eau a été effectué par dérivation avec la 2,4-DNPH, suivie d'une étape de préconcentration sur une cartouche en C18, puis séparation et quantification par HPLC-UV. L'analyse des composés SV a été réalisée par SPME-GC-FID sur une fibre en PDMS/DVB. L'analyse des BTEX et du styrène a été réalisée par HS-Trap-GC-FID. Ces méthodes ont été validées conformément à la norme NF T 90-210 avec des LQ inférieures aux LMA. L'analyse des COV dans l'eau embouteillée et dans le PET a été réalisée par HSTrap- GC-MS. La présence de FA, d'AA, de DEP et des BTEX dans l'eau embouteillée en PET a été confirmée. Il a été mis en évidence que la température et l'exposition solaire influencent la migration des aldéhydes. Les concentrations d'AA trouvées après incubation à 40ºC ne sont pas négligeables et peuvent modifier les propriétés organoleptiques de l'eau. L'analyse du PET montre la présence des hydrocarbures, des aldéhydes, des alcools, et des cétones / The inertness’ value of plastics that are on contact with foodstuffs is governed by the European rule N°10/2011. In Lebanon, the lack of the Rules and Regulations of plastics’ usage led for a non-controlled packing in production as in storage, which will cause a sanitary risk to the consumer and to the population. The purpose of this research was to appreciate the migration’s phenomena of contaminants from the PET to the water. For this, there was an analysis of potential migrants such as VOC and SV to verify their tenor’s conformity relative to the MCL in international regulations. The aldehydes’ dosage in the water was performed by the derivation with the 2,4-DNPH, followed by a concentration on a C18 cartridge, then separation and quantification was done by HPLC-UV. The SV compounds’ analysis was realized by SPME-GC-FID method on a PDMS / DVB fiber. The BTEX and the styrene’s analysis was done by HS-trap-GC-FID method. These methods were validated in accordance with standard NF T 90-210 with a LQ less than the MCL. The volatile compounds in the bottled water and in the PET were analyzed by HS-trap-GC-MS method. The presence of FA, AA, DEP and BTEX in the bottled water with the PET was confirmed. It was highlighted that temperature and sun exposition influence the aldehydes’ migration. The acetaldehyde’s concentration after incubation at 40ºC wasn’t negligible and can modify the water’s organoleptic properties. Les phénols et les phtalates incluent des composés très largement utilisés dans des produits de la vie quotidienne. Une grande partie de la population générale y est donc largement exposée. Ces composés sont suspectés d’être des perturbateurs endocriniens et des effets sur la santé chez l’Homme ont été rapportés, notamment après une exposition périnatale. Les études épidémiologiques sur les effets sur la santé humaine reposent généralement sur un faible nombre de biospécimens pour estimer l’exposition. Cependant, la variabilité intra-individuelle des phénols et des phtalates est potentiellement forte, ce qui peut entraîner une mauvaise classification de l’exposition dans les études sur les effets des phénols et des phtalates et limite leurs conclusions. La variabilité intra-individuelle des phénols et des phtalates au cours de la grossesse n’est pas très bien caractérisée à l’heure actuelle.L’objectif de cette thèse est d’explorer l’exposition aux phénols et aux phtalates et plus précisément : 1) d’étudier les associations entre une telle exposition pendant la grossesse et la santé respiratoire de l’enfant au cours de ses premières années de vie ; 2) de caractériser la variabilité temporelle intra-individuelle de ces composés au cours de la grossesse ; et 3) d’évaluer l’efficacité d’une approche basée sur le pooling intra-sujet d’un nombre réduit d’échantillons journaliers pour estimer l’exposition.Les associations entre l’exposition aux phénols et phtalates et la santé respiratoire reposent sur n = 587 couples mèresenfants de la cohorte prospective française EDEN. Les développements sur l’estimation de l’exposition au cours de la grossesse s’appuient sur n = 16 femmes enceintes ayant participé à l’étude de faisabilité de la cohorte SEPAGES.Les travaux de cette thèse quantifient la variabilité intra-individuelle des concentrations urinaires des biomarqueurs d’exposition aux phénols et des phtalates au cours de la grossesse pour des échelles de temps variées (du jour à plusieurs mois). Ils confirment empiriquement que cette variabilité peut biaiser fortement les fonctions doses-réponses dans les études épidémiologiques explorant les effets de l’exposition fœtale à ces composés chez l’Homme.Les résultats de cette thèse enrichissent la littérature émergente sur les effets des expositions précoces aux phénols et phtalates sur la santé respiratoire de l’Homme. Cependant, notre étude ainsi que la plupart des recherches précédentes sont potentiellement limitées par les problématiques liées à la mesure de l’exposition. Ce travail souligne l’importance de stratégies d’échantillonnage des biomarqueurs d’exposition plus élaborées pour l’étude de ces composés dans de futures études épidémiologiques. Ces résultats sont aussi pertinents en dehors du contexte de la grossesse et pour d’autres composés non-persistants. De nouvelles approches, telles que le pooling répété pour chaque sujet d’un petit nombre de biospécimens journaliers, validé dans cette thèse, sont nécessaires pour caractériser efficacement l’impact des composés non-persistants sur la santé de l’Homme. / Phenols and phthalates include chemicals widely used in daily-life products, resulting in ubiquitous exposure of the general population. There is growing concern