[pt] Este estudo investiga o envolvimento do córtex pré-frontal medial ventral nos
comportamentos de defesa inatos e aprendidos em paradigmas de
condicionamento de medo e estimulação elétrica intracraniana em ratos. A lesão
cortical aumentou significativamente o comportamento defensivo condicionado.
No comportamento defensivo incondicionado, a lesão cortical diminuiu
significativamente o congelamento pós-fuga dos animais. Os resultados replicam
os dados da literatura científica a respeito do papel do córtex infralímbico como
uma estrutura inibitória do estímulo condicionado em um circuito amidaloide de
medo condicionado, e indicam uma participação do córtex pré-frontal na
modulação dos comportamentos de defesa originários da estimulação da MCPd,
em especial a sustentação do congelamento motor pós fuga. / [en] This study investigates the role of the prefrontal cortex in the innate and
conditioned defensive behaviors in rats during classical conditioning and
intracranial electrical stimulation procedurals. It was found that the cortical lesion
augmented the conditioned freezing behavior to contextual fear cues. On the
other hand, the lesions impaired the motor freezing presented after the escaping
provoked by dPAG stimulation. These results replicate the findings from the
literature about a prefrontal cortex role as an inhibitory structure in the aversive
classic conditioning circuitry, as well as presenting a role for it in modulating
freezing behavior in a panic circuitry involving the dPAG, especially regarding its
function as a possible short term memory device for innate fear expression.

[pt] Os transtornos de ansiedade afetam milhares de pessoas em todo o mundo, sendo representados como um dos principais distúrbios mentais. A ansiedade é acompanhada por uma série de respostas comportamentais e fisiológicas, quando na presença de estímulos aversivos. Essas respostas provocam ações neuroendócrinas envolvendo o sistema nervoso simpático (SNS), o eixo hipotálamo-pituitária-adrenal (HPA) e o sistema renina-angiotensina (SRA). Pesquisas feitas em modelos animais possibilitam uma melhor compreensão dos mecanismos neurofisiológicos e comportamentais associados à patologias observadas em humanos. Os animais Cariocas com Alto Congelamento (CAC) e Cariocas com Baixo Congelamento (CBC) são duas linhagens condicionadas de ratos que apresentam, respectivamente, níveis altos ou baixos de respostas semelhantes à ansiedade. O presente estudo investigou os efeitos da Losartana e Valsartana, sendo ambas da classe antagonistas do receptor de angiotensina II, na modulação da ansiedade nos ratos CAC, CBC e ratos controle. O tratamento crônico com Losartana e o tratamento agudo com Valsartana não produziram efeitos significativos nas respostas comportamentais associadas à ansiedade no condicionamento de medo contextual, campo aberto e labirinto em cruz elevado. Nossos dados sugerem que nas doses e duração dos tratamentos, a administração destes medicamentos anti-hipertensivos não é capaz de modular a ansiedade nos animais CAC e CBC. / [en] Anxiety disorders affect thousands of people all around the world, being represented as the most common of mental disorders. Anxiety is associated with a number of behavioral and physiological responses when faced with aversive stimuli. These responses provoke neuroendocrine actions involving the activation of the sympathetic nervous system (SNS), the hypothalamic-pituitary-adrenal axis (HPA) and the renin-angiotensin system (RAS). Studies on animal models allow for a better understanding of the neurophysiological and behavioral mechanisms associated with pathologies observed in humans. The Carioca High Freezing (CHF) and Carioca Low Freezing (CLF) are two conditioned strains of rats that present, respectively, high or low levels of anxiety-like responses. The present study investigated the effects of Losartan and Valsartan, both angiotensin II receptor blockers, on the modulation of anxiety-like behaviors of CHF, CLF and control rats. Neither chronic treatment of Losartan nor acute treatment of Valsartan yielded significant effects on anxiety measurements in the contextual fear conditioning, open field and elevated plus maze tests. Thus, our findings suggest that at the doses and durations of treatment tested, administration of these antihypertensive drugs did not play a modulating role of anxiety-like behaviors in CHF and CLF animals.

