El presente proyecto se ha realizado para analizar la viabilidad de ejecución de un modelo de negocio basado en la creación de una Página Web que tiene como finalidad la búsqueda de talleres mecánicos para personas con vehículos que requieran algún servicio técnico nos muestra. Nuestras hipótesis principales se enfocan en dos grupos principales. Por un lado, los talleres y su disposición a pertenecer a la red Tutaller mediante un pago mensual. Por otro lado, si es que las personas dueñas de vehículos estarían dispuestas a crear una cuenta en nuestra plataforma. Estas dos hipótesis se validaron a través de diversas entrevistas en distritos como Chorrillos, Surquillo, Barranco y Miraflores. Hoy en día, los administradores de los talleres se basan principalmente en el marketing tradicional y en ocasiones realizan pagos para promocionarse en “páginas amarillas”. Son solo algunos los que invierten en publicidad en Facebook o Página Web. Esto genera una complicación para los talleres y para los dueños de vehículos, ya que estos últimos no tienen la información necesaria para poder asistir a un taller; lo cual, ocasiona que los talleres no generen de manera rápida nuevos clientes. La conclusión final del proyecto es que el modelo de negocio genera rentabilidad para los inversionistas. De manera más específica, se estima que en el año 2 la utilidad neta sea positiva. / The present project has been carried out to analyze the feasibility of executing a business model based on the creation of a Website, whose purpose is to search for mechanical workshops for people with vehicles that require a technical service. Our main hypotheses focus on two main groups. On the one hand, the workshops and their willingness to belong to the Tutaller network through a monthly payment. On the other hand, if the owners of vehicles would be willing to create an account on our platform. These two hypotheses were validated through various interviews in districts such as Chorrillos, Surquillo, Barranco and Miraflores. Today, the administrators of the workshops are mainly based on traditional marketing and sometimes make payments to promote themselves in "yellow pages". There are only a few who invest in advertising on Facebook or Websites. This generates a complication for the workshops and for the owners of vehicles, since the latter do not have the necessary information to be able to attend a workshop; which, causes that the workshops do not aquire new clients quickly The final conclusion of the project is that the business model generates profitability for investors. Transitioning towards a Single Page Application for a fashion e-commerce : Qualitative and quantitative analysis of the transition's impact on business performance / Övergången till en Single Page Application för en elektronisk modehandel

Over the course of the last few years, the fashion industry hasbegun to focus more resources on their digital transition. For afashion e-commerce business, it is essential to know whether ornot to invest money and time in building a modern webapplication. This master's thesis aims at finding practical resultson how transitioning from a multi-page website towards a SinglePage Application can have an impact on the businessperformances of the company, as measured by ConversionRate, Page Views and Gross Sales. In collaboration with thedevelopment and product team of a fashion company, thismaster's thesis is based on the six-months development of a newSingle Page Application using the Javascript framework React.js,building on known User-Experience Design principles andHuman-Computer Interface heuristics. The live data collectionfrom the website's audience allowed a quantitative analysis ofthe transition's effect on business performance, which showedpositive impact on business performance. A qualitative usersurvey was then conducted in order to further elaborate on thecauses of the aforementioned impact: all respondents praisedthe Single Page Application as compared to the multi-pagewebsite, and noted lower Response Time, efficient filter-andsearchsystem and high user interaction as advantages thatplayed in favour of their browsing experience and their will-tobuya product. The impact of lowering Response Time evenmore was discussed, as well as the different limitations due tothe scope of this thesis. A list of user suggestions for furtherimprovements was also compiled. / Under de senaste åren har modebranschen lagt mer resurser på digital omställning. För en modebutik inom e-handeln är det viktigt attveta om det är värt att investera pengar och tid i en modern webbapplikation. Detta examensarbete syftar till att söka praktiska resultatom hur övergången från en flersidig webbplats till en Single Page Application kan ha betydande inverkan på en modebutik inom ehandelnsekonomiska resultat. Detta examensarbete grundar sig i sex månader av utveckling av en Single Page application för ettfranskt startup-företag inom modebranschen. Applikationen byggdes med Javascript-ramverket React.js. Designen grundas på kändaprinciper inom användarupplevelsedesign och människa-datorinteraktionsheuristik. Den dynamiska datainsamlingen från webbplatsensbesökare användes till en detaljerad kvantitativ analys av övergångens inverkan på de ekonomiska resultaten: Det fanns en signifikantökning på 284% för Conversion Rate. En kvalitativ användarundersökning genomfördes i syfte att undersöka orsakerna bakom dennaeffekt. Alla respondenter svarade att Single Page Application var bättre än den flersidiga webbplatsen. De noterade att lägre svarstid,effektiva filter och söksystem och hög användarupplevelse var viktiga aspekter för deras vilja att köpa en produkt. Effekten av att minskasvarstiden ytterligare diskuterades, så även de begränsningar som finns inom ramarna för det här examensarbetet. En lista medanvändarförslag för fortsatta förbättringar sattes också samman.

Identification of Chlamydial Iron-Responsive Proteins during Intracellular Growth.