regarding the effects on human health of these compounds, suspected to be endocrine disruptors, particularly during early life. Epidemiological research on the health effects of phenols and phthalates in offspring generally rely on a few biospecimens to assess exposure. These studies are limited by the possibly strong within-subject variability, which may result in exposure misclassification. The within-subject variability in the context of pregnancy and its possible impact on dose-response functions are poorly characterized.The aim of this thesis was to study the exposure to several phenols and phthalates during pregnancy by: 1) investigating the possible associations between this exposure and respiratory outcomes in childhood; 2) characterizing the temporal within-subject variability of these compounds during pregnancy; and finally 3) studying the efficiency of a within-subject pooling approach using a small number of daily biospecimens for exposure assessment.Associations between exposure to phenols and phthalates and respiratory health relied on n = 587 mother-child pairs from the French EDEN prospective cohort. Developments about the assessment of exposure during pregnancy relied on n = 16 pregnant participants of the SEPAGES-feasibility study who had collected all their urine samples for three weeks.This work quantified the within-subject variability of phenol and phthalate biomarker concentrations during pregnancy over various time scales (day to months), and confirmed empirically that this variability is likely to strongly bias the doseresponse functions in human-based epidemiological studies exploring the effects of gestational exposure to these chemicals.This thesis adds to the emerging literature on respiratory health impacts of early-life exposure to several phenols and phthalates. However, as for most studies on the human health effects of phenol and phthalate exposure, it is potentially challenged by this exposure assessment issue. Thus, this work emphasizes the relevance of more elaborate sampling strategies for exposure biomarkers in future epidemiological studies. These results have relevance for studies outside the context of pregnancy, and also for other nonpersistent compounds. New designs, such as the within-subject pooling of biospecimens validated in this study, are needed so as to efficiently characterize the health impact of nonpersistent chemicals.

Childhood and adolescent exposure to chemicals found in personal care products

Contexte : Les produits de soins personnels contiennent plusieurs substances chimiques dont l'exposition est préoccupante pour les jeunes vu leur plus grande susceptibilité aux expositions chimiques. Aucune revue publiée n’a étudié l'exposition des jeunes aux substances chimiques dans ces produits. Objectif : Cette revue de la portée vise à synthétiser des études de biosurveillance décrivant les concentrations de parabènes, phénols, phtalates et substances per- et polyfluoroalkylées (PFAS) après l'utilisation de produits de soins personnels chez les jeunes âgés de 5 à 19 ans. Méthodes : La recherche a été effectuée dans MEDLINE, Embase et Global Health. L’éligibilité des articles a été évaluée. Les résultats des études éligibles ont été résumées et décrites. Résultats : Trente-sept études ont été incluses, la première étant publiée en 2013. La majorité des études étaient transversales (n=35) et incluaient plus de 100 participants (n=27). Les substances chimiques les plus fréquemment étudiées étaient les phtalates (n=23), suivies des parabènes et des phénols (chacun n=18), puis des PFAS (n=1). La matrice biologique principalement utilisée était l'urine (n=36). Plusieurs études (n=31) ont rapporté au moins une association positive entre les produits de soins personnels et certaines classes chimiques, comme entre le maquillage et les parabènes; la lotion et les phénols; et le parfum et les phtalates. Conclusion : Plus d'études sur ce sujet sont nécessaires pour documenter l'importance relative de l'utilisation des produits de soins personnels par rapport à l'exposition totale aux substances chimiques. L’uniformisation des questionnaires de collecte de données pourrait aider à réduire l'hétérogénéité des résultats. / Background: Personal care products (PCPs) contain a variety of chemicals including parabens, phenols, phthalates, and per- and polyfluoroalkyl substances (PFAS). Exposure to these chemicals is of concern for children and adolescents, as they are known possess an increased susceptibility to chemical exposures. To date, no published review has explored youth exposure to chemicals found in PCPs. Objective: This scoping review aimed to provide an overview of biomonitoring studies that describe the concentrations of parabens, phenols, phthalates, and PFAS following PCP use among children and adolescents of any age from 5 to 19 years old. Methods: The search was conducted in MEDLINE, Embase, and Global Health. Articles were screened and assessed for eligibility. From eligible studies, data were extracted to summarize and describe the findings. Results: Thirty-seven studies were included, the first of which appeared in 2013. The majority of studies were cross-sectional (n=35) and included more than 100 participants (n=27). The most frequently studied chemicals were phthalates (n=23), followed by parabens and phenols (each n=18), and then PFAS (n=1). The biological matrix mainly used was urine (n=36). Thirty-one studies reported at least one positive association between personal care products and certain chemical classes, such as between makeup and parabens; lotion and phenols; and perfume and phthalates.