[pt] A suscetibilidade ao estresse é ditada por fatores externos e internos. Fatores externos estão associados a variáveis ambientais, enquanto os internos estão relacionados à função cerebral, cognição, constituição genética e epigenética e ao microbioma. Nesta tese, investigamos a influência das respostas individuais de enfrentamento ao medo sobre os efeitos do estresse crônico e sua modulação por uma dieta rica em calorias. Nesse intento, foram usadas as linhagens de cruzamento bidirecional Carioca como modelos de respostas opostas ao medo contextual condicionado. Inicialmente, caracterizamos essas linhagens quanto às suas respostas de congelamento após a aprendizagem e a extinção de medo. Em seguida, descrevemos em uma meta-análise o envolvimento da via serotoninérgica, importante mecanismo nos transtornos de ansiedade e medo, nos efeitos do estresse crônico. Finalmente, demonstramos que o estresse crônico tem efeitos mais fortes em ratos Cariocas de alto congelamento (CHF), alterando o equilíbrio entre as respostas de enfrentamento ativas e passivas em vários paradigmas comportamentais; por outro lado, os ratos Carioca de baixo congelamento (CLF) são minimamente afetados. Além disso, uma dieta suplementada com alimentos ricos em açúcar e gordura pode proteger os ratos CHF contra os efeitos do estresse crônico. Assim, demonstramos os efeitos adversos do estresse crônico em um modelo de transtorno de ansiedade generalizada e propomos que a suscetibilidade ao estresse está ligada à características individuais e inatas de medo e de transtorno de ansiedade, podendo ser alvo de novas terapias para transtornos de humor. / [en] Susceptibility to stress is dictated by external and internal factors. External factors are associated with environmental variables, whereas the internals are related to brain function, cognition, genetic and epigenetic constitution and the microbiome. In this thesis, we investigated the influence of the individual background underlying fear-coping responses on the effects of chronic stress and its modulation by a palatable diet. We used the Carioca bidirectional breeding lines as models of opposing conditioned contextual fear responses. At first, we characterize these lines concerning their freezing responses after fear learning and extinction. Then, we describe in a meta-analysis the involvement of the serotoninergic pathway, an important mechanism in anxiety and fear disorders, in the effects of chronic stress. Following, we demonstrate that chronic stress has stronger effects in the Carioca high-conditioned freezing (CHF) rats, altering the balance between active and passive coping responses in several behavioral paradigms; conversely, the Carioca low-conditioned freezing (CLF) rats are barely affected. Moreover, a diet supplemented with high-sugar and high-fat food can protect the CHF against the effects of chronic stress. Thus, we demonstrate the increased harmful effects of chronic stress in a model of generalized anxiety disorder and propose that susceptibility to stress is linked to fear and anxiety individual innate characteristics, which can be the target of novel therapies of mood disorders.