Chlamydia trachomatis is an obligate intracellular bacterium and the most prevalent cause of bacterial sexually transmitted disease. Genital chlamydial infections, marked by chronic, intense inflammation, can lead to genital tissue scarring and infertility and is a contributing factor to development of pelvic inflammatory disease and ectopic pregnancy. Iron is required as a cofactor for numerous highly conserved pathways, and nearly all studied organisms rely on iron for growth. In response to iron restriction, the chlamydial developmental cycle arrests at the intracellular reticulate body stage, resulting in a phenomenon termed persistence. Persistence likely plays a role in chlamydial pathogenesis through the expression of virulence factors and antigens in addition to sustaining chronic infection; however, little is known concerning how chlamydiae respond to iron limitation at the molecular level, and no systems for iron acquisition have been identified in Chlamydia. This dissertation presents an investigation into the chlamydial response to iron restriction. Chlamydial heat shock protein 60 (cHsp60) has been implicated in development of the more severe disease sequelae and has been found to increase in expression following iron restriction; however, three cHsp60 homologues were identified following the sequencing of the chlamydial genome. Here, iron restriction is shown to increase expression of cHsp60-2 but not the two other homologs, cHsp60-1 or -3. Next, in order to investigate an alternate model for restricting iron availability to chlamydiae, a cell line with inducible expression of recombinant ferroportin, a eukaryotic iron efflux protein, was examined. Lastly, 10 chlamydial proteins differentially expressed during growth in iron-restricted host cells were identified by proteomic analysis of radiolabeled proteins followed by mass spectrometry analysis; transcripts encoding 5 iron responsive proteins were examined across a timecourse of infection and revealed increased transcript levels at 18 and/or 24 hours post infection. Together, these studies have examined the molecular response of chlamydiae to reduced iron availability and have underlined the importance for pathways involved in protection against oxidative damage and adaptation to stress.

Measuring Influence on Linear Dynamical Networks

Influence has been studied across many different domains including sociology, statistics, marketing, network theory, psychology, social media, politics, and web search. In each of these domains, being able to measure and rank various degrees of influence has useful applications. For example, measuring influence in web search allows internet users to discover useful content more quickly. However, many of these algorithms measure influence across networks and graphs that are mathematically static. This project explores influence measurement within the context of linear time invariant (LTI) systems. While dynamical networks do have mathematical models for quantifying influence on a node-to-node basis, to the best of our knowledge, there are no proposed mathematical formulations that measure aggregate level influence across an entire dynamical network. The dynamics associated with each link, which can differ from one link to another, add additional complexity to the problem. Because of this complexity, many of the static-graph approaches used in web search do not achieve the desired outcome for dynamical networks. In this work we build upon concepts from PageRank and systems theory introduce two new methods for measuring influence within dynamical networks: 1) Dynamical Responsive Page Rank (DRPR) and 2) Aggregated Targeted Reachability (ATR). We then compare and analyze and compare results with these new methods.

Introduction: Cellular membrane proteins, such as the cytochrome P450 enzyme superfamily (P450), have important roles in the physiology of the cell. P450s are important in metabolising endogenous molecules, as well as metabolising xenobiotic substances for detoxification and excretion. P450s are also implicated in cancer as they can act to ¿negatively¿ de-activate or ¿positively¿ activate cancer therapeutics. Identifying specific P450s that are highly up-regulated at the tumour site could be used to predict drug response and formulate targeted cancer therapy to help diminish systemic side-effects. Methods: Previous enrichment strategies have been unable to isolate the full complement of the P450 superfamily. To develop enrichment procedures for the P450s, a proteomic strategy was developed so that compounds could be screened for their effectiveness as general P450 probes. A standardised work-flow was created, encompassing affinity chromatography, protein concentration/desalting, followed by sodium dodecyl sulphate polyacrylamide gel electrophoresis (SDS-PAGE) and high performance liquid chromatography-mass spectrometry (HPLC-MS). A ketoconazole analogue and a 2-EN analogue, with known P450 inhibition, were immobilised on a solid support for comparison to immobilised histamine. Co-factor removal, competitive elution and DTT cleavage of disulfide bonds of probes were utilised to elute bound proteins. Results/Discussion: Inhibitor-beads bound a large range of proteins, including P450¿s, of which some were eluted by co-factor removal, some by competitive elution. Specificity of binding was improved by optimising buffer conditions and solid supports, however non-specific binding was not totally eradicated. All human P450s from spiked samples and 18 P450s from more complex mouse liver samples were recovered using one or more ligands. / Bruker Daltonics

Detection of a Peptide Hormone - Somatostatin - Label-free Split-aptameric Probes

Peptide hormones are important biomolecules that transduce downstream effects such as cell proliferation, regulation, and gene expression. Their levels have been upregulated in various disorders such as cancer, yet detection methods are lacking. We designed two split aptamer-based assays for the detection of a peptide hormone – Somatostatin (SST) – with different signal readouts: fluorescent readout based on light-up aptamers and the colorimetric readout of ABTS peroxidation from a G-quadruplex. We used an already selected split-aptamer –SSTA5–for SST for our designs and we had expected the developed detection systems to exhibit detection and quantification capabilities that would hopefully allow their use for SST monitoring in clinical samples. However, our experiments did not support the hypothesis of this project and SST was not able to be detected using either of our fluorescent or colorimetric methods. To determine if the SSTA5 aptamer could bind SST appropriately, Förster resonance energy transfer (FRET) was used. We verified that there was no energy transfer between two covalently-attached light-sensitive molecules (one attached to each part of the split SSTA5 aptamer); thus, we theorize that the aptamer does not hybridize in the presence of the tetra decapeptide SST. Therefore selection of another, more appropriate, aptamer for SST will be needed for further aptameric-based detection methods. Once this is accomplished, our methodologies could be re-applied for detection of SST which could lead to real-time detection of essential hormonal levels in patients.