THE ROLE OF PXR AND IKKβ SIGNALING IN CARDIOMETABOLIC DISEASE

Cardiovascular disease (CVD) is the leading cause of death worldwide and is partially attributed to perturbations in lipid metabolism. Xenobiotics, such as pharmaceutical drugs and environmental chemicals, have been associated with increased risk of CVD in multiple large-scale human population studies, but the underlying mechanisms remain poorly defined. We and others have identified several xenobiotics as potent agonists for the pregnane X receptor (PXR), a nuclear receptor that can be activated by numerous drugs as well as environmental and dietary chemicals. However, the role of PXR in mediating the pathophysiological effects of xenobiotic exposure in humans and animals remains elusive. The work herein identified several widely used pharmaceutical agents and endocrine disrupting chemicals as PXR-selective agonists such as drugs involved in HIV therapy and phthalates/phthalate substitutes, respectively. We investigated the role of amprenavir, an HIV protease inhibitor, and tributyl citrate, a phthalate substitute, on PXR-dependent alterations in lipoprotein metabolism. Acute exposure with either xenobiotic in mice elicited increases in the proatherogenic LDL-cholesterol levels in a PXR-dependent manner. PXR activation significantly induced expression of genes involved in intestinal lipid metabolism. Further, we went on to identify the intestinal cholesterol transporter, Niemann-Pick C1-Like 1 (NPC1L1), as a direct PXR-target gene. PXR activation also stimulated cholesterol uptake in both murine and human intestinal cells. Moreover, we provide evidence that the microsomal triglyceride transfer protein (MTP) may be a direct PXR-target gene. Taken together, these findings provide critical mechanistic insight into the role of xenobiotic-mediated PXR activation on lipid homeostasis and demonstrate a potential role of PXR in mediating adverse effects of xenobiotics on CVD risk in humans. In addition to PXR signaling, we investigated the role of IκB kinase β (IKKβ), a central coordinator of inflammation, in adipocyte progenitor cells. Targeting IKKβ in adipose progenitor cells resulted in decreased high fat diet (HFD)-elicited adipogenesis, while protecting mice from inflammation and associated insulin resistance. Consistently, we discovered that IKKβ inhibition by antisense oligonucleotides ablated HFD-induced adiposity, while protecting mice against associated metabolic disorders. In conclusion, targeting IKKβ with antisense therapy may present as a novel therapeutic approach to combat obesity and metabolic dysfunctions.