[pt] Assunto: O ingresso na universidade é um momento de transição que muitas
vezes contribui para o desenvolvimento de sintomas psicológicos ou para um
agravamento de sintomas pré-existentes em jovens. Objetivo: Esta dissertação é
composta por dois estudos realizados durante a pandemia da covid-19. O primeiro
com o objetivo de investigar, por meio de Revisão Sistemática da literatura, as
prevalências de sintomas de ansiedade e depressão em estudantes universitários em
diferentes continentes do mundo. O segundo estudo teve como objetivo rastrear os
sintomas psicológicos apresentados por alunos da graduação da PUC-Rio. Método:
A Revisão sistemática foi feita a partir de 9 bases de dados acessados em agosto de
2021. Após aplicados os critérios de inclusão e exclusão, foram selecionados 77
estudos para a pesquisa. A coleta de dados para a pesquisa com alunos de graduação
foi conduzida entre junho e setembro de 2021, com amostra composta por 1.189
alunos através da aplicação de questionário online com dados sociodemográficos,
dados acadêmicos, histórico de saúde mental e duas escalas de Rastreio: Escala
Transversal de Sintomas de Nível 1 do DSM-5 e a Escala de Medo da Covid-19.
Resultado: No estudo de revisão, o resultado aponta maior índice de prevalência de
sintomas de ansiedade (52,7 por cento) e depressão (81,4 por cento) na América do Sul. Os
resultados do estudo quantitativo sugerem grau moderado para sintomas de
ansiedade (M=3,08) e leve a moderado para sintomas depressivos (M=2,65).
Conclusão: Nos dois estudos foi possível identificar a prevalência de sintomas de
ansiedade e depressão na população universitária. / [en] Subject: The admission to university is a time of transition that often
contributes to the development of psychological symptoms or to a worsening of
pre-existing symptoms in young people. Objective: This dissertation is composed
of two studies carried out during the COVID-19 pandemic. The first aimed to
investigate, through a Systematic Review of the literature, the prevalence of
symptoms of anxiety and depression in university students in different continents
of the world. The second study aimed to track the psychological symptoms
presented by undergraduate students at PUC-Rio. Method: The systematic review
was carried out from 9 databases accessed in August 2021. After applying the
inclusion and exclusion criteria, 77 studies were selected for the research. Data
collection for the survey with undergraduate students was conducted between June
and September 2021, with a sample composed of 1,189 students through the
application of an online questionnaire with sociodemographic data, academic data,
mental health history and two Screening scales: Scale Cross-sectional DSM-5 Level
1 Symptoms and the COVID-19 Fear Scale. Result: In the review study, the result
points to a higher prevalence rate of symptoms of anxiety (52.7 percent) and depression
(81.4 percent) in South America. The results of the quantitative study suggest a moderate
degree for anxiety symptoms (M=3.08) and mild to moderate for depressive
symptoms (M=2.65). Conclusion: In both studies it was possible to identify the

[pt] Os adultos emergentes de 18 a 29 anos estão na fase do desenvolvimento
que precede a vida adulta marcada pela autonomia e independência. Além dos
desafios peculiares, a pandemia da COVID 19 os expôs à maior tragédia
humanitária desde a segunda guerra mundial. O estudo propôs avaliar a
espiritualidade, religiosidade, materialismo e ansiedade perante a morte como
preditores para presença e busca de sentido neste contexto sociocultural com
intensas ameaças psíquicas. A pesquisa obteve 434 respondentes brasileiros e 615
portugueses. O objetivo da investigação foi verificar o quanto religiosidade,
espiritualidade, materialismo e ansiedade perante a morte podem explicar a
presença e a busca de sentido de vida. Após averiguar os dados de cada país, fez-se
uma comparação dos resultados. O estudo utilizou-se dos instrumentos: Escala de
Sentido de Vida (Steger et al., 2006); Escala de Não-Religiosidade e Não-Espiritualidade (Cragun, 2015); Escala de Materialismo (Richins, 2004) e o
Questionário de Ansiedade Perante a Morte (Conte et al., 1982). Os resultados
demonstraram seguintes os resultados tanto para os respondentes brasileiros quanto
portugueses: (1) a religiosidade predisse a presença de sentido na vida. Uma menor

unique challenges, the COVID 19 pandemic has exposed them to the greatest

spirituality, religiosity, materialism and anxiety before death as predictors for
presence and search for meaning in this sociocultural context with intense psychic

presence of meaning in life demonstrated the possibility of greater psychological

[pt] Os ratos Carioca com alto condicionamento (CAC) e Carioca com baixo condicionamento (CBC) são duas linhagens que são criadas seletivamente para altos e baixos níveis de congelamento, em resposta a sugestões contextuais previamente associadas ao estímulos elétricos nos membros. Essas duas linhas
têm traços significativamente diferentes de respostas semelhantes à ansiedade, em diferentes protocolos comportamentais. O presente estudo utilizou ratos machos e fêmeas de CAC e CBC para investigar possíveis associações entre comportamento relacionado à ansiedade e ingestão de álcool. Quinina e sacarina foram utilizadas como soluções de controle do paladar. Os resultados indicaram que os ratos CAC
tinham um fenótipo ansioso mais alto e os animais CBC tinham um fenótipo ansioso mais baixo, quando as duas linhagens foram comparadas com uma linhagem, selecionada aleatoriamente, que foi usada como grupo controle (CTR). Os ratos machos exibiram consistentemente maior congelamento condicionado do
que as fêmeas. Os ratos CAC consumiram mais concentrações de álcool de 6 porcento e 10 porcento do que os ratos CBC em um teste de livre escolha de consumo e maior consumo na concentração de álcool 10 porcentp em um teste de escolha forçada. Ratos fêmeas CAC exibiram a maior quantidade de ingestão de álcool durante essas três condições, em comparação com ratos machos CAC e também consumiram mais
quinina que ratos machos. Finalmente, ratos CAC exibiram menor consumo de sacarina em comparação com CBC e animais controle. No total, esses resultados revelaram diferenças entre os sexos no comportamento semelhante ao da ansiedade e no consumo de álcool, corroborando a hipótese de que existe uma relação positiva entre ansiedade e ingestão de álcool. / [en] Carioca High-conditioned Freezing (CHF) and Carioca Low-conditioned Freezing (CLF) rats are two linesthat are selectively bred for high and low levels of freezing in response to contextual cues that are previously associated with footshock. These two lines have significantly different traces of anxiety-like responses in different behavioral protocols. The present study used male and female CHF and CLF rats to investigate possible associations between anxietyrelated behavior and alcohol intake. Quinine and saccharin were used as taste control solutions.The results indicated that CHF rats had a higher anxious phenotype and CLF animals had a lower anxious phenotype when both breeding lines were compared with a randomly selected line that was used as a control group. Male rats consistently exhibitedmore conditioned freezing than females. CHF rats consumed more 6 percent and 10 percent alcohol concentrations than CLF rats in a free-choice test and more of a 10 percent alcohol concentration in a forcedchoice test. Female CHF rats exhibited the highest amount of alcohol intake during these three conditions compared with male CHF rats. Female rats also consumed more quinine than male rats. Finally, CHF rats exhibited lower saccharin consumption compared with CLF and control animals. Altogether, these results revealed sex differences in anxiety-like behavior and alcohol consumption,supporting the hypothesis that there is a positive relationship between anxiety and alcohol intake. The Carioca breeding lines were developed for high and low defensive behavior in response to diffusecontextual cues that are associated with footshock but not to well-defined danger-related stimuli, corroborating the view that anxiety but not fear is associated with alcohol consumption.