Exposition cumulée aux contaminants de l'air intérieur susceptibles d'induire des affections respiratoires chroniques de l'enfant / Cumulative exposure to indoor air contaminants known or suspected to induce chronic respiratory affections in children

Depuis quatre décennies, la prévalence des affections respiratoires chroniques de l'enfant a considérablement augmenté dans les pays développés. Les conditions de survenue de ces affections sont complexes, mais de nombreux travaux suggèrent la contribution importante de l'exposition par inhalation aux polluants de l'air intérieur. Dans ce contexte, cette thèse vise à évaluer l’exposition cumulée à une gamme de polluants chimiques et biologiques de l’air intérieur dans un échantillon donné de logements. Il a également pour objectif de créer une typologie des logements en fonction de leur multi-contamination, et vise à construire des modèles explicatifs des concentrations des polluants en fonction des caractéristiques de l’habitat et des habitudes de vie des occupants.Une enquête environnementale a été menée dans 150 logements issus de la cohorte Pélagie, suivie en Bretagne depuis 2002. Des prélèvements ont permis de mesurer la concentration de 8 aldéhydes, 4 THM, 22 autres COV, 9 COSV et 4 genres de moisissures dans l’air de ces logements. Celles-ci, ainsi que 4 allergènes ont également été dosés dans des échantillons de poussières. Les paramètres d’ambiance (température, humidité relative et dioxyde de carbone) ont été mesurés. Un questionnaire renseigné par les familles a permis de collecter des informations sur les logements et leurs occupants : structure et historique du bâtiment, revêtements, ménage, chauffage, aération, utilisation de certains produits ou réalisation d’activités particulières. Ces données ont été analysées par des approches statistiques multivariées, et des modèles de régression linéaire et logistique ont été mis en oeuvre pour relier les concentrations des contaminants aux caractéristiques des logements. Ces mesures ont mis en évidence une contamination importante et systématique des logements par une grande part des contaminants chimiques et biologiques, à des niveaux parfois élevés au regard d’études comparables et des valeurs guides lorsqu’elles existent. Des analyses en composantes principales ont permis de mettre en évidence des sous-groupes de composés qui ont pu être interprétés en termes de sources, et de sélectionner un certain nombre de composés traceurs représentatifs de chaque sous-groupe. Une analyse factorielle multiple a permis de répartir les logements en 7 classes, chacune présentant un profil de multi-contamination particulier. Enfin, les modèles de régression linéaire et logistique construits pour les composés traceurs permettent d’expliquer entre 5 et 60% de la variabilité des concentrations, et mettent en évidence la multiplicité des sources, l’importance de la description précise des environnements intérieurs, et l’impact des paramètres d’ambiance sur ces concentrations. Ce travail décrit donc une contribution importante à l’évaluation des expositions aux contaminants de l’air intérieur et fournit un certain nombre d’éléments quant à la prédiction des expositions dans les environnements intérieurs. / For the last four decades, the prevalence of chronic respiratory affections in children has increased dramatically in developed countries. Occurring conditions of these affections are complex, but many studies suggest the important contribution of inhalation exposure to indoor air pollutants. In this context, this thesis aims to assess the cumulative exposure to a range of chemical and biological pollutants in indoor air in a given sample of dwellings. It also aims to create a typology of these dwellings based on their multi-contamination, and to build explanatory models for concentrations of pollutants based on characteristics of the dwellings and lifestyle of the occupants. An environmental survey was conducted in 150 dwellings from the Pelagie cohort, followed in Brittany since 2002. We measured the concentration of 8 aldehydes, 4 THMs, 22 other VOCs, 9 SVOCs and 4 mold genera in the air of these dwellings. Molds as well as four allergens were also measured in dust samples. Ambient parameters (temperature, relative humidity and carbon dioxide) were also measured. A questionnaire completed by families allowed collecting information on dwellings and their occupants: structure and history of the building, wall and floor coatings, cleaning, heating and ventilation habits, use of certain products or performing specific activities. These data were analyzed by multivariate statistical approaches, and linear and logistic regression models were used to link the concentrations of the contaminants with the housing characteristics. These measures showed an important and systematic contamination of the dwellings by a large amount of both chemical and biological contaminants, sometimes at relatively high levels regarding comparable studies and guideline values when they exist. Principal components analysis allowed to identify subgroups of compounds that could be interpreted in terms of sources, and to select representative compounds of each subgroup. A multiple factor analysis was used to classify the dwellings into 7 categories, each with a special multi-contamination profile. Finally, linear and logistic regression models built for the representative compounds explained between 5 and 60% of the variability of the concentrations, and highlighted the multiplicity of sources, the importance of a precise description of indoor environments, and the impact of the ambient parameters on these concentrations. This work thus describes an important contribution to the exposure assessment to indoor air contaminants and provides elements for prediction of exposures in indoor environments.