[pt] CRENÇAS DISFUNCIONAIS SOBRE EMOÇÕES NEGATIVAS E REPERCUSSÕES PARA A SAÚDE MENTAL / [en] DYSFUNCTIONAL BELIEFS ABOUT NEGATIVE EMOTIONS AND MENTAL HEALTH OUTCOMESBRUNA IRIS ARRUDA 20 December 2019 (has links)
[pt] Crenças disfuncionais sobre emoções negativas estão associadas à presença de sintomas como ansiedade, depressão, fadiga, medo de avaliação negativa, menor busca por apoio social e supressão emocional. A supressão é uma estratégia de regulação das emoções contraproducente, relacionada com dificuldades como menores níveis de entendimento emocional e aumento dos estados afetivos indesejados. Estas crenças estão relacionadas, ainda, com diferentes quadros clínicos, e podem se originar a partir de fatores ambientais, que exercem um papel central no aparecimento de sintomas referentes à presença das crenças. Elas atuam, também, como fator de risco e vulnerabilidade para certos grupos, já que podem levar a maiores taxas desses sintomas, com os quais parece haver uma relação bidirecional, conduzindo a desregulação emocional e estresse. O primeiro estudo endereçou participantes expostos a estes estressores ambientais, policiais militares, que constituem grupo de risco. Para verificação de dados de saúde mental nessa população, eles foram avaliados com relação às crenças sobre emoções, ansiedade, depressão, fadiga e medo de avaliação negativa. Diferenças significativas foram encontradas entre o grupo policial e o controle, com o grupo de policiais apresentando índices elevados de sintomas de fadiga, depressão e ansiedade com relação aos escores totais das escalas. Também, altas taxas de prevalência foram encontradas para a população policial para esses mesmos sintomas, que dobraram em comparação aos controles, o que aponta para a gravidade clínica destes sintomas. Acreditamos ser a primeira vez que dados de prevalência são relatados em forças policias, constituindo uma importante contribuição do presente estudo. A partir desses resultados, é possível sugerir programas para policiais que enderecem a saúde mental, em especial intervenções que contemplem a regulação das emoções, visando
gerenciamento dos sintomas mais graves. O segundo estudo buscou por meio da validação brasileira do Questionário de Regulação Emocional para a população adulta, investigar a relação entre estratégias de regulação emocional, crenças sobre emoções e alexitimia, fatores emocionais que podem ter uma forte associação. A supressão emocional se apresentou uma estratégia pouco adaptativa pela associação com maiores níveis de crenças sobre emoções e alexitimia, merecendo maior atenção nos tratamentos pelos resultados encontrados no presente estudo, que corroboram com a literatura, e ainda, pela intensidade de sua presença em diferentes grupos clínicos. Além disso, acreditamos ser a primeira vez que é observada a relação entre os contructos de crenças disfuncionais sobre emoções negativas e alexitimia, e que as propriedades psicométricas do instrumento são investigadas na população adulta, o que representa a relevância do segundo estudo.
Assim, de maneira geral, os resultados apontam para uma correlação entre as variáveis clínicas citadas, com uma influência mais sutil que o esperado das crenças no primeiro estudo, mas com associações em ambos que apontam para a presença das crenças como disfuncionais pela vinculação com maiores níveis de depressão, medo de avaliação negativa, supressão emocional e alexitimia. / [en] Dysfunctional beliefs about negative emotions are associated with symptoms such as anxiety, depression, fatigue, fear of negative evaluation, less search for social support and emotional suppression. Emotional suppression is a counterproductive emotion regulation strategy related to difficulties such as lower levels of understanding and greater undesirable emotional states. Unhelpful beliefs are also related to different clinical conditions, and may also come from environmental factors. These beliefs act as a risk factor and vulnerability for certain groups, leading to higher rates of symptoms, emotional dysregulation and stress. The first study addressed participants exposed to such environmental stressors, namely military policemen. To investigate mental health data in this population, participants were assessed for beliefs about emotions, anxiety, depression, fatigue and fear of negative assessment. Results indicates high scores in the scales for fatigue, depression and anxiety, with high prevalence rates of these same symptoms in the at-risk group. To the best of out knowlwdge, this is the first time that fatigue prevalence rates are described in the literature. Considering what has been exposed so far, it is possible to suggest programs for policemen that address mental health, especially interventions that contemplate emotion regulation. The second study sought to validate the Emotion Regulation Questionnaire for the Brazilian adult population, investigating the relationship between emotion regulation strategies, beliefs about emotions and alexithymia. The results point to a correlation between the mentioned clinical variables, albeit with a more subtle influence than expected in the first study.
Furthermore, suppression is a strategy that deserves greater attention due to the high association with variables present in different clinical groups and with emotional difficulties, such as alexithymia and high levels of unhelpful beliefs. Emotional suppression was shown to be a non adaptative emotion regulation
strategy, presenting correlations with high levels of beliefs about emotions and alexithymia. Considering the present results as well as the existing literature, emotional suppression should be adressed in future clinical trials. In addition, this is the first time that the relation between beliefs about emotions and alexithymia is observed and that the psychometic properties of the Emotion Regulation Questionnaire are investigated in adult population. Thus, in general, results points to a correlation between the clinical variables cited above, with a more subtle influence of beliefs about emotions in the first study, but in both with associations that point to the dysfunctional nature of these beliefs, by presenting associations with high levels of depression, fear of negative evaluation, emotional suppression and alexithymia.