Apesar de serem eficazes na função de reduzir a rigidez do PVC, os plastificantes adicionados podem migrar por difusão passiva das embalagens poliméricas para alimentos e para o meio ambiente. Dentre os ftalatos largamente usados como plastificantes do PVC, o ftalato di(2-etil hexila) ou DEHP, vem apresentando-se como tóxico, sendo objeto de restrição de diversas agências de controle sanitário em todo o mundo. Diante de diversos estudos relatando a ligação do DEHP com distúrbios hormonais, infertilidade masculina, puberdade precoce em meninas, potencial carcinogênico e hepatotoxicidade bem como a possibilidade real de migração do ftalato das embalagens aos alimentos, a Agência Nacional de Vigilância Sanitária, ANVISA, desde 1999 e posteriormente em 2008 limita o uso do DEHP em embalagens que entram em contato com alimentos. Apesar da limitação ao uso do DEHP, estudos recentes demonstram que o uso indiscriminado do aditivo em embalagens de alimentos no Brasil continua, o que torna necessário um olhar mais atento das autoridades para a fiscalização a fim de evitar a exposição da população ao risco da contaminação dos alimentos pelo plastificante. Esse trabalho foi destinado a avaliar filmes de PVC, quanto à presença de DEHP, em embalagens de alimentos no Distrito Federal-DF. Foram obtidas nove amostras de uso profissional dos filmes de PVC diretamente nos locais de uso em supermercados e lanchonetes de três áreas do DF, bem como três amostras destinadas ao uso residencial compradas em supermercados para avaliação quanto a presença do DEHP e confronto com o normativo atual para verificar a conformidade das embalagens. Foi feita a caracterização das amostras por meio dos ensaios de, Espectroscopia por Energia Dispersiva de Raios X, Difração de Raios X, Espectroscopia na Região do Infravermelho com Transformada de Fourier (FTIR), Calorimetria Exploratória Diferencial, Termogravimetria e Ensaios de Resistência Mecânica à Tração. Os resultados apontaram que as amostras foram similares quanto à composição química básica, bem como apresentaram resultados semelhantes nos ensaios mecânicos e calorimétricos. O FTIR indicou bandas relativas ao DEHP em todas as amostras, indiferente da área de coleta ou da finalidade, uso profissional ou residencial. Foram observados indicativos de DEHP em maiores quantidades nas amostras de uso residencial. Os achados de DEHP em todas as amostras contrariam o disposto no normativo estabelecido pela ANVISA que limita o uso de DEHP em 1,5 mg/Kg em embalagens de alimentos não gordurosos e veta a possibilidade de uso em embalagens de alimentos gordurosos. O estudo conclui que o amadurecimento do conhecimento do normativo não levou a resultados distintos em relação a estudos similares executados pouco tempo após a publicação das normas de limitação de uso do DEHP. Os resultados expõem que o plastificante derivado de petróleo continua presente indiscriminadamente nas embalagens de alimentos e o consumidor está sujeito aos riscos já conhecido, inerente da exposição ao plastificante DEHP. Torna-se necessário maior controle e rigor na fiscalização de embalagens de alimentos a fim de minimizar os efeitos deletérios à saúde da população consumidora desse tipo de alimento. / Phthalates are widely used as plasticizers for polyvinyl chloride or PVC which gave this large polymer application in various areas ranging from construction to food packaging. Although they are effective in the function of reduce the stiffness of PVC, plasticizers can migrate added by passive diffusion form polymeric packaging to food and the environment. Among the phthalates widely used as PVC plasticizers, the phthalate, di (2-ethyl hexyl) or DEHP, has been presented as toxic, and restriction object of various sanitary control agencies worldwide. Faced with several studies reporting the DEHP connection with hormonal disorders, male infertility, early puberty in girls, potential carcinogen and hepatotoxicity and the real possibility of migration of phthalate form packaging to the food, the National Health Surveillance Agency, ANVISA, since 1999 and later in 2008 limits the use of DEHP in packaging that come into contact with food. Despite the limitation to the use of DEHP, recent studies show that the indiscriminate use of the additive in food packaging in Brazil continues, what a close look at the authorities for surveillance is necessary in order to avoid exposure of the population to the risk of contamination food by the plasticizer. This study was designed to assess PVC films, for the presence of DEHP in food packaging in the Federal District-DF. Nine samples of professional use were obtained from PVC films directly at points of use in supermarkets and fast food three areas of DF and three samples intended for residential use purchased from supermarkets to assess for the presence of DEHP and confrontation with the current regulatory to check the conformity of packaging. The characterization of samples through the test, spectroscopy Energy Dispersive X-ray, X-ray diffraction, infrared spectroscopy in the region Fourier Transform was made (FTIR), Differential Scanning Calorimetry, Thermogravimetry and Testing of Mechanical Resistance to traction . The results showed that the samples were similar in basic chemical composition and showed similar results in mechanical and colorimetric assays. Spectroscopy in the Infrared Region Fourier Transform indicated bands on the DEHP in all samples, regardless of the collection area or purpose, professional or residential use. DEHP indicative were observed in larger quantities in the residential samples. DEHP findings in all samples contrary to the provisions of normative established by ANVISA which limits the use of DEHP in 1.5 mg / kg in non-fatty food packaging and veto the possibility of use in fatty food packaging. The study concludes that the maturing of normative knowledge did not lead to different results compared to similar studies performed shortly after the publication of standards of DEHP use limitation. The results state that the plasticizer derived from oil is still present indiscriminately in food packaging and the consumer is subject to known risks inherent exposure to the plasticizer DEHP. It is most necessary control and rigorous supervision of food packaging in order to minimize the deleterious health effects of consumers of this type of food.
